        Interleukin-1B (IL-1B-31 and IL-1B-511) and interleukin-1 receptor antagonist (IL-1Ra) gene polymorphisms in primary immune thrombocytopenia

        Background: Immune thrombocytopenia (ITP) is an immune-mediated disease caused by autoanti-bodies against platelets membrane glycoproteins GPIIb/IIIa and GPIb/IX. The etiology of ITP remains unclear. This study evaluated the association of polymorphisms in interleukin (IL)-1B-31, IL-1B-511, and IL-1Ra with ITP. Methods: Genotyping of IL-1B-31, IL-1B-511, and IL-1Ra was performed in 118 ITP patients and 100 controls by polymerase chain reaction restriction fragment length polymorphism and detection of variable number tandem repeats. Results: Genotype differences in IL-1B-31 and IL-1Ra were significantly associated with ITP. Patients showed a higher frequency of the IL-1B-31 variant allele (T) and a 1.52-fold greater risk of susceptibility to ITP (odds ratio [OR]=1.52, 95% confidence interval [CI]=1.04‒2.22, P=0.034). The frequencies of both homozygous and heterozygous variant geno-types of IL-1B-31 were higher (OR=2.33, 95% CI=1.069‒5.09, P=0.033 and OR=2.044, 95% CI=1.068‒39, P=0.034) among patients and were significantly associated with ITP susceptibility. Both homozygous and heterozygous variant genotypes of IL-1Ra were also more frequent (OR=4.48, 95% CI=1.17‒17.05, P=0.0230 and OR=1.80, 95% CI=1.03‒3.14, P=0.0494) among patients and were associated with ITP risk. IL-1B-31 and IL-1Ra also showed significant association with severe ITP. However, IL-1B-511 was not asso-ciated with ITP. Conclusion: IL-1B-31 and IL-1Ra polymorphisms may significantly impact ITP risk, and they could be associated with disease severity, which may contribute to the pathogenesis of ITP.

      • Seroprevalence of Hepatitis B Virus and Hepatitis C Virus Among Patients Visiting Tertiary Care Hospital of Western Nepal

        Aims: Hepatitis viruses including hepatitis B and hepatitis C are major public health problems and significant cause of mortality and morbidity especially in developing countries like Nepal. Patients with hepatitis B and hepatitis C virus infections are important sources for spread of infection to close contact population. This study was aimed to determine the sero-prevalence of hepatitis B virus (HBV) and hepatitis C virus (HCV) infection among the patients visiting Manipal Teaching hospital, Pokhara, Nepal. Co infection among Human Immunodeficiency Virus (HIV) infected patients was also studied. Methods: This retrospective study was conducted over a period of four years from 2014 to 2018. A total of 32410 serum samples were tested. Screening of HBV, HCV and HIV was carried out by rapid diagnostic test kits and confirmed by Enzyme Linked Immunosorbent Assay (ELISA). Results: The overall sero-prevalence of HBV and HCV was 0.96% (314/32410) and 0.29% (96/32410) respectively. Infection of both hepatitis B and hepatitis C virus was detected in 9 patients. Hepatitis B and hepatitis C co-infection among HIV positive cases was found in 8 patients. Prevalence of hepatitis B and Hepatitis C was found higher among male patients as compared to females, however differences were statistically insignificant. Most of the cases of HBV and HCV were detected among the age group of 25 to 35 years. Conclusions: Despite availability of effective vaccines, HBsAg carrier rate was found high as compared to HCV. Regular screening of patients for HBV and HCV would minimize the transmission among close contact group and healthcare workers. Effective vaccination program at national level would be effective to reduce the consequences.

        <P><B>Abstract</B></P> <P>Present study reports the synthesis of Ag<SUB>3</SUB>PO<SUB>4</SUB> nano/mircoparticles by facile precipitation and colloidal synthesis approach in aqueous solution. Importantly, the presented approach does not require complicated instruments, difficult processes, and harsh conditions such as high temperature and high pressure. The obtained samples were characterized using SEM with EDS, XRD, TEM with HR-TEM and SAED, and FTIR. Their optical properties were studied using UV–vis diffusive reflectance spectra. Size dependent antibacterial activity towards Gram-positive (S. aureus) and Gram-negative (E. coli, K. pneumoniae and P. aeruginosa) bacteria. Experimental results showed that the antibacterial performances of nanoparticles obtained by mixing 0.01M of each reactant (colloidal synthesis approach) were found to be much higher than that of other formulation under identical conditions.</P>
