<i>CYP2A6</i> and <i>ERCC1</i> polymorphisms correlate with efficacy of S-1 plus cisplatin in metastatic gastric cancer patients

<P><B>Background:</B></P><P>We evaluated the association between polymorphisms of cytochrome P450 2A6 (<I>CYP2A6</I>)/excision repair cross-complementation group 1 (<I>ERCC1</I>)/X-ray repair cross-complementing group 1(<I>XRCC1</I>) and treatment outcomes of metastatic gastric cancer (MGC) patients treated with S-1/cisplatin.</P><P><B>Methods:</B></P><P>Among MGC patients (<I>n</I>=108), who received S-1 (40 mg m<SUP>−2</SUP> b.i.d., days 1–14) and cisplatin (60 mg m<SUP>−2</SUP>, day 1) every 3 weeks, we analysed the wild-type allele (<I>W</I>) and variants (<I>V</I>) of <I>CYP2A6</I> (<I>*4</I>, <I>*7, *9, *10</I>), and the polymorphisms of <I>ERCC1</I> (rs11615, rs3212986) and <I>XRCC1</I> (rs25487).</P><P><B>Results:</B></P><P>Patients having fewer <I>CYP2A6</I> variants had better response rates (<I>W</I>/<I>W vs W</I>/<I>V</I> other than <I>*1/*4 vs V</I>/<I>V</I> or <I>*1/*4</I>=66.7 <I>vs</I> 58.3 <I>vs</I> 32.3% <I>P</I>=0.008), time to progression (TTP) (7.2 <I>vs</I> 6.1 <I>vs</I> 3.5 months, <I>P</I>=0.021), and overall survival (23.2 <I>vs</I> 15.4 <I>vs</I> 12.0 months, <I>P</I>=0.004). <I>ERCC1 19442C</I>><I>A</I> (rs3212986) was also associated with response rate (<I>C/C</I>, 46.7% <I>vs C/A</I>, 55.3% <I>vs A/A</I>, 87.5%) (<I>P</I>=0.048) and TTP (4.4 <I>vs</I> 7.6 <I>vs</I> 7.9 months) (<I>P</I>=0.012). Patients carrying both risk genotypes of <I>CYP2A6</I> (<I>V</I>/<I>V</I> or <I>1/*4</I>) and <I>ERCC1 19442C</I>><I>A</I> (<I>C/C</I>) <I>vs</I> those carrying none showed an adjusted odds ratio of 0.113 (<I>P</I>=0.004) for response, and adjusted hazard ratios of 3.748 (<I>P</I>=0.0001) for TTP and 2.961 (<I>P</I>=0.006) for death.</P><P><B>Conclusion:</B></P><P>Polymorphisms of <I>CYP2A6</I> and <I>ERCC1 19442C</I>><I>A</I> correlated with the efficacy of S-1/cisplatin.</P>

<P><B>Abstract</B></P><P>Let QA denote the class of bounded linear Hilbert space operators <I>T</I> which satisfy the operator inequality <SUP>T∗</SUP>|<SUP>T2</SUP>|T⩾<SUP>T∗</SUP>|T<SUP>|2</SUP>T. It is proved that if T∈QA is a contraction, then either <I>T</I> has a nontrivial invariant subspace or <I>T</I> is a proper contraction and the nonnegative operator D=<SUP>T∗</SUP>(|<SUP>T2</SUP>|-|T<SUP>|2</SUP>)T is strongly stable. It is shown that if T∈QA is a contraction with Hilbert–Schmidt defect operator such that <SUP>T-1</SUP>(0)⊆<SUP><SUP>T∗</SUP>-1</SUP>(0), then <I>T</I> is completely non–normal if and only if T∈<SUB>C10</SUB>, and a commutativity theorem is proved for contractions T∈QA. Let <SUB>Tu</SUB> and <SUB>Tc</SUB> denote the unitary part and the cnu part of a contraction <I>T</I>, respectively. We prove that if A=<SUB>Au</SUB>⊕<SUB>Ac</SUB> and B=<SUB>Bu</SUB>⊕<SUB>Bc</SUB> are QA-contractions such that <SUB>μ<SUB>Ac</SUB></SUB><∞, then <I>A</I> and <I>B</I> are quasi-similar if and only <SUB>Au</SUB> and <SUB>Bu</SUB> are unitarily equivalent and <SUB>Ac</SUB> and <SUB>Bc</SUB> are quasi-similar.</P>

