        다시마, 청매실, 동충하초 및 키토산이 첨가된 멸치액젓의 저장 중 품질특성

        소비자의 기호 및 상품성을 높이고 다시마, 청매실, 동충하초, 키토산의 여러 성분과 영양 및 기능성이 향상된 저염 멸치액젓을 개발하고자, 5년간 숙성시킨 멸치액젓(식염 26%)에 끓인 물로 염 농도를 14 및 17%으로 조정한 후 다시마, 청매실, 동충하초, 키토산을 첨가하여 150일 동안 후 숙성하면서 나타나는 제품의 품질특성을 분석하였다. 저장기간 동안에 pH는 서서히 감소하였고, 아미노질소 및 휘발성 염기질소량은 저장 50일째까지 증가하였다가, 일정한 값을 유지하였으며, TBA 값은 저장 50일째까지 증가하였다가 감소하였다. 생균수를 비롯한 젖산균, 단백질분해균 및 곰팡이 균은 후 숙성이 진행됨에 따라 다소 일정한 증가율을 나타내었다. 다시마, 청매실, 동충하초 및 키토산 첨가에 따른 제품 열화현상은 저장 150일째까지는 나타나지 않아서, 저염 멸치액젓으로 상품화의 개발이 가능함을 확인하였다. 하지만 숙성 150일부터는 관능적으로 다소 감소함을 볼 수 있어서 유통기한에 대한 실험과 제조된 저염 멸치액젓에 대한 기능성 검증에 대한 실험이 추가적으로 필요하다고 본다. Fish sauce is one of the most popular fermented fish products over the world. But it is usually manufactured with high salt concentration (>25%) and long periods of elaboration. In order to increase the consumption of fish sauce, the functional anchovy sauces with low salt concentrations (14 and 17%) were manufactured by adding sea tangle (Kjellamaniealla crassifolia), ume (Prunus mume Sieb. et Zucc), tochukaso (Paecilomyces japonica), and chitosan. On 50 days of storage, pH of all treatments decreased to 5.1, while the amount of lactic acid increased continuously as storage period increased. The amounts of VBN, amino-N, and TBA were highest on 50 days of storage and then kept constantly or decreased a little thereafter. The numbers of total viable cell, lactic acid bacteria, proteolytic bacteria, and fungi increased very slowly as storage period increased.

        멸치액젓 유래 저분자 물질의 항산화활성

        Antioxidative activities of low molecular weight biocompounds purified from anchovy (Engraulis japonicus) sauce fermented at $15\pm3^{\circ}C$ for 5 years were investigated. The fermented anchovy sauce showed 5 peaks on gel chromatography pattern. Antioxidant activity of peak 2 was $82.7\%$ followed by $42.6\%$ of peak 1. Main antioxidant compounds of peak 1 were glutamic acid and lysine, but those of peak 2 were not confirmed by amino acid sequence analysis.

