Background: Asiatic acid is a putative anticancer agent in several types of cancer cells. Researches of its biological role in negative regulation of cell growth have focused on the extent of induction of apoptosis in a cell-type-specific manner. Objectives: To identify the mechanisms underlying asiatic acid-induced downregulation of BCL2. Methods: MicroRNA microarray analysis was used to investigate the effect of asiatic acid treatment of A549 cells on miRNA expression profiles. We used quantitative RT-PCR to confirm the asiatic acid-induced increase in miR-1290 level with primers specific for the mature miR-1290 sequence. We also assessed the effect of asiatic acid on the levels of BCL2 in A549 cells using western blot analysis. Results: We identified an important regulator of asiatic acid-induced cell death, microRNA (miR)-1290, which sensitizes cells to Asiatic acid-induced cytotoxicity and negatively regulates BCL2 expression. Asiatic acid significantly upregulated miR-1290, and asiatic acid-induced cell death was shown to be dependent on miR-1290 activity. Molecular assays demonstrated that BCL2 mRNA is a direct target of miR-1290-mediated RNA interference. Conclusion: The findings of the present study suggest that miR-1290 suppresses cell viability and cell cycle progression. Our results provide insight into miR-1290-mediated cellular mechanisms in asiatic acid-treated A549 non-small cell lung carcinoma cells.

Background: Previous investigations of differences in the serum levels of cytokines relative to the clinical psoriatic phenotype have yielded conflicting results. Objectives: To investigate the levels of circulating inflammatory cytokines and the LL-37 relative to the morphological phenotype in the patients with psoriasis. In addition, correlations between disease severity and inflammatory cytokines were compared. Methods: Age-matched 74 patients with psoriasis (32 guttate and 42 plaque) and 12 healthy controls were included. A multiplex cytokine assay and ELISA were used to measured Th1- and Th17-derived cytokines and LL-37, respectively. Results: Circulating levels of interferon-γ, interleukin (IL)-1 receptor antagonist (IL-1RA), IL-2, IL-23, and LL-37 were significantly higher in patients with psoriasis than in healthy controls. However, the serum levels of inflammatory cytokines and LL-37 did not differ significantly between patients with the guttate and plaque phenotypes. There were positive correlations between serum inflammatory cytokine levels and the Psoriasis Area and Severity Index score, but no significant correlation between LL-37 level and PASI score. Conclusion: The findings of this study suggest that the serum levels of inflammatory cytokines reflect disease activityrather than determine the morphological phenotype. Further studies are needed to elucidate the relationship between serum LL-37 and cytokines in the pathogenesis of psoriasis, and clinical implications.

Background: Arctiin is an active lignin isolated from Arctium lappa and has anti-inflammation, antimicrobial, and anti-carcinogenic effects. Objectives: To find that arctiin exerts antioxidative effects on human hair dermal papilla cells (HHDPCs). Methods: To better understand the mechanism, we analyzed the level of hydrogen peroxide (H2O2)-induced cytotoxicity, cell death, ROS production and senescence after arctiin pretreatment of HHDPCs. Results: The results showed that arctiin pretreatment significantly inhibited the H2O2-induced reduction in cell viability. Moreover, H2O2-induced sub-G1 phase accumulation and G2 cell cycle arrest were also downregulated by arctiin pretreatment. The increase in intracellular ROS mediated by H2O2 was drastically decreased in HHDPCs cultured in the presence of arctiin. This effect was confirmed by senescence associated-beta galactosidase (SA-β-gal) assay results; we found that arctiin pretreatment impaired H2O2-induced senescence in HHDPCs. Using microRNA (miRNA) microarray and bioinformatic analysis, we showed that this anti-oxidative effect of arctiin in HHDPCs was related with mitogen-activated protein kinase (MAPK) and Wnt signaling pathways. Conclusion: Our data suggest that arctiin has a protective effect on ROS-induced cell dysfunction in HHDPCs and may therefore be useful for alopecia prevention and treatment strategies.

