        선모(仙茅) 열수(熱水) 추출물의 Collagen 유발 관절염에 대한 약리 효능 연구

        Objectives:In Korean medicine, Curculiginis Rhizoma was treated for arthritis in remedy. But efficacy of Curculiginis Rhizoma on collagen induced arthritis was not revealed. Methods:Anti inflammatory effect of Curculiginis Rhizoma was researched in vitro with RAW264.7 cell and cell toxicity, levels of proinflammatory cytokines (TNF-α, IL-1β, IL-6 and IL-12) and PGE2 were analyzed by ELISA assay. Inflammatory protein were analyzed by western blotting assay (JNK, ERK, COX-2, TNF-α and IL-1β). In vivo, collagen induced arthritis mice model was used to evaluate anti-inflammation effect through arthritis index, immune cell number and cytokine levels (TNF-α, IL-6 and IL-1β) in serum. Results:ECR(Extract of Curculiginis Rhizoma) has not shown cell toxicity in 200 ㎍/㎖ on RAW264.7 cell. ECR suppressed releases of NO, TNF-α, IL-1β, IL-6, IL-12 and PGE2 on RAW264.7 cell treated with lipopolysacharide (1 ㎍/㎖). And ECR inhibited regulation of TNF-α, IL-1β and IL-6 mRNA, reduced protein release of JNK, ERK, iNOS, COX-2, IL-1β and TNF-α. AI of group treated with ECR 200 ㎎/㎏ and 100 ㎎/㎏ were significantly decreased compared to vihicle arthritis mice, the number of immune cell in foot joint was increased on control mice but those of group treated with ECR 200 ㎎/㎏ and 100 ㎎/㎏ were significantly reduced. This results correspond with contens of cytokines (TNF-α, IL-1β and IL-6) in serum. Conclusions:Curculiginis Rhizoma has anti-inflammation effect on RAW264.7 cell in vitro and collagen induced arthritis in vivo . So it is necessary to research more mechanism for cascade imfact.

      • Monosodium Iodoacetate로 유발한 골관절염 흰쥐에 대한 여지초(荔枝草)의 효능 평가

        Salviae Plebeiae Herba (SP) contains a numberous chemical components and reported to exert various pharmacological activities such as antioxidant, anti-inflammatory, antimicrobial, analgesic, hypoglycemic, antiviral, anti-tumour, and hepatoprotective effects. Even though it has these various effects, its scientific proof remains uncertain the effect and underlying mechanism about joint damage. Accordingly, the present study was designed to evaluate the therapeutic effects and possible underlying mechanism of SP on osteoarthritis. OA models were induced via intra-joint injection of monosodium iodoacetate (MIA) 50 ㎕ with 80 ㎎/㎖ in rats. Excluding the normal group, OA-induced mice were divided into 4 groups (control, INDO, SPL, SPH). The drug concentrations were indomethacin 5 ㎎/㎏, SP 500 ㎎/㎏, and SP 1000 ㎎/㎏, and were administered orally once a day for 2 weeks. After drug supplementation, the effects of SP were measured with serum analysis, western blotting, and histopathological staining. SP administration improved hind limb overload and it led to relieve pain. Moreover, it suppressed oxidative related factors. The NF-κB inactivation though the inhibition of IκBα phosphorlyation significantly decreased both inflammatory mediators such as COX-2 and iNOS and cytokines such as TNF-α, IL-1β, and IL-6. Whereas, anti-inflammatory cytokines like IL-4 and IL-10 were shown to increase. In addition, SP administration exerted a protective effect on ECM by reducing the expression of collagen degradation factors including MMP-1, -3, -13 and by increasing TIMP-1. Taken together, SP may possess the effect of delaying the progression of osteoarthritis and protecting cartilage. Accordingly, the present study may provide a scientific basis the chondroprotective effect of SP on MIA induced osteoarthritis.

        갈근이 뇌허혈 손상 흰쥐의 해마신경세포 손상에 미치는 영향

        Objective : This study investigated a neuroprotective effect of herariae Radix on cerebral ischemia. Method : The global cerebral ischemia was induced by bilateral common carotid arteries occlusion under hypotension (40 mmHg) in Sprague-Dawley rats. After the treatment of Puerariae Radix extract, changes of the pyramidal neurons in CAl region of the hippocampus were observed histologically. Results :l. The significant decrease of numbers of pyramidal neurons in CA1 region of the hippocampus resulted by cerebral ischemia. Then herariae Radix treatment demonstrated significant increase of numbers of pyramidal neurons in CA1 region as compared with control group. 2. The significant decrease of pyramidal layer thickness in CA1 region of the hippocampus resulted by cerebral ischemia. Then herariae Radix treatment demonstrated significant increase of pyramidal layer thickness in CAI region as compared with control group. Conclusion : These results suggested that Puerariae Radix reveals the neuroprotective effect through the control of noxious stress stimulations to neurons.

