        Mechanical Behaviors on T-shaped Hook-connected Structure Made of 2.5D Woven Composites and TC4 Alloy

        This work designed a T-shaped hook-connected structure composed of a bottom plate made of 2.5D woven composites and a T-shaped plate made of TC4 alloy. In order to explore to its bending and tensile properties, the corresponding experiments were designed and performed. Then, based on the progressive damage model and an improved Hashin’s failure criteria, numeral calculations were conducted in ANSYS to study its bending and tensile damage propagations. Compared with experimental results, the maximum error is less than 20 %, and the damage modes are similar, which proves the rationality of numerical calculation. In addition, both the experimental and simulation results show that the fatal damage both occurs at the constrained plate of the bottom plate whether in bending or tensile processes. Specifically, the fatal failure occurs at the installed edges in bending process or at the sides of groove-middle to groove-edge in tensile process. This work provides a design reference for typical T-shaped hook-connected composite structure, especially the connector of blade and casing, which appear in engineering and has good engineering application value.

        EXTINCTION AND PERMANENCE OF A KIND OF PEST-PREDATOR MODELS WITH IMPULSIVE EFFECT AND INFINITE DELAY

        In this paper, a kind of pest-predator model with impulsive effect and infinite delay is considered by the method of chain transform. By using Floquet's theorem, it is shown that there exists a globally asymptotically stable periodic pest eradication solution when the impulsive period is less than or equal to some critical value which is a directly proportional function with respect to the population of release. Furthermore, it is proved that the system is permanent if the impulsive period is larger than some critical value. Finally, the results of the corresponding systems are compared, those results obtained in this paper are confirmed by numerical simulation.

        The Associations of Family Functioning, General Well-Being, and Exercise with Mental Health among End-Stage Renal Disease Patients

        Objective This study aims to explore the relationships of family functioning, general well-being, and exercise with psychological distress. Furthermore, we investigated the special roles of general well-being and exercise on the association between family functioning and psychological distress. Methods Of 769 end-stage renal disease (ESRD) patients participated in the cross-sectional study which consisted of the 12-item General Health Questionnaire (GHQ-12), the Family APGAR Scales, and the General Well-Being Schedule. The collected data were analyzed using multiple linear regression analysis and path analysis. Results The prevalence of psychological distress was 72.3%. Family functioning, general well-being and exercise were associated factors of psychological distress (p<0.05). The indirect effect of family functioning on psychological distress was partially mediated by general well-being (Effect=-0.08, 95% CI=-0.11, -0.04). In addition, the effect of family functioning on general well-being was moderated by exercise (Index=-0.092, SE=0.033, 95% CI=-0.159, -0.029). Conclusion The prevalence of psychological distress among ESRD patients was high. Family functioning, general well-being and exercise were associated with psychological distress. Family functioning could affect psychological distress partially by affecting general wellbeing. Furthermore, exercise had a significant moderating effect on the relationship between family functioning and general well-being.

        A Novel Effect of Lipids Extracted from Vernix Caseosa on Regulation of Filaggrin Expression in Human Epidermal Keratinocytes

        Background: Vernix caseosa (VC), which is known as a unique human substance, is a biofilm that covers the skin of most human newborns. VC has many biological functions including anti-infective, skin cleansing and skin barrier repair. Objective: In the study, we purpose to investigate the novel effect of lipids extracted from VC on the regulation of filaggrin (FLG) expression and anti-inflammation in normal human epidermal keratinocyte (NHEK) cells. Methods: The lipids were extracted by chloroform/methanol (Folch method) and the major properties of fatty acid methyl esters were determined with gas chromatography-mass spectrometer. The relative viability of NHEK cells was evaluated by Cell Counting Kit 8 assay. The related expression of skin barrier protein was accessed with real-time quantitative polymerase chain reaction, Western blot and Immunofluorescence in NHEK cells with or without poly (I:C). Meanwhile, the changes of thymic stromal lymphopoietin (TSLP) and tumor necrosis factor alpha (TNF-α) are analyzed by enzyme- linked immunosorbent assay. Results: VC lipids mostly contained saturated and branched chains fatty acids. The expression of mRNA and protein of FLG were significantly increased after the supplement with lipid in NHEK cells. Meanwhile, lipids reversed the inhibition of poly (I:C) on FLG. Moreover, lipids suppressed the over secretion of TSLP and TNF-α induced by poly (I:C). Conclusion: These results indicate that lipids extracted from VC has positive effects on the expression of FLG and anti-inflammation, suggesting that lipids of VC may be used for a reference for novel therapeutic method in reducing and remedying skin disease like atopic disease.         Identification of putative olfactory genes in newly hatched larvae of a Coleopteran ectoparasitoid Dastarcus helophoroides (Fairmaire) (Coleoptera: Bothrideridae) by transcriptome analysis