Background: Several studies have been conducted to develop various ingredients, examine the viscosity, and determine the proper density of anti-aging cosmetics; however, additional systematic clinical trials and analysis of the results are necessary in this field. Objectives: To evaluate the effects of moisturizer, ‘TIME CONTROL Day Cream and TIME CONTROL Eye Cream’ on facial wrinkles, elasticity, dermal density, and skin tone. Methods: Twenty healthy volunteers participated in this clinical study and were asked to apply the test materials on face twice per day for 4 weeks. For evaluation, PRIMOS Lite, DermaLab, DUB-Skin Scanner, USB elasticity probe and spectrophotometer was applied. Facial skin condition characteristics, skin improvement before and after using the test materials were surveyed. Results: Crow’s feet wrinkles were decreased 5.97% after two weeks and 14.07% after four weeks in comparison of pre-application. Skin elasticity was increased 6.81% after 2 weeks and 8.79% after 4 weeks. Dermal density was increased 16.74% after 2 weeks and 27.63% after 4 weeks. Skin brightness was increased 1.70% after 2 weeks and 2.14% after 4 weeks, and erythema was decreased 10.45% after 2 weeks and 22.39% after 4 weeks. There were no abnormal skin responses during the trial period. Conclusion: The findings of this study suggest that ‘TIME CONTROL Day Cream and TIME CONTROL Eye Cream’ were found to improve facial wrinkles, elasticity, dermal density and skin tone.

N/A We have reviewed 34 cases of ulcerative colitis, experenced in the Seoul National University Hospital from January 1982 to August 1988. 1) The disease was most prevalent in fourth decades and the sex ratio (Male:Female) was 1:2.4. 2) The duration of symptoms was less than 5 years in 89.4% 3) The most common symptoms were hematochezia, abdominal pain, weight loss and bloody diarrhea in the order of frequency. 4) In anatomical distribution, left sided colitis were seen in 35.3%, pancolits in 26.5%, proctosigmoiditis in 23.5%, back-wash ileitis in 8.8%, and proctitis in 5.9%. 5) Medical treatment with TPN, steroid administration, steroid enema, were effecfive in 37.5%, 48 %, 63.6%, 50% respectively. 6) In all 8 proctocolectomy cases continent ileostomy were done in 7 cases and conventional ileostomy in 1 case. 1 case of continent ileostomy cases was converted to conventional ileostomy due to ileal pouch perforation. 7) Complications were 2 intestinal obstructions, 1 pouch perforation and 1 wound infection without any mortality. If treated early with accurate surgical indication, ulcerative colitis can be treated surgically with less morbidity and mortality

Background: Despite the physical disfigurement, bothersome symptoms and psychological problemsfrom the proliferative scarring process of keloids and hypertrophic scars, there is currently no consensus regarding the best management of these lesions. Objective: The purpose of this study was to determine the therapeutic efficacy and safety of intralesional corticosteroid injection combined with 5-fluorouracil (5-FU) and/or pulsed dye laser (PDL) in Koreans with keloids or hypertrophic scars. Methods: Twenty-five patients with keloids and/or hypertrophic scars over 2 cm in size or who had more than 3 separated lesions were included in this study. Each scar was divided equally into 2 or more segments and these were treated with 1 of the following 3 different modalities: (1) intralesional injection of triamcinolone acetonide (TCA: 20 mg/ml weekly), (2) intralesional injection of TCA 0.1 ml (40 mg/ml) mixed with 5-FU 0.9 ml (50 mg/ml) weekly and (3) treatment #2 combined with PDL (585 nm, 5∼7.5 J/cm2) at baseline and at the 3rd and 7th weeks. We estimated the change of the height, the erythema, the pliability, the pruritus, the pain/tenderness scores, the patient-self assessment and the complications. This study was an 8-week prospective study and the assessment of relapse was done at 4 and 12 weeks after the last treatment. Results: There was a statistically significant improvement of the clinical signs and symptoms in all the treated groups. However, the 5-FU-treated groups had more sustained effects compared to the group that underwent TCA monotherapy with respect to flattening and pliability. Complications such as ulcerations, crusts and hyperpigmentation were more frequent in the 5-FU-treated groups. Conclusion: Although 5-FU may be preferable to TCA for improving the symptoms of keloids and hypertrophic scars, we believe it is prone to induce more complications in Koreans.
