        Anti-IL-1beta 단일클론 항체를 이용해서 발열환자의 뇨중 IL-1beta inhibitor의 확인

        To effectively purify of IL-1 inhibitor from human febrile urine, we have established monoclonal antibody that reacts with human recombinant interleukin 1beta(IL-1beta). The antibody, designated ON-1, was highly specific to IL-1beta and no cross-reaction with other cytokines(IL-1alpha and IL-4) was observed. As the results of ELISA inhibition assay and Western blotting method, it was further identified that ON-1 had high binding specificity with IL-1beta. IL-1 receptor binding material from febrile patient urine was effectively purified with affinitv column chromatography which conjugated with ON-1. This urinary material inhibited the thymocyte proliferation in a dose-dependent manner. IL-1beta induced thymocyte proliferation activity was inhibited to 67.3% at 6mcg of the purified urinary material. The result may suggest that this urinary material the purified urinary material. The result may suggest that this urinary material will have antagonic effect on IL-1 action mechanism and act IL-1beta inhibitor.

        온라인 쇼핑몰의 재이용 의도에 영향을 미치는 요인

        The use of online shopping mall has been increasing rapidly, which reflects the convenience and economic benefits associated with its usage. As with traditional offline businesses. internet businesses have to manage customer relations to provide better services in order to ensure their business sustainability. In this regard. identifying the influencing factors to reuse intention of online shopping mal l is very important. In this paper. we aim to examine the effects of perceived and individual variables on intention to reuse in the context of online shopping mall. In order to ana lyze the antecedents, we selected perceived and individual factors such as shopping enjoyment, concentration. perceived variety and maximizing tendency. For empirical analysis. we surveyed real users of on line shopping mall. As a result, all antecedents showed significant influence on intention to reuse except perceived variety and among the variables the concentration has top standardized coefficient. In addition. perceived variety has significant effect on shopping enjoyment but not on concentration. Maximizing tendency is related to shopping enjoyment and concentration significantly. The results of this research can be useful guidelines for online shopping mall providers.

        개인화 서비스의 수용에 있어서 인지된 개인화와 이해의 역할

        Many online companies have installed various tools enabling them to provide personalized offerings and unique experiences to individual users based on their personal information. By offering a higher level of personalization with greater accuracy and on a wider range of product or service attributes, the companies are trying to improve their value propositions for their consumers, and accordingly increase profitability. However, linking personalization to profits can be challenging, if the firms do not understand psychological factors and subjective attributions that matter to the users when they consider the adoption of the personalization. For example, recent studies based on behavioral decision making have shown that customers perceive the level of fit between their preferences and personalized offerings differently even when the same personalized offerings are provided, depending on their involvement in the personalization process. Also it has been found that when a customer perceives a higher level of fit for the same personalized offering, she has a higher willingness to pay for the offering. These imply that there is a lack of congruency between the actual level of personalization and users` perception and valuation of the personalization. Therefore, users` subjective perception regarding personalization may play a critical role in the adoption of personalization services. To investigate the role of users` psychological factors in the adoption of personalization, this article theoretically develops a conceptual model of personalization adoption with a focus on two psychological factors, perceived personalization and understanding of personalization. Grounded on SOR (Stimulus-Organism-Response) model and schema theory adapted to the peculiar features of personalization, we formulate six hypotheses describing the underlying mechanism of personalization adoption as follows. First, we identify the mediating role of perceived personalization in the relationship between actual level of personalization and intention to adopt personalization (Hypotheses 1 through 3). According to the SOR paradigm, an external stimulus (S) to the individual is transformed with an internal process (O), and both S and O (or only O sometimes) elicit(s) a response (R). By applying the paradigm into the case of personalization service adoption, the influence of actual level of personalization (S) on intention to adopt the personalization (R) is expected to be mediated by perceived personalization (O). Second, based on the schema theory and the literature on intelligent information systems, we hypothesize that valid and relevant explanation from a personalization system on the process and results of personalization can facilitate the user`s understanding of personalization (Hypothesis 4), which, in turn, can increase the user`s intention to adopt the personalization as well as her perceived personalization (Hypotheses 5 and 6). The proposed model is tested using data gathered from 211 Internet users through an online experiment. Each participant was randomly assigned to one of four recommendation systems on TV, which were generated based on the combination of two manipulation factors: the actual level of personalization (high vs. low) and the provision of explanations on the process and results of recommendation (provided vs. not provided). The two systems with the higher level of personalization recommended three TV models out of sixty-four models to each participant, based on the participant`s responses to eight questions regarding his or her lifestyle. On the other hand, thirty-two TV models and four questions were used for recommendations in the other two systems with the lower level of personalization. Independently of the personalization level, explanation about personalization process and results was provided in the two systems while it was not offered in the remaining two systems.         Streptomyces 배양액에서의 Interleukin-1 유사물질의 동정

        Streptomyces의 배앙액이 쥐의 흉선세포(thymocyte)를 활성화시켰는데 그것은 농도에 의존적이었고 receptor binding 분석법에서는 C_3H/HeJ의 흉선세포에 있는 IL-1 receptor에 IL-1β-FITC의 결합을 경쟁적으로 억제하였다. Streptomyces의 배앙액에 있는 단백질을 분리하기 위해서 ultrogel AcA44 column chromatography를 하여 흉선세포의 증식이 제일 많은 분획을 모아서 FPLC용 column인 superose 12에 loading한 결과 흉선세포의 증식이 제일 높은 분획은 21번 분획이었다. Affinity chemical cross-linking 실험에서 미생물 배양액의 활성인자는 IL-1 receptor를 통하여 흉선세포의 활성을 유발하는 것으로 생각된다. 미생물 배양액에서 분리정제된 단백질은 anti-rIL-1β 항체와 반응하며, western blotting 결과 그 분자량은 약 21 kd으로 추정되었다. We have identified a T cell-activating material in the culture supernatant of Streptomyces species. The factor in microbial culture supernatant (MCS) induced thymocyte proliferation in a dose dependent fashion and it could be detected by immunoblot analysis using anti-interleukin1(IL-1) antibody. The factor in MCS was slightly larger(about 21 kd) in its molecular weight than IL-1 on SDS-PAGE. When ^125I-MCS was covalently coupled with homo-bifunctional cross-linking agent, disuccinimidyl-propionate to IL-1 receptor(IL-1R) on mouse thymoma cell(EL-4) and immunoprecipitated with anti-IL-1R antibody the molecular weight of this complex of 110 kd was observed. Interaction between IL-1 receptor and the factor in MCS could be inhibited by addition of excess IL- lβ. These results suggest that Streptomyces species may produce a factor which is closely related to IL-1β in its functional and biochemical characteristics.
