Objectives : Portulaca oleracea (PO) have been used as a traditional medicine to treat inflammatory diseases in Korea. However, the anti-inflammatory effect of PO ethanol extract on lipopolysaccharide (LPS)-induced inflammation is not well-known. Therefore, this study was performed to identify the anti-inflammatory effect of PO on LPS induced inflammatory. Methods : Identification of PO was conducted by comparison with purified standards by HPLC. To measure out the cytotoxicity of PO, author performed the MTT assay. To evaluate the anti-inflammatory effects of PO, author examined the inflammatory mediators such as nitric oxide (NO) and pro-inflammatory cytokines (tumor necrosis factor (TNF)-α, interleukin, (IL)-1β and IL-6) on RAW 264.7 cells. Author also examined molecular mechanisms such as mitogen-activated protein kinases (MAPKs) and nuclear factor-B (NF-κB) activation by western blot. Results : Three major components (peaks 1, 2, 3) were detected in both varieties and peak 1 was characterized as caffeic acid, peak 2 as p-coumaric acid, and peak 3 as ferulic acid by comparison of chromatographic properties with authentic standards. Extract from PO itself did not have any cytotoxic effect in RAW 264.7 cells. PO inhibited LPS-induced productions of inflammatory mediators such as NO and pro-inflammatory cytokines in RAW 264.7cells. In addition, PO inhibited the phosphorylation of extracellular signal-regulated kinase1/2 (ERK1/2), c-Jun NH2-terminal kinase (JNK) and NF-κB activation in RAW 264.7 cells. Conclusions : Above experiment data can be an important indicator for the identification of PO and this study suggest that treatment of PO could reduce the LPS-induced inflammation. Thereby, PO could be used as a protective agent against inflammation.

Objective : This study was performed to estimate the effects of OMC-2010 extract on cytokine production in mouse spleen cells. Methods : Mouse spleen cells were pre-treated with ethanol and water extract of OMC-2010 for 1 h, then stimulated with lipopolysaccharide (LPS, 1 μg/ml) for 48 h. Then the cells were harvested for real-time reverse transcription polymerase chain reaction to detect cytokines. Results : OMC-2010 ethanol extract significantly inhibited the LPS-induced interleukin (IL)-1beta, tumor necrosis factor (TNF)-alpha and IL-5 mRNA expressions, but not shown such changes in IL-6, IL-4, IL-13. OMC-2010 water extract significantly inhibited the LPS-induced TNF-alpha, and IL-5 mRNA expressions, but not shown such changes in IL-1beta, IL-6, IL-4, IL-13. Conclusions : Theses results could suggest that both ethanol and water OMC-2010 extract could inhibit the TNF-alpha and IL-5 mRNA expression.
본 연구는 하이힐을 신은 기간이 하지 근 활성화에 미치는 영향을 알아보기 위해 수행되었다. 대상자는 항공서비스학과에 재학 중인 여학생 34명이었으며 2그룹으로 분류되었다 : 대조군 - 16명으로 하이힐을 한 번도 신지 않은 자, 실험군 - 18명으로 하이힐을 1년간 꾸준히 신은 자. 근 활성화는 EMG를 사용하여 앞정강근, 장딴지근, 넙다리곧은근, 척추세움근에 패드를 붙이고 맨발로 걸을 때와 하이힐을 착용하고 걸을 때 측정되었다. 연구결과, 평지 걷기에서는 대조군과 실험군 사이에 통계학적으로 유의한 차이가 없었다. 계단 오르기에서 하이힐을 신었을 때 앞정강근(p<.05)과 장딴지근(p<.001)에서 실험군에 비해 대조군에서 통계학적으로 유의하게 근 활성화가 증가하였다. 계단 내려오기에서 맨발일 때 척추세움근(p<.05)에서 대조군에 비해 실험군에서 통계학적으로 유의하게 근 활성화가 증가하였다. 계단 내려오기에서 하이힐을 착용했을 때 앞정강근(p<.01)과 장딴지근(p<.05)에서 실험군에 비해 대조군에서 통계학적으로 유의하게 근 활성화가 증가하였다. 본 연구의 결과를 통해 하이힐을 처음 신는 경우가 하이힐을 1년 동안 신은 경우보다 보행 시 하지 근 활성화에 영향을 미친다는 것을 알 수 있었다. The purpose of this study to determine the effect of the period wearing high-heeled shoes on muscle activity of lower extremity during walking. The subjects of this study were 34 female students attending aviation services department and divided into two groups : control group – sixteen students in do not wear high-heeled shoes, experiment group – eighteen students in do wear high-heeled shoes. To achieve this purpose, muscle activity was measured when walking barefoot and wearing high-heeled shoes with EMG pad attached on tibialis anterior, gastrocnemius, rectus femoris, elector spinae. When flat-ground walking there was no significant difference muscle activity between control and experiment group. When going up stairs wearing high-heeled shoes, tibialis anterior(p<.05) and gastrocnemius(p<.001) muscle activity of control group was significantly increased than experiment group. When going down stairs barefoot, elector spinae(p<.05) muscle activity of experiment group was significantly increased than control group. When going down stairs wearing high-heeled shoes, tibialis anterior(p<.01) and gastrocnemius(p<.05) muscle activity of control group was significantly increased than experiment group. The results found that wearing high-heeled shoes for the first time affects the lower extremity muscles activation during walking than wearing high-heeled shoes for one year.
