        Aberrant Promoter Methylation at CpG Cytosines Induce the Upregulation of the E2F5 Gene in Breast Cancer

        Purpose: The promoter methylation status of cell cycle regulatory genes plays a crucial role in the regulation of the eukaryotic cell cycle. CpG cytosines are actively subjected to methylation during tumorigenesis, resulting in gain/loss of function. E2F5 gene has growth repressive activities; various studies suggest its involvement in tumorigenesis. This study aims to investigate the epigenetic regulation of E2F5 in breast cancer to better understand tumor biology. Methods: The promoter methylation status of 50 breast tumor tissues and adjacent normal control tissues was analyzed. mRNA expression was determined using SYBR® green quantitative polymerase chain reaction (PCR), and methylation- specific PCR was performed for bisulfite-modified genomic DNA using E2F5-specific primers to assess promoter methylation. Data was statistically analyzed. Results: Significant (p<0.001) upregulation was observed in E2F5 expression among tumor tissues, relative to the control group. These samples were hypo-methylated at the E2F5 promoter region in the tumor tissues, compared to the control. Change in the methylation status (Δmeth) was significantly lower (p=0.022) in the tumor samples, indicating possible involvement in tumorigenesis. Patients at the postmenopausal stage showed higher methylation (75%) than those at the premenopausal stage (23.1%). Interestingly, methylation levels gradually increased from the early to the advanced stages of the disease (p<0.001), which suggests a putative role of E2F5 methylation in disease progression that can significantly modulate tumor biology at more advanced stage and at postmenopausal age (Pearson’s r=0.99 and 0.86, respectively). Among tissues with different histological status, methylation frequency was higher in invasive lobular carcinoma (80.0%), followed by invasive ductal carcinoma (46.7%) and ductal carcinoma in situ (20.0%). Conclusion: Methylation is an important epigenetic factor that might be involved in the upregulation of E2F5 gene in tumor tissues, which can be used as a prognostic marker for breast cancer.

        Pretreatment of Albino Rats with Methanolic Fruit Extract of Randia Dumetorum (L.) Protects against Alcohol Induced Liver Damage

        Alcohol abuse and its medical and social consequences are a major health problem in many areas of the world. The present study was conducted to evaluate the protective effect of methanolic fruit extract of Randia dumetorum (L.) on alcohol-induced liver damage in rats. Rats were divided into five different groups (n=6), group I served as a control, group II received ethanol (3 ml/100 g/day p.o.), group III served as standard group and received silymarin (50 mg/kg p.o.), group IV and V served as extract treatment groups and received 50 & 100 mg/kg methanolic extract of R. dumetorum. All the treatment protocols followed 30 days and after rats were sacrificed blood and liver were used for biochemical and histological studies, respectively. The activities serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), triglyceride (TG), direct bilirubin (DB), total bilirubin (TB) and lipid peroxidation were statistically increased in rats exposed to alcohol while total protein and glutathione decreased compared to control rats. Treatment with R. dumetorum significantly decreased the elevated levels of ALT, AST, TG, DB, TB and lipid peroxidation compared to the group exposed to alcohol only. R. dumetorum significantly resulted in increased levels of total protein and reduced glutathione compared to the group that received alcohol only. Histology of the liver section of the animals treated with R. dumetorum improved the hepatotoxicity caused by alcohol. Hence the study concluded that R. dumetorum has potential hepatoprotective activity.

        A simplified design approach for high-speed wind tunnels: part II. Diffuser optimization and complete duct design

        This study presents part-II of the work-in-progress for the development of a high-speed wind tunnel under the EU Commission (ERDF) project. This study was carried out after the inlet section of the supersonic wind tunnel prototype had been designed in part-I [1] by introducing a simplified design approach of “table of inclination”. The design optimization of the exhaust section, i.e., diffuser of the wind tunnel is the main objective of the current study. Diffuser design optimization was carried out in three steps; at first, the selection of the diffuser type was carried out and the initial estimated design was created. In the next step, 2D optimization of the wind tunnel design was carried out, and finally, 3D optimization of the complete wind tunnel model was performed. The main geometric parameters of diffuser design were optimized using CFD analysis. Due to the proposed design optimization strategy, the conventionally long diffuser length was successfully reduced. Additionally, 3D optimization further improved the design and helped to resolve the problem of higher operating power. This compact wind tunnel duct will have relatively lower manufacturing costs as well as the overall testing facility will occupy a smaller space. The optimized design of the diffuser and its integration with the inlet section of the wind tunnel marked the completion of the design of the full wind tunnel duct.

