        Histopathological changes and cadmium accumulations in the gills were investigated in crucian carp (Carassius auratus) exposed to 0, 0.01, 0.1, 0.5 mg/l cadmium (Cd)concentrations for 25 days. After 25 days of exposure, cadmium accumulations increased in each of the different exposure concentrations. Histopathological changes in the gills of curcian carp exposed to cadmium included the acidification of mucous cell, the terminal clubbing of lamellae, the hyperplasia of epitherial cell and the curved of secondary lamellae. The similar histopathological changes were observed once the accumulations of cadmium reached the similar levels found in the present study with the different exposure schemes. Also the acidification of mucous cell in gills appeared when cadmium accumulations reached 5 ㎍/g and more. These results suggested that histopathological changes in the gills, an appearance of mucous cell acidification, be used to estimate the relation between toxic effect and cadmium accumulations in the gills.

        In present study, we fractionated polysaccharides from Croton tiglium and investigated their hepatoprotective effects on CCI₄ intoxication. Polysaccharide fraction of which molecular weight is over 300,000(HP) showed the most potent hepatoprotective effects on CC1₄ intoxication. Lipid peroxidation, BAST and sALT were used as parameters to evaluate the liver damage. Glucose, xylose and arabinose were found to be monosaccharides composing sugar moiety of HP.

      • Effects of DK-002, a synthesized (6aS,cis)-9,10-Dimethoxy-7,11b-dihydro-indeno[2,1-c]chromene-3,6a-diol, on platelet activity

        In the present study, the mechanism of antiplatelet activity of DK.-002, a synthesized (6aS,cis)-9.10-Dimethoxy-7,11b-dihydro-indeno|2,1-c ]chroniene-3.6a-diol, was investigated. DK-002 inhibited the thrombin, collagen, and ADP-induced rat platelet aggregation in a concentration- dependent manner, with 1C_(50) values of 120. 27, and 47 μM, respectively. DK-002 also inhibited thrombin-induced dense granule secretion, thromboxane A_(2) synthesis, and [Ca^(2+)], elevation in platelets. DK-002 did not show any significant effect on ADP-induced inhibition of cyclic AMP elevation by prostaglandin E_(1). but DK-002 was confirmed to inhibit ADP-induced [Ca^(2+)] elevation and shape change. DK-002 inhibited 4- bronw-A23187-induced [Ca^(2+)], elevation in the presence of creatine phosphate/creatine phosphokinase (CP/CPK, a ADP scavenging system) and indomethacin (a specific inhibitor of cyclooxygenase). DK-002 also inhibited Ca^(2+) mobilization in thrombin- or 4-bromo-A23187-stimulated platelets through its inhibitory effects on both Ca^(2+) release from intracellular stores and Ca^(2+) influx, in the presence of CP/CPK and indomethacin. Taken together, the present study shows that DK-002 has inhibitory effects on stimulation of platelets, and suggests that its antiplatelet activity might be related to the inhibitory mechanism on Ca^(2+) mobilization in stimulated platelets. ⓒ 2005 Elsevier Inc.         다양한 골 이식재의 임상 효과에 대한 고찰

        Purpose: Various bone graft materials are being used for periodontal tissue regeneration. Th materials are being developed continuously for ideal clinical effects. Therefore, it is necessary to identify the clinical characteristics of each bone graft material through comparing the various bone graft materials statistically and in doing so, proposing a more efficient bone graft material. In this study, the following results were attained through comparing the clinical effects among the bone graft materials, using the statistical method based on the clinical studies published at the department of periodontology of Yonsei hospital. Materials and Method: 6 selected studies of department of Periodontology at Yonsei University Hospital were based on clinical study of bone grafting in intrabony defects. It was compared the clinical parameters among the 6 clinical studies, using the statistical META analysis. Result: When comparing the probing depth reduction, there was a relatively great amount of decease when using the xenograft, Anorganic Bovine Derived Hydroxapatite Bone Matrix/Cell Binding Peptide(ABM/P-15: PepGen $P-15^{(R)}$) and the autogenous bone and absorbable membrane, d, 1-alctide/glycolide copolymer(GC: $Biomesh^{(R)}$). The allogfrafts showed a relatively low decrease in the probing depth and clinical attachment change. It also showed a slight decrease in the bone probing depth. The allografts showed various results according to different bone graft materials. When comparing the ABM/P-15 and bovine bone $powder(BBP^{(R)})$, ABM/P-15 showed a relatively high clinical attachment level and the bovine bone powder showed a relatively high clinical attachment level. The probing depth change and gingival recession change showed a lower value than the mean value between the two bone graft materials. The synthetic bone showed a relatively high decrease in clinical attachment level and periodontal probing depth change. There was a relatively larger amount of gingival recession when using Bioactive Glass(BG) but a relatively low bone regeneration effect was seen. Conclusion: Good restorative results of the periodontal tissue can be attained by applying the various bone graft materials being used today after identifying the accurate clinical effects.

