      • Effects of subacute lead exposure on [3H]MK-801 binding in hippocampus and cerebral cortex in the adult rat

        We used the NMDA receptor non-competitive antagonist, [³H]MK-801, as a ligand for an autoradiographic study to determine the effects of lead on NMDA receptor in the rat brain. Adult male rats were administered lead acetate, 100 ㎎/㎏, or sodium acetate, 36 ㎎/㎏ (control), by i.p. for 7 days. High lead levels were detected in blood (41.1 ㎍/㎗) and in brain (16.7-29.4 ㎍/g). Concentrations of lead in brain regions were not significantly different. the [³H]MK-801 binding was heterogeneously distributed throughout the rat brain with the following order of binding densities: hippocampal formation $gt; cortex $gt; caudate-putamen $gt; thalamus $gt; brainstem. Lead exposure produced a significant decrease in [³H]MK-801 binding to the NMDA receptor in the hippocampal formation including CA2 stratum radiatum, CA3 stratum radiatun, hilus dentate gyrus and presubiculum, and in the cerebral cortex including agranular insular, cingulate, entorhinal, orbital, parietal and perirhinal areas. The hippocampal formation is known as a critical neural structure for learning and memory processes, whereas, cortical and subcortical regions have been demonstrated to be involved in the modulation of complex behavioral processes. The NMDA receptor has been demonstrated to play a key role in synaptic plasticity underlying learning and memory. Lead-induced alterations of NMDA receptors in the hippocampal formation and cortical areas may play a role in lead-induced neurotoxicity.

        Studies on the Processing Properties and Interactions Between Porcine Blood Proteins and Waxy Rice Starch During Making Porcine Blood Cake

        The physiochemical properties and interactions between porcine blood and waxy rice were determined. Addition of calcium chloride (0.15%) improved acceptability of blood cake and increased the gelatinization degree of waxy rice. The water-holding capacity of porcine blood gel (blood/water=60/40, v/v), extent of absorption and gelatinization of waxy rice, and scanning electron microscopy showed that blood protein matrix and waxy rice are competitors for holding water in the cooking procedure. Non-haem iron content increased linearly (R=0.95) when heating temperature rose. The presence of blood proteins caused increasing of peak temperature (Tp) of gelatinization in differential scanning calorimetric thermal gram, The microstnlcture of plasma proteins and haemoglobin appeared continuous changes, and interacted with surface of waxy rice flour in terms of network and mosaic form, respectively. The electrophoretic patterns revealed an interaction between plasma proteins and waxy rice glutelin and haemoglobin when heated could be found at temperatures above $60^{\circ}C$.

      • Sustainable Development Strategy for Casino Industry of Hualien Area

        Been long a controversial issue. Gambling proponents usually took the gambling industry in Las Vegas as example and argued that casino liberalization at Las Vegas, has attracted tremendous tourism benefits and brought economic prosperity to the surrounding region. They questioned why Hualien shouldn’t follow the path to success in Las Vegas to resolve poverty and economical depression commonly occurred in Hualien area. In Taiwan, Hualien enjoys the best development in infrastructure and various industries, visiting crowd of millions of every year, match every large-scale activity that the government of Hualien County plans elaborately (such as the main road of star light, the activity that the aboriginal holds a memorial ceremony for in bumper harvest year .etc.) Due to tourism downturn in recent years, as a result aggravating the budget deficit of local government, casino establishment is urgently considered as a way to prosper local economic activities. The policy and legislation of Taiwan government towards this issue remain conservative. Howe, most advanced countries in the world view liberalization and legalization of gambling as an effective method to regulate the long-existing, problematic industry. It is believed that casino liberalization in Taiwan is about a matter of time. Casinos will be legally established at the time of ripeness in the near future, Based on this assumption, this study summarizes preliminarily a number issues potentially present on the development of gaming business at Hualien with respect to infrastructure and tourism development at Hualien according to the analyses of reviewing the literature en the definition and categories cf casino, influence of gaming industry in all aspects, and current situation and perspective of tourism development at Hualien. This study firstly employed Participatory Local Appraisal (PLA) to form a consensus from local resident for sustainable development of Hualien area. Then, SWOT, TOWS matrix analysis, expert judged Fuzzy Delphi method to determine the most valuable issues on the development of casino industry at Hua1ien in both respects.

        Melatonin acts synergistically with pazopanib against renal cell carcinoma cells through p38 mitogen-activated protein kinase-mediated mitochondrial and autophagic apoptosis

        Background: Mounting evidence indicates that melatonin has possible activity against different tumors. Pazopanib is an anticancer drug used to treat renal cell carcinoma (RCC). This study tested the anticancer activity of melatonin combined with pazopanib on RCC cells and explored the underlying mechanistic pathways of its action. Methods: The 786-O and A-498 human RCC cell lines were used as cell models. Cell viability and tumorigenesis were detected with the MTT and colony formation assays, respectively. Apoptosis and autophagy were assessed using TUNEL, annexin V/propidium iodide, and acridine orange staining with flow cytometry. The expression of cellular signaling proteins was investigated with western blotting. The in vivo growth of tumors derived from RCC cells was evaluated using a xenograft mouse model. Results: Together, melatonin and pazopanib reduced cell viability and colony formation and promoted the apoptosis of RCC cells. Furthermore, the combination of melatonin and pazopanib triggered more mitochondrial, caspase-mediated, and LC3-II-mediated autophagic apoptosis than melatonin or pazopanib alone. The combination also induced higher activation of the p38 mitogen-activated protein kinase (p38MAPK) in the promotion of autophagy and apoptosis by RCC cells than melatonin or pazopanib alone. Finally, tumor xenograft experiments confirmed that melatonin and pazopanib cooperatively inhibited RCC growth in vivo and predicted a possible interaction between melatonin/pazopanib and LC3-II. Conclusion: The combination of melatonin and pazopanib inhibits the growth of RCC cells by inducing p38MAPK-mediated mitochondrial and autophagic apoptosis. Therefore, melatonin might be a potential adjuvant that could act synergistically with pazopanib for RCC treatment.

      • Geographic Information System (GIS) Technology Analysis and Implementation A Query System for Hakka Area Tourist Attraction of Southern Taiwan

        Tourism Bureau, Ministry of Transportation and Communication(MOTC) and The Council for Hakka Affairs (CHA) are promoting tourist attraction resources of hakka areas actively, and they also expect the data can be shared through the Internet. Furthermore, Geographic Information System are not only a system to show geographic graphic but also the best database to save space data. Nowadays, the applications of Geographic Information System in non-hakka areas are much more than in hakka areas, so this is value to advance. This research will investigate the tourist attraction resources of hakka area deeply, and establish a complete database with a query system through the Geographic Information System. Then it can supply for the tourists getting the traveling information and tourist attraction of hakka area fastly.
