        소 c-KIT Receptor 유전자의 다형성에 관한 연구

        소의 흰 반점 관련 후보유전자로 c-KIT receptor 유전자를 선정하여, c-KIT receptor 유전자내의 변이를 탐색하고 변이가 흰반점 표현형과 연관성이 있는지를 분석하였다. 한우, Angus, Brown Swiss, Charolais, Hereford, Holstein, Limousin 및 Simmental 등 8개 품종의 DNA 시료를 사용하여 c-KIT receptor 유전자의 intron 6번 영역에서 다형성을 조사하고 분석하였다. c-KIT receptor 유전자의 intron 6번 영역에서는 4개의 염기치환이 발견되어, MspⅠ, BsrBⅠ 및 NdeⅠ 제한효소를 이용하여 PCR-RFLP 분석을 실시하였다. Intron 6번을 포함하는 영역의 PCR 산물 크기는 2,440 bp 이었다. MspⅠ다형성은 PCR-RFLP 분석 결과 3개의 대립유전자가 존재하였으며, 한우품종에서는 3개의 대립유전자 모두가 발견되었고, CC 형태이 유전자형을 제외한 5개의 유전자형 (AA, AB, AC, BC 및 BB)을 확인하였다. Angus, Brown Swiss, Hereford, Holstein 및 Simmental 품종에서는 A 대립유전자만을 갖는 것으로 조사되었고, 한우는 44%만 AA 유전자형을 나타내었다. BsrBⅠ 다형성은 2개의 대립유전자로서 3개의 유전자형이 나타나는 것을 확인하였으며, Charolais 및 Hereford 품종이 다른 소 품종에 비하여 A 대립유전자의 빈도가 높게 나타났다. NdeⅠ 다형성을 분석한 결과 Brown Swiss 품종에서는 NdeⅠ에 의해 절단되는 형태인 A 대립유전자만 관찰되었으며, Holstein 품종은 92%, Simmental 품종은 72%가 절단되는 형태를 나타내어, 모색이 흰색을 띠는 소 품종에서 절단되는 형태가 많았다. 소 c-KIT receptor 유전자의 intron 6번 영역에서 확인된 4개의 염기치환은 품종에 따라 다른 빈도를 보였으나, 이들 염기치환과 흰 반점과의 연관성에 대한 증거는 발견하지 못하였다. 그러므로 소의 흰 반점과 c-KIT receptor 유전자 내의 변이와의 관련성은 다른 영역에 대한 추가적인 분석과, 이미 보고된 다른 모색관련 유전자의 다형성과의 연관성 분석 등과 같은 연구가 필요한 것으로 판단된다. We considered KIT gene as a candidate gene for the white-spotting pattern in cattle. This study was carried out to detect genetic variation of c-KIT receptor gene and to investigate association between the mutation and the white-spotting pattern in cattle. PCR-RFLP analysis within intron 6 of c-KIT receptor gene were performed with 8 cattle breeds including Hanwoo, Angus, Brown Swiss, Charolais, Hereford, Holstein, Limousin and Simmental. When PCR product of approximately 2,440 bp including intron 6 of c-KIT receptor gene was sequenced, four nucleotide substitutions were found within intron 6 of the bovine c-KIT receptor gene. In PCR-RFLP analysis, three alleles (A, B and C), two alleles (A and B) and two alleles (A and B) at each locus were identified by Msp Ⅰ, BsrBⅠ and NdeⅠ, respectively. Although frequencies of allele at each locus were different among cattle breeds, we could not get any evidence related with white or white spotting phenotypes in these mutations on intron 6 of c-KIT receptor gene. However, we can not entirely exclude the possibility that c-KIT receptor gene is responsible for white spotting phenotype in cattle. Thus, further studies need to detect other mutations in c-KIT receptor gene and to test association of those mutations and coat color phenotypes in cattle.

        Carbon-coated boron using low-cost naphthalene for substantial enhancement of J<sub>c</sub> in MgB<sub>2</sub> superconductor

        Carbon coating approach is used to prepare carbon-doped $MgB_2$ bulk samples using low-cost naphthalene ($C_{10}H_8$) as a carbon source. The coating of carbon (C) on boron (B) powders was achieved by direct pyrolysis of naphthalene at $120^{\circ}C$ and then the C-coated B powders were mixed well with appropriate amount of Mg by solid state reaction method. X-ray diffraction analysis revealed that there is a noticeable shift in (100) and (110) Bragg reflections towards higher angles, while no shift was observed in (002) reflections for $MgB_2$ doped with carbon. As compared to un-doped $MgB_2$, a systematic enhancement in $J_c(H)$ properties with increasing carbon doping level was observed for naphthalene-derived C-doped $MgB_2$ samples. The substantial enhancement in $J_c$ is most likely due to the incorporation of C into $MgB_2$ lattice and the reduction in crystallite size, as evidenced by the increase in the FWHM values for doped samples.