      • 디젤분진의 수용성 추출물에 의한 메탈로치오닌 유전자 발현

        Metallothionein gene expression of diesel exhaust particles(DEP) was investigated in human lung epithelial cell line. DEP was collected from diesel motor bus and soluble fraction in water was obtained. Cells, grown to near confluence, were exposed to 5~50 ppm DEP for 6 hours. Regarding the metallothionein gene expressions., M1 and MT-2 were induced in the DEP-treated cell by using RT-PCR and real-time PCR. However, MT-3 which is known to be brain specific, and another isoform MT-4 were not expressed in cadmium-treated groups as well as control group. Heavy metal of DEP was also analyzed and Zn was found as the major component of heavy metals in DEP used in this study.

      • Alloxan 유도 당뇨병상태에서 과산화지질생성에 미치는 Brazilin의 효과

        Brazilin was tested for its ability to inhibit alloxan induced lipidperoxidation. Lipid peroxide contents in liver, kidney and serum were measured by the TBA method. ICR mice receiving alloxan at a dose of 43mg/kg via the tail vein after a 24hrs starvation showed significantly increased lipid peroxide contents as compared to untreated control. Lipid peroxide contents in liver, kidney and serum of alloxan-induced diabetic mice were dose-dependently decreased by the treatment of brazilin at a dose of 10mg/kg, 50mg/kg, 100 mg/kg for 5 days.

      • 디젤분진이 사람 동맥 평활근 세포(VSMC)에 미치는 영향

        The purpose of the present study was to examine the effect of diesel exhaust particles on human aortic vascular smooth muscle cells(VSMCs). DNA synthesis, cell viability and morphology of VSMCs after treatment of diesel exhaust particles(DEP) and fine particulate matter(PM_(2.5)) were assayed. PM_(2.5) inhibited the DNA synthesis of VSMCs in a concentration-dependent manner, whereas DEP did not affect VSMCs up to 50㎍/mL . These results were confirmed by morphological examination of VSMCs. PM_(2.5) showed a dose-dependent cytotoxicity of VSMCs by MTT assay. Fraction 4(organic acids) and fraction8(moderately polar compounds) showed the most potent inhibition of DNA synthesis of VSMCs, and fraction7(slightly polar compounds), fraction9(higher polar compounds), and fraction6(aromatic compounds) were next order. These results were confirmed by morphological examination of VSMCs. These results suggest that PM_(2.5) inhibits the DAN synthesis of VSMCs through the cytotoxicity.

        Diesel exhaust panicle (<25μm, DEP_(2.5)) is known to be probarbly carcinogenie (IARC group 2A) DEP_(2.5) contains organic compounds such as polycyclicatomatic hydrocarbon (PAH), heterocyclic compounds, phenols, and nitroarenes Reactive oxygen species (ROS) are generated by DEP_(2.5) without any biological activation system Therefore, an alternative mechanism by which DEP_(2.5) could be carcinogenic is known by the generation of oxidative DNA damage The aim of this study was to investigate genotoxic effects of DEP_(2.5) using single cell gel electrophoresis In order to evaluate the mechanisms of DEP_(2.5) genotoxicity, the rat microsome mediated and DNA repan enzyme treated comet assays together with routine comet assay were performed DEP_(2.5) was collected from diesel engine bus and dichloiomethane extract was obtained The organic extract of DEP_(2.5) revealed DNA damage itself in NIH/3T3 cells And it showed both oxidative and microsome mediated DNA damages Vitamin C as an model antioxidant reduced DNA damage in endonuclase Ⅲ treated comet assay One of flavonoid, galangin as a CYP[A] inhibitor reduced DNA damage in the presence of S-9 mixture Our results show that DEP_(2.5) ate genotoxic and a great source of oxidative stress, but antioxidants can significantly reduce oxidative DNA damages And DEP_(2.5) may contain indirect mutagens which can be inhibited by CYP inhibitors.

        2-[(4-Cyanophenyl)amino]-3-chloro-1, 4-naphthalenedione (NQ-Y15) was asssayed for its genotoxic potential by using Salmonella typhimurium reversion assay and in vitro chromosome aberration test on Chinese hamster lung cells. In the Ames test, NQ-Y15 induced his+revertants of Salmonella typhimurium TA 98 and TA 1537, reaching levels twice the negative control values. But, NQ-Y15 induced only his+revertants of Salmonella typhimurium TA 1537 more than twice the control values under the condition with metabolic activation system. In the cytogenetic test on chinese hamster lung cells. NQ-Y15 showed significant chromosomal aberrations, but the incidence was significantly reduced in the presence of metabolic activation.