        Dastarcus helophoroides (Fairmaire) (Coleoptera: Bothrideridae) is an effective ectoparasitoid of pine sawyer beetle Monochamus alternatus Hope (Coleoptera: Cerambycidae) which is the major median insect for transmission of pine wood nematode, Bursaphelenchus xylophilus (Steiner et Buhrer) Nickle (Aphelenchida: Parasitaphelenchidae). A sensitive and acute olfactory system is extremely essential for newly hatched larva of D. helophoroides to forage for a host. Therefore, we established the transcriptome of newly hatched D. helophoroides larvae and characterized the expression profiles of olfactory genes. We identified 20 transcripts encoding putative odorant binding proteins (OBPs), 11 putative chemosensory proteins (CSPs), 9 putative odorant receptors (ORs), 11 putative ionotropic receptors (IRs), 7 putative gustatory receptors (GRs) and 4 putative sensory neuron membrane proteins (SNMPs) olfactory genes from the transcriptome of newly hatched D. helophoroides larvae. According to the phylogenetic analysis, 3 OBPs (DhelOBP6, DhelOBP14 and DhelOBP19), 1 CSP (DhelCSP11), 1 OR (DhelOrco1), 3 IRs (DhelIR3, DhelIR6 and DhelIR8), 3 GRs (DhelGR2, DhelGR4 and DhelGR5) and 2 SNMPs (DhelSNMP1 and DhelSNMP3) had high similarities with reported Coleopteran species. The highest expressed olfactory gene was DhelCSP3, followed by DhelOBP8 and DhelCSP2. The expression levels of the majority of OR, IR, GR and SNMP genes were low. These results indicate that CSPs and OBPs may play important roles in the olfactory recognition process in newly hatched D. helophoroides larvae. The findings from the present study are helpful in unraveling the profound mystery between D. helophoroides and M. alternatus, as well as improving the parasitism efficiency of D. helophoroides larvae.

        Arginine Supplementation Recovered the IFN-gamma-Mediated Decrease in Milk Protein and Fat Synthesis by Inhibiting the GCN2/eIF2alpha Pathway, Which Induces Autophagy in Primary Bovine Mammary Epithelial Cells

        During the lactation cycle of the bovine mammary gland, autophagy is induced in bovine mammary epithelial cells (BMECs) as a cellular homeostasis and survival mecha-nism. Interferon gamma (IFN-) is an important antiproliferative and apoptogenic factor that has been shown to induce autophagy in multiple cell lines in vitro. However, it remains unclear whether IFN- can induce autophagy and whether autophagy affects milk synthesis in BMECs. To understand whether IFN- affects milk synthesis, we isolated and purified primary BMECs and investigated the effect of IFN- on milk synthesis in primary BMECs in vitro. The results showed that IFN- significantly inhibits milk synthesis and that autophagy was clearly induced in primary BMECs in vitro within 24 h. Interestingly, autophagy was observed following IFN- treatment, and the inhibition of autophagy can improve milk protein and milk fat syn-thesis. Conversely, upregulation of autophagy decreased milk synthesis. Furthermore, mechanistic analysis con-firmed that IFN- mediated autophagy by depleting argi-nine and inhibiting the general control nonderepressible-2 kinase (GCN2)/eukaryotic initiation factor 2 (eIF2) signaling pathway in BMECs. Then, it was found that arginine supplementation could attenuate IFN--induced autophagy and recover milk synthesis to some extent. These findings may not only provide a novel measure for preventing the IFN--induced decrease in milk quality but also a useful therapeutic approach for IFN--associated breast diseases in other animals and humans.
