Actomyosin $Ca^{++}$ Activated Adenosinetriphosphatase 활성도에 대한 pH 및 온도의 영향

The activity of the $Ca^{++}$ activated adenosinetriphosphatase (Ca-ATPase) of actomyosin systeme of rabbit and frog skeletal muscle has been studied at varying pH and temperature. The PH optima of the Ca-ATPase activity of the rabbit actomyosin was rather broad. Over the temperature range of $16-36^{\circ}C$ activity of the enzyme was not appreciably changed between pH 6.4-8.5; below and above which it rapidly reduced. The pH at the inflection point of the enzyme activity increased as temperature decreased, showing the ${\bigtriangleup}pH\;inflection/{\bigtriangleup}T$ of approximately $-0.018\;unit/^{\circ}C$. Consequently, $(OH^-)/(H^+)$ ratio at the inflection point was constant regardless of assay temperature. In the frog actomyosin systems the Ca-ATPase activity was not apparently altered between PH 6.4-7.0 when the incubation temperature was $15{\sim}30^{\circ}C$. Outside of this range of pH, however, the enzyme activity was dramatically decreased. The pH of the inflection point changed inversely with temperature. ${\bigtriangleup}pH\;inflection/{\bigtriangleup}T$ at the acidic side was approximately $-0.018\;unit/^{\circ}C$, whereas that at the alkaline side it was about $-0.037\;unit/^{\circ}C$. The Arrhenius Plot on the Ca-ATPase activity at constant $(OH^-)/(H^+)$ ratio of 1.0 was not linear, but showed break at arround $20^{\circ}C$ for both rabbit and frog actomyosin Preparations. From these results it was speculated that pH dependence of Ca-ATPase activity of rabbit actomyosin systems might reflect titrations of histidine-imidazole and -SH groups, and that of the frog actomyosin represents titrations of histidine-imidazole and lysyllysine ${\alpha}-NH_2$ groups.
CO<sub>2</sub> laser조사가 성견치수에 미치는 영향에 관한 실험적 연구

The object of this paper was to investigate the histopatological changes on dog's pulp under cavitation by irradiation of the $CO_2$ laser. The subjects were derived from four dogs, and irradiated 113.23 J/$mm^2$, 283.09 J/$mm^2$, 566.08 J/$mm^2$ in Group I, II, and III respectively. The dogs were sacrificed immediately, 24 hour, 72 hour and 1 week after $CO_2$ laser treatment. For light microscopic examination, routine H-E and PAS stains were employed. For electron microscopic observation, the teeth were fixed in 1% paraformaldehyde and 1% glutaraldehyde, decalcified teeth in 10% EDTA were stained by uranyl acetate and lead citrate. The observation was made with a Hitachi H-500 model electron microscope. The following results were obtained in this study: 1. At the early stage of the experimental sub-groups-immediately, 24 hour, 72 hour samples of Group I, II and III-coagulation necrosis and hyperemia were observed in odontoblastic and subodontoblastic pulpal layer. 2. At the 1 week sub-group of Group I, II, regenerative hyperplasia of the odontoblasts without coagulation necrosis were revealed, in addition to thickened predentin. On he other hand coagulation necrosis and atrophic change accompanying with hyperplasia were found at the 1 week sub-group of Group III. 3. Ultrastructurally, the odontoblasts appeared nuclear degeneration, vacuolar change of cytoplasmic organelles and rupture of plasma membrane at the early stage of the experimental period of all groups. 4. Under spectrohelioscopic examination, regenerative odontobalsts were seen at the 1 week specimens of Group I, II and III. 5. The pulpal response occured at 113-566 J/$mm^2$. The pathologic change of pulp tissue occured at the early experimental period but regeneration of odontoblasts could be seen after 1 week.

The structural members under seismic loading actually show inelastic behavior, so the inelastic responses should be calculated for the seismic design of structures or estimating the structural damage level. Although direct time history analysis may calculate the exact dynamic nonlinear responses for given ground motions, this approach involves a high computational cost and long period. Therefore, it should be developed the approach to estimate nonlinear responses for the practical purpose. The artificial earthquake accelerograms were generated to obtain the smoothed responses spectra, and the samples of generated accelerogram for each seismic event was used to examine average nonlinear response spectra. The stabilized response spectra for each earthquake event was used to evaluate the effects of various yield strength ratios, damping values and nonlinear hysteretic models. The approach, which can simply predict the nonlinear seismic responses of structures, was shown in this study.
입자성 유기물의 안정동위원소비를 이용한 팔당호 수계내의 유기물 기원 연구

Organic carbon and nitrogen stable isotope ratios of particulate organic matter (POM) were determined at 7 stations (6 stations in Han river and Paldang dam outflow station) from May to October 2013, in order to understand the origin of POM at the Han river. δ<sup>13</sup>C values of POM in 7 stations were no significant changes season, but δ<sup>15</sup>N values were enriched in around Kyeongan stream (K). POC, PN and Chl-a concentration showed a similar seasonal pattern in Kyeongan stream, with an apparent decrease from July to August. POC and PN concentration has a higher correlation with Chl-a concentration in Kyeongan stream (K). δ<sup>13</sup>C and δ<sup>15</sup>N values of POM has a lighter value during lower Chl-a concentration period, compared to other seasons. Our results revealed that Kyeongan stream (K) seemed to be influenced by substantial amount of organic manure or fertilizer input in 2013, compared to the previous year (2012). These results suggest that the analysis of stable isotope ratios is a simple but useful tool for the identification of organic matter origin in aquatic environments.

N/A Background:Autoantibodies of heterogeneous specificities can be found in patients with SLE. Objective:We planned to examine the presence and frequency of anti-epidermal protein autoantibodies among patients with systemic lupus erythematosus (SLE). Patients:Thirty consecutive patients of SLE seen at the rheumatology section with no blistering lesions, who showed positive serological findings for anti-dsDNA antibody and high titers of antinuclear antibodies, were enrolled in this study. Methods:Indirect immunofluorescence study with normal human skin substrates and immunoblot assay using antigens obtained from A431 cell-extracts were performed with SLE sera. Results:Among the 30 patients of SLE, 2 had IgG autoantibodies that bound along the basement membrane zone of the skin in IIF. In the immunoblot assay, IgG bands reacting to 230kD and/or 180kD antigens were observed in 3 cases among the 30 SLE sera; additional IgA band reactivities against 230kD and 180kD were also seen in one of the above 3. Besides these autoantibodies, they had no other autoantibodies reacting to epidermal antigens. Conclusion:In this study, the frequency of serum autoantibodies to the epidermal proteins (bullous pemphigoid antigens and so on) among the patients with non-bullous SLE was estimated to be 10%-positivity. This result suggests that some patients with SLE may have autoantibodies to the epidermal basement membrane zone antigens with no pathogenecity. However, possible clinical significances should further be clarified. (Korean J Dermatol 2000;38(4):457~461)