        유산균 발효를 통한 황금 추출물의 항염증 효과 증진

        본 연구에서는 소염, 항염 작용이 뛰어난 작물로 알려진 황금을 유산균인 Lactobacillus rhamnosus를 이용해 발효를 통한 황금의 추출물에 대한 추출공정별 항염증 효과 증진을 측정하였다. 추출 공정으로는 일반 열수추출(WE), 일반 70% 에탄올 추출(EE), L. rhamnosus 발효 열수 추출(FWE), L. rhamnosus 발효 70% 에탄올 추출(FEE)로 나누어 진행하였다. 각각의 추출 공정별 수율 측정결과 발효 70% 에탄올 추출이 25.9%로 최대 수율을 얻었으며, 발효 열수 추출, 일반 70% 에탄올 추출, 일반 열수 추출 순으로 높은 수율을 얻었다. 추출 공정별 세포독성 측정에서 발효 황금 에탄올 추출물의 농도가 1.0 mg/mL일때 9.8%로 가장 낮은 세포독성을 나타냈으며, 일반 에탄올 추출의 경우 22.6%로 가장 높은 세포 독성을 가졌다. Hyaluronidase 저해 효과는 발효 황금 에탄올 추출물의 농도가 1.0 mg/mL에서 최대 46.8%의 hyaluronidase 저해 활성을 보였으며, NO 생성량 및 PGE<sub>2</sub> 생성량 측정도 1.0 mg/mL 농도의 발효 황금 에탄올 추출물이 각각 36%의 최대 NO 생성량 감소 및 810 pg/mL의 낮은 PGE<sub>2</sub> 값을 나타냈다. 따라서 황금의 항염증 관련 효과를 위한 최적 추출 공정으로는 발효를 통한 황금의 70% 에탄올 추출 수율이 가장 높았으며, 이에 따라 항염증 관련 생리활성 물질이 보다 더 많이 용출됨을 확인할 수 있다. This study was performed to investigate the anti-inflammatory activities of fermented Scutellaria baicalensis extracts using Lactobacillus rhamnosus. The extracts were WE (water extract at 100 ℃ for 24 h), EE (70% ethanol extract at 60 ℃ for 24 h), FWE (fermented and water extract at 60 ℃ for 24 h), FEE (fermented and 70% ethanol extract at 60 ℃ for 24 h). The cytotoxicity of the extracts was in the range of 11.2 ∼ 15.6 % at 1.0 mg/mL concentratioin. The FEE showed the lowest activity at 1.0 mg/mL concentratioin. Compared to the WE, hyaluronidase inhibitory activity contents in the FEE increased to 9.2% at 1.0 mg/mL concentratioin. Nitric oxide production of WE, EE, FWE and FEE at 1.0 mg/mL concentration was mesured as 7.6, 7.9, 6.9, 6.4 μM, respectively. PGE<sub>2</sub> secretion of the human fibroblast of the FEE were lower than 810 pg/mL. Our results suggested that the extracts from fermentation process after 70% ethanol extraction had relatively high anti-inflammatory activities and that the Scutellaria baicalensis could be more extracted in FEE than others.

        구멍갈파래의 고압 균질 전처리 공정을 통한 바이오에탄올 생산용 당화수율 증진

        This study was investigated to improve the saccharification yield of Ulva pertusa Kjellman by the high pressure homogenization process. It was found that the high pressure homogenization pretreatment effectively destructed the cell wall structures only by using water. The high pressure homogenization process was operated under various conditions such as 10000, 20000 or 30000 psi with different recycling numbers. The optimal condition was determined as 30000 psi and 2 pass of recycling numbers and the sugar conversion yields were 16.02 (%, w/w) of glucose and 14.70 (%,w/w) of xylose, respectively. In the case of enzymatic treating the hydrolyzates with 5 FPU/glucan of celullase and 100 units/mL of amyloglucosidase, 65.8% of carbohydrates was converted into glucose. Using the hydrolysates of Ulva pertusa Kjellman, 48.7% of ethanol was obtained in the culture S.cerevisiae. These results showed that the high pressure homogenization process could efficiently hydrolyze the marine resource by using only water for bioethanol production.

