      • Therapeutic Profile of T11TS vs. T11TS+MiADMSA: A Hunt for a More Effective Therapeutic Regimen for Arsenic Exposure

        Arsenic exposure is a serious health hazard worldwide. We have previously established that it may result in immune suppression by upregulating Th2 cytokines while downregulating Th1 cytokines and causing lymphocytic death. Treatment modalities for arsenic poisoning have mainly been restricted to the use of chelating agents in the past. Only recently have combination therapies using a chelating agent in conjunction with other compounds such as anti-oxidants, micronutrients and various plant products, been introduced. In the present study, we used T11TS, a novel immune potentiating glycopeptide alone and in combination with the sulfhydryl-containing chelator, mono-iso-amyl-dimarcaptosuccinic acid (MiADMSA) as a therapeutic regimen to combat arsenic toxicity in a mouse model. Results indicated that Th1 cytokines such as TNF-${\alpha}$, $IFN{\gamma}$, IL12 and the Th2 cytokines such as IL4, IL6, IL10 which were respectively downregulated and upregulated following arsenic induction were more efficiently restored to their near normal levels by T11TS alone in comparison with the combined regimen. Similar results were obtained with the apoptotic proteins studied, FasL, BAX, BCL2 and the caspases 3, 8 and 9, where again T11TS proved more potent than in combination with MiADMSA in preventing lymphocyte death. The results thus indicate that T11TS alone is more efficient in immune re-establishment after arsenic exposureas compared to combination therapy with T11TS+MiADMSA.

        Evaluating the contribution of urban ecosystem services in regulating thermal comfort

        The study focuses on spatio-temporal dynamics of urban ecosystem services (UES) and their contribution in maintaining thermal comfort in Bhubaneswar city, India. An extensive increase in UES demand (grey) patches (187.95%) was observed during 1992–2016 in contrast to significant decline (47.94%) in UES supply (blue–green) patches, primarily in the northern and south–western directions. Also, a drastic rise in area under thermally highly uncomfortable zone (35–40 C) from 0.005 to 56.68 km2 and a decrease in area of thermally comfortable zone (B 26 C) from 0.46 km2 to zero during 1992–2016 exhibiting deteriorating natural urban living condition. Although, the land surface temperature (LST) was remained higher in urban areas, the peri-urban and neighbouring rural areas (27.31–33.98 C) of Bhubaneswar city recorded a high increase in mean LST as compared to the urban areas (31.19–34.69 C). In both the cases, UHI intensities were less as compared to other growing cities of India. The MODIS based time series analysis depicted similar trends with minor increase in LST (30.55–30.76 C) during 2000–16. The study proves the intrinsic linkages of UES with thermal comfort and necessitates to adopt sustainable measures to make the city green and habitable.

        Starvation Induced Changes of Some Biomolecules in Eggs and Hatched Larvae of Indigenous Strain of Bombyx mori (Lepidoptera : Bombycidae)

        Variations in protein and nucleic acid concentrations were observed in 24 hrs old eggs and hatched larvae of Nistari strain, Bombyx mori, exposed to starvation. Three starvation treatments of 24,48 and 60 hrs were given separately from 0 hr old fifth instar larvae. Biochemical variations were studied in the resultant hatched larvae of one time starved parent, while the eggs obtained from parents receiving starvation in two successive generations were considered for the study. In hatched larvae, protein levers in 24 hrs starvation groups remained significantly higher over control (never starved) while the same was found to be lower in 48 and 60 hrs starvation individuals. The RNA concentration remained significantly higher in all the treated lots. However, DNA content was not found to be significantly altered in hatched larvae after exposure to feeding stress. Protein, RNA and DNA concentration of 24 hrs old eggs produced by all the starved groups of Nistari, which had deceived two consecutive starvation during parental generations, showed higher concentrations of these biomolecules over control. Hence, starvation induced alterations in protein and nucleic acids in eggs and hatched Iarvae are indicative of a preparatory phase adopted by the insect to acclimatise itself and its progeny to stress situations.

        Biomolecular Variations in Poly and Bivoltine Strains and Their Hybrids of Bombyx mori during Embryonic Development

        Biometabolic assessment was made in early and late embryonic stage (just before hatching) of one poly, one bi and their hybrids (DP, YPe, DP ${\times} Ype, and YPe ${\times} DP respectively ) of Bombyx mori to observe the racial differences. Protein and nucleic acid (RNA and DNA) concentrations were recorded to be significantly higher in bivoltine breed (YPe) and also in the hybrid than the polyvoltine (DP) strain in both the stages of embryonic development. The single egg weight of polyvoltine race was lower as compared to that of bivoltine and the hybrid studied. Age specific changes in all the biomolecules were evident where protein and RNA concentrations were elevated sharply in prehatched larvae while in case of DNA it was observed to be just reverse. The differences in protein, RNA and DNA composition between breeds and hybrids reflect the racial variations in biometabolic demands responsible for differential growth and development of the breeds and hybrids.

