        EBV-miR-BHRF1-1 Targets p53 Gene: Potential Role in Epstein-Barr Virus Associated Chronic Lymphocytic Leukemia

        Purpose The purpose of this study was to investigate the prognostic impact of Epstein-Barr virus (EBV)–microRNA (miRNA, miR)-BHRF1-1 with chronic lymphocytic leukemia (CLL) as well as role of EBV-miR-BHRF1-1 in p53 gene. Materials and Methods Quantitative reverse transcription–polymerase chain reaction and western blotting were used to quantify EBV-miR-BHRF1-1 and p53 expression in cultured CLL. Results p53 aberration was associated with the higher expression level of EBV-miR-BHRF1-1 (p < 0.001) which was also an independent prognostic marker for overall survival (p=0.028; hazard ratio, 5.335; 95% confidence interval, 1.193 to 23.846) in 97 newly-diagnosed CLL patients after adjusted with International Prognostic Index for patients with CLL. We identified EBV-miR-BHRF1-1 as a viral miRNA regulator of p53. EBV-miR-BHRF1-1 repressed luciferase reporter activity by specific interaction with the seed region within the p53 3- untranslated region. Discordance of p53 messenger RNA and protein expression was associated with high EBV-miR-BHRF1-1 levels in CLL patients and cell lines. EBV-miR-BHRF1- 1 inhibition upregulated p53 protein expression, induced cell cycle arrest and apoptosis and decreased cell proliferation in cell lines. EBV-miR-BHRF1-1 mimics downregulated p53 protein expression, decreased cell cycle arrest and apoptosis, and induced cell proliferation in cell lines. Conclusion This study supported the role of EBV-miR-BHRF1-1 in p53 regulation in vitro. Our results support the potential of EBV-miR-BHRF1-1 as a therapeutic target in EBV-associated CLL with p53 gene aberration.

        밀로 라우의 뉴리얼리즘 -재연을 통한 허구와 실재의 작용-

        밀로 라우는 2018년 겐트 선언문을 통해서 연극은 더 이상 세상을 보여주는 게 아니라 세상을 바꿔야 한다고 주장한다. 연극은 세상의 진실을 보여줄 수 있어야 하고, 이를 통해 변화의 가능성들이 제기되어야 한다는 것이다. 그는 논쟁의 여지가 많은 역사 속 사건을 선택하고 그것을 재연이라는 방식을 통해 현재로 소환한다. 이때 재연은 과거의 사건을 현실에서 연극이라는 허구의 세계로 옮겨오는 역할을 수행한다. 리얼리즘이 재현을 통해 연극의 허구성을 감추고 완결된 환상을 지향한다면, 라우는 재연을 통해 연극의 허구성을 인정하고 환상이 아닌 실재를 연출하는 뉴리얼리즘을 주장한다. 허구와 실재라는 서로 상반되는 개념이 충돌하고 공존하면서 연극 안에서 과거의 사건은 현재로 소환된다. 관객은 역사적 사건에 감정적으로 이입되는 동시에, 지금 관객의 눈앞에서 다시 반복되는 사건의 현재화를 통해 퍼포먼스적인 측면에서의 현장성을 경험한다. 무엇보다 재연은 과거 사건에 대한 하나의 입장 혹은 태도를 전제하기 때문에, 연출가의 예술적 해석을 반영한다. 본 논문에서는 라우의 독특한 연극 미학을 고찰하고, 그가 2016년에 연출한 <다섯 개의 소품들>을 중심으로 재연이라는 방식이 어떻게 작동하고 어떠한 의미를 산출하는지에 집중한다. 무엇보다 그가 일관되게 사용하는 재연 기법이 초기 작품에서 현재에 이르기까지 어떻게 변주되는지 그 양상을 추적하고 이에 따른 효과를 분석하고자 한다. 이를 통해 아직 국내에서 활발하게 다뤄지지 않은 라우의 연극 미학에 대한 관심을 촉발하고 이후 연구를 위한 초석을 마련하고자 한다. In 2018, Milo Rau proclaims in his Ghent Manifesto that theater is not just about portraying the world anymore. It’s about changing it. Theatre should be able to reveal the truths of the world and the possibilities of change must be raised through this. He chooses controversial events from the past and summons them to our present by using the form of ‘reenactment’. This reenactment plays a pivotal role of transferring the reality of the past events into the fictional world of theater. In other words, if ‘Realism’ pursues the complete illusion which hides the fictionality of theater through the representation of reality, then what Rau suggests is that he accepts the fictional aspects of theater, but instead of directing illusion he directs the reality. Which is what he calls the ‘New Realism’. In Rau's plays, the past events become our present as the two of the opposing ideas, so called fiction and reality clashes and co-exists. As the audiences make the emotional connection to the past events, they also witness and experience the liveness as in the form of performance through the past events being repeated in front of their present eyes. Most of all, the reenactment premises one of many stances or opinions about the past events, so it reflects the director’s artistic interpretation. In this dissertation, I will be contemplating Rau’s distinctive theatrical aesthetics, and focus on the mechanism of reenactment, and what meanings it produces by studying the play Five Easy Pieces which he directed in 2016. Above all, I will be tracking the variations of his consistent use of this reenactment method from his early plays to the most recent ones and analyze the effects that follows. Through this, I wish to trigger the interests toward Rau’s aesthetics of theatre, which hasn’t been actively mentioned in Korea. And thus create a foundation for a further study of Milo Rau and his theatre.

