        선학초(仙鶴草)가 선조체내출혈(線條體內出血) 흰쥐의 뇌조직 손상에 미치는 영향

        Objects: This study was performed in order to evaluate the effects of Agrimoniae herba (AH) ethanol extract on intrastriatal hemorrhage (ISH). Method: ISH was induced by the stereotaxic intrastriatal injection of bacterial collagenase type IV in Sprague-Dawley rats. AH was orally given once a day for 3 days after ISH. Hematoma volume and percentage edema were examined. Immunohistochemistry was processed for iNOS, c-Fos, MMP-9, and MMP-12 expressions in the brain sections and each immuno-labeling were calculated with image analysis. Results: results are as follows; 1. AH reduced the hematoma volume and percentage edema of the ISH-induced rat brain. 2. AH swollen apoptotic bodies and neurons in the peri-hematoma regions of the ISH-induced rat brain. 3. AH significantly reduced c-Fos, MMP-9 and MMP-12 positive cells in the peri-hematoma regions of the ISH-induced rat brain. 4. AH swollen iNOS expressions in the peri-hematoma regions of the ISH-induced rat brain. Conclusion: These results suggest that AH plays an anti-apoptotic neuroprotective effect through control of ISH, suppression of c-Fos, and down-regulation of MMP-9 and MMP-12 expressions in the brain tissues.

        퇴예명목(退翳明目) 약재에 관한 문헌적 고찰과 활용연구

        Objectives : This study was planned to help clinical treatment by examining the efficacy and frequency of use of medicines with removing nebula and improving vision action, centering on historical literature. Methods : Chinese literature centered on CNKI published after 2000 and ophthalmic books of the Beijing University of Chinese Medicine Library, regardless of the publication period. Domestic literature was searched using NDSL and OASIS, and literature published after 2000 was referenced. Results : According to traditional medical classics, as medicine with removing nebula and improving vision action, medicine with dispelling wind and clearing heat action accounted for the majority. However, it is desirable to use these medicine in the early stages of the disease. Conclusions : 1. Among the external contacts that induce corneal opacity, the wind-heat pattern was the most common. Among the visceral manifestation patterns that induce corneal opacity, liver or spleen was the most common. Cold-tempered medicine which is dispelling wind, clearing heat and bottling up the liver were most commonly used as therapeutic medicine. 2. As removing nebula and improving vision medicine, Cassiae Semen, Chrysanthmi Flos, Eriocauli Flos, and Buddlejae Flos have been widely used. 3. It has been investigated that removing nebula and improving vision medicine can be effectively used in the early stages of various diseases that cause corneal opacity.

        계지작약지모탕(桂枝芍藥知母湯)이 Acetic Acid에 의한 통증에 미치는 영향

        Objectives : We want to know the effect of Gyejakjimo-tang in mice model of acute pain.Methods : We investigated writhing reflex in mice with acetic acid-induced abdominal pain using mice, we observed c-Fos protien expression by immunohistochemistry dyeing method in the paraventricular nucleus(PVN) and supraoptic nucleus(SON) of the hypothalamus.Results : All of Gyejakjimo-tang treated group suppressed acetic acid-induced writhing response as acetic acid injuction group, but in 100㎎/㎏ Gyejakjimo-tang treated group and 200㎎/㎏ Gyejakjimo-tang treated groups represernted significance. All of Gyejakjimo-tang treated groups(50, 100 and 200㎎/㎏ Gyejakjimo-tang-treated group), Fos-positive cells in PNV significantly decreased as acetic acid injuction group, and All of Gyejakjimo-tang-treated groups Fos-positive cells in SON significantly decreased as acetic acid injuction group.Conclusions : The present results showed that the mice pre-treated with the aqueous extract of Gyejakjimo-tang showed analgesic effect on acetic acid-induced abdominal pain.

        행인(杏仁)이 전립선 암세포의 Bax, Bcl-2 및 Caspase-3에 미치는 영향

        Prostate cancer is one of the most common non-skin cancers in men. Armeniacae Amarum Semen has traditionally been used for the treatment of inflammation diseases, leprosy, leucoderma, and tumors. Apoptosis, which is also known as programmed cell death, is an important mechanism in cancer treatment.Objectives : We observed whether an aqueous extract of Armeniacae Amarum Semen induces apoptotic cell death in human DU145 prostate cancer cells.Methods : We treated DU145 cells with Armeniacae Amarum Semen extract and investigated characteristics of apoptosis. And investigated whether treated with Armeniacae Amarum Semen extract increased Bax mRNA expression, Bcl-2 mRNA expression, caspase-3 enzyme activity and their protein level.Results : We have shown that Armeniacae Amarum Semen extract can induce apoptotic cell death in human DU145 prostate cancer cells by caspase-3 activation through the down-regulation on Bcl-2 expression and the up-regulation on Bax expression.Conclusions : It can be expected that an aqueous extract of Armeniacae Amarum Semen may offer a valuable means for the treatment of prostate cancers.

