        Mix LPS-ELISA법을 이용한 살모넬라균 복합감염항체 동시 모니터링

        Salmonellosis caused by a number of serotypes of Salmonella is an infectious, acute or chronic, zoonotic disease and characterized by enteritis and diarrhea, septicemia in animal. In these studies we investigated the prevalent serotypes of Salmonella causing animal salmonellosis in Korea and the 71 strains of Salmonella spp. were isolated from materials such as mesenteric lymph nodes, fecal samples from slaughtered animal. With the identification test results, the most prevalent serotypes were, in order, S stanley 31 strains (43.7%), S typhimurium 19 strains (26.8%) and S montevideo 11 strains (15.5%), respectively. And we could establish the method for detection of antibodies to broad variety of Salmonella serotypes. Lipopolysaccharide(LPS) antigen extracted from Salmonella was more sensitive and specific than outer membrane protein antigen from that for detection of Salmonella antibody by using an indirect ELISA. The optimal concentration of antigen was 100ng/ml of LPS, the dilutions of conjugate and serum were 1 : 1,000~2,000 and 1 : 200~400, respectively. The mix LPS-ELISA which was used by mixing LPS from S typhimurium (group B), S choleraesuis (group C) and S enteritidis (group D) were more rapid and effective than that used LPS from individual strain for detection of Salmonella serogroup O4, O7 and O9 antibody at the same time. We could obtain the high values of optical density ($0.73{\pm}0.32$) by mix LPS-ELISA on the farm which had occurred salmonellosis, but very low values of $0.17{\pm}0.06$ on the negative farm of salmonellosis. So, the mix LPS-ELISA may be used to monitor the serological surveillance for the presence of infection with a number of serotypes of Salmonella and would be useful for prevention and control of salmonellosis in animal.

        국내 분리주 Salmonella gallinarum의 닭에 대한 병원성

        Fowl typhoid(FT) caused by Salmonella gallinarum is an infectious, egg-transmitted disease and characterized by swollen bronze liver, greenish-yellow diarrhea and high mortality in growing and adult chickens. Since 1992 the outbreak of FT has been increased. Several problems have been occurred such as absence of appropriate vaccines and lack of useful therapeutic methods. In these studies we investigated the pathogenicity of S gallinarum isolated in chickens. To compare the pathogenicity among the species of chickens, all chickens were challenged intramuscularly or orally with $1{\times}10^7$ CFU of S gallinarum. The brown-colored layers were more susceptible and white leghone chickens were more resistant than other species. In the brown layer chickens orally challenged, lethal doses ($LD_{50}$) of the isolates were inoculated at 1 day, 2 weeks, 4 weeks and 8 weeks old chickens with amount of $10^{4.2}$, $10^{4.7}$, $10^{7.0}$ and $10^{7.6}$ CFU, respectively. The chickens which were intramuscularly challenged with the less amount than $10^2$ CFU showed higher mortality than that of the chickens orally inoculated with same dose. Also, we investigated the recovery rates of bacteria from various organs of survival chickens which were challenged orally with $5{\times}l0^7$ CFU of S gallinarum. The bacteria was more frequently and isolated earlier from the liver and spleen than from any other ogans. In the pathogenicity test, the white-leghorn chickens which were known as resistant-strain against Salmonella were artificially immunosuppressed using bursectomy and/or dexamethasone treatment. Mortality of chickens with both bursectomized and treated with dexamethasone was higher(90%) than that of the control group(10%), the bursectomized chickens(10%) and the dexamethasone only treated group(20%). It was suggested that the protective mechanism in chickens against S gallinarum may be required both the functions of B-cells and T-cells.

        Salmonella gallinarum 분리주로부터 추출한 세포외막 단백질의 닭에 대한 면역원성

        Fowl typhoid caused by Salmonella gallinarum has increased dramatically since 1992 and has caused a great economic losses in chicken industry by characterizing with high mortality. In these studies, we investigated the immunogenicity and protectivity in chickens which were immunized with outer membrane protein(OMP) extracted from isolates of S gallinarum against challenge with live microorganism. Outer membrane proteins were composed of various sizes of molecular weight including 14K, 22K, 31K, 36K, 40K and 55K and the most of them responded strongly against rabbit antisera in immunoblot analysis. The chickens vaccinated with OMP or vaccinated with whole-cell combined with OMP($200{\mu}g$/chickens) complex showed higher delayed type hypersensitivity(DTH) response than that of whole-cell vaccinated group. The protective rates of OMP or whole-cell combined with OMP complex group against challenge of S gallinarum were higher (above 75%) than those (45~50%) of whole-cell vaccinated group. All vaccines were safe and the body weight-gains of all vaccinated groups were not significantly different (p<0.05) from those of nonvaccinated control group. In vitro tests, OMP stimulated both the proliferation of lymphocytes and T-lymphocytes, and OMP-induced lymphocyte proliferation was higher in the cells of the immunized chickens with OMP than in those from the control chickens.

