        Adrenal myelolipoma is an unusual, nonfunctiomng benign tumor composed of mature fat and hematopoietic tissue. Most cases of adrenal myelolipomas are clinically silent and found incidentally at autopsy or radiological examinations done for other purposes. Their size is usually less than 5 ㎝ and they are managed non-operatively. We report a case of a surgically treated, giant adrenal myelolipoma in a 60-year-old man who presented with abdominal discomfort and we present a review of the literature.

      • Glomus Tumor of the Stomach : a case report

        Glomus tumor, also known as glomangioma, originates in the neuromyoarterial glomus, a normal arteriovenous shunt abundantly supplied with nerve fibers and fulfilling a temperature-regulating function. The classical location of the glomus tumor is the subungual region, but it can occur elsewhere in the skin, soft tissues, nerves, nasal cavity, trachea, and stomach. Here, we report a case of a 38-year-old female patient in whom glomus tumor of the stomach was detected at the routine health examination, and didn’t show any previous tumor-related signs and symptoms. The tumor was 2.5 cm in diameter and located in the muscularis proper. Gastrointestinal glomus tumors occur almost exclusively in the stomach, and there is a strong female predominance. Although most gastrointestinal glomus tumors are clinically benign, and have a good overall prognosis, a small possibility of malignant behavior can not be ruled out.

        Pathological significance of connexin 26 expression in colorectal adenocarcinoma.

        <P>This study was conducted to determine the level of expression and cellular localization of connexin 26 (Cx26) and the expression of p53 in colorectal adenocarcinoma as well as their relationship to clinicopathological features. Immunohistochemical staining was performed in 130 colorectal adenocarcinoma cases. A correlation between the expression levels of the two proteins and an analysis of the clinicopathological features of the samples was performed. There was a statistical significant difference in the Cx26 expression level among normal epithelium (NE), adenomas and adenocarcinomas (p<0.001). Of the 130 adenocarcinomas, 48.5% were positive for Cx26. All of the adenoma and NE samples were positive for Cx26 expression; however, the level of expression of Cx26 in adenomas was smaller than the level of expression for NE. Cytoplasmic staining for Cx26 was observed in the adenocarcinomas (23.8%), but was not observed in the adenoma and NE samples. A positive correlation between a reduction in intercellular Cx26 and tumor invasion was statistically significant (p<0.05). Expression of p53 was positive for 50% of the adenocarcinomas, and the level of p53 was increased in a reverse proportion to the level of Cx26 intercellular staining. In conclusion, loss of intercellular and gain of intracytoplasmic Cx26 expression may play a role in colorectal carcinogenesis and tumor progression.</P>

        Extracellular acidity enhances tumor necrosis factor-related apoptosis-inducing ligand (TRAIL)-mediated apoptosis via DR5 in gastric cancer cells

        The tumor microenvironment greatly influences cancer cell characteristics, and acidic extracellular pH has been implicated as an essential factor in tumor malignancy and the induction of drug resistance. Here, we examined the characteristics of gastric carcinoma (GC) cells under conditions of extracellular acidity and attempted to identify a means of enhancing treatment efficacy. Acidic conditions caused several changes in GC cells adversely affecting chemotherapeutic treatment. Extracellular acidity did inhibit GC cell growth by inducing cell cycle arrest, but did not induce cell death at pH values down to 6.2, which was consistent with down-regulated cyclin D1 and up-regulated p21 mRNA expression. Additionally, an acidic environment altered the expression of atg5, HSPA1B, collagen XIII, collagen XXAI, slug, snail, and zeb1 genes which are related to regulation of cell resistance to cytotoxicity and malignancy, and as expected, resulted in increased resistance of cells to multiple chemotherapeutic drugs including etoposide, doxorubicin, daunorubicin, cisplatin, oxaliplatin and 5-FU. Interestingly, however, acidic environment dramatically sensitized GC cells to apoptosis induced by tumor necrosis factor-related apoptosis-inducing ligand (TRAIL). Consistently, the acidity at pH 6.5 increased mRNA levels of DR4 and DR5 genes, and also elevated protein expression of both death receptors as detected by immunoblotting. Gene silencing analysis showed that of these two receptors, the major role in this effect was played by DR5. Therefore, these results suggest that extracellular acidity can sensitize TRAIL-mediated apoptosis at least partially via DR5 in GCs while it confers resistance to various type of chemotherapeutic drugs.

