        RAPD typing of Lactobacillus brevis isolated from various food products from Korea

        In the present study, the fingerprinting technique, random amplified polymorphic DNA-PCR was evaluated to characterize 13 strains of Lactobacillus brevis, isolated from different vegetable products of South Korea. Two primers i.e. 239 and KAY3 were used. The primer 239 produced bands ranged from 500-4,000 bp and KAY3 primer produced bands with sizes from 600-4,000 bp. Both primers produced thirteen different RAPD profiles. Phylogenetic dendrogram showed that all the isolates could be divided into six major clusters both the primers. However, a few strains of L. brevis had similar profiles and were not well differentiated by RAPD.

        Molecular typing tools for identifying and characterizing lactic acid bacteria: a review

        Identification and classification of beneficialmicrobes is of the highest significance in food science andrelated industries. Conventional phenotypic approachespose many challenges, and they may misidentify a target,limiting their use. Genotyping tools show comparativelybetter prospects, and they are widely used for distinguishingmicroorganisms. The techniques already employed ingenotyping of lactic acid bacteria (LAB) are slightly differentfrom one another, and each tool has its ownadvantages and disadvantages. This review paper compilesthe comprehensive details of several fingerprinting toolsthat have been used for identifying and characterizing LABat the species, sub-species, and strain levels. Notably, mostof these approaches are based on restriction digestion,amplification using polymerase chain reaction, andsequencing. Nowadays, DNA sequencing technologieshave made considerable progress in terms of cost,throughput, and methodology. A research journey todevelop improved versions of generally applicable andeconomically viable tools for fingerprinting analysis isongoing globally.

        Tracking of deliberately inoculated Leuconostoc mesenteroides and Lactobacillus brevis in kimchi

        The objective of this study was to track intentionallyinoculated Leuconostoc mesenteroides (11251) andLactobacillus brevis (B151) strains in kimchi using randomamplified polymorphic DNA (RAPD), repetitive elementpalindromic PCR (rep-PCR), and comparative housekeepinggene sequences analysis. The 16S rRNA gene providedspecies-level information for 30 colonies randomly pickedfrom kimchi inoculated with strains 11251 and B151. Outof 30 colonies, one colony was matched to strain 11251,and two colonies were found identical to strain B151 referencestrain in inoculated kimchi. Notably, among thethree tools, strain 11251 was best tracked by comparativegene sequence analysis, while strain B151 tracked by allthree tools. Our results suggest that the gene sequenceanalysis is a more reliable tool for tracking of desiredstrains than RAPD and rep-PCR. Based on the findings, itis recommended that gene sequence analysis could be usedto avoid misuse of industrially useful strains within thegrowing food industry.

        Analysis of Leuconostoc citreum strains using multilocus sequence typing

        The objective of this study was to perform genetic diversity analysis of 13 strains isolated from South Korean foods by multilocus sequence typing (MLST). For typing, seven housekeeping loci (atpA, dnaA, dnaK, gyrB, pheS, pyrG, and rpoA) were selected, amplified and analyzed. Fifty-one polymorphic sites varying from 1 to 22 in each species were identified. Thirteen sequence types were generated with allele numbers ranged from 2 to 10. The overall relationship between strains was assessed by unweighted pair group method with arithmetic mean dendrogram and minimum spanning tree. In addition, combined spits tree analysis revealed intragenic recombination. No clear relationship was observed between the isolation sources and strains. The developed MLST scheme enhanced our knowledge of the population diversity of Leu. citreum strains and will be used further for the selection of industrially important strain.