        Immunoprophylatic effect of IgY against B. bronchispetica was proven with 100% preventive rate in mice administrated with IgY with antibody titer 1:640~1:2,560. Intramuscular administration was more efficient than oral administration. This phenomenon was also observed in the therapeutic effects of IgY after challenge with B. bronchseptica in mice. In the field trials with the egg yolk antibodies from hens immunized with combined antigens with B. bronchiseptica and parvovirus, curing rates in dogs with severe clnical signs such as bloody diarrhea were 81.6% and 86.7% by intramuscular or subcutaneous administration of IgY, respectively. Safety of the antibodies in dogs was proven without any side effects such as vomiting, edema, fever, etc. by adminstration of double doses for 7 days. These results indicated that the egg yolk antibodies could be used as effective prevention and treatment of alimentary and respiratory diseases in dogs.

        This study was carried out to produce IgY against B. bronchiseptica, parvovirus and distemper virus that are major pathogens in alimentary and/or respiratory diseases of dogs. In the comparison of adjuvants, ISA70 was the best in the rapid induction and maintence of antibody titers. Agglutination antibody titers against B. bronchiseptica were 1:1,280 ~ 1:10,240 in sera and 1:160 ~ 1:1,280 in egg yolk. Hemagglutination inhibition(HI) titers against parvovirus in sera and egg yolk were 1:80 ~ 1:320 and 1:64 ~ 1:256, respectively. Virus neutralization titers against canine distemper was 1:8 ~ 1:64 in sera and egg yolk. These results suggested that egg yolk antibody titers could be variable according to a sort of adjuvant and antigens of the pathogens.

        살모넬라 세포외막단백질 혼합백신을 이용한 돼지 및 닭에서의 살모넬라균감염증 방어효과

        We investigated the safety, immunogenicity and protectivity of mix-crude outer membrane protein (cOMP) vaccine against salmonellosis in animals. The mix-cOMP vaccine was extracted from Salmonella enterica serovar Typhimurium (ST) and Salmonella enterica serovar Enteritidis (SE) and Salmonella enterica serovar Braenderup (SB) isolated from pigs. The mix-cOMP vaccine gave significantly higher antibody response than ST-bacterin and ST-cOMP vaccine in guinea pigs. The survival rates of mix-cOMP vaccinated groups showed significantly higher (100%) than those (0-20%) of unvaccinated control group, challenged with 3 species of Salmonella (ST, SE and SB) in mice. Vaccinated groups in pigs showed reduction of clinical signs, increase of average weight gains, decrease of bacterial recovery rates, compared with unvaccinated groups. Especially, the survival rates (100%) of vaccinated groups in chickens showed higher than that (0%) of unvaccinated group. Based on these results, we suggest that the mix-cOMP Salmonella vaccine developed in this study will be effective for the protection against Salmonellosis caused by the various serotypes Salmonella species in animals.

        Salmonella Typhimurium의 돼지 호중구내 연속노출에 따른 특성변화

        To develop a live vaccine candidate using an attenuated strain of Salmonella Typhimurium (ST), biochemical properties, plasmid profile, PFGE patterns and pathogenic analysis of the ST isolate were carried out after sequential passage of the ST isolate in porcine neutrophils. By the passage, the ability of the neutrophil-adapted isolate to utilize d-xylose was lost, while the ability of the strain to ferment trehalose was delayed after 2 or more days of the culture. Also, changes including deletion of the gene fragments were observed in PFGE analysis of the neutrophil-adapted isolates. Two plasmids, 105kb and 50kb, were cured in the strain passaged over 15 times in porcine neutrophils. The 50% of lethal dose ($LD_{50}$) of the parent strain was changed from $1{\times}10^5\;LD_{50}$ to $6{\times}10^6\;LD_{50}$ by the passage in intraperitoneal injection of the strains into mice. These results suggested that bacterial genotypic and phenotypic responses might be globally altered depending on the inside environment of neutrophils.

        高壓下의 水熱反應에 依한 $CaO-SiO_2-H_2O$ 系 硬化體에 關한 基礎的 硏究

        It is a fundamental study for the hardened bodies of $CaO-SiO_2-H_2O$ system to clear up various physical properties and structures of the products, using the Seosan quartz and $Ca(OH)_2$(C. P. grade) as raw materials. Various samples were obtained by varying $CaO/SiO_2$ mole ratio (0.3∼2.1) and hydrothermal conditions ($100∼220^{\circ}C$ and 2∼14hr.) within the given limit. It was found that tobermorite phase as hydrate is contained in the hardened bodies and that the development of crystal has a great influence on the strengths and other physical properties of hardened bodies.

        금융시장 전염 동적 검정

        본 연구에서는 금융시장 통합화에 따른 금융 시장 전염을 생물학적 전염개념에 기초하여 분석하는 검정 방법론을 제시하였다. 금융 시장 통합화를 측정하기 위하여 U-통계량을 사용하였고, 금융 시장 전염 검정을 위하여 단일방정식 오차수정 모형을 중심으로 잠재 요인모형, 분위수 회귀모형과 런검정을 사용하였다. 시뮬레이션결과 단일방정식 오차수정 모형이 자기상관을 갖는 오차항을 포함한 선형 회귀모형에서 비교적 높은 수준의 적합도를 일관성 있게 보여 주고 있다. We propose methodology to analyze the dynamic mechanisms of financial market contagion under market integration using a biological contagion analytical approach. We employ U-statistic to measure market integration, and a dynamic model based on an error correction mechanism (single equation error correction model) and latent factor model to examine market contagion. We also use quantile regression and Wald-Wolfowitz runs test to test market contagion. This methodology is designed to effectively handle heteroscedasticity and correlated errors. Our simulation results show that the single equation error correction model fits well with the linear regression model with a stationary predictor and correlated errors.

