      • cDNA Cloning and Genomic Structure of Glyceraldehyde-3-Phosphate Dehydrogenase from the Entomopathogenic fungus, Paecilomyces tenuipes-Jocheon

        Fungi belonging to the Paecilomyces spp. have recently been used as food and herbal medicines in Korea and are greatly popular as commercially available powdered supplement or dried fruiting body. Despite this acceptance and its use, little is known of the genes related to its reactive agents. Presently, We have constructed an olig-d(T) primed directional cDNA library from the silkworm Dongchunghacho, an entomopathogenic fungus, of which species is belonging to Paecilomyces spp. based on the previous identification of ITS1 and ITS2 at the molecular level and collected from Jocheon Miryang, Korea. To isolate and screen genes in the fungus, 626 expressed sequence tags(ESTs) were generated by a partial sequencing from the cDNA library. cDNA encoding the glyceraldehyde-3-phosphate dehydrogenase(Pt-GAPDH) of Paecilomyces tenuipes- Jocheon was cloned from the above cDNA library. The complete cDNA sequence of Pt-GAPDH is comprised of 1,014bp encoding 338 amino acid residues. The deduced protein sequence of Pt-GAPDH showed higher homology with Beauberia bassiana-GAPDH(93% amino acid identity). Hydropathy analysis revealed that Pt-GAPDH protein is hydrophilic. The major three amino acids in its composition of amino acid residues were alanine(11.54%), valine(9.47%) and glycine(8.88%). The Pt-GAPDH gene of Paecilomyces tenuipes entomopathogenic fungus consisted of three exons and two introns coding for 338 amino acid residues, and the genomic DNA length of the gene spans 1302bp. The accession number of the gene in GenBank are GU997099 for Pt-GAPDH cDNA and GU997102 for Pt-GAPDH genomic DNA. More investigation works including gene expression, immunological analysis etc. will be carried continuously without hesitation after this presentation.

The Pt-GAPDH gene of Paecilomyces tenuipes entomopathogenic fungus consisted of three exons and two introns coding for 338 amino acid residues, and the genomic DNA length of the gene spans 1302bp. The accession number of the gene in GenBank are GU997099 for Pt-GAPDH cDNA and GU997102 for Pt-GAPDH genomic DNA.

      • 인슐린 비의존형 당뇨병환자에 있어서 Multitestⓚ Kit 를 이용한 면역반응에 관한 연구

        인슐린 비의존형 당뇨병환자에서 세포성 면역반응 및 감염과의 관련성 유무를 알아 보고져 1986년 1월부터 1986년 6월까지 원광대학병원 내과에 입원한 당뇨병환자 25명을 대상으로 Multitest CMI 방법을 이용하여 지연성 피부과민 반응을 측정 비교한바 정상 대조군과 환자군에 있어서 면역반응의 평균 득점치는 대조군 16.9 ±8.41 ㎜, 환자군 7.7±4.24 ㎜로 환자군에서 대조군에 비해 현저히 저하된 면역반응 ( P < 0.005) 을 보였으며, 환자군에서 면역반응과 당화 혈색소치와의 관련성은 유의한 음의 상관관계 (r = -0.50, P < 0.05) 를 보였으며 환자군중 감염이 있는 군의 면역반응 평균 득점치는 5.5 ±3.55 ㎜ , 비감염군은 9.4 ±4.02 ㎜로 감염군에서 유의한 감소 ( P<0.01)를 보였다. 결론적으로 Multitest CMI 방법은 편리하게 환자의 세포성 면역반응을 알아볼 수 있었으며, 혈당을 정상으로 유지하므로서 면역능을 강화하고 감염을 방지할 수 있음을 시사하였다. To assess cell mediated immunity in NIDDM, 25 NIDDM patients were evaluated by Multitest device. Delayed cutaneous hypersensitivity (DCH) responsiveness is quantified by a score ( the sum of the diameters of induration)at 48 hours. The results were summarized as follows 1) Compaired to male, significantly lower DCH response were observed in female of the control group ( P < 0.005 ), for the average scores (22.0 ㎜ for male, 10.8 for female). 2) In all ages classes, the patients had an average score significantly lower than that of the control subject(16.9 ㎜ for controls, 7.7 ㎜ for patients. P<0.005). 3) In the oldest group of the control subjects, DCH responses were decreased (8.4 ㎜) 4) There were significant cerrelations between DCH responses and glycosylated hemoglobins in diabetics ( r = -0.50, P < 0.05). 5) Compaired to patients without infection, Patients with infection had significant reductions in response to skin test (9.4 ㎜ for patients without infection, 5.5 ㎜ for patients with infection. P<0.01) 6) As the complications, no adverse reation was observed in both groups. Local reactions such as itching sensation and vesicle formation particularly to the tuberculin antigen were obsered 7 cases for the control groups and 2 cases for the patients. But these reactions disappeared within 8 to days without secondary infection or scar formation. In conclusion, the Multitest system appears to be safe, rapid and reproducible means of evaluating DCH and it suggested that better diabetic control may have a role in bolstering or augmenting cell mediated immunity and protecting against infections.