        Anti-diabetic effects of blue honeyberry on high-fed-diet-induced type II diabetic mouse

        BACKGROUND/OBJECTIVE: The blue honeysuckle berry (Lonicera caerulea var. edulis L.) is a small deciduous shrub belonging to the Caprifoliaceae family that is native to Russia, China, Japan, and Korea. The berry of this shrub is edible, sweet and juicy and is commonly known as the blue honeyberry (BHB). This study examined the anti-diabetic potential of BHB on high-fat-diet-induced mild diabetic mice. The hypoglycemic, and nephroprotective effects of the 12-week oral administration of blue honeyberry extract were analyzed. MATERIALS/METHODS: The hypoglycemic effects were based on the observed changes in insulin, blood glucose, and glycated hemoglobin (HbA1c). Furthermore, the changes in the weight of the pancreas, including its histopathology and immunohistochemical investigation were also performed. Moreover, the nephroprotective effects were analyzed by observing the changes in kidney weight, its histopathology, blood urea nitrogen (BUN), and serum creatinine levels. RESULTS: The results showed that the high-fat diet (HFD)-induced control mice showed a noticeable increase in blood glucose, insulin, HbA1c, BUN, and creatinine levels. Furthermore, growth was observed in lipid droplet deposition related to the degenerative lesions in the vacuolated renal tubules with the evident enlargement and hyperplasia of the pancreatic islets. In addition, in the endocrine pancreas, there was an increase in the insulin-and glucagon-producing cells, as well as in the insulin/glucagon cell ratios. On the other hand, compared to the HFD-treated mice group, all these diabetic and related complications were ameliorated significantly in a dose-dependent manner after 84 days of the continuous oral administration of BHBe at 400, 200 and 100 mg/kg, and a dramatic resettlement in the hepatic glucose-regulating enzyme activities was observed. CONCLUSIONS: By assessing the key parameters for T2DM, the present study showed that the BHBe could act as a potential herbal agent to cure diabetes (type II) and associated ailments in HFD-induced mice.

        Molecular discrimination of <i>Lactobacillus brevis</i> strains isolated from food products in South Korea using multilocus sequence typing

        <P><B>Abstract</B></P> <P> <I>Lactobacillus brevis</I> plays an important role in the manufacture of fermented foods. The aim of the present study was to know the genetic relationships among <I>L. brevis</I> isolates using multi-locus sequence typing of seven different housekeeping genes. 13 isolates were collected from different food products of South Korea. The results of the sequence analysis indicated 13 different sequence types (STs), each represented by a single strain. Among the seven loci, a total of 83 polymorphic sites were detected. The number of polymorphic sites varied from 4 (<I>rpoB</I>) to 24 (<I>recA</I>), whereas nucleotide diversity per site among the seven genes varied from 0.00161 (<I>rpoB</I>) to 0.01585 (<I>recA</I>). The number of alleles varied from 2 (<I>dnaK</I>) to 13 (<I>recA</I>). SplitsTree recombination was also performed for all seven loci and concatenated sequences. Phylogenetic analysis divided the 13 strains into three major groups based on their relationships. The minimum spanning tree revealed no clear relationship between the and isolation source or location and <I>L. brevis</I> isolates.         Efficacy and Safety of Sinetrol-XPur on Weight and Body Fat Reduction in Overweight or Obese Adults: A 12-Week, Randomized, Double-Blind, Parallel, Placebo-Controlled Trial

        This study investigated the effect of Sinetrol-XPur on weight and body fat reduction in overweight or obese Korean participants. Among 100 overweight or obese participants enrolled in a 12-week randomized, double-blinded, controlled study, 86 participants completed the trial. Participants took either two Sinetrol-XPur tablets (450 mg per tablet) or two placebo tablets once a day. Bodyweight, body fat percentage, body mass index (BMI), body fat mass, waist circumference, and various safety parameters were measured. After the 12-week intervention, a significant reduction was observed in the body fat mass (P = .030) by dual-energy X-ray absorptiometry (DEXA), body weight (P = .002), and BMI (P = .002) compared to the placebo. Body fat percentage (P = .007) by DEXA showed a significant reduction in the Sinetrol-XPur group, but no difference compared to the control group. Abdominal metabolic risks by computed tomography and blood biochemistry analysis were significantly decreased in the Sinetrol-XPur group, but there were no differences between the Sinetrol-XPur and placebo groups. Safety profiles were not different between the two groups. These results suggested that Sinetrol-XPur significantly reduced body weight, body fat mass, and BMI in obese Korean subjects, which confirms the antiobesity effect of Sinetrol-XPur in the Korean population.
