유방염 치료제로부터 유래되어 우유에 내재하는 항생물질 검출방법에 있어서 각 유가공업체에서는 실험시간이 오래 소요되는 (약 3시간) 진부한 TTC(2,3,5-triphenyl-2H-tetrazoli-um chloride)시험방법을 아직도 사용하고 있는 실정이다. 그래서 본 실험에서는 이의 단점을 보완하여 발효유제품과 우유의 품질향상, 생산 효율성 증가 등을 위하여 TTC 시험방법과 최근에 발전된 속성검출방법(β-lactam screening test, sulfamethazine screening test)의 비교시험을 실시하였으며 그 결과는 다음과 같다. 1. β-Lactam 계 항생물질 속성검출법을 사용한 경우 실험시간은 단지 15분이 소모되었고 penicillin G는 TTC 방법보다 10배 낮은 농도인 0.003 IU/㎖까지 검출되었고 ampicillin은 TTC방법의 검출 농도보다 500배 낮은 농도인 1.0×10^(-10) g/㎖까지 검출되었다. 2. TTC 시험방법의 경우 penicillin G는 0.03IU/㎖, ampicillin은 5.0×10^(-10) g/㎖ 농도까지 검출이 가능하였고 sulfamethazine은 1.0×10^-3 g/㎖ 농도 이상만 검출되었다. 3. Sulfamethazine 속성 검출법의 경우 sulfamethazine은 1.0×10^(-10) g/㎖까지 신속 정확하게 검출되었다. 4. 결과적으로 이들 속성검출법은 신속성(fastness), 특이성(specificity), 민감도(sensitivity), 정확도(precision), 간편함(easy handling), 그리고 경제성(inexpense)등의 장점들이 있음이 실험결과 나타났다. Dairy industry in Korea has been using time-consuming and insensitive method(TTC test) for the detection of antibiotics in milk derived from mastitis treatment. So this study was carried out to improve detection methods in raw and pasteurized milk. We compared rapid screening tests(β-lactam screening test and sulfamethazine screening test) and TTC test for quality improvement, increase of productivity in milk and fermented dairy products. The results obtained are summarized as follows : 1. In β-lactam screening test, it took only 15 mins per 5 samples and was detceted 0.003 IU/㎖ of penicillin G and 1.0×10^(-10)g/㎖ of ampicillin in reconstituted skim milk. 2. In TTC test, it took at least 3 hrs and was detected 0.03 IU/㎖ of ampicillin and more than 1.0×10^(-3)g/㎖ of sulfamethazine. 3. In sulfamethazine screening test, 1.0×10^(-10)g/㎖ of sulfamethazine was detected but almost wasn't detected in TTC test. 4. In conclusions, this study showed some advantages such as fastness, specificity, sensitivity, precision, easy handling and inexpensiveness with rapid screening tests.