        초고압 가공이 조미오징어의 저장성에 미치는 영향

        본 연구는 조미오징어의 저장성을 향상시키기 위한 초고압 기술의 응용 가능성을 평가하였다. 포장된 조미오징어를 400 ㎫에서 20분간 처리한 후 초고압 처리되지 않은 대조군과 21일간 냉장보관을 하였다. 냉장기간 동안 미생물의 생육, DMA 함량, TMA 함량, 총 BA 함량 및 효소활성을 0, 7, 14, 21에 각각 평가하였다. 미생물의 수는 초고압 처리 후 대조군에 비교하여 효과적으로 2.77 log CFU/g을 감소시켰다(p<0.05). 대조군에서 DMA와 TMA의 함량은 냉장저장 7일 후 각각 15.99 ㎎/g과 42.82 ㎎/g까지 증가되었다. 초고압 처리된 조미오징어의 효소활성은 냉장저장 7일 후 4.32 nkat/g으로 대조군(7.13 nkat/g) 비교하여 유의성 있게 감소되었다. 그러므로 초고압의 응용은 조미오징어의 미생물학적 안전성과 저장성 향상시키기 위한 가공방법으로 이용될 수 있을 것으로 사료된다. This study was designed to evaluate the potential of using high pressure processing (HPP) for extending shelf life of seasoned squid during refrigerated storage. The vacuum-packed seasoned squid samples were subjected to 400 ㎫ for 20 min using a custom-made high pressure processor. Microbial counts, dimethylamine (DMA), trimethylamine (TMA), total biogenic amine, autolytic activity were determined on days 0, 7, 14, and 21 of refrigerated storage. The numbers of indigenous bacteria were effectively reduced by 2.77 log CFU/g after HPP treatment. The amounts of DMA and TMA produced in the control samples increased up to 15.99 and 42.82 ㎎/g after 7 days of refrigerated storage when compared to 5.27 and 10.21 ㎎/g the HPP-treated samples, respectively. The autolytic activity of the HPP-treated sample (4.32 nkat/g) significantly lower than that of the control (7.13 nkat/g) after 7 days of refrigerated storage. Therefore, HPP can be applied as a potential squid processing method microbiological safety and shelf life.

        감마선 조사 오렌지의 급성 및 아만성 독성 평가

        본 연구에서는 방사선 조사식품의 규정을 확립하고 안전성을 입증하기 위해 1 kGy 감마선 조사 오렌지를 ICR 마우스에 투여 또는 섭취시켜 급성 및 아만성 독성시험을 실시하였다. 급성독성의 경우 시험물질을 0, 1,000, 2,000 mg/kg의 용량으로 설정하여 단회 투여한 후 14일간 사망동물, 일반증상, 체중 변화, 부검소견, 혈액학적 및 혈액생화학적 검사를 수행한 결과 어떠한 이상도 관찰되지 않았으며, 통계학적으로도 유의적인 차이를 나타내지 않았다. 따라서 1 kGy 감마선 조사 오렌지의 대략의 치사량(approximate lethal dose)은 암수 ICR 마우스 모두 2,000 mg/kg을 상회하는 것으로 판명되었다. 아만성독성은 시험물질을 식이에 함유하도록 하여 3개월간 섭취시킨 후 사망동물, 일반증상, 체중변화, 사료 섭취량, 장기 무게, 조직학적 검사, 혈액학적 및 혈액생화학적 검사를 수행하였다. 시험기간 동안 암수 ICR 마우스 모두에서 사망동물이나 이상증상은 발견되지 않았으며, 체중 변화, 사료 섭취량 및 장기 무게에서도 통계학적으로 유의적인 차이를 보이지 않았다. 혈액학적 및 혈액생화학적 검사에서도 암수 ICR 마우스 모두 정상적인 수치를 나타냈다. 또한 조직학적 검사에서 간, 신장 조직은 모두 정상적인 구조를 유지하고 있었으며, 염증, 괴사 등의 유의할만한 병적 변화도 관찰되지 않았다. 따라서 1 kGy 감마선 조사 오렌지를 ICR 마우스에 3개월간 섭취시켜도 본 시험조건에서는 독성이 없는 것으로 판명되었다. The acute and subchronic toxicity of 1 kGy gamma-irradiated orange was evaluated in ICR mice. For acute toxicity, groups of 30 male and 30 female ICR mice were orally administered 1 kGy gamma-irradiated orange (0, 1,000, and 2,000 mg/kg). The mortality, clinical sign, body weight changes, and necropsy findings of ICR mice were observed for 14 days. No significant changes in body weight or abnormal gross findings were observed in relation to 1 kGy gamma-irradiated orange. Hematological and serum biochemical parameters were within normal ranges. According to the results, 1 kGy gamma-irradiated orange had no special toxic effects in male and female ICR mice at 2,000 mg/kg. For subchronic toxicity, groups of 36 male and 36 female ICR mice were given a diet of 1 kGy gamma-irradiated orange for 13 weeks (control, non-irradiated, and irradiated imported orange). During the experimental period, mortality, clinical signs, body weight change, food consumption, organ weight, and histopathological examination did not show any changes in comparison to the control group. Several hematological and serum biochemical parameters showed statistically significant changes, but these changes were within normal range. These results indicate that 1 kGy gamma-irradiated orange did not cause any toxic effects in male and female ICR mice and therefore can be considered as safe.

