The effect of protein constituents of six-year old fresh Panax ginseng root on chick embryonic brain, spinal cord and skeletal muscle dissociation cultures was studied. The protein constituents showed the enhancing effect on cultured brain, spinal cord and skeletal muscle cells. The neurite formation from brain and spinal cord cells and the outgrowth of neurite seemed to be enhanced by almost all of the protein constituents employed for this study. The maturation of skeletal muscle cells was stimulated by the protein constituents. This enhancing effect of the protein constituents was more vivid when brain, spinal cord and skeletal muscle cells were cultured with a medium which did not contain chick embryonic extracts known as an essential component for primary cell culture. The protein fraction having molecular weight range of 1,000 to 5,000 out of all the protein fractions employed for this study showed the most stimulatory effect on cultured brain, spinal cord and skeletal muscle cells.

Mice were fed on normal diet(18% casein) or protein deficient diet(5% casein) with supplementation of the total saponin fraction of Panax ginseng for 2weeks, 4weeks and 8weeks, respecaively. The amounts of norepinephrine in the mouse brain were increased by the supplementation of the total saponin fraction of Panax ginseng in both normal and protein deficient diet fed groups. The amounts of dopamine in the mouse brain were only increased in the mouse fed with normal diet supplemented with the total saponin fraction of Panax ginseng for 8weeks. Norepinephrinergic neurons were more influenced than dopaminergic neurons by Panax ginseng and this influence was more amplified in the case of protein deficient diet.

The changes of thymus weight, spleen weight, total number of peritoneal exudate cells, contents of total serum protein and albumin were measured and the change on the electrophoregram pattern of serum protein was observed in order to study on the effects of the saponin fraction of Panax ginseng as a tonic on the immune system of the mouse fed with protein deficient diet. The involution of the thymus of the mouse was induced by feeding protein deficient diet and the saponin fraction on the thymus weight was diminished after four weeks. The saponin fraction of Panax ginseng showed no effect on the spleen weight of the mouse. The supplement of the saponin fraction enhanced total number of peritoneal exudate cells and contents of total serum protein and albumin of the mouse, and overcame the effects caused by protein deficiency. The electrophoregram of serum protein of the mouse fed with protein deficient diet was different from that of the mouse fed with normal diet, but this difference seemed to be overcome by the supplement of the saponin fraction of Panax ginseng.

The effect of saponin fractions of Panax ginseng root on the interactions of human plasma lipoproteins and lecithin dispersions with dextran sulfate were studied in order to examine the effect of Panax ginseng on the lipid accumulation in the aorta. The total saponin fraction and protopanaxadiol glycosides of Panax ginseng root seemed to slightly enhance the interaction of low density lipoproteins with dextran sulfate in the absence of divalent metal ions. Protopanaxatriol glycosides remarkably inhibited the interaction of low density lipoproteins with dextran sulfate. However, all of these three saponin fractions of Panax ginseng root showed the tendency of inhibition to the interaction of high density lipoproteins with dextran sulfate in the presence of divalent metal ions by the order of protopanxatriol glycosides, protopanaxadiol glycosides and total saponin. Three saponin fractions of Panax ginseng exerted almost same tendency to the interaction of lecithin dispersions with dextran sulfate in the presence of divalent metal ions as the interaction of low density lipoproteins with dextran sulfate absence of divalent metal ions.

Atherosclerosis is associatod with the presence of extracellular lipid droplets and large fatty deposits, both of which are to be covered at the surface mainly by zwitterionic phospholipids. The development of atherosclerosis is often associated with the accumulation of calcium. Furthermore, the presence of glycosaminoglycans directly underlying fatty deposits in human aorta has been demonstrated. Also, the possible involvement of the interaction between sulfated polysaccharide and lipoprotein in the development of atherosclerosis has been suggested in view of the presence of both low density lipoproteins and glycosaminoglycans, as well as their complexes, in atherosclerotic aortas. Therefore interactions of sulfated polysaccharides with low density lipoproteins which serve as a vehicle for cholesterol and cholesterol ester and with zwitterionic phospholipids have been studied extensively by a number of workers to provide mechanisms. In this paper, the mechanism of the interaction of sulfated polysaccharides with low density lipoproteins and the mechanism of the interaction between sulfated polysaccharides and zwitterionic phospholipids are reviewed. The possibility of the occurence of these interactions in the body are also considered.
한국산 백삼과 수삼의 Polyamine 성분에 관한 연구

Polyamine constituents of fresh Panax ginseng root and dried white ginseng were extracted with 5% trichloroacetic acid, respectively. The isolation of polyamine constituents was conducted by the cation exchange chromatography using Dowex-50W×8 resin and the detection was performed with TLC. Identification of the polyamine was carried out by the methods of IR, NMR, MS spectroscopy and GLC. Polyamine constituents of white and fresh ginseng root were identical and composed of five different polyamines. The major polyamine in white and fresh ginseng root was determined as putrescine.

Effects of water extracts of Torilis Fructus, Rubus hructus, Schizandrae Fructus, Lycii Fructus and Cuscutae Semen on the immune system of ICR mice fed with normal diet and fed with protein deficient were investigated. In control group, body weight, the ratio of spleen per body weight, the ratio of thymus per body weight, peritoneal exudate cell number, total serum protein and albumin content were decreased in case of mice fed with protein deficient diet as compared to the mice fed with normal diet. Without regard to kinds of diet, water extracts of Torilis Fructus, Rubus the ratio of spleen per body weight and total serum protein and albumin content. Characteristic electrophoresis patterns of serum protein were obtained according to kinds of diet and sample.

Effects of the methanol extract of Bupleuri Radix on primary cultured. chicken embryonic brain cells, dorsal root ganglia (DRG) and rat hepatocytes were studied. The methanol extract of Bupleuri Radix at the concentration ranging from 10 ㎍/㎖ to 100 ㎍/㎖ significantly recovered the cytotoxicity of rat hepatocytes induced by the treatment of galactosamine; at the concentration of 100 ㎍/㎖, values of GOT and GPT in the culture medium were reduced by the 60% and 75%, respectively of those in the absence of the methanol extract of Bupleuri Radix. The addition of the methanol extract of Bupleuri Radix, into chicken embryonic brain cells which were cultured with a deficient medium significantly increased the number of cells promoting the neurite outgrowth. However, the methanol extract of Bupleuri Radix showed no effect on the activities of PDHC and acetylcholinesterase in primary cultured brain cells.

Effects of betaine, a major component of Lycii Fructus, on carbon tetrachloride intoxicated rats were evaluated. Histological studies showed that betaine had improved the typical necrosis around centrilobular area in liver tissue due to the carbon tetrachloride intoxication. Betaine, whether it was administrated simultaneously or prior to carbon tetrachloride, prevented or retarded the elevation of liver-weight/body-weight ratio due to the carbon tetrachloride intoxication. Betaine also significantly elevated the reduced activities of some enzymes, cytochrome P-450, 7-ethoxycoumarin-O-deethylase and glutathione-S-transferase, involved in xenobiotic metabolism due to carbon tetrachloride intoxication. These results demonstrate a possible hepato-protective role of betaine against fatty liver that could be easily induced by carbon tetrachloride in rat.