        성향정기산이 흰쥐의 MCAO에 의한 국소뇌허혈에 미치는 영향

        This study evaluated neuroprotective effect of Sunghyangjungki-San (SHS) on the focal cerebral ischemia. The rats were induced infarct in cerebral cortex and caudoputamen by using temporal occlussion of the middle cerebral artery (MCAO), then water extract of SHS was treated for MCAO rats. Neuroprotective effect was evaluated by neurological score, infarct sizes and total volume, positive neurons against Bax, Caspase-3, HSP-72, and $HIF-1{\alpha}$ in infarct area with immunohistochemistry. The results obtained were as follows: Treatment of SHS improved neurological score of MCAO rats, but there was not a statistical significance. Treatment of SHS reduced significantly infarct sizes in the brain sections of MCAO rats. Treatment of SHS reduced significantly total volume of infarct of MCAO rats. Treatment of SHS reduced significantly Bax positive neurons in penumbra of cerebral cortex of MCAO rats. Treatment of SHS reduced significantly Caspase-3 positive neurons in caudoputamen and penumbra of cerebral cortex of MCAO rats. Treatment of SHS reduced significantly HSP-72 positive neurons in penumbra of cerebral cortex of MCAO rats. Treatment of SHS reduced significantly $IF-1{\alpha}$ positive neurons in penumbra of cerebral cortex of MCAO rats.

        관절염 백서의 염증과 간기능에 대한 목과.백굴채.위영선(木瓜.白屈菜.威靈仙)의 효능연구

        The purpose of this study is to observe the effects of Chaenomelis Fructus Herba Chelidonii Clematis Florida Thunb mixed water extract on the inflammatory edema induced by Found's complete adjuvant and on different blood volume, and then to demonstrate its efficiencies as well as its safety. We injected Chaenomelis Fructus Herba Chelidonii Clematis Florida Thunb mixed water extract on the inflammatory edema induced by Found's complete adjuvant once daily at the same time for a week, then observed the paw-edema, the counts of the blood WBC, Serum total protein, Serum total bilirrubin, SGOT and SGPT The result obtained is as follows : Inflammatory edema on the right plantar in both sample groups showed a significant reduce compared with that in the control group. The counts of WBC in both sample groups showed a significant decrease compared with that in the control group. The volume of serum total protein in the Sample A group showed a significant decrease compared with that in the control group. The volume of serum total bilirrubin, GOT and GPT in both sample groups was not changed compared with that in the control group. As a result of this experiment, it is concluded that Chaenomelis Fructus Herba Chelidonii Clematis Florida Thunb mixed water extract showed that therapeutic effect on the anti-inflammation on arthritis; otherwise it showed non-toxic effect on the liver function.

        『영소침구경』 보사법 분석

        Objectives : Although the tonification and sedation method such as mountain-burning fire method and heaven-penetrating cooling method, is one of the important techniques that have been claimed to increase the effect of acupuncture, research on this has been sparse. The aim of this study is to provide an understanding of tonification and sedation methods in the "Yeongsochimgugyeong". Methods : We analyzed the similarities and differences by comparing the contents of the tonification and sedation method described in books "Yeongsochimgugyeon" and "Complete Compendium of Acupuncture and Moxibustion" Results : A total of 28 out of the 33 tonification and sedation methods described in "Yeongsochimgugyeon" were compared with those in "Complete Compendium of Acupuncture and Moxibustion". As a result, we found that many of the tonification and sedation methods recorded in "Yeongsochimgugyeon" were based on more detailed and specific clinical cases. Conclusions : The tonification and sedation methods in "Yeongsochimgugyeon" were not only descriptive and consistent in describing that methods, but also contains valuable information for clinical use. In the future, based on this research, we will be able to clinically implement these tonification and sedation methods and quantitatively evaluate them.

        치자(梔子)가 JNK와 NF-kB를 통한 항산화와 항염증의 대사과정에 미치는 영향

        Objectives : The purpose of this study is to observe the effects of Gardeniae Fuctus(GF) on the metabolic process of antioxidant and anti-inflammation. Methods : 4-HNE was injected into PC-12 cell to cause oxidative stress-induced inflammatory response, and then a western blot was conducted to observe the expression of Nuclear Factor-kB (NF-kB) and c-Jun N-terminal kinase (JNK) protein that are important factors involved with inflammation. Results : 1. The Gardeniae Fuctus water extract 50 ㎍ and 100 ㎍ significantly suppressed the increase in JNK protein expression in PC-12 cell. 2. The Gardeniae Fuctus water extract 50 ㎍, 100 ㎍ and 200 ㎍ significantly suppressed the increase in NF-kB protein expression in PC-12 cell. Conclusion : These results suggest that the Gardeniae Fuctus water extract has antioxidative and anti-inflammatory activity through suppressing the activity of JNK and NF-kB.
