        Zeitgeschichte im Fernsehen. Zur Darstellung der Wiedervereinigung in Hans-Christoph Blumenbergs Dokudrama Deutschlandspiel

        최근 독일의 역사적 사건을 다루는 텔레비전 프로그램은 독일에서 약 20년 전부터 높은 인기를 누리고 있다. 이러한 프로그램은 폭넓은 관객을 만날 수 있기 때문에 사회의 문화적 기억을 확고히 하는데 중요한 역할을 수행한다. 텔레비전의 역사프로그램은 특정 역사적 사건이나 상황이 사람들에게 기억되고,역사적 사건들에 어떤 의미가 부여되는지에 영향을 끼치는 것이다. 다큐드라마영화는 한 극영화에서 다큐적 기록과 허구의 영화적 요소들을 연결시키는 흥미로운 장르이다. 본 논문은 독일의 통일 과정을 담은 한스-크리스토프 블루 멘베르크의 2000년 영화 「독일게임」을 두 가지 관점에서 고찰하고자 했다.우선 다큐드라마영화가 형식적이고 생산미학적인 관점에서 어떻게 다큐기 록물이나 인터뷰장면들을 영화의 사건들과 서로 연결시키는지에 대해 분석하고자 했다. 영화의 몽타주기법을 통해 이 세 가지의 상이한 영화재료들은 서로 교차되어 하나의 완성된 영화로 만들어지는데, 이를 통해 영화 속의 허구로 창작된 부분들도 다큐적인 부분만이 지니고 있는 신빙성과 진정성을 획득하게 됨을 볼 수 있었다. 다른 한편으로는 영화의 내용적인 부분으로 통일의 이야기를 어떻게 보여주는지, 즉 역사적 사건에 대해 어떠한 해석을 관객들에게 전달 하는지를 분석하고자 했다. 이 작업을 통해 독일 통일의 과정이 특정 정치인의 이야기를 중심으로 설명되고 극적인 첨예화를 통해 몇몇 정치인들의 단독적인 결정의 결과물로 나타나는 것을 확인할 수 있었다. 또한 정치인들의 인간적인차원이 강조되면서 관객들은 그들과 또한 그들이 내린 결정에 동일시할 수 있게 된다.본 논문은 『독일게임』의 예를 통해 다큐드라마라는 영화장르가 역사적 사건 에 있어 특정 읽기 방식만을 보여줌에도 불구하고, 형식적인 특징 때문에 신빙 성을 획득하여 역사적 사건이 실제로 어떻게 일어났는지를 보여주는 것처럼 기능하는 장르의 문제점을 밝히고자 했다.

        68Ga-PSMA PET/CT Imaging Predicting Intraprostatic Tumor Extent, Extracapsular Extension and Seminal Vesicle Invasion Prior to Radical Prostatectomy in Patients with Prostate Cancer

        Purpose 68Ga-labeled prostate-specific membrane antigen (PSMA) ligand positron emission tomography/computed tomography (PET/CT) has shown promising results in patients with biochemical recurrence after primary therapy for prostate cancer. In this study, we evaluated the usefulness of PSMA I&T (imaging and therapy) PET/CT prior to radical prostatectomy. Methods The study population consisted of 21 patients with prostate cancer who underwent 68Ga-PSMA I&T PET/CT before either open or laparoscopic radical prostatectomy. Intraprostatic tumor extent, extracapsular extension (ECE) and seminal vesicle invasion (SVI) were assessed on the PET/CT scans. Tracer uptake was quantified in terms of standardized uptake values (SUVs). Imaging findings were correlated with final whole-gland histopathology. Results Of the 21 patients, two had T stage 2b disease, nine stage 2c, six stage 3a and four stage 3b. The median Gleason score was 7. The SUVmean of the primary tumors was 9.5 ± 8.8. SUVmean was higher in tumors with ECE than in organconfined tumors (13.8 ± 11.0 vs. 5.6 ± 3.2, p = 0.029). Peak tracer uptake was significantly positively correlated with Gleason score (rs = 0.49, p = 0.025). Sensitivity, specificity, positive predictive value and negative predictive value were, respectively, 94.7%, 75.0%, 97.3% and 60.0% for tumor infiltration of an individual prostate lobe, 75.0%, 100.0%, 100.0% and 97.4% for SVI, and 90.0%, 90.9%, 90.0% and 90.9% for ECE, using an angulated contour of the prostate as the criterion. Tumor volume derived from 68Ga-PSMA I&T PET/CT was significantly correlated with preoperative prostate-specific antigen value (rp = 0.75, p < 0.001) and tumor volume on histopathology (rp = 0.45, p = 0.039). Conclusions 68Ga-PSMA I&T PET/CT prior to radical prostatectomy can contribute to presurgical local staging of prostate cancer. In this pilot study, 68Ga-PSMA I&T PET/CT showed promising results for prediction of lobe infiltration, ECE and SVI.