V994 Herculis: the multiple system with a quadruple-lined spectrum and a double eclipsing feature

<P>ABSTRACT</P><P>We report the discovery of a multiple system with a quadruple-lined spectrum and a double eclipsing feature. Our photometric and high-resolution spectroscopic data show that V994 Herculis (V994 Her; ADS 11373 AB) is composed of two pairs of double-lined eclipsing binaries, which we designate as A and B. System A consists of a B8V+A0V binary with an orbital period of 2.083 264 d and system B of a A2V+A4V binary with 1.420 033 d. Our light curves show that both of them have a detached binary configuration. We derive masses and radii of four components (Aa, Ab, Ba and Bb) from the synthetic analyses of light curves and radial velocity curves. The masses of systems A and B are <I>M</I><SUB>Aa</SUB>= 2.83 ± 0.20 M<SUB>⊙</SUB>, <I>M</I><SUB>Ab</SUB>= 2.30 ± 0.16 M<SUB>⊙</SUB>, <I>M</I><SUB>Ba</SUB>= 1.87 ± 0.12 M<SUB>⊙</SUB> and <I>M</I><SUB>Bb</SUB>= 1.86 ± 0.12 M<SUB>⊙</SUB>, with radii <I>R</I><SUB>Aa</SUB>= 2.15 ± 0.05 R<SUB>⊙</SUB>, <I>R</I><SUB>Ab</SUB>= 1.71 ± 0.04 R<SUB>⊙</SUB>, <I>R</I><SUB>Ba</SUB>= 1.59 ± 0.08 R<SUB>⊙</SUB> and <I>R</I><SUB>Bb</SUB>= 1.50 ± 0.08 R<SUB>⊙</SUB>, respectively. These masses and radii are well consistent with the empirical relation for double-lined eclipsing binaries.</P>

Cytogenetic assessment of <i>Lilium longiflorum × L. hansonii</i> revealed by genomic in situ hybridization (GISH)

<P> Martagon (<I>Lilium hansonii;</I> MM) and <I>Longiflorum</I> (LL) are two major groups under the family <I>Liliaceae</I>, used for modern breeding to introduce new inter-genomic lily cultivars. Interspecific F<SUB>1</SUB> hybrids (LM) introduced through cut-style method between two diploid <I>Lilium longiflorum (2n=2x=24)</I> and <I>Lilium hansonii (2n=2x=24)</I> were evaluated cytogenetically by genomic in situ hybridization (GISH) technique. However, GISH analysis of F<SUB>1</SUB> interspecific (LM) hybrids showed equal chromosomal contribution from both female <I>Lilium longiflorum</I> (LL) and male <I>Lilium hansonii</I> (MM). Each of the parent contributed 12 chromosomes except three crosses i.e., two of <I>L. longiflorum</I> 'White Tower' × <I>L. hansonii;</I> (2x-1) and one of <I>L. longiflorum</I> 'Bright Tower' × <I>L. hansonii;</I> (2x-1). Among 11 inter-genomic crosses, 3 crosses failed (False hybrid) and 8 crosses (True hybrids) showed different ploidy level i.e., 2n=2x=24, 2n=2x-1=23 and 2n=2x-1=23 respectively. Recombinant chromosome usually not found in F<SUB>1</SUB> interspecific lily hybrids. Most often, genomic recombination occurred in the cross between two genetically different parents. Chromosome pairing and crossing over normally occurred during meiosis in backcross progenies. However, in this study, genome analysis (GISH) of F<SUB>1</SUB> hybrids (<I>L. longiflorum</I> 'White Tower' × <I>L. hansonii</I>) showed four recombinant sites including two M/L and two L/M recombinant chromosomes that denotes high genetic relationship between <I>L. longiflorum</I> and <I>L. hansonii.</I> </P>

Charmonium and <i>e</i> ⁺ <i>e</i> ⁻ pair photoproduction at mid-rapidity in ultra-peripheral Pb–Pb collisions at [FORMULA OMISSION]