      • EDI 서비스의 데이터 웨어하우스 구현에 관한 연구

        데이터 웨어하우스의 데이터 통합은 정보가 미리 추출되고 번역되어 저장되며, 질의가 요구될 때 저장소에서 평가된다. EDI 서비스의 의사 결정 지원을 위해서는 데이터 웨어하우스의 데이터 통합이 요구된다. 본 논문에서는 EDI 메시지 중계 시스템과 데이터 웨어하우스의 개념을 살펴보고, 실제로 거래되는 EDI 송수신 데이터를 이용하여 데이터 웨어하우스를 구현하고, 트랙픽 유형 및 주요 특성을 분석한다. 데이터 웨어하우스의 일반적인 모형인 정보의 생성, 가공, 활용 단계에 맞추어 TANDEM에서 EDI 원천 데이터 생성, SUN에서 통계 데이터 가공, P/C에서 활용하는 구성을 갖는다. 트래픽 분석 결과, 배달할 메지기가 있을 때 중계시스템에서 먼저 접속을 개설하는 Store & Forwarding 방식이 필요하고, EDI는 온라인성 작업임으로 접속절차의 간소화가 요구되며, 일괄 처리 시간의 단축이 필수적임을 알았다. For the integration in data warehouse, information is extracted, translated, and stored, and when we request the query, it is estimated through the stored information. To support the decision of EDI service, it is necessary to implement data integration in data warehouse. In this paper, we explain the concept of EDI message transfer system and data warehouse, implement data warehouse using EDI data which is exchanged in real field, and analyze the attribute of EDI traffic. According to the general model of data warehouse - information creation, processing, utilizing - EDI source data is created in TANDEM, in SUN statistics data is made from source data, and P/C display the attribute of EDI traffic graphically. As a result, we get to know the followings. "Store & Forwarding" mechanism in which a center initiate the session as soon as it can deliver some messages is important. Since EDI is a on-line job, it is efficient to simplify the connection procedure. The time to process batch jobs must to be decreased.

        Temporal bone high resolution computed tomogram(TBHRCT)is a essential diagnostic tool in considering for cochlear implantation in patients with profound sensorineural hearing loss. This study was designed for an effort to provide and efficacy of TBHRCT in determination of inner ear malformations for the patients who have profound sensorineural hearing loss and to examine the relationships between TBHRCT findings and other preoperative evalutions for cochlear implantation in that patients. Authors examined 68 deaf patients to provide fundamental data for cochlear implantation. Inner ear abnormalities were founded in 13(19%) patients, among which 10(14.7%) patients had Mondini'dysplasia in which 5 patients were hypoplastic type and 5 patients were hydrops type. Other inner ear abnormalities were as follows : lateral semicircular canal hypoplasia ; internal auditory canal abnormality ; cochlear aqueduct widening ; vestibular aqueduct widening. No relationships were found between TBHRCT findings and other test results. : promontory test, K-WISC cognitive test etc.(Korean J Otolaryngol 39 : 8, 1996)