He is also an associate lecturer at PSB Academy of Singapore. He has won the Bronze Award in 2005 at the prestigious International Institute of Insolvency for his outstanding research work in cross-border insolvency.

        Towards Methionine Overproduction in Corynebacterium glutamicum -Methanethiol and Dimethyldisulfide as Reduced Sulfur Sources

        In the present work, methanethiol and dimethyldisulfide were investigated as sulfur sources for methionine synthesis in Corynebacterium glutamicum. In silico pathway analysis predicted a high methionine yield for these reduced compounds, provided that they could be utilized. Wild-type cells were able to grow on both methanethiol and dimethyldisulfide as sole sulfur sources. Isotope labeling studies with mutant strains, exhibiting targeted modification of methionine biosynthesis, gave detailed insight into the underlying pathways involved in the assimilation of methanethiol and dimethyldisulfide. Both sulfur compounds are incorporated as an entire molecule, adding the terminal S-CH3 group to O-acetylhomoserine. In this reaction, methionine is directly formed. MetY (O-acetylhomoserine sulfhydrylase) was identified as the enzyme catalyzing the reaction. The deletion of metY resulted in methionine auxotrophic strains grown on methanethiol or dimethyldisulfide as sole sulfur sources. Plasmid-based overexpression of metY in the △metY background restored the capacity to grow on methanethiol or dimethyldisulfide as sole sulfur sources. In vitro studies with the C. glutamicum wild type revealed a relatively low activity of MetY for methanethiol (63 mU/mg) and dimethyldisulfide (61 mU/mg). Overexpression of metY increased the in vitro activity to 1,780 mU/mg and was beneficial for methionine production, since the intracellular methionine pool was increased 2-fold in the engineered strain. This positive effect was limited by a depletion of the metY substrate O-acetylhomoserine, suggesting a need for further metabolic engineering targets towards competitive production strains.

        This review aims to provide an overview and update of current literature on the use of intranasal (IN) drug delivery in pediatric emergency medicine (PEM), in terms of the anatomy, physiology, pharmacokinetics, limitations, drug delivery methods, necessary training, safety, contraindications, effectiveness, current indications and trends, and implications for clinical practice and future developments in IN drug administration. We evaluate how IN medication use in PEM has recently evolved, what recent research has revealed about the utility of IN drug delivery in PEM, and what the future of IN drug delivery might look like.

        Gene Expression Profiling and Assessment of Vitamin D and Serotonin Pathway Variations in Patients With Irritable Bowel Syndrome