<P>The ALICE Collaboration at the LHC has measured the J/<I>ψ</I> and <I>ψ</I>′ photoproduction at mid-rapidity in ultra-peripheral Pb–Pb collisions at [FORMULA OMISSION].</P><P>The charmonium is identified via its leptonic decay for events where the hadronic activity is required to be minimal. The analysis is based on an event sample corresponding to an integrated luminosity of about 23 μb<SUP>−1</SUP>. The cross section for coherent and incoherent J/<I>ψ</I> production in the rapidity interval −0.9<<I>y</I><0.9, are [FORMULA OMISSION] and [FORMULA OMISSION], respectively. The results are compared to theoretical models for J/<I>ψ</I> production and the coherent cross section is found to be in good agreement with those models incorporating moderate nuclear gluon shadowing at Bjorken-<I>x</I> around 10<SUP>−3</SUP>, such as EPS09 parametrization. In addition the cross section for the process <I>γγ</I>→<I>e</I><SUP>+</SUP><I>e</I><SUP>−</SUP> has been measured and found to be in agreement with models implementing QED at leading order.</P>

Molecular characterization and expression analysis of IκB from Haliotis discus discus

Innate immune system relies on the recognition of pathogen associated molecular patterns present in the microbes by the pattern recognition receptors leading to the activation of signaling cascade and subsequent synthesis of cytokines. NF-κB is a major stimulus activated transcription factor, which regulates the expression of a diverse array of genes. IκB is an inhibitor of NF-κB, retaining NF-κB in an inactive state in the cytoplasm. In this study, we have reported the characterization of first abalone IκB (HdIκB). The cDNA possessed an ORF of 1200 bp coding for a protein of 400 amino acids with molecular mass of 45 kDa and isoelectric point of 4.7. HdIκB protein possessed a conserved phosphorylation site <SUP>58</SUP>DSGIFS<SUP>63</SUP> in the N-terminal region, six ankyrin repeats, and a PEST sequence in the C-terminal region. A casein kinase II phosphorylation site could also be observed in the PEST sequence. Constitutive expression of HdIκB revealed its physiological significance since NF-κB is known to be activated by various stimuli. Elevated expression of HdIκB transcripts could be observed in abalones challenged with various mitogens and live microbes. This novel characterization of abalone IκB would further be a positive approach in the affirmation of evolutionary conservation and significance of this protein as a repressor/inhibitor of a pleiotropic transcription factor like NF-κB.

Measurements of the absolute branching fractions of B+→Xcc¯K+ and B+→D¯(*)0π+ at Belle

<P>We present the measurement of the absolute branching fractions of B+ -> Xc (c) over barK+ and B+ -> (D) over bar ((*)0)pi(+) decays, using a data sample of 772 x 10(6) B (B) over bar pairs collected at the gamma(4S) resonance with the Belle detector at the KEKB asymmetric-energy e(+)e(-) collider. Here, X-c (c) over bar denotes eta(c), J/psi, chi(c0), chi(c1), eta(c) (2S), psi(2S), psi(3770), X(3872), and X(3915). We do not observe significant signals for X(3872) or X(3915) and set the 90% confidence level upper limits at B(B+ -> X(3872)K+) < 2.6 x 10(-4) and B(B+ -> X(3915)K+) < 2.8 x 10(-4). These represent the most stringent upper limit for B(B+ -> X(3872)K+) to date and the first limit for B(B+ -> X(3915)K+). The measured branching fractions for eta(c) and eta(c)(2S) are the most precise to date, B(B+ -> eta K-c(+)) = (12.0 +/- 0.8 +/- 0.7) x 10(-4) and B(B+ -> eta(c)(2S)K+) = (4.8 +/- 1.1 +/- 0.3) x 10(-4), where the first and second uncertainties are statistical and systematic, respectively.</P>

