蜂毒藥鍼이 脊髓內 痛症關聯 神經細胞의 活性에 미치는 影響

In spite of the use of Bee Venom aqua-acupuncture in the clinics, the scientific evaluation of effects is not enough. Bee Venom aqua-acupuncture is used according to the stimulation of acupuncture point and the chemical effects of Bee Venom. The aims of this study is to investigate the analgegic effects of the Bee Venom aqua-acupuncture, through the change of writhing reflex and the change of c-fos in secondary neurons in the spinal cord. Materials and Methods : Pain animal model was used acetic acid method. The changes of writhing reflex of the mice which were derived pain by injecting acetic acid into the abdomen, after stimulating Bee Venom aqua-acupuncture on Chungwan(CV12) were measured. We used Fos immunohistochemical technique to study the neuronal activity in the spinal cord. Results : 1.Expression of c-fos in superficial dosal horn(SDH), nucleus proprius(NP) and neck of dorsal horn(N) on 6~9th thoracic spine decreased significantly at 2.5×10-4g/kg Bee Venom aqua-acupuncture, compared with saline-acetic acid group. 2.The numeral change of Fos-LI neurons on the NP, N, and ventral gray(V) on 6~9th thoracic spine, SDH on 9~11th thoracic spine, and SDH and V on 11~13th thoracic spine decreased significantly at Chungwan(CV12) Bee Venom aqua-acupuncture, compared with saline-acetic acid group. 3.The correlation between the numbers of writhing reflex and Fos-LI neurons in T6-13 segment was statistically statistically significant at Chungwan(CV12) Bee Venom aqua-acupuncture. Conclusion : This study shows that the Bee Venom aqua-acupuncture on Chungwan(CV12) decreases the numbers of Fos-LI neurons. As the analgegic effects of Bee Venom aqua-acupuncture is recognized. Bee Venom aqua-acupuncture treatment is expected for pain modulation. In order to use it in many ways. more researches are needed for the dose and stability of Bee Venom aqua-acupuncture.

To study anti-cancer effect and molecular biological mechanism of bee venom for aqua-acupuncture, the effects of bee venom on cell viability, apoptosis, and cell cycle were analyzed using MTT assay, tryphan blue assay, [3H]thymidnine release assay, flow cytometric analysis, activity of caspase-3 protease activity assay, and immunocytometric analysis of PCNA. To explore whether anti-cancer effects of bee venom are associated with the transcriptional control of gene expression, quantitative RT-PCR analysis of apoptosis- and cell cycle-related genes was performed. The obtained results are summarized as follows: 1.The MTT assay demonstrated that cell viability was decreased by bee venom in a dose-dependant manner. 2.Significant induction of apoptosis was identified using tryphan blue assay. [³H]thymidine release assay, and flow cytometric analysis of sub G₁fraction. 3.In analysis of caspase-3 protease activity, the activity had increased significantly, in a dose-dependant manner. 4.Quantitative RT-PCR analysis of the apoptosis-related genes showed that Bcl-2 and Bcl-X□ were down-regulated whereas Bax was up-regulated by bee venom treatment. 5.In flow cytometric analysis of cell cycle and immunocytometric analysis of PCNA expression, cell numbers of G₁phase was increased by a dose-dependant manner. 6.In quantitative RT-PCR analysis of the cell cycle-related genes, p21, p27, and p57 were increased, while Cyclin D1, CDK4, c-Myc, c-Fos, and Histone H3 were decreased. In contrast, there were no remarkable changes in expression levels of CDC2 and c-Jun.
高頻度電鍼刺戟의 刺戟時間에 따른 中樞神經系 神經細胞의 活性變化에 미치는 影響

The present study was designed to investigate the effect of different stimulation-duration of high frequency electroacupuncture(EA) treatment on the neuronal activities in the spinal cord brainstem using Fos immunohistochemical technique. Three different stimulus-duration was used in this experiment: 30minutes, 1 hour and 2 hours. The summerized results were summerized as follow : 1. The number of Fos expression was significantly increased in the spinal cord dorsal horn depending upon the increase of stimulus-duration (P<0.05). Otherwise, there was no significant difference between 30 minutes EA treated group and anesthetic control. 2. High frequency EA biphasic stimulation significantly enhanced the Fos expression in the middle and rostral portion of PAG LD, and caudal PAG LV after 1 hour and 2 hours treatment, The number of Fos immunoreactive neuron in the brainstem was increased according to the length of stimulus-duration. Those results indicate that at least 1 hour EA treatment was necessary to increase the neuronal activities in the spinal cord and brainstem. Those basic data from this study can be applied to establish the effective treatment of EA for pain control in the clinical field.

Although Korean influenza virus isolates have been genetically associated with the vaccine strains of the corresponding year, influenza B viruses have prevailed almost every year in Korea during the past decades. We have 'analyzed the genetic characteristics and evolutionary patterns of the haemagglutinin (HA) 1 domains of influenza B viruses isolated during 1988-1999 using direct RT-PCR and sequencing. Phylogenetic analysis of influenza B viruses isolated in Korea indicated that antigenically and genetically distinguishable strains of the lineage II and lineage III variants had been cocirculated. Variants prevailed in early 1990s are represented in 1996/97 and 1998/99 and some different variants have been co circulated geographically and prevailed concurrently in Korea. All HA1s of Korean isolates have amino acid substitutions mainly in the region between position 124 and 310, which was previously proposed an immunodominant region. Insertion-deletion patterns of the HA gene revealed that Korean influenza B viruses were evolved from Lee40 with different manner between lineage II and III viruses. Lineage III viruses were also divided into two groups as conserved group and inserted group, in relation to Lee40. But lineage II viruses had evolved with directional pattern. Antigenic index proposed that influenza B isolates prevailed since 1996/97 seasons might had emerged from the antigenic variants of a Seo1697-like virus and that new variants might appear from the lineage II viruses resulting in persistent prevalence in Korea.