        Fabrication of Photopolymer Hierarchical Micronanostructures by Coupling Electrospinning and Photolithography for SERS Substrates

        Reported here is the fabrication of photopolymer hierarchical micronanostructures through a combinative process of electrospinning and subsequent photolithography. Electrospun SU-8 (epoxy-based negative photoresist)nanofiber films have been patterned into gratings with periods of 100, 200, 300, and 400 μm, respectively. Deposition of a silver nanolayer on these interlaced nanofiber films would lead to the formation of various plasmonic nanostructures,and therefore, giving rise to abundant surface-enhanced Raman scattering (SERS) “hot spots”. In the detection of Rhodamine 6G (R6G), probing molecule, the resultant SERS substrates show both high sensitivity and good reproducibility. The SERS enhancement factor could reach as high as ~108, indicating high efficiency. The fabrication of patterned, highly efficient SERS substrates may hold a great promise for the integration of SERS substrates in various microdevices such as microfluidic chips.

      • Distinct Pro-Apoptotic Properties of Zhejiang Saffron against Human Lung Cancer Via a Caspase-8-9-3 Cascade

        Lung cancer is the leading cause of cancer-related death worldwide. Here we investigated the antitumor effect and mechanism of Zhejiang (Huzhou and Jiande) saffron against lung cancer cell lines, A549 and H446. Using high performance liquid chromatography (HPLC), the contents of crocin I and II were determined. In vitro, MTT assay and annexin-V FITC/PI staining showed cell proliferation activity and apoptosis to be changed in a dose- and time-dependent manner. The inhibition effect of Jiande saffron was the strongest. In vivo, when mice were orally administered saffron extracts at dose of 100mg/kg/d for 28 days, xenograft tumor size was reduced, and ELISA and Western blotting analysis of caspase-3, -8 and -9 exhibited stronger expression and activity than in the control. In summary, saffron from Zhejiang has significant antitumor effects in vitro and in vivo through caspase-8-caspase-9-caspase-3 mediated cell apoptosis. It thus appears to have more potential as a therapeutic agent.

        커뮤니케이션 플랫폼의 상호작용이 동일시와 추천 의도에 미치는 영향

        코로나19의 영향이 장기화되면서 오프라인 활동은 중단되었고 온라인 소통이 더욱 큰 비중을 차지하게 되었다. 한류와 인터넷 정보 기술의 비약적인 발전과 함께 다양한 엔터테인먼트 커뮤니케이션 플랫폼이 탄생했다. 팬들은 플랫폼을 통해 스타 및 다른 팬들과 소통하고 정보를 공유할 수 있게 되었다. 이러한 방식은 엔터테인먼트 커뮤니케이션 플랫폼에 대한 사용자의 가치 지각을 향상하고 공감을 자아내며, 플랫폼 추천 의도를 강화할 수 있다. 본 연구는 설문조사를 통해 엔터테인먼트 커뮤니케이션 플랫폼 내 사용자 간의 상호작용, 동일시와 추천 의도의 영향에 대해 실증분석을 실시하였다. 연구 결과는 다음과 같다. 첫째, 팬과 콘텐츠 상호작용은 심리 동일시와 행위 동일시에 긍정적인 영향을 미치는 것으로 나타났다. 둘째, 팬과 팬의 상호작용은 심리 동일시와 행위 동일시에 영향을 미치지 않는 것으로 나타났다. 셋째, 팬과 스타의 상호작용은 심리 동일시와 행위 동일시에 긍정적인 영향을 미치는 것으로 나타났다. 넷째, 심리 동일시와 행위 동일시는 커뮤니티 구성원의 추천 의도에 긍정적인 영향을 미치며, 행위 동일시는 심리 동일시와 추천 의도의 관계 중에서 부분 매개 효과가 있는 것으로 나타났다. 이상의 분석 결과를 토대로 연구의 시사점과 한계점 및 향후 연구방향을 제시하였다. Under the long-term influence of COVID-19, offline activities were interrupted and online communication became the main way. With the rapid development of Korean Wave and network information technology, there have been many entertainment communication platforms. Fans can communicate with stars and other fans and share information through entertainment and communication platforms. This can improve users" perception of the value of entertainment communication platforms, arouse emotional resonance and have a positive impact on users" platform recommendation intention. In this study, the influence of user interaction, identity and recommendation intention of entertainment communication platforms was investigated by questionnaire. The results are as follows: First, the interaction between fans and content has a positive effect on psychological and behavioral identity. Second, the interaction between fans does not affect their psychological and behavioral identity. Third, the interaction between fans and stars has a positive impact on psychological identity and behavior identity. Fourth, psychological identity and behavioral identity have a positive impact on community members" willingness to recommend. Behavioral identity plays a partial mediating role between psychological identity and recommendation intention. Based on the above analysis results, the present situation, limitations and future research directions of this study are put forward.