        Dynamics of a robust photo-induced insulator–metal transition driven by coherent and broad-band light in epitaxial films of La<sub>0.625 − <i>y</i></sub>Pr<sub><i>y</i></sub>Ca<sub>0.375</sub>MnO<sub>3</sub>

        <P>A dramatic drop of ≈5 orders of magnitude in the resistance (<I>R</I>) of La<SUB>0.175</SUB>Pr<SUB>0.45</SUB>Ca<SUB>0.375</SUB>MnO<SUB>3</SUB> epitaxial films upon exposure to optical photons derived from both continuous and pulsed lasers, as well as broad-band sources at temperatures (<I>T</I>) < 30 K is reported. The strength of change is a sensitive function of both the incident photon flux and temperature. Under isothermal conditions the photo-generated low resistance state persists eternally after removal of light. This non-equilibrium state is metallic, as revealed by the positive d<I>R</I>/d<I>T</I> for <I>T</I> ≤ <I>T</I><SUB><I>p</I></SUB> (≈120 K). This electrically conducting state is presumably ferromagnetic as <I>T</I><SUB><I>p</I></SUB> coincides with the temperature where a weak ferromagnetism sets in on cooling the insulating film from room temperature. To rule out the possibility of photon-induced local heating of the sample as a mechanism of the observed effects, photo-illumination experiments were performed under identical conditions on thin films of two non-charge-ordered manganites deposited on substrates of similar thermal conductivity. Our model for the observed transition encompasses a global charge-ordered state in which ferromagnetic metallic clusters of fraction <I>p</I> much less than the critical fraction <I>p</I><SUB>c</SUB> for percolation exists at low temperatures. Photo-induced melting of the charge-ordered state increases this fraction beyond <I>p</I><SUB>c</SUB> in a cumulative manner as successive pulses of light fall on the sample. </P>

        Free Education Policy and Trade Liberalizaiton : Consequences on Child and Adult Labour Markets in a Small Open Economy

        The paper purports ot examind the implocations of a free education policy and trade liberalizaion on the child and adult labout markets in the set-up of a Harris-Todaro type general equilibriym model. It has been found that a hike in the education subsidy or an infliw of foreign calital may produce counterproductive results ont he supply of child labour in the urban area. Moreover, these policies may raise the level of urban unemployment of adult labour even when two types of labour are not substitutes to each other. The average income of the urban poor families may also decrease as a consequence.

        Foreign Direct Investment in a Small Open Economy and Global Trade Liberalization in Agriculture : A Note

        In ap production structure reasonsble for a developing economy this note shows that there may arise a conglict between the worldwide liberalized trade policies in agriculture, which raise the price of the economy's primary qxportable commodity, adn the inflow of foreign capital into the economy. However, if the evonomy strictly, e.g. the removal of the indirect farm subsidies, the paper argues that the pissible conflict may be avoided, The paper provides a theoretical basis for the removal of the farm suvsidies if the economy wants to develop its technologically more advanced sectors with an adequate supply of foreign capital.

        Induction of Shikonin Production in Hairy Root Cultures Of Arnebia hispidissima via Agrobacterium rhizogenes-mediated Genetic Transformation

        Data presented herein provides a rapid and efficient method for Agrobacterium rhizogenes-mediated genetic transformation of Arnebia hispidissima for hairy root cultures as well as for enhancing Shikonin production. Etiolated explants viz. shoot tip, nodal, leaf and internodal segments were co-cultivated with Agrobacterium rhizogenes for induction of hairy root. Among the various explants employed, leaf explant showed maximum 70.7% response followed by shoot tip 48.3%, nodal segment 38.7% and internodal segment 9.3%. Integration of Ri plasmid rolB gene in the transformed hairy root cultures was confirmed by PCR analysis using forward (FrolB) and reverse (RrolB) primers of rolB gene resulting in the amplification of 0 ~ 0.8 kb fragments. Medium compositions have been optimized for in vitro induction of Shikonin in hairy root cultures of Arnebia hispidissima. Hairy roots on hormone-free MS medium showed red spots in the older part of the tissues which turned white after a second subculture. Whereas hairy roots cultured on RC medium showed faster growth and produced large amount of Shikonin. The Shikonin content in transformed hairy root culture was estimated by recording absorbance at 620 nm and quantified against authentic sample of Shikonin. Shikonin content was estimated to be $0.85\;mg\;g^{-1}$ fresh weight of tissue at the end of the 50 days of culture. The results presented herein will help to design strategies for bridging the gap between ever increasing demand and supply of raw products necessary for obtaining Shikonin for cosmetic, dyeing, food, medicinal, and pharmaceutical industries.

        High Frequency Direct Plant Regeneration from Leaf and Petals of Cape Primrose (Streptocarpus)

        High frequency direct plant regeneration from leaf and petal explants was accomplished for the first time in Streptocarpus varieties. The shoot induction frequency varied with respect to the benzylaminopurine (BAP) concentration added to the Murashige and Skoog (MS) medium. MS medium with $0.5\;mg\;l^{-1}$ BAP exhibited the highest (69.9%) plant regeneration frequency with an average of 186 shoots per explant. A higher concentration of BAP inhibited shoot bud induction and plant regeneration along with necrosis of explants. Petal explants derived from the varieties 'Branwen' (pink and white) and 'Chorus Line' (violet and white) displayed plant regeneration frequency of 22.2-47.4% (within a total of 12 weeks) on MS medium containing $2.0\;mg\;l^{-1}$ $\alpha$-naphthaleneacetic acid and $0.5\;mg\;l^{-1}$ BAP for 8 weeks followed by 4 weeks on MS medium with $1.0\;mg\;l^{-1}$ BAP. Scanning electron microscopy confirmed direct plant regeneration without callus. Regenerated plants from leaf explants with well-developed leaves and roots were hardened and successfully transferred to pots in glasshouse exhibiting 86% survival at the end of 4-6 weeks. Whereas, regenerated plants from flower petal explants upon transfer to pots in glasshouse exhibited 75-82% survival at the end of 4-6 weeks.