        Background/Aims Irritable bowel syndrome (IBS) is a multifaceted disorder that afflicts millions of individuals worldwide. IBS is currently diagnosed based on the presence/duration of symptoms and systematic exclusion of other conditions. A more direct manner to identify IBS is needed to reduce healthcare costs and the time required for accurate diagnosis. The overarching objective of this work is to identify gene expression-based biological signatures and biomarkers of IBS. Methods Gene transcripts from 24 tissue biopsy samples were hybridized to microarrays for gene expression profiling. A combination of multiple statistical analyses was utilized to narrow the raw microarray data to the top 200 differentially expressed genes between IBS versus control subjects. In addition, quantitative polymerase chain reaction was employed for validation of the DNA microarray data. Gene ontology/pathway enrichment analysis was performed to investigate gene expression patterns in biochemical pathways. Finally, since vitamin D has been shown to modulate serotonin production in some models, the relationship between serum vitamin D and IBS was investigated via 25-hydroxyvitamin D (25[OH]D) chemiluminescence immunoassay. Results A total of 858 genetic features were identified with differential expression levels between IBS and asymptomatic populations. Gene ontology enrichment analysis revealed the serotonergic pathway as most prevalent among the differentially expressed genes. Further analysis via real-time polymerase chain reaction suggested that IBS patient-derived RNA exhibited lower levels of tryptophan hydroxylase-1 expression, the enzyme that catalyzes the rate-limiting step in serotonin biosynthesis. Finally, mean values for 25(OH)D were lower in IBS patients relative to non-IBS controls. Conclusions Values for serum 25(OH)D concentrations exhibited a trend towards lower vitamin D levels within the IBS cohort. In addition, the expression of select IBS genetic biomarkers, including tryptophan hydroxylase 1, was modulated by vitamin D. Strikingly, the direction of gene regulation elicited by vitamin D in colonic cells is “opposite” to the gene expression profile observed in IBS patients, suggesting that vitamin D may help “reverse” the pathological direction of biomarker gene expression in IBS. Thus, our results intimate that IBS pathogenesis and pathophysiology may involve dysregulated serotonin production and/or vitamin D insufficiency. (J Neurogastroenterol Motil 2018;24:96-106)

        A Methodology for Customized Prediction of Energy Consumption in Manufacturing Industries

        Regulative measures and rising energy cost foster the trend towards energy efficient manufacturing. However, companies are facing hurdles such as high effort and little expertise when implementing energy efficiency measures. Moreover, the embodied energy of products cannot be determined accurately in eco-assessments of factories. This paper presents a methodology for the reliable prediction of energy consumption of arbitrary manufacturing processes. It is based on minimal measurements and requires little effort and previous knowledge due to precise guidelines. Consumption models help to allocate energy cost to products, to calculate the product carbon footprint and to derive and validate measures to improve energy efficiency in production. The methodology has been applied in a medium size company with a large number of different products and machines.

        Investigation of the Central Carbon Metabolism of Sorangium cellulosum: Metabolic Network Reconstruction and Quantification of Pathway Fluxes

        In the present work, the metabolic network of primary metabolism of the slow-growing myxobacterium Sorangium cellulosum was reconstructed from the annotated genome sequence of the type strain So ce56. During growth on glucose as the carbon source and asparagine as the nitrogen source, So ce56 showed a very low growth rate of 0.23 d-1, equivalent to a doubling time of 3 days. Based on a complete stoichiometric and isotopomer model of the central metabolism, 13C metabolic flux analysis was carried out for growth with glucose as carbon and asparagine as nitrogen sources. Normalized to the uptake flux for glucose (100%), cells recruited glycolysis (51%) and the pentose phosphate pathway (48%) as major catabolic pathways. The Entner-Doudoroff pathway and glyoxylate shunt were not active. A high flux through the TCA cycle (118%) enabled a strong formation of ATP, but cells revealed a rather low yield for biomass. Inspection of fluxes linked to energy metabolism revealed that S. cellulosum utilized only 10% of the ATP formed for growth, whereas 90% is required for maintenance. This explains the apparent discrepancy between the relatively low biomass yield and the high flux through the energy-delivering TCA cycle. The total flux of NADPH supply (216%) was higher than the demand for anabolism (156%), indicating additional reactions for balancing of NADPH. The cells further exhibited a highly active metabolic cycle, interconverting C3 and C4 metabolites of glycolysis and the TCA cycle. The present work provides the first insight into fluxes of the primary metabolism of myxobacteria, especially for future investigation on the supply of cofactors, building blocks, and energy in myxobacteria, producing natural compounds of biotechnological interest.
