<P><B>Abstract</B></P> <P>Climate change gives rise to rapid degradation of rural soils in sloping subtropical and tropical areas and might further threaten environmental sustainability. In this study, we conducted an integrated evaluation of the effects of wood biochar (WB) application mixed with a green waste dreg compost (GWC) on runoff quality, soil losses, and agricultural productivity for a highly weathered tropical soil. A conventional agriculture method, in which soils are treated with anionic polyacrylamide (PAM), was also conducted for comparison. The amounts of runoff and soil loss, and nutrient retention were evaluated a year after WB application. Soil fertility was also investigated through a year pot experiment with rape (<I>Brassica campestris</I> L.) cultivation. Our results showed that the WB application not only effectively increased soil pH, soil organic carbon (SOC) and exchangeable K<SUP>+</SUP> but also increased the production of rape plants. Significant reduction of runoff and the increases of inorganic nitrogen (IN) and total phosphorus (TP) were found in the WB-treated soil. Compared to the control, the co-application of WB and GWC, particularly for the WB at 4%, decreased runoff by 16.8%, soil loss by 25%, and IN loss (via runoff) by 41.8%. Meanwhile, compared to the control and PAM treatments, the co-application of WB and GWC improved soil acidity and the contents of SOC, IN, TP, and exchangeable K<SUP>+</SUP>. The co-application of WB and GWC could be an alternative agricultural strategy to obtain benefits to agricultural productivity and environmental sustainability.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Co-applying biochar and compost notably reduced runoff and soil loss by 16%–25% in a tropical soil. </LI> <LI> In-situ biochar applying increased inorganic N, available P and K by 1.5–2.5 times in slopelands. </LI> <LI> Crop productivity was enhanced by biochar applying 1.5–2 times at least in a tropical slopeland. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

Background/Aims Both sexual and physical abuse history have been reported to be associated with irritable bowel syndrome (IBS) in Western countries. The impact of abuse history in IBS patients in Asia remains unclear. We aim to determine the prevalence of abuse history, its associated psychological profiles, and sleep problems among IBS patients in Taiwan. Methods In total, 194 Rome III-defined IBS patients were invited to participate. Age- and sex- matched healthy carriers of chronic hepatitis B or hepatitis C without chronic abdominal symptoms were identified as disease-controls. We administered a validated questionnaire to evaluate bowel symptoms, physical/sexual abuse history, anxiety/depression (Hospital Anxiety and Depression Scale [HADS]), and sleep quality. Results IBS patients had a significantly higher prevalence of sexual abuse history than the disease-control group both before (16.5% vs 6.7%, P < 0.05) and after (16.0% vs 6.6%, P < 0.05) adolescence. These significant differences were mainly observed in women (13.4% vs 3.4%, P < 0.05). No difference was noted in history of physical abuse between the 2 groups. IBS patients with a history of sexual abuse had significantly higher HADS scores and higher frequencies of sleep difficulty than those without. Conclusions In Taiwan, sexual abuse history was more prevalent in female IBS patients than controls. Sexual abuse history may contribute to higher anxiety/depression levels and sleep difficulties, which are commonly experienced in IBS patients. In Asia, abuse history should be obtained when approaching IBS patients to facilitate better management. (J Neurogastroenterol Motil 2018;24:79-86)
IgE-Binding Epitope Mapping and Tissue Localization of the Major American Cockroach Allergen Per a 2

Purpose: Cockroaches are the second leading allergen in Taiwan. Sensitization to Per a 2, the major American cockroach allergen, correlates with clinical severity among patients with airway allergy, but there is limited information on IgE epitopes and tissue localization of Per a 2. This study aimed to identify Per a 2 linear IgE-binding epitopes and its distribution in the body of a cockroach. Methods: The cDNA of Per a 2 was used as a template and combined with oligonucleotide primers specific to the target areas with appropriate restriction enzyme sites. Eleven overlapping fragments of Per a 2 covering the whole allergen molecule, except 20 residues of signal peptide, were generated by PCR. Mature Per a 2 and overlapping deletion mutants were affinity-purified and assayed for IgE reactivity by immunoblotting. Three synthetic peptides comprising the B cell epitopes were evaluated by direct binding ELISA. Rabbit anti-Per a 2 antibody was used for immunohistochemistry. Results: Human linear IgE-binding epitopes of Per a 2 were located at the amino acid sequences 57-86, 200-211, and 299-309. There was positive IgE binding to 10 tested Per a 2-allergic sera in 3 synthetic peptides, but none in the controls. Immunostaining revealed that Per a 2 was localized partly in the mouth and midgut of the cockroach, with the most intense staining observed in the hindgut, suggesting that the Per a 2 allergen might be excreted through the feces. Conclusions: Information on the IgE-binding epitope of Per a 2 may be used for designing more specific diagnostic and therapeutic approaches to cockroach allergy.
