<P>Signal transduction studies have indicated that Akt is essential for transducing the signals originating from extracellular stimuli. An exploration of the Akt signal transduction mechanism depends on the ability to assay its activation states by determining the ability of Akt to phosphorylate various substrates. This paper describes a CE-based kinase assay for Akt using a UV detection method. The RPRAATF peptide was used as the specific substrate to determine the Akt activity. Under the CE separation conditions used, the phosphorylated and nonphosphorylated forms of the RPRAATF peptide were rapidly resolved in the Akt reaction mixture within 20 min. Using this method for measuring the Akt activity, the incubation time for the Akt reactions as well as the kinetic parameters (K<SUB>M</SUB>) were examined. Furthermore, the developed method was applied to a PC12 cell system to assess the dynamics of the Akt activity by examining the effectiveness of the RPRAATF peptide substrate under various cytokine-stimulated environments. These results highlight the feasibility of the CE method, which is a simple and reliable technique for determining and characterizing various enzyme reactions particularly kinase enzymes.</P>

<P><B>Abstract</B></P><P>Immunoassays are commonly used in bioresearch for the detection and quantification of small proteins and macromolecules in biological fluids and other complex matrices. In this report, a competitive immunoassay using capillary electrophoresis (CE) with laser-induced fluorescence was developed for methionine-enkephalin (ME). The method is based on the competitive reaction between the ME and fluorescein conjugated ME (ME-F) with anti-ME antibody, capillary electrophoresis separation of the ME-antibody bound and free ME-F, followed by the laser-induced fluorescence detection of the fluorescent species. With the optimized separation conditions, it was possible to separate the antibody bound and free fluorescien conjugated ME by a capillary electrophoresis–laser-induced fluorescence (CE–LIF) analysis using an uncoated fused-silica capillaries. The results concluded that the assay specificity, selectivity and accuracy were excellent.</P>

The present study elucidates the influence of reinforcement particles in friction stir welded aluminium (Al 6063) matrixcomposites on mechanical properties of the metal matrix composition. Initially composites were successfully fabricated withdifferent weight percentage and combination of reinforcements (graphite (gr), B4C and SiC) through stir casting technique. All sets of composite specimen were welded through solid state joining process of friction stir welding technique, theparameters such as tool rotational speed (800, 1000 and 1200 rpm), welding speed (20 and 40 mm/min) and axial loading (10and 20 kN) are taken for characterization of fabricated composites. The effects of friction stir welding parameters wereexamined by mechanical and microstructural characterizations. The composites microstructure and dispersion of particlereinforcements were analysed through optical microscope and also the mechanical properties of yield strength, ultimatestrength and elongation were analysed using universal testing machine. The optimized friction stir weld parameters wereidentified for 20% weight fraction reinforced hybrid composites.

<P>Emerging systems biology aims at integrating the enormous amount of existing omics data in order to better understand their functional relationships at a whole systems level. These huge datasets can be obtained through advances in high-throughput, sensitive, precise, and accurate analytical instrumentation and technological innovation. Separation sciences play an important role in revealing biological processes at various omic levels. From the perspective of systems biology, CE is a strong candidate for high-throughput, sensitive data generation which is capable of tackling the challenges in acquiring qualitative and quantitative knowledge through a system-level study. This review focuses on the applicability of CE to systems-based analytical data at the genomic, transcriptomic, proteomic, and metabolomic levels.</P>

Actinobacteria are prolific producers of a large number of natural products with diverse biological activities. In the present study, an actinobacterium isolated from sunflower rhizosphere soil sample collected from Medak, Andhra Pradesh, South India was identified as Amycolatopsis thermoflava strain SFMA-103. A pigmented secondary metabolite in culture broth was extracted by using methanol and it was further purified by silica gel column chromatography with methanol-chloroform solvent system. Structural elucidation studies based on UV-visible, 1D and 2D-NMR, FT-IR, and mass spectroscopic analyses confirmed the structure as 1-methoxy-3-methyl-8-hydroxy-anthraquinone. It showed significant in vitro anticancer activity against lung cancer and lymphoblastic leukemia cells with IC<sub>50</sub> values of 10.3 and 16.98 μM, respectively. In addition, 1-methoxy-3-methyl-8-hydroxy-anthraquinone showed good free radical scavenging activity by DPPH method with an EC<sub>50</sub> of 18.2 μg/ml. It also showed other promising superoxide radical scavenging, nitric oxide radical scavenging and inhibition of lipid peroxidation activities. This is a first report of antiproliferative and antioxidant activities of 1-methoxy-3-methyl-8-hydroxy-anthraquinone isolated from A. thermoflava strain SFMA-103 which may find potential application in biotechnological and pharmaceutical fields.
Study on effect of welding speed on friction stir welded aluminium metal matrix hybrid composite

The development of aluminum series matrix hybrid composites with ceramic reinforcements has recently caught the interestof new researchers. The addition of more than one or two reinforcements in aluminium alloy leads to the fabrication of theAluminium Metal Matrix Hybrid Composite (AMMHC). The present investigation is aimed at the development of newlydeveloped AMMHC followed by friction stir welding for obtaining the improved mechanical properties. The stir castedAA6063 with reinforcement of 5%, 10% and 15% wt. Silicon Carbide (SiC) and 10% wt. Boron Carbide (B4C) werefabricated by stir casting technique and the AMMHC with 10% each of SiC and B4C showed higher mechanical properties. Further the AMMHC are joined by friction stir welding (FSW) route. A prime experimental work has been tried out in theFSW process by varying welding speed (20, 30, 40, 50 and 60 mm/min). The effect of welding speed on AMMHCs were studiedon macro and microstructural observations followed by micro hardness and ultimate tensile. Strength, in which 40 mm/minof welding speed gives better properties than other processing condition. The weldment reveals the better joint strength withthe presence of fine particle dispersion in the microstructure.