Effect of Carbohydrate Sources in Phase I and Phase II Pig Starter Diets

Previous research in our laboratory has demonstrated the importance of lactose in phase I and II pig starter diets. Two experiments were conducted to evaluate the use of a carbohydrate by-product (food by-products) as a replacement for lactose. In Exp. I, 120 weaned pigs ($14{\pm}2d$ and 5.65kg) were allotted in a randomized complete block design (RCBD) to 10 replications with four pigs per pen. This experiment evaluated three carbohydrate sources (lactose, carbohydrate by-product, and 50-50 blend of the carbohydrate by-product and lactose). The carbohydrate sources were added at 26% in the phase I diets and 15% in the phase II diets. Phase I diets contained 7.5% spray dried plasma protein (SDP). The phase I diets were fed from d 0 to 14 and the phase II diets from d 15 to 28. There were no significant differences between carbohydrate sources on pig performance in phase I. However, during phase II pigs fed the diet with lactose had an improved gain/feed ratio (G/F) (p=0.06) compared to pigs fed the carbohydrate by-product. For the entire 28 d trial ADG, ADFI and G/F were similar for the 50-50 blend and those fed lactose. Total replacement of lactose with the carbohydrate byproduct resulted in a reduced G/F (p=0.09). Exp. 2 used 100 weaned pigs ($17{\pm}2d$ and 4.75kg) with five replications with five pigs per pen. This experiment evaluated four carbohydrate treatments (lactose, carbohydrate by-products, 50-50 blend, and corn). All phase I diets contained 3.5% SDP with the carbohydrate sources included at 15%, and were fed d 0 to 14. The phase II diets contained 7.5% of the carbohydrate sources and were fed d 15 to 27. A common phase III diet was fed d 28 to 42. During all phases pigs fed com tended to have a lower ADG than pigs fed the other carbohydrate sources with the 50-50 blend resulting in the highest ADG. The results of both experiments suggest that this carbohydrate by-product can replace at least 50% of the lactose in phase I and phase II pig starter diets.

Carcinoma is one of the most frequently observed malignant tumor in Korean patients. Result of surgery, for instance, in early stomach carcinomais encouraging and it is reported to be about 45% of five year survival rate but cases are few and rare. The majority of cases are that of either unresectable or metastatic regional lymphnode involvement of carcinoma. Their prognosis is poor. In those who had regional lymph node metastasis two year and three year survival rates are reported tobe each 37.1% and 17.9% respectively. Development of tumor immunology of the malignant disease made us greatly interested on the immunotheraly on the post resection stomach carcinoma. Studies on immunological competency by way of D.N.C.B. skin test started from 1971 and few cases those who may understand the new development of additional immuno therapy were selecter for B.C.G. innoculation after the subtotal gastrectomy. Recently monitering of peripheral thymus depentent lymphocytes (T-cell) count were undertaken to correlate the clinical course of the patients with the immunotherapy. In order to evaluate the immunological competency of healthy and all kind of malignant deseases of different stages, study with D.N.C.B. sensitization was undertaken in 264 cases. In healthy men of 53, only one man showed anergy, this anergic man is still healthy after three year observation. Table 1 further demonstrate the incedence of D.N.C.B. anergy in each group of patients, it notes that the unresectable malignant patients showed anergy 77 out of 105 cases(73.3%). From the D.N.C.B. sensirtization studies it may be clear to state that patient's immunological competency inthe malignant disease is severly deteriorated in the advanced tumo patients. The peripheral T-cell count also showed that the advanced malignant patients had lower T-cell count and it is progressively decreasing at the terminal stage(Table 2). In few cases who has B.C.G. after the subtotal gastrectomy the T-cell count are increasing after the treatment(Table 4). In 1971 five partients received B.C.G. therapy after the subtotal gastrectomy and three patients are sill living well. These 5 had all regional lymph node metastasis. The usual three year survival ratein similar patients is about 17 to 20%. Number of B.C.G. cases are not sufficient to a statistical significance but it may suggest a good deal of implication. Chance of two year survival after the B.C.G. therapy may be also far better than non-B.C.G.group(Table 5). The immunotherapy on malignant patients may be controversial, and yet we understand that the malignant tumor patients had impared immunity or immnological ability and that B.C.G. or immunotherapy may be effective on microscopic malignant lesion. In a selected case an immunotherapy after subtotal gastrectomy for gastric carcinoma may offer a beneficial effects on clinical course of the disease.

Search for B→hνν¯ decays with semileptonic tagging at Belle

<P>We present the results of a search for the rare decays B -> h nu(nu) over bar, where h stands for K+, K-S(0), K*(+); K*(0); pi(+); pi(0), rho(+) and rho(0). The results are obtained with 772 x 10(6) B (B) over bar pairs collected with the Belle detector at the KEKB e(+)e(-) collider. We reconstruct one B meson in a semileptonic decay and require a single h meson but nothing else on the signal side. We observe no significant signal and set upper limits on the branching fractions. The limits set on the B-0 -> K-S(0)nu(nu) over bar, B-0 -> K*(0)nu(nu) over bar, B+ -> pi(+)nu(nu) over bar, B-0 -> pi(0)nu(nu) over bar, B+ -> rho(+)nu(nu) over bar, and B-0 -> rho(0)nu(nu) over bar channels are the world's most stringent.</P>
