<P><I>In situ</I> hybridization and immunohistochemistry with different types of antibody (monoclonal vs. polyclonal, natural vs. synthetic) was compared to detect porcine circovirus 2 (PCV2) in formalin-fixed, paraffin-embedded tissues from pigs with experimentally and naturally occurring postweaning multisystemic wasting syndrome. PCV2 DNA and antigen was detected in tissues from both experimentally and naturally infected pigs by <I>in situ</I> hybridization and immunohistochemistry, respectively. Statistical evaluation revealed that more PCV2 positive signals were significantly detected in both experimentally and naturally infected pigs by <I>in situ</I> hybridization compared with immunohistochemistry (<I>P</I><0.05). The results of this study demonstrated that<I> in situ</I> hybridization proved more sensitive than immunohistochemistry for the detection of PCV2 in formalin-fixed, paraffin-embedded lymph node tissues.</P>

<P>The aim of the current study was to develop a nonradioactive in situ hybridization assay that can differentiate between genotypes 2a and 2b of Porcine circovirus-2 (PCV-2) in formalin-fixed, paraffin-embedded lymph node tissues from pigs with postweaning multisystemic wasting syndrome. Two different digoxigenin-labeled oligonucleotide probes were designed from the PCV-2 open reading frame 2 sequences. The PCV-2a-specific probe did not hybridize with formalin-fixed, paraffin-embedded lymph nodes from naturally PCV-2b-infected pigs and vice versa. Both PCV-2a-specific and PCV-2b-specific probes gave consistent negative signals in lymph nodes from naturally PCV-1-infected pigs. The in situ hybridization assay described in the present study represents a diagnostic tool that can differentiate between the 2 genotypes of PCV-2.</P>

<P>The objective of this study was to determine the capsular serotypes and potential virulence factors of Streptococcus suis isolated from pigs with polyserositis. Among the 24 isolates evaluated, serotype 3 [7 (29%) of the isolates] and serotype 4 [5 (21%)] were the most common. The isolates were also studied for the presence of the genes mrp, epf, and sly, which encode muramidase-released protein (MRP), extracellular factor (EF), and suilysin (SLY), respectively. Of the 24 isolates, 8 carried mrp: 4 of serotype 3, 2 of serotype 2, and 2 of serotype 4. One mrp(+) isolate (serotype 2) also carried the epf gene. All 24 isolates carried the sly gene. The serotype and genotype distribution greatly differed from that reported for isolates from pigs with other clinical manifestations of S. suis infection in other countries.</P>

염분 스트레스에 노출된 옥수수(Zea mays) 유식물의 원뿌리는 길이생장이 억제되는데, 본 연구에서는 위와 같은 조건에서 뿌리의 굴중성 반응을 조사하였다. 고농도의 NaCl과 KCl에 의해 스트레스를 받은 옥수수 원뿌리는 수평 상태에서 중력 자극을 받기 시작한 지 2시간 이후부터 대조구에 비하여 현저한 굴중성 반응을 나타내었다. 이러한 굴중성 촉진 현상은 Na?가 아니라 K?에 의한 것이었으며, Cl?도 굴중성반응 촉진에 기여하였다. 이로부터 이들 K?와 Cl? 이온들이 세포 팽창에 필요한 삼투 조절자로 기능한다는 결론을 얻었다. 또한 염분 스트레스에 의하여 뿌리의 길이 생장이 억제되더라도 중력장 내에서의 위치 결정은 억제되지 않고 오히려 촉진된다는 점도 확인하였다. We investigated gravitropic responses in the primary root of maize (Zea mays) seedlings which were exposed to salt stress. The maize roots salt-stressed with higher than 100 mM NaCl or KCl started to reveal enhanced gravitropic curvature after 2 hours form the gravi-stimulation. Such a promotion was not caused by sodium phosphate, but invoked by potassium phosphate, indicating the active component is K?. Because NaCl increased gravitropic curvature, despite that Na? did not played any role, we evaluated the role for Cl? by comparing the effects of MgCl₂ and MgSO₄. The enhancement of the curvature only with MgCl₂ revealed that Cl? played a role in the gravi-response, indicating the involvement of anion channels. These results suggest that both of K? and Cl? play roles in the regulation of osmosis that is required for cell expansion in gravitropism as well as in nyctinasty and stomatal opening.
Unilateral noncystic renal dysplasia in a Sprague Dawley rat

Renal dysplasia is a developmental disorder of the renal parenchyma involving anomalous differentiation. It is characterized by persistent metanephric ducts surrounded by primitive mesenchyme, fetal or immature glomeruli, fetal or immature tubules, interstitial fibrosis, and dysontogenic metaplasia involving tissues such as cartilage. Renal dysplasia has been rarely reported in rats. Here, we observed a small left kidney in a rat used in a short-term repeat toxicity study. The rat showed no clinical signs throughout the study. All parameters, including those reflecting kidney functions, were normal upon hematological examination and urinalysis. Grossly, the kidney was small (5 × 8 mm) and its surface appeared normal. Histological examination revealed that the cortex and medulla were poorly demarcated and contained immature/atrophic glomeruli, immature renal tubules, and mesenchymal cells. The cortex contained immature glomeruli, atrophic glomeruli with cystic dilatation of Bowman’s capsular space, and some atypical tubules. Primitive metanephric tubules in the medulla were larger in diameter than normal collecting ducts, lined by a tall columnar epithelium with pale cytoplasm and basal nucleus, and surrounded by loose mesenchymal cells. Occasional tubules contained pale eosinophilic homogenous material in the lumen. Thus, this was diagnosed as a case of renal dysplasia on the basis of histologic features and is the first reported case of renal dysplasia in Sprague Dawley rats.