        A Study on Biophilia Applied in Landscape Design of Chinese Old-age Care Community

        인구의 고령화는 이미 중국의 중요한 사회 문제가 되었다. 이러한 고령화 문제를 개선하기 위한 방법으로 중국정부는 의료를 기본으로 요양을 결합한 ‘의양’ 커뮤니티 건설을 제안했다. ‘의양’ 커뮤니티는 도시 경관의 기본 단위로서 조경을 계획하며, 노년층의 안락한 거주성을 제공하며 또한 사회적 관계를 유지할 수 있도록 하는 환경이다. 본 연구는 이를 바탕으로 문학 테이더 방식을 사용하여 중국의 고령화 환경, 중국의 현재 양로 커뮤니티의 조경 디자인, 고령자의 정신 건강을 분석하였다. 그 결과 현행 양로 커뮤니티는 경관과 정신 건강 사이의 긍정적인 효과를 간과하고 있음을 발견했다. 이를 바이오필리아 관점으로 경관의 긍정적 효과를 결합시켜 ‘의양 결합’ 커뮤니티의 조경 디자인 개념을 도출하였다. 미래 ‘의양 결합’ 커뮤니티의 조경 디자인을 논의하고 ‘의양 결합’ 커뮤니티의 경관 환경에서 인간과 자연 사이의 긍정적인 관계를 구성한 설계 방법을 제안하고자 한다. 본 논문의 목적은 ‘의양 결합’ 커뮤니티가 노인의 정신 건강에 긍정적인 영향을 줘 생활환경의 질을 향상시킬 수 있는 경관 디자인을 제안하는 것이다. The aging of the population has become an important social issue in China. In order to improve the growing aging problem, China has proposed to construct a community with the combination of medical and health care, which is based on the medical treatment with recuperation as the core to solve social problems. As the basic unit of urban landscape, the community landscape can not only provide the space and place for the elderly, but also meet the psychological and social needs of the elderly. On this background, through the data study, analyze the China’s aging environment, the landscape design of Medical and Health Care Community, and the Psychological Status of Elderly. It is found that there is less research on the positive effects between landscape and mental health in the old-age care community in China. Therefore, this paper makes a summary of the concept of biophilia landscape design in medical and health care community through the positive effects of the natural landscape of biophilia hypothesis. What’s more, it discusses the landscape design of the medical and health care community in the future, and puts forward the design methods for reconstructing the positive relation between man and nature in the landscape environment of such a community. This paper aims to make the landscape environment of the medical and health care community in the future play a positive role in the mental health of the elderly people, so that the mental health of the aged will be effectively improved and the quality of their living space will be enhanced.

        Five new anthraquinones from the seed of Cassia obtusifolia

        A phytochemical investigation on the seeds ofCassia obtusifolia led to the isolation of five new anthraquinones,including one aglycon and four glycosides. Thestructures were elucidated by analysis of extensive spectroscopicdata and the results of acid hydrolysis. All theseisolates were evaluated for their inhibitory effects againsta-glucosidase, and 1 showed potent activity with IC50value of 185 ± 15 lM.

        Operational modal analysis of structures by stochastic subspace identification with a delay index

        Practical ambient excitations of engineering structures usually do not comply with the stationary-white-noise assumption in traditional operational modal analysis methods due to heavy traffic, wind guests, and other disturbances. In order to eliminate spurious modes induced by non-white noise inputs, the improved stochastic subspace identification based on a delay index is proposed in this paper for a representative kind of stationary non-white noise ambient excitations, which have nonzero autocorrelation values near the vertical axis. It relaxes the stationary-white-noise assumption of inputs by avoiding corresponding unqualified elements in the Hankel matrix. Details of the improved stochastic subspace identification algorithms and determination of the delay index are discussed. Numerical simulations on a four-story frame and laboratory vibration experiments on a simply supported beam have demonstrated the accuracy and reliability of the proposed method in eliminating spurious modes under non-white noise ambient excitations.

        In power system, the voltage pulsation and deficiency or surplus of reactive power will have a great effect on the quality of electric power supply. Dynamicly local reactive power compensation is one of the best methods to reduce the power network loss and improve the efficiency of transmission and distribution.Through the effective methods of compensation, we can keep the balance of reactive power in the maximum degree by the maintenance of acceptable voltage. This paper firstly describes the principles of synthetic compensation of reactive power and voltage then analyzes the principle of magnetically controlled reactor-MCR in detail and derives the characteristics, secondly analyses the disadvantages of the conventional reactive power compensation devices and the advantages of MCR, thirdly on this basis designs a control strategy of reactive power and voltage which is based on MCR,so as to achieve the goal of dynamic reactive power compensation. At the end of paper, simulator and experimental research are carried out for the model of compensation method; the results accord with the theoretical analyses done before. So the compensation method is proved to be accurate in theory and feasible in practice.