        직업훈련 전문기업 ㈜지아이티아카데미의 환경대응 전략 메커니즘에 관한 연구

        국가 차원의 인적자원개발은 4차 산업혁명 시대와 펜데믹 이후 시대에 적합한 국가경쟁력 확보와 개인 발전을 위한 핵심 인력양성 국가 정책이다. 고용노동부가 주관하고 있는 직업능력개발사업은 연간 1조원이 넘는 예산을 투입하고 있고, 약 6,800개의 직업훈련을 직업훈련기관이 위탁운영하고 있다. 특히, 4차 산업혁명과 COVID19로 인하여 사회가 급변하고 있고, 국가와 사회는 다양한 사업의 혁신과 발전에 관심이 매우 높아지고 있다. 하지만 급변하는 환경에 적합한 인재를 양성하기 위한 직업능력개발 분야의 산업과 직업훈련기관에 대한 연구들은 미비한 상황이다. 본 연구는 직업훈련기관 중에서 국내 최대 규모의 훈련기관을 운영 중이며, 훈련참여자 점유율 1위를 차지하고 있고, 직영점 전체가 고용노동부 선정 우수훈련기관이며, 연매출액 300억 원 이상인 ㈜지아이티아카데미를 대상으로 기업의 성장과 발전전략에 대한 사례 연구를 진행하였다. 본 사례 연구에서는 외부환경의 변화와 정책적 변화에 따른 ㈜지아이티아카데미의 성장과 발전과정을 시기별 4단계로 구분하고, 성공과 발전전략을 메커니즘 관점에서 분석하였다. 주체의 강력한 비전과 함께 뉴노멀 시대의 새로운 환경을 창출하는 창조적 메커니즘을 기반으로 ㈜지아이티아카데미는 다시 발전과 긍정적인 변화를 만들어 가고 있다. 메커니즘 기반 관점에서 환경창조(S-E-R)-환경혁신(R-S-E)-자원혁신(E-R-S)-환경창조(S-E-R)메커니즘 유형으로의 성공적인 변화 과정을 이어가고 있다. ㈜지아이티아카데미의 메커니즘 순열을 기반으로 나타난 기업의 성장과 발전과정은 경영자들에게 메커니즘 순열의 구축과 체계적인 전략 실행에 대한 중요한 시사점을 줄 것이다. 본 연구는 직업훈련기관에 대해 성장과 발전전략을 메커니즘 관점에서 살펴본 연구로서 직업훈련기관의 성장과 발전과정을 경영전략과 노하우 축적 과정의 특성, 정책적 기회를 활용하는 경영전략을 중점적으로 분석하여 직업훈련기관의 육성을 위한 성공 전략과 직업훈련 정책에 새로운 정책적 관점을 제시할 것이다. Human resource development at the national level is a national policy for securing national competitiveness suitable for the era of the 4th industrial revolution and post-pandemic era and fostering key human resources for individual development. The vocational competency development project organized by the Ministry of Employment and Labor invests more than 1 trillion won a year, and about 6,800 vocational training institutions are commissioned and operated. In particular, society is rapidly changing due to the 4th Industrial Revolution and COVID-19, and the national and society are very interested in innovation and development of various projects. However, studies on management strategies in the field of vocational skills development industry and vocational training institutions to develop talented people who are suitable for a rapidly changing environment are insufficient. This study will present a success strategy for developing vocational training institutions and a new policy perspective on vocational training policy through case studies on corporate growth and development strategies about GIT Academy Co., Ltd. operating the largest training institution in Korea among vocational training institutions, ranked first in training participants market share, having more than 30 billion won in annual sales and more than 200 full-time workers, with the entire direct management store being an excellent training institution selected by the Ministry of Employment and Labor. This study shows the growth and the development process of GIT Academy according to changes in the external environment and policy changes in four stages by period and analyzes the success and development strategies using a mechanism perspective. Based on the creative mechanism that creates a new environment in the New Normal era with the strong vision, GIT Academy is again developing and making positive changes. As a mechanism perspective, the process of successful change is continuing to environmental creation(S-E-R)-environmental innovation(R-S-E)-resource innovation(E-R-S)-environmental creation(S-E-R) mechanism type. The growth and development process of a company based on the mechanism permutation of GIT Academy will have important implications to CEO what kind of mechanism permutation should be construct and what efforts should be made to. This study examines growth and development strategies for vocational training institutions using a mechanism perspective, It focuses on analyzing the growth and development process of vocational training institutions, the characteristics of the management strategy and know-how accumulation process, and management strategies that utilize policy opportunities and it will provide a practical perspective from a strategic aspect.
