        골막기원세포에서 strontium에 의한 조골세포 표현형의 활성

        This study investigated the effects of strontium on osteoblastic phenotypes of cultured human periostealderived cells. Periosteal tissues were harvested from mandible during surgical extraction of lower impacted third molar. Periosteal-derived cells were introduced into cell culture. After passage 3, the periostealderived cells were further cultured for 28 days in an osteogenic induction DMEM medium supplemented with fetal bovine serum, ascorbic acid 2-phosphate, dexamethasone and at a density of $3{\times}10^4$ cells/well in a 6-well plate. In this culture medium, strontium at different concentrations (1, 5, 10, and 100 ${\mu}g$/mL) was added. The medium was changed every 3 days during the incubation period. We examined the cellular proliferation, histochemical detection and biochemical measurements of alkaline phosphatase (ALP), the RT-PCR analysis for ALP and osteocalcin, and von Kossa staining and calcium contents in the periostealderived cells. Cell proliferation was not associated with the addition of strontium in periosteal-derived cells. The ALP activity in the periosteal-derived cells was higher in 5, 10, and 100 ${\mu}g$/ml strontium-treated cells than in untreated cells at day 14 of culture. Among the strontium-treated cells, the ALP activity was appreciably higher in 100 ${\mu}g$/ml strontium-treated cells than in 5 and 10 ${\mu}g$/ml strontium-treated cells. The levels of ALP and osteocalcin mRNA in the periosteal-derived cells was also higher in strontium-treated cells than in untreated cells at day 14 of culture. Their levels were increased in a dose-dependent manner. Von Kossa-positive mineralization nodules were strongly observed in the 1 ${\mu}g$/ml strontium-treated cells at day 21 and 28 of culture. The calcium content in the periosteal-derived cells was also higher in 1 ${\mu}g$/ml strontium-treated cells at day 28 of culture. These results suggest that low concentration of strontium stimulates the osteoblastic phenotypes of more differentiated periosteal-derived cells, whereas high concentration of strontium stimulates the osteoblastic phenotypes of less differentiated periosteal-derived cells. The effects of strontium on osteoblastic phenotypes of periosteal-derived cells appear to be associated with differentiation-extent.

        서울시 마을마당의 이용후 평가

        The purpose of this study is to provide the basic data for planning and designing Village Parks in Seoul in the future, which result from analyzing post-occupancy evaluation. For conducting this study, 10 Village Parks among 45, built between 1996 and 1997, were selected and analyzed. The results of this study are as follows. The degree of satisfaction of 10 Village Parks in Seoul has been evaluated on a scale of 10, to be a 6.68 average. This result means that the most respondents are satisfied with the existing Village Parks in Seoul. The correlation of the degree of satisfaction with major planning factors for 3 types of Village Parks shows that accessibility is the most important factor. As for future directions of planning and designing Village Parks, many kinds of sitting, leaning and resting place need to be provided. It is also necessary to improve the visual quality of the parks with various spatial elements. The research also shows that various facilities for people's use should be provided in Village Parks.

        형태주제 분석을 통한 마을마당의 설계특성 -서울시의 사례-

        In this study, design characteristics of Village Parks in Seoul are investigated through an analysis of structuring themes expressed in spatial design. The goals and objectives of this study are: to emphasize the significance of design themes; and to demonstrate an approach to the design of landscapes through an analysis of structuring themes. For conducting this study, geometric forms and naturalistic forms were examined first. That is, the following 15 structuring themes were examined, as guiding themes: the 90$^{\circ}$rectangular theme; the 45$^{\circ}$/90$^{\circ}$angular theme; the 30$^{\circ}$/60$^{\circ}$ angular theme; circles on circle; concentric circles and radii; arcs and tangents; circle segments; the ellipse; the spiral; the meander; the free ellipse and scallops; the free spiral; the irregular polygon; the organic edge; and clustering and fragmentation. Forty five Village Parks in Seoul, built between 1996 and 1997, were analyzed through these 15 structuring themes. An analysis of Village Parks was conducted by the following two categories: land shape and structuring themes; and design directions and structuring themes. The research results are as follows; 1) Geometric forms are more frequently applied than naturalistic forms. 2) Regardless of land shape, geometric forms derived from simple primary shapes, the square and the circle, are frequently used. And the meander and the irregular polygon among naturalistic forms are frequently used. 3) Regarding design directions and structuring themes, design concepts, symbolic meanings and spatial forms are, to some degree, integrated. 4) The spiral is not applied among geometric forms. And the meander and the irregular polygon, as naturalistic forms, are frequently utilized. Research findings obtained from this study could be used in the designing of future Village Parks. For a profound study, future, research is needed in two-dimensional plans and three-dimensional elements of Village Parks.

