9012AB is a recessive genic male sterility (RGMS) line in rapeseed, of which the male sterility is controlled by two pairs of recessive duplicate male sterile genes (Bnms3 and Bnms4) interacting with a recessive epistatic suppressor gene (esp). The recessive homozygosity at the esp locus (espesp) can suppress the expression of the recessive male sterility trait in homozygous plants (Bnms3Bnms3Bnms4Bnms4) and result in fertility restoration. A F2 population of 188 plants, derived from self-pollinated progenies of a 9012 AB fertile plants (BnMs3Bnms3Bnms4Bnms4EspEsp), was conducted to identify molecular markers linked to the recessive male sterility gene (Bnms3). By amplified fragment length polymorphism (AFLP) assay combining with bulked segregant analysis (BSA), 13 markers linked to Bnms3 were identified. Linkage analysis indicated that 13 AFLP markers were tightly linked to the Bnms3 gene with a genetic distance varying from 1.3 cM to 7.1 cM. Among them, one marker was co-dominant marker, 6 markers were in coupling phase with Bnms3, and the others were in repulsion phase with Bnms3 gene, One AFLP marker with a genetic distance of 1.4cM was further converted into a SCAR marker successfully, which have been applied in marker-assisted selection of RGMS lines and their temporary maintainers effectively.

Background and Aims: Scutellaria is one of the most popular traditional Chinese herbal remedies against various human diseases, including cancer. In this study, we examined the active effects of Scutellaria extract and its main flavonoid constituents on the proportion of side population cells within human multiple myeloma cell line RPMI8226 in vitro and explored the potential molecular mechanisms involved. Materials and Methods: The contents of flavonoids in ethanolic extract of Scutellaria baicalensis Georgi were determined using high performance liquid chromatography. The antiproliferative effect of the ethanolic extract on RPMI-8226 was determined by CCK assay. Apoptosis was measured by annexin combining with propidium iodide in a flow cytometer. Cell cycle analysis was performed by propidium iodide staining in combination with flow cytometry analysis. Hoechst 33342 exclusion assay was used for the identification of side population within RPMI8226 cells. The expression of ABCG2 protein was assessed by Western blotting assay. Results: The content of major flavonoids constitutents of Scutellaria extract was baicalin (10.2%), wogonoside (2.50%), baicalein (2.29%), and wogonin (0.99%), respectively. The crude Scutellaria extract did not show significant anti-proliferative effect, apoptosis induction and cell cycle arrest in RPMI-8226 within the concentrations of $1-75{\mu}g/mL$. However, the ethanolic extract, baicalein, wogonin and baicalin reduced the side population cells in RPMI-8226, and data showed that baicalein and wogonin had stronger inhibitory effects. Correspondingly, they also exhibited significant effects on decreasing the expression level of ABCG2 protein in RPMI-8226 in vitro. Conclusions: Our results for the first time demonstrated a novel mechanism of action for Scutellaria extract and its main active flavonoids, namely targeting SP cells by modulating the expression of ABCG2 protein. This study provides an insight for new therapeutic strategies targeting cancer stem cells of multiple myeloma.

Because of the unsatisfactory treatment options for breast cancer (BC), there is a need to develop novel therapeutic approaches for this malignancy. One such strategy is chemotherapy using non-toxic dietary substances and botanical products. Studies have shown that Panduratin A (PA) possesses many health benefits, including anti-inflammatory, anti-bacterial, anti-oxidant and anticancer activities. In the present study, we provide evidence that PA treatment of MCF-7 BC cells resulted in a time- and dosedependent inhibition of cell growth with an IC< SUB >50< /SUB > of 15 μM and no to little effect on normal human MCF-10A breast cells. To define the mechanism of these anti-proliferative effects of PA, we determined its effect critical molecular events known to regulate the cell cycle and apoptotic machinery. Immunofluorescence and flow cytometric analysis of Annexin V-FITC staining provided evidence for the induction of apoptosis. PA treatment of BC cells resulted in increased activity/expression of mitochondrial cytochrome C, caspases 7, 8 and 9 with a significant increase in the Bax:Bcl-2 ratio, suggesting the involvement of a mitochondrial-dependent apoptotic pathway. Furthermore, cell cycle analysis using flow cytometry showed that PA treatment of cells resulted in G0/G1 arrest in a dose-dependent manner. Immunoblot analysis data revealed that, in MCF-7 cell lines, PA treatment resulted in the dosedependent (i) induction of p21<sup>WAF1/Cip1</sup> and p27Kip1, (ii) downregulation of Cyclin dependent kinase (CDK) 4 and (iii) decrease in cyclin D1. These findings suggest that PA may be an effective therapeutic agent against BC.
Triterpenoids from Schisandra henryi with Cytotoxic Effect on Leukemia and Hela Cells In Vitro

Four known lanostane triterpenoids, schiprolactone A (1), schisanlactone B (2), nigranoic acid (3) and schisandronic acid (4) Were isolated from the stems of Schisandra henryi for the first time. Their structures were characterized by IR, MS and NMR techniques. Compounds 1, 2 and 4 showed moderate cytotoxic activity against Leukemia cells in vitro. Cytotoxic activity of compounds 1-4 showed $IC_{50}$ of 0.0097, 0.01, 0.097 and 0.0099 $\mu$ mol/mL respectively toward Leukemia cells and $IC_{50}$ of 0.097, 0.1, 0.097 and 0.099 $\mu$mol/mL toward Hela cells respectively. It is the first report that these compounds possess cytotoxic activity on Leukemia and Hela cells.