        해당화 뿌리 추출물의 면역 중진 효과

        The biological activities of extracts from Rosa rugosae Radix were compared. About 78% of the growth of human hepato-carcinoma and 68% of human gastric cancer cell was inhibited in adding 0.5% ㎎/ml of the extracts of Rosa rugosae Radix respectively. The growth of human breast cancer cells was also inhibited in adding 0.5㎎/ml of the extracts as well as 66% of the human cancer cells. It was proved that the growth of human normal lung cell, scored as 20% for the extracts. Overall selectivity of the extracts on several human cancer cell line was over 4, which in higher than those from the Rosa rugosae Radix. The growth of both human immune B and T cells was enhanced up to 1.2 to 1.5 times by adding the extracts, compared to the controls. The secretion of tumor necrosis factor-alpha(TNF-α) from T cell was also increased up to 61.9pg/ml in adding the ethanol extract (0.5㎎/ml). Ethanol extract also increased up to about 61.3pg/ml of interleukin-6(IL-6) from B cell.

        한국산 겨우살이 분획물의 면역세포의 생육증진 및 세포독성

        The biological activities on human immune and cancer cell lines of the four kinds of Korean mistletoes (Korean Viscum Album var. coloratum, : Korean Viscum sp. in Quercus acutissima Carr., Korean Viscum sp. in Castanea crenata, Korean Viscum sp. in Betula platyphylla, and Korean Viscum sp. in Salix Koreensis) extracts were investigated. the extracts were preparated with ethanol, and fractionated with n-butanol, ethyl acetate, chloroform, hexane, and second distilled water. Cytotoxic potencies of the fractions on human normal lung cell line (HEL 299) showed under 28% in the concentration of 0.5㎎/ml. Growth inhibition effect of the Korean mistletoe extracts on the several human cancer cell lines depends on the concentration of the extracts, and extracting solvent. The hexane, chloroform, and ethyl acetate fractions indicated a strong anticancer on the cancer cell lines. Stimulation on the growth of human immuno cell lines(B cell : Raji, T cell : Jurkat) of the extracts were confirmed in the ethyl acetate, chloroform, hexane fractions, but not in aqueous system.

        복분자 열매 추출물의 유용 생리활성 탐색

        The biological activities of extracts from Rubus coreanus Miq. ere compared, About 70% of the growth of human hepatocarcinoma and 79% of human gastric cancer call was inhibited in adding 1.0㎎/ml of the extracts of Rubus coreanus Miq. respectively. The growth of human breast cancer cells was also inhibited in adding 1.0 ㎎/ml of the extracts as well as 78% of the human cancer cells. It was proved that the growth of human normal lung cell, scored as 15% for the extracts. Overall selectivity of the extracts on several human cancer cell line was over 5, which is higher than those from the Rubus coreanus Miq. The growth of both human immune B and T cells was enhanced up to 1.4 to 1.8 times by adding the extracts, compared to the controls. The secretion of tumor necrosis factor-alpha(TNF-α) from T cell was also increased up to 78.8pg/ml in adding the ethanol extract (0.5 ㎎/ml). Ethanol extracts also increased up to about 70 pg/ml of interleukin-6(IL-6) from B cell. For screening regulate function of blood pressure, angiotensin converting enzyme(ACE) activity was inhibited up to 25% by adding ethanol extract (1.0 ㎎/ml). In testing the hypoglycemic activity, 20% of α-glucosidase activity was inhibited for the extracts (0.5 ㎎/ml). GST activity was increased in the range of 1.2 to 1.6 times by adding extracts.