        Tofisopam is a new type of tranquilizer valuable for the relief of anxiety and tension in a wide range of emotional disorders. Tofisopam has the therapeutic characteristics of a minor tranquilzer and a mild stimulatory effect. The purpose of the present study was to evaluate the bioequivalence of two tofisopam tablets, Grandaxin^TM (Hwan In Pharmaceutical Co., Ltd.) and Tofim^TM (Kyung Dong Pharmaceutical Co., Ltd.), according to the guidelines of Korea Food and Drug Administration (KFDA). Eighteen normal male volunteers, 23.11±2.83 years in age and 65.43±7.64㎏ in body weight, were divided into two groups and a randomized 2 x 2 cross-over study was employed. After one tablet containing 50㎎ of tofisopam was orally administered, blood was taken at predetermined time intervals and the concentrations of tofisopam in serum were determined using HPLC method with UV detector. The pharmacokinetic parameters such as AUC_t, C_max and T_max were calculated and ANOVA test was utilized for the statistical analysis of the parameters. The results showed that the differences in AUC_t, C_max and T_max between two tablets based on the Grandaxin^TM were -5.59%, 2.22% and -13.18%, respectively. Minimum detectable differences (Δ) at α=0.10 and 1-β=0.8 were less than 20% (e.g., 14.95% and 19.34% for AUC_t and C_max, respectively). The powers (1-β) at α=0.10, Δ=0.2 for AUC_t and C_max were 95.21% and 81.93%, respectively. The 90% confidence intervals were within ±20% (e.g., -15.64∼4.45 and -10.77∼-15.21 for AUC_t, and C_max, respectively). Two parameters met the criteria of KFDA for bioequivalence, indicating that Tofim^TM tablet is bioequivalent to Grandaxin^TM tablet.

      • 直立葉型의 一般系와 統一系 水稻品種의 生育 및 收量性 比較

        Light interception efficiency, parameters during ripening period, yield response to fertilization and optimum leaf area index and leaf area duration for maximum grain yield were compared between high yielding variety, Nampung with long culm and erect leaves, and Japonica variety, Seonam with short culm and erect leaves under six fertilization levels and two planting densities. 1. Light extinction coefficient was estimated at 0.56 in Seonam and 0.58 in Nampung, showing no difference between the two varieties. 2. Relative growth rate, net assimilation rate and crop growth rate during 30 day after heading were greater in Nampung than in Seonam.] 3. Sink capacity determined before heading showed no difference between the two varieties, but the yield was higher in Nampung than in Seonam. This suggests that Seonam is inferior to Nampung in photosynthetic and translocating efficiency during ripening period. 4. The optimum fertilization level for maximum grain yield ranged 20 to 24㎏N/10a in Nampung and 16 to 19kgN/10a with planting densities. 5. The optimum LAIs for maximum grain yield showed the range of 7.6 to 8.3 in Seonam and of 8.6 to over 10.2 in Nampung with year. These were greater than those reported previously for Tongil and Japonica Varieties. 6. The optimum LADs for maximum grain yield were estimated to be in the range of 176 to 180 day in Seonam and 245 to over 238 day in Nampung with year.

      • Polysulfone 블럭 공중합체를 이용한 음이온 교환막의 제막 및 전기화학적 특성

        It was prepared an anion exchange membrane using block copolymer of Polysulfone(PSf) and polyphenylene sulfide sulfone(PPSS) by a two step synthesis: the chloromethylation and amination by triethylamine, and studied the effect of the preparation condition on the electrochemical properties such as the electrical area resistance, the ion exchange capacity, and the static transport number. The optimum chloromethylation condition were mole ratio of BPSf to chloromethyl methyl ether 1:20, reaction time 6 hours, and reaction temperature 25℃. The best electrochemical properties were measured, with area resistance 120.9 Ω·㎠, ion exchange capacity 1.324 meq./ g dry membrane, water content 0.204 g H₂O/g dry membrane, and fixed ion concentration 6.49 meq./g H₂O.