        Morphological engineering via processing additive in thin film bulk-heterojunction photovoltaic cells: A systematic understanding of crystal size and charge transport

        <P>In this research, organic photovoltaic devices based on bulk-heterojunction (BHJ) from Poly(3-hexylthiophene-2,5-diyl) (P3HT)/[6,6]-phenyl C-71 butyric acid methyl ester (PC71BM) were successfully fabricated to determine the optimization of solvent additive of 1,8-diiodooctane (DIO). The BHJ devices with DIO show improved short circuit current (J(SC)) correlated to the improved device performance, since the DIO in P3HT: PC71BM BHJ affects the morphology in terms of the surface roughness and crystal size. Furthermore, the DIO contributes to the enhanced charge carrier transport of the BHJ device, because the decreased fluorescence and increased hole mobility as found through Raman spectra analysis and SCLC measurement, respectively. The device with optimized concentration (1 vol %) of DIO accomplishes 5% improved photovoltaic performance due to the favorable structural property and enhanced carrier mobility.       • CPC (Collaborative Product Commerce) 개념의 설계 정보시스템 개발

        research shows that a new category of software, Collaborative Product Commerce (CPC), is now emerging, allowing discrete manufacturers to once again distinguish themselves on their products and innovations. CPC permits discrete manufacturers to significantly improve the core processes around the management functions associated with the complete product life cycle that are the basis of their existence. As a way to develop computing tools of CPC to support a design process of product, a web-based design information system was constructed in the paper. The system consists of C-Product system and Net Meeting Communication system to improve communications between designers and persons for verification of design. The product data files of C-Product system were designed by Pro-Engineer and converted to 3D Viewer format for being used in the web browser. Also, Net Meeting Communication system and Database were developed using ASP and Microsoft SQL 2000 Server to share diverse files that can be utilized to design on the web in real time.

        Versatile functionalization of electron rich-fused heterocyclic arenes via electrophilic aromatic addition reaction and their applications

        <P>Divergent functionalized 8-methoxyquinaldines were synthesized via regioselective debromination, 1,3 bromine shift process, aromatization with treatment of a strong base, nucleophilic substitution reaction at the C7 position using amines and cyanide as a nucleophile in the absence of a metal source and a catalyst from an unusual electrophilic aromatic addition (Ad(E)Ar) reaction products 7 and 8. In addition, quinaldine-7,8-dione was prepared by presence of CAN (ceric ammonium nitrate) in AcOH and H2O for 10 min at room temperature from N-(alkylamino)-8-methoxyquinalidines. During the Ad(E)Ar reaction, new stereoselective dearomatized addition products were generated via discriminative reaction routes depending on the methoxy and bromine occupancy position. The Ad(E)Ar reaction not only allowed for the functionalization of electron rich-fused heterocyclic arenes, but also provided a new synthetic route to an alternative mechanism for electrophilic aromatic substitution reactions.         Antiviral activity of KR-23502 targeting nuclear export of influenza B virus ribonucleoproteins

        <P>The spiro compound 5,6-dimethyl-3H,3'H-spiro(benzofuran-2,1'-isobenzofuran)-3,3'-dione (KR-23502) has antiviral activity against influenza A and more potently B viruses. The aim of this study is to elucidate its mechanism of action. Subcellular localization and time-course expression of influenza B viral proteins, nucleoprotein (NP) and matrix protein 1 (M1), showed that KR-23502 reduced their amounts within 5 h post-infection. Early steps of virus life cycle, including virus entry, nuclear localization of NP and viral RNA-dependent RNA replication, were not affected by KR-23502. Instead it interrupted a later event corresponding to nuclear export of NP and M1 proteins. Delivery of viral ribonucleoprotein (vRNP)-M1 complex has been known to be mediated by the viral nuclear export protein (NEP) through interaction with cellular chromosomal maintenance 1 (CRM1) protein. In this study, we experimentally demonstrated that the compound targets the nuclear export of vRNP. Moreover, a single mutation (aspartate to glycine) at amino acid position 54 in M1 [M1(D54G)] was detected after 18 passages in the presence of KR-23502 with a 2-fold increase in 50% effective concentration indicating that this compound has a relatively high genetic barrier to resistance. Interestingly, it was observed that proteasome-mediated degradation of M1 (D54G) was attenuated by KR-23502. In conclusion, we suggest that KR-23502 shows its anti-influenza activity by downregulating NEP/CRM1-mediated nuclear export of influenza vRNP and M1. KR-23502 provides a core chemical skeleton for further structure-based design of novel antivirals against influenza viruses. (C) 2016 Elsevier B.V. All rights reserved.</P>