        Biomonitoring of Toxic Effects of Pesticides in Occupationally Exposed Individuals

        Background: Workers in pesticide manufacturing industries are constantly exposed to pesticides. Genetic biomonitoring provides an early identification of potential cancer and genetic diseases in exposed populations. The objectives of this biomonitoring study were to assess DNA damage through comet assay in blood samples collected from industry workers and compare these results with those of classical analytical techniques used for complete blood count analysis. Methods: Samples from controls (n = 20) and exposed workers (n = 38) from an industrial area in Multan, Pakistan, were subjected to various tests. Malathion residues in blood samples were measured by gas chromatography. Results: The exposed workers who were employed in the pesticide manufacturing industry for a longer period (i.e., 13-25 years) had significantly higher DNA tail length ($7.04{\mu}m$) than the controls ($0.94{\mu}m$). Workers in the exposed group also had higher white blood cell and red blood cell counts, and lower levels of mean corpuscular hemoglobin (MCH), MCH concentration, and mean corpuscular volume in comparison with normal levels for these parameters. Malathion was not detected in the control group. However, in the exposed group, 72% of whole blood samples had malathion with a mean value of 0.14 mg/L (range 0.01-0.31 mg/L). Conclusion: We found a strong correlation ($R^2=0.91$) between DNA damage in terms of tail length and malathion concentration in blood. Intensive efforts and trainings are thus required to build awareness about safety practices and to change industrial workers' attitude to prevent harmful environmental and anthropogenic effects.

      • Dynamic Interaction between Conditional Stock Market Volatility and Macroeconomic Uncertainty of Bangladesh

        Purpose: The aim of this study is to explore the dynamic linkage between conditional stock market volatility and macroeconomic uncertainty of Bangladesh. Research design, data, and methodology: This study uses monthly data covering the time period from January 2005 to December 2018. A comprehensive set of macroeconomic variables, namely industrial production index (IP), consumer price index (CPI), broad money supply (M2), 91-day treasury bill rate (TB), treasury bond yield (GB), exchange rate (EX), inflow of foreign remittance (RT) and stock market index of DSEX are used for analysis. Symmetric and asymmetric univariate GARCH family of models and multivariate VAR model, along with block exogeneity and impulse response functions, are implemented on conditional volatility series to discover the possible interactions and causal relations between macroeconomic forces and stock return. Results: The analysis of the study exhibits time-varying volatility and volatility persistence in all the variables of interest. Moreover, the asymmetric effect is found significant in the stock return and most of the growth series of macroeconomic fundamentals. Results from the multivariate VAR model indicate that only short-term interest rate significantly influence the stock market volatility, while conditional stock return volatility is significant in explaining the volatility of industrial production, inflation, and treasury bill rate. Conclusion: The findings suggest an increasing interdependence between the money market and equity market as well as the macroeconomic fundamentals of Bangladesh.

        Control and prevention of foot and mouth disease (FMD) by vaccination remains unsatisfactory in endemic countries. Indeed, consistent and new FMD epidemics in previously disease-free countries have precipitated the need for a worldwide control strategy. Outbreaks in vaccinated animals require that a new and safe vaccine be developed against foot and mouth virus (FMDV). FMDV can be eradicated worldwide based on previous scientific information about its spread using existing and modern control strategies.