Glutathione peroxidase (GPX) is one of the important members of the antioxidant enzyme family. It can catalyze the reduction of hydroperoxides with glutathione to protect cells against oxidative damage. Single-chain variable fragment (scFv) can be converted into seleniumcontaining single-chain variable fragment (Se-scFv) by chemical modification of the hydroxyl groups in scFv, thus Se-scFv possesses GPX activity and becomes a prodrug. To improve the expression of scFv and simplify its purification steps, Single-protein production (SPP) system was used to express scFv and chemical modification was used to synthesize Se-scFv. Therefore, we must construct a new scFv-WCD1-lessACA gene, which can express its mRNA not containing any ACA sequences and express its amino acid sequence of target protein (scFv) being same to scFv-WCD1. In this way, the scFv-WCD1-lessACA can be only expressed in SPP system and no other background proteins in the cells could be expressed. The expression results showed that high level of scFv-WCD1-lessACA synthesis was at least sustained for 96 h in the virtual absence of background protein synthesis. Then, selenocysteine (Sec) was incorporated into the scFv-WCD1-lessACA by chemical modification and resulted in Se-scFv-WCD1-lessACA. The enzymatic characteristics of Se-scFv-WCD1-lessACA were determined. GPX activity was 2,563 U/μmol,its binding constant for GSH was 0.687 ×105/mol. Moreover,Se-scFv-WCD1-lessACA was confirmed to have a strong antioxidant ability to protect mitochondria against oxidative damage induced by Vc/Fe2+ (mitochondrial damage model),suggesting that Se-scFv-WCD1-lessACA has potential application for protection of mitochondrial damage induced by reactive oxygen species (ROS).

According to the China tumor registry 2013 annual report, breast cancer, lung cancer, and ovarian cancer are three common cancers in China nowadays, with high mortality due to the absence of early diagnosis technology. However, proteomics has been widespreadly implanted into every field of life science and medicine as an important part of post-genomics era research. The development of theory and technology in proteomics has provided new ideas and research fields for cancer research. Proteomics can be used not only for elucidating the mechanisms of carcinogenesis focussing on whole proteins of the tissue or cell, but also seeking the biomarkers for diagnosis and therapy of cancer. In this review, we introduce proteomics principles, covering current technology used in exploring early diagnosis biomarkers of breast cancer, lung cancer and ovarian cancer.

Activated carbon(AC) was prepared using brewing yeast as a precursor by chemical activation; meanwhile,cobalt, copper, ferrum and manganese supported on activated carbon(Co/AC, Cu/AC, Fe/AC, Mn/AC) were preparedby adsorption-activation method. The characterizations of prepared AC, Co/AC, Cu/AC, Fe/AC, Mn/AC and theirperformance as ozonation catalysts were tested. The total BET surface areas of prepared AC, Co/AC, Cu/AC, Fe/AC,Mn/AC were found to be 957.7, 789.7, 485.3, 486.1 and 529.8m2/g. Absorption capacities of methylene blue (MB) weredetermined to 407.77, 206.52, 121.25, 123.01, 170.94mg/g, respectively. The presence of AC was advantageous for TOCreduction of MB compared with UV/O3 system, and metal Co, Cu, Fe, Mn play an important role in the degradationprocess. The greatest TOC removal efficiency was obtained in the presence of Co/AC.

A study on submerged rotating MBR for wastewater treatment and membrane cleaning

A submerged rotating membrane bioreactor (SRMBR), with a rotatable, rounded, flat-sheet Poly(vinyldiene fluoride) (PVDF) membrane module fixed on hollow axes and moved by an electromotor, was used for wastewater reclamation. It was found that the effluent COD became stable and lower than 20 mg/L after one day running. The equilibrium permeate flux increased from 42.5 to 47.5 L/m2·h with the rotation speed increasing from 15 r/min to 25 r/min. Prolonging relaxation time could alleviate membrane fouling and enhance the flux. Finally, membrane cleaning was studied. The results showed that flushing the membrane surface with water, water/NaOH and water/NaOH/HCl recovered permeate flux to 48.4%, 83.5% and 90.2% of that of the initial operation, respectively.