        조형원리에 의한 옥상정원 디자인 분석 연구

        인구의 도시집중과 고층, 고밀도의 건축행위로 열악해진 도시생태환경을 복원하고 사람들이 가까이에서 접할 수 있는 녹지공간을 확보하기 위한 대안으로 옥상녹화를 주목하고 있다. 서울특별시는 2002년부터 옥상녹화지원 사업을 실시하면서 2008년부터 옥상을 도시경관의 향상과 휴식공간의 제공 및 생태환경을 개선할 수 있는 공간으로 활용하기 위하여 명칭을 옥상녹화사업에서 옥상공원화사업으로 바꾸고 지원을 확대하고 있다. 이에 본 연구는 옥상정원의 디자인 경향을 알아보기 위하여 2007년도 서울특별시에서 지원하여 조성된 옥상정원을 대상으로 조형원리를 적용하여 시각적으로 디자인을 분석하고 향후 옥상정원의 디자인을 위한 기초자료를 제공하기 위하여 수행되었다. 연구 대상지를 옥상정원의 공간구성요소인 식생, 시설물, 수공간, 포장으로 나누고 통일, 비례, 대비, 반복, 균형, 율동, 대칭, 강조 등 여덟 개의 조형원리를 적용하여 조사표를 만들고, 현장조사와 사진촬영을 하여 자료를 수집·분석하였다. 이에 대한 연구결과를 요약하면 다음과 같다. 첫째, 공간구성요소별 조형원리 적용분석 결과 식생에서는 반복이 많이 적용되었고, 시설물에 적용된 조형원리 빈도는 반복과 강조로 나타났다. 수공간은 열세 개소 중에서 여덟 개소가 설치되었으며, 적용된 조형원리는 자유곡선을 사용한 율동이었다. 포장에 적용된 조형원리는 대비와 반복이 많이 사용되었다. 둘째, 조형원리의 공간구성요소별 적용 분석 결과 가장 높은 빈도를 보인 것은 반복이었고 통일, 대비와 강조, 대칭, 율동, 균형, 비례 순으로 나타났다. 세째, 건물유형별 공간구성요소 특징 분석결과 계성여고는 텃밭을 만들어 학생들이 야생화와 채소를 직접 심고 가꾸면서 체험하는 학습장으로 활용하고 있었으며, 장로회 신학교-A는 신학교의 특성을 살려서 개인 기도실을 만들어 학생들이 자유롭게 이용할 수 있게 하였다. 또 장로회 신학대-A와 이대목동병원은 공연을 할 수 있는 무대 만들어 작은 공연을 할 수 있게 하였다. 본 연구를 통한 시사점으로는 옥상정원은 주이용자 및 이용목적에 따라서 차별화된 설계가 되어야 하며, 시민들이 도심 속에서 자연을 접하고 휴식과 여가 활동을 할 수 있도록 옥상정원의 이용도를 높이는 다양한 시도가 필요하다고 여겨진다. The increase in population and in the number and height of buildings result in the poor condition of city environment. In order to restore the environment and let people come in contact with nature, the concept of green roof is considered to be the most effective solution. In particular, Seoul Metropolitan Government has been supporting the green roof project since 2002. In 2008, the `green roof project` was renamed to `green garden project`, because the project involves improving city scenery and making the roof gardens wherever possible, for people can come to and rest. Roof gardens, created with the support from the government of Seoul in 2007, were analyzed according to their visual and aesthetic qualities. This research will provide design data to make roof gardens more useful and artistic. The subject of research has been separated into four different factors: planting, facility, water space, and pavement. It has been separated into eight different principles of design: unity, proportion, contrast, repetition, balance, rhythm, symmetry, and emphasis. A field study and photographs have been collected as data. A summary of the study results are as follows, 1. The following analysis is the application of the principle of modeling in different spaces. In many facilities, repetition of plant occurs. Regarding equipment, repetition and emphasis occurred frequently. Out of thirteen facilities, eight facilities provided water space, showing rhythm with a free curved line technique. Regarding pavement, contrast and repetition occurred frequently. 2. The following is an analysis of space composition factors regarding different principle of modeling. In thirteen facilities, repetition is being used the most, the second is unity, then contrast and emphasis, symmetry, rhythm, balance, and proportion. 3. The following is an analysis of modeling in different building usage. In Gye Seong Girls` High School, there is a field where students can plant wildflowers and grow vegetables. In Presbyterian College and Theological Seminary-A(PCTS-A), there is a prayer room that the students can use freely. Also there are stages at PCTS-A and Ewha Woman`s University Mokdong Hospital, where performances take place. In the future, designers should come up with designs that are specialized for their users and differentiated from other services. Also designers should develop many more palces where people and nature meet and exist in one place together.