      • 盛土下部 軟弱地盤의 沈下 및 變位 擧動分析

        As embankment constructions on the soft ground proceed, many geotechnical problems concerning the ground stability and settlements begin to occur. Generally, soil has nonlinear stress-strain relation and its behavior depends on initial stress condition and stress history. There are many uncertain factors in soil. So, it is very difficult to predict the constitutive relation precisely. Results of the field data obtained by geotechnical investigations and field instrumentations were performed to investigate the deformational characteristics of the soft grounds. Theoritical calculations and numerical anaylsis were also performed. Many constitutive models on soil are proposed by several scientists. This study uses Modified Cam-Clay model among these models to analysis of the settlement and displacement of the soft ground under the embankment. Numerical analysis is conducted using CRISP which made by Cambridge University. Analysis of the settlement and displacement of the soft ground under the embankment with the finite element tenchnique using CRISP program made good predictions of the settlement and lateral displacement.

      • 慶南 梁山地方의 工業化에 따른 氣溫上昇에 대한 考察

        The temperature variations due to industrialization in Yangsan area are investigated using the 10 years(1980∼1989) temperature data and compared with those of Jinju. The major results are as follows: (1) The rates of increase of mean and minimum temperatures were more pronounced in Yangsan area than in Jinju area, and the increasing rates were always positive for every month. (2) There exists close correlation between the growth of population and increase of minimum temperature, and between the growth of population and the increase of mean temperature. Furtheromore, close correlations between the increase of those temperature and other industrial factors are also indentified.

        夏枯草의 免疫調節作用 및 알레르기 低減化에 關한 硏究

        The results of Prunellae Spica on immune response and anti-allergic reaction were as follows. 1. In Prunellae Spica group, hemagglutinin titer and hemolysin titer were increased in comparison with control group. But the results were not recognized as having significance. 2. The number of Plaque forming cells were increased in case of Prunellae Spica. But the results were not recognized as having significance. 3. The number of Rosette forming cells were increased in case of Prunellae Spica group, and the results were significant. 4. In experimentation of phagocytic activity in peritoneal exudate cells and monolymphocytus cells, Prunellae Spica group showed increase. But Prunellae Spica group in spleen cells showed decrease on the contrary. But these results were not recognized as having significance. 5. In Prunellae Spica group, results of PCA response were increased in case of 2 week, 4 week, 6 week on the contrary. 6. In experimentation on histamine contents, Prunellae Spica group showed derease at all agent contact. But these results were of no importance. From above these results, Prunellae Spica group showed good effects on immunoreaction. And Prunellae Spica group showed effects on anti-allergic reaction Ⅰ.

        韓國産 紅花子와 中國産 紅花子가 鎭痛 , 消炎 및 關節炎에 미치는 效果

        This experimental study was done to compare the efficacy of Korean and Chinese Carthami Semen on analgesic effect and remedy of inflammation and arthritis. The results were as follows; 1. In analgesic action on the writhing syndrome in mice induced by 0.7% acetic acid, Korean and Chinese Carthami Semen extract group showed significant decrease in comparison with control group. And in analgesic action on the writhing syndrome in mice induced by Acetylcholine HC1, Korean and chinese Carthami Semen extract group showed significant decrease in comparison with control group. 2. In Anti-inflammatory effects, Chinese Carthami Semen extract group showed significant decrease at 150 and 180 minutes after inducing paw-edema in comparison with control group. 3. In the method of adjuvant arthritis, orally administered for 21 days, Korean Carthami Semen extract group showed significant inhibition of the hind paw edema after 1 day and 21 days. Whereas Chinese Carthami Semen extract group showed some reduction, but the results didn's show significant decrease. 4. In the level of leucocytes count, Korean and Chinese Carthami Semen extract group showed significant decrease in comparison with control group. And in the level of total protein. CRP and glucose, Korean and Chinese Carthami Semen extract group didn't showed significant change in comparison with control group.

        산사가 알콜 투여로 유발된 흰쥐의 고지혈증과 간 손상의 예방에 미치는 영향

        Objectives : This study was performed to prove the preventive effect of water extracts from Crataegi Fructus on hyperlipiderma and liver damage induced by alcohol. Methods : Except for the normal group, we fed rat on 25% alcohol for 55 days. And Crataegi Fructus extract was administrated for the same period. We measured the serum component in rat's blood, body weight and weight of liver. Results : 1. At first, we observed preventive effects of Crataegi Fructus on hyperlipiderma induced by alcohol. Crataegi Fructus group showed significant decrease of total cholesterol and triglyceride in comparison with those of the control group. Crataegi Fructus group showed significant increase of HDL(high-density lipoprotein) cholesterol in comparison with those of the control group. Crataegi Fructus group showed significant increase of body weight in comparison with those of the control group in 4weeks and 8weeks. 2. At second, we observed preventive effects of Crataegi Fructus on liver damage induced by alcohol. Crataegi Fructus group showed significant decrease of GOT, GPT, ALP and LDH in comparison with those of the control group. Crataegi Fructus group showed significant increase of liver weight in comparison with those of the control group. Conclusions : Reviewing these experimental results, it appears that water extracts from Crataegi Fructus have pharmaceutical preventive efficacy on hyperlipiderma and liver damage induced by alcohol. Therefore further additional study should be conducted to elucidate in depth the pharmaceutical efficacy of these.