        A simplified design approach for high-speed wind tunnels. Part I: Table of inclination

        This study aims to develop a simplified approach for the design of high-speed wind tunnels, operating within the range of Mach 1-4. The study is conducted in two stages;design stage-I consists of the basic parts of a high-speed wind tunnel, i.e. test section and nozzle throat sizes along with the calculations of the minimum pressure ratios necessary for the starting and running of the wind tunnel. Design stage-II is the aerodynamic design of the inlet nozzle contour with the focus on the divergent nozzle section only. In this stage, the fundamental method of characteristics (MOC) is employed by using region-to-region solution approach. Due to the long and complicated procedure of classical MOC, a simpler approach of “table of inclination” is proposed. This inclination table approach is simple in use, highly precise, and efficient which reduces more than 75 % of the calculation time of the MOC. Validation of the nozzle contour design is performed with the Area-Mach ratio relation followed by the results verification obtained from the MATLAB programming. The contour design results of the study are highly accurate (less than 1 % error) which manifests the future possible significance of the inclination table approach for supersonic wind tunnel designs.

        Design Optimization and Investigation of Aerodynamic Characteristics of Low Reynolds Number Airfoils

        This study aims to develop a simple and efficient design optimization methodology for the low Reynolds number airfoils. XFOIL is used as an aerodynamic solver while modeFRONTIER workflow is employed for the design optimization purpose. The airfoil SG6043 is used as the reference airfoil for optimization due to its common applications when long-endurance characteristics are desired. A simple design optimization methodology with the integration of XFOIL in the modeFRONTIER workflow environment is proposed in this study. The proposed “software integration methodology” demonstrated up to 10% improvement in the optimization parameter which makes it more efficient by reducing the optimization time and steps without unnecessary user intervention which are the limitations conventionally associated with the optimization process. The optimization results are further compared with the results of the numerical simulations. The use of the transition-sensitive turbulence model allowed the evaluation of the behavior of the laminar separation bubble for different angles of attack, observing that it shifts towards the leading edge and has its length reduced as the angle of attack increases. The newly generated airfoil exhibits improved aerodynamic characteristics as compared to the base airfoil. The optimized airfoil can be used in the applications of UAVs as well as in general aviation. Further validation of the airfoil using wind tunnel testing is recommended and planned.

        Relation between the Irreducible Polynomials that Generates the Same Binary Sequence Over Odd Characteristic Field

        A pseudo-random sequence generated by using a primitive polynomial, trace function, and Legendre symbol has been researched in our previous work. Our previous sequence has some interesting features such as period, autocorrelation, and linear complexity. A pseudo-random sequence widely used in cryptography. However, from the aspect of the practical use in cryptographic systems sequence needs to generate swiftly. Our previous sequence generated by utilizing a primitive polynomial, however, finding a primitive polynomial requires high calculating cost when the degree or the characteristic is large. It’s a shortcoming of our previous work. The main contribution of this work is to find some relation between the generated sequence and irreducible polynomials. The purpose of this relationship is to generate the same sequence without utilizing a primitive polynomial. From the experimental observation, it is found that there are (p - 1)/2 kinds of polynomial, which generates the same sequence. In addition, some of these polynomials are non-primitive polynomial. In this paper, these relationships between the sequence and the polynomials are shown by some examples. Furthermore, these relationships are proven theoretically also.

        Determination of Bergenin in Different Parts of Bergenia ciliata using a Validated RP-HPLC Method

        Bergenia ciliata (Family: Saxifragaceae) is a folklore remedy for the treatment of various ailments in Asian countries. Bergenin (1) has been isolated as an active constituent in many studies, however, the amount of bergenin has not been determined in all parts of the plant. A simple RP-HPLC method was developed to determine the amount of bergenin in methanol extracts of leaves, rhizomes and roots of the plant. Separation was achieved on an Agilent Eclipse XDB-C18 column maintained at 25 oC using isocratic solvent system (water: methanol: acetic acid; 62.5:37:0.5 v/v/v) adjusted at pH 2 0 at a flow rate of 1.0 mL/min. and detected at 275 nm. Correlation coefficient (0.9952) showed linearity of concentration (5-200 μg/mL) and response. The values of LOD (0.00947 μg/mL) and LOQ (0.02869 μg/mL) indicated that method was sensitive. The recovery of bergenin was 99.99-100% indicating accuracy of method. The methanol extract of rhizomes contained higher amount of bergenin (19.4%) than roots (9.2%) and leaves (6.9%). It is concluded that methanol extract of rhizomes is a better source of bergenin than other parts of the plant. The findings are useful for standardization of bergenin containing extracts and herbal preparations.
