시티딘은 장미조직이 가지고 있는 시티딘 디아미나제에 의해 우리딘과 암모니아 기체를 생성한다. 이 장미조직 50mg을 암모니아 기체감응센서에 고정시켜 장미조직센서를 제조하고, 수용액 중의 시티딘을 정량하는데 최적 실험조건을 조사하였다. pH8.4, 온도 37$^{\circ}C$, 0.2M 인산 완충용액을 사용하였을 때 시티딘의 농도가 $7.0 {\times}10^{-4}$에서 $1.0{\times}10^{-2}$M까지 직선이었고, 53mV/decade의 감응기울기를 나타내었다. 검출한계는 $3.0 {\times} 10^{-4}$ M 상대표준편차는 3.4%였다. 또한 이 조직센서는 선택성이 탁월하였다. Rose tissue containing cytidine deaminase converts cytidine to uridine and ammonia gas. Rose tissue sensor was constructed by immobilizing 50mg of a rose petal tissue on an NH3 gas sensor and the optimum condition of the sensor for the determination of cytidine was investigated. The tissue sensor showed a linear range of$7.0 {\times} 10^{-4}$∼$1.0{\times} 10^{-2}$M cytidine with a slope of 53 mV/decade in 0.2 M phosphate buffer, pH 8.4 at 37$^{\circ}C$. The detection limits were $3.0{\times}10^{-4}$ M and relative standard deviation was 3.4%. This sensor showed an excellent selectivity among various nucleosides and amino acids.
새로운 백금착물 항암제 SKI 2053R 의 토끼 최기형성시험

SKI 2053R, cis-Malonato[(4R,5R)-4,5-bis(aminomethyl)-2-isopropyl-1,3-dioxolane]platinum(II), is a newly developed antitumor platinum complex derived from cisplatin. Preclinical studies suggest that it may have greater antitumor activity and lower toxicity than cisplatin. Effects of test agent on general toxicity of does and embryonic development of F1 fetuses were investigated in rabbits. Sixty eight New Zealand white rabbits were distributed among three treated groups and a control group. SKI 2053R was administered intravenously to pregnant rabbits from days 6 to 18 of gestation at dose levels of 0, 0.67, 2.0, or 6.0 ㎎/㎏/day. The pregnant does were subjected to the caesarean section on day 28 of gestation. No treatment-related changes in clinical signs, body weight, food consumption, and necropsy findings were observed in all groups. F1 fetuses showed no changes related to the treatment of SKI 2053R, except that an increase in the incidence of skeletal variations were observed at 6.0 ㎎/㎏. There were no signs of maternal toxicity or embryotoxicity at 0.67 and 2.0 ㎎/㎏. The results show that the administration of 6.0 ㎎/㎏ SKI 2053R induces skeletal variations in fetuses and that the no observed adverse effect levels(NOAELs) of SKI 2053R are considered to be over 6.0 ㎎/㎏ for does and 2.0 ㎎/㎏ for F1 fetuses in rabbits.

QCM Study of β-Casein Adsorption on the Hydrophobic Surface: Effect of Ionic Strength and Cations

The adsorption kinetics of ${\beta}$-casein on a hydrophobic surface has been studied by means of the quartz crystal microbalance (QCM). The self assembled monolayer of 1-octadecanethiol on a gold coated quartz crystal was used as a hydrophobic surface for adsorption. The adsorption kinetics was monitored in different solution conditions. Formation of monolayer is observed in most cases. At high concentration of protein, micelle formation which is interrupted by high ionic strength of solution is observed. Casein binding cations such as $Ca^{2+},\;Ba^{2+}\;and\;Al^{3+}$ increase the hydrophobicity of the protein and the multiple layer adsorption occurs. The strong and weak points of the QCM method in the study of protein adsorption are discussed.
On-line Monitoring of Cellulase Treatment by Differential Refractometer

Cellulase treatment of cellulose fibers needs to be monitored to give proper weight loss without significant strength loss. On-line monitoring of cellulase treatment is presented which can monitor the weight loss of cotton fabrics by measuring differential refractive index in real time. On-line monitoring was tried under the condition where the cellulase treatment of scoured 100% cotton knits would give weight loss of about 6.6% during one hour cellulase treatment. For comparison, processing solutions with treatment time of 5, 10, 20, 30, 45 and 60 minutes were taken out, and subjected to sugar analyses using total organic carbon (TOC), and dinitrosalicylic acid (DNS) method as well as HPLC analyses. HPLC analyses showed that cellobiose and glucose were the major components resulting from cellulase treatment of cotton fabric. TOC and differential refractive index measurement proved to be linear to total sugar concentration while DNS method result was dependent on the type of sugar. Various sugar concentrations of the processing solutions measured by these methods were compared with HPLC measurements and correlated with the actual weight loss of the cotton fabric processed. The on-line monitoring device using differential refractometer gave a real time signal which was independent of the ratio of cellobiose and glucose but dependent on the total sugar concentration. The device was also non-destructive.