      • Cell Death and Intracellular Distribution of Hematoporphyrin in a KB Cell Line

        <P>OBJECTIVE: The objective of this study is to investigate the effect of intracellular photosensitizer distribution on tumor cell death after photodynamic therapy (PDT). BACKGROUND DATA: The photosensitizer accumulates in tumor tissue during PDT, and generates intracellular reactive oxygen species (ROS), resulting in tumor cell death. MATERIALS AND METHODS: This study was carried out to elucidate the effects of PDT in a KB oral cancer cell line using hematoporphyrin with irradiation at 635 nm and 5 mW/cm(2). After irradiation, the MTT reduction method, agarose gel electrophoresis, flow cytometry, and Diff-Quick staining were performed. The intracellular ROS level was measured by DCF-DA. Intracellular hematoporphyrin was monitored with a confocal microscope, and Western blot and caspase activity assays were performed. RESULTS: In our study, cell survival was reduced by about 50% after 3 h of hematoporphyrin incubation time. In DNA fragmentation, flow cytometry, and Diff-Quick assay, necrosis was identified within 12 h and apoptosis soon thereafter. Confocal microscopy revealed that hematoporphyrin was localized in the cell membrane, cytoplasm, and nucleus as time passed. The quantities of intracellular ROS correlated with the time of hematoporphyrin accumulation. Additionally, Western blot analysis of Bcl-2/Bax, the release of cytochrome C, and activity of caspase-3 and caspase-9 showed that apoptosis followed the mitochondria-dependent pathway. CONCLUSION: PDT with hematoporphyrin in the KB cell line showed morphological changes of cell necrosis and apoptosis, which were associated with the time of distribution and localization of hematoporphyrin. Also, the apoptosis evoked followed the mitochondria-dependent pathway.</P>

        1990년대 후반에 들어서면서 많은 기업들이 기업의 생산성 향상 및 고객 서비스를 위해 IT투자를 확대해 감에 따라 기업 내에서도 다수의 이질적인 시스템 도입을 추진하게 되었다. 그 결과 많은 기업 내에는 호환성이 없는 시스템이 산재해 업무추진에 어려움을 주고 있으며, 이러한 복잡하고 까다로운 분산화된 시스템을 통합하고자 업계에서는 많은 노력을 기울이게 되었다. 그 결과로 등장한 기술이 웹서비스이며 이것은 분산화된 시스템을 통합하는 기존의 통합기술들의 문제점을 해결하고 분산된 애플리케이션을 저비용 고효율로 통합하기 위한 대안이 되었다. 본 논문에서는 웹서비스기반 시스템 개발을 위한 웹서비스 개발 절차를 제시하고 비즈니스 도메인에서 요구사항 분석을 통해 생성된 유스케이스모델을 기반으로 하는 웹 서비스 식별 방법을 제안한다. 유스케이스를 기반으로 서비스를 식별하므로 서비스에 도메인의 비즈니스가 잘 반영되도록 할 수 있으며 적절한 입도(granularity)을 갖는 서비스를 식별할 수 있다.

        소프트웨어 아키텍처는 소프트웨어 시스템의 중요한 요소를 정의하고 이들 요소 간의 상호관계를 나타낸 전체 시스템의 기본 구조이다. 시스템의 변경과 확장에 용이하고 시스템의 상호 연동성을 확보하는데 있어 아키 텍처기반의 기술 접근은 필수적인 요소이다. 이러한 소프트웨어 아키텍처를 평가하기 위한 다양한 방법들이 제시되고 있으며, 이 방법들은 비즈니스요구를 반영하고 품질속성에 따른 시나리오를 만들어 그것을 기반으로 소프트웨어아키텍처를 평가하게 된다. 이에 본 논문에서는 ISO9126에서 제시한 소프트웨어의 품질을 기반으로 아키텍처 평가에 적용되어질 품질속성을 도출하기 위하여, 웹기반 전자상거래시스템인 B2C에서의 사용자요구사항을 분석해 보고자 한다.

        <P>Human gingival fibroblasts (hGFs) play an important role in the inflammatory reaction to lipopolysaccharide (LPS) from P. gingivalis, which infects periodontal connective tissue. In addition, although light-emitting diode (LED) irradiation has been reported to have biostimulatory effects, including anti-inflammatory activity, the pathological mechanisms of these effects are unclear. This study examined the effects of 635-nm irradiation of P. gingivalis LPS-treated human gingival fibroblasts on inflammatory cytokine profiles and the mitogen-activated protein kinase (MAPK) pathway, which is involved in cytokine production. Gingival fibroblasts treated or not treated with P. gingivalis LPS were irradiated with 635-nm LED light, and cytokine profiles in the supernatant were assessed using a human inflammation antibody array. Expression of cyclooxyginase-2 (COX-2) protein and phosphorylation of extracellular signal-regulated kinase (ERK 1/2), p38, and c-Jun-N-terminal kinase (JNK) were assessed by Western-blot analysis to determine the effects on the MAPK pathway, and prostaglandin E(2) (PGE(2)) in the supernatant was measured using an enzyme-linked immunoassay. COX-2 protein expression and PGE(2) production were significantly increased in the LPS-treated group and decreased by LED irradiation. LPS treatment of gingival fibroblasts led to the increased release of the pro-inflammatory-related cytokines interleukin-6 (IL-6) and IL-8, whereas LED irradiation inhibited their release. Analysis of MAPK signal transduction revealed a considerable decrease in p38 phosphorylation in response to 635-nm radiation either in the presence or absence of LPS. In addition, 635-nm LED irradiation significantly promoted JNK phosphorylation in the presence of LPS. LED irradiation can inhibit activation of pro-inflammatory cytokines, mediate the MAPK signaling pathway, and may be clinically useful as an anti-inflammatory tool.</P>
