Evaluation of the Feasibility and Efficacy of Forward-Viewing Endoscopic Ultrasound

( Seohyun Lee ),( Dong Wan Seo ),( Jun Ho Choi ),( Do Hyun Park ),( Sang Soo Lee ),( Sung Koo Lee ),( Myung Hwan Kim ) 대한소화기학회 2015 Gut and Liver Vol.9 No.5

Background/Aims: We aimed to evaluate the feasibility and efficacy of a forward-viewing linear endoscopic ultrasound (FV-EUS) in diagnostic EUS procedures compared to standard oblique-viewing EUS (OV-EUS). Methods: This study was a prospective, randomized study that permitted cross-over. Fifty-one patients with subepithelial pancreatobiliary and upper gastrointestinal lesions underwent FV-EUS and OV-EUS sequentially, in random order. The EUS visualization was performed by a novice endosonographer, and the image quality of specific lesions was scored by an expert endosonographer. If fine-needle aspiration (FNA) was indicated, it was performed using both echoendoscopes by an expert endosonographer. Results: Both of the EUS procedures had similar visualization times and image quality. In general, the visualization time was inversely related to the diameter of the specific lesions. In subepithelial lesions of the stomach and duodenum, the visualization time (98.8±62.2 seconds vs 139.0±66.6 seconds, p=0.008) and image quality (4.1±1.3 vs 3.3±1.7, p=0.02) of FV-EUS were significantly superior to OV-EUS. FV-EUS-guided FNA of pancreatic masses was successful in seven patients (87.5%). Conclusions: FV-EUS may increase the ease of access to gastrointestinal subepithelial lesions compared to conventional OV-EUS. The performance of FV-EUS for evaluating pancreatobiliary diseases and performing interventions was comparable to conventional OV-EUS. (Gut Liver 2015;9:679-684)

Partial Replacement of High-Fat Diet with Beef Tallow Attenuates Dyslipidemia and Endoplasmic Reticulum Stress in db/db Mice

        Ji-Su Lee,Jennifer K. Lee,Jae-Joon Lee,Seohyun Park,Sunyoon Jung,Hyun-Joo Lee,Jung-Heun Ha 한국식품영양과학회 2022 Journal of medicinal food Vol.25 No.6

High-fat diet (HFD) consumption is closely associated with an increased risk of metabolic syndromes (MetS), such as obesity, type 2 diabetes, and cardiovascular diseases (CVDs). Therefore, the consumption of alternative and functional fatty acids to replace saturated fatty acids and/or trans-fatty acids with polyunsaturated fatty acids has become an important dietary strategy for the prevention of MetS. Consumption of omega-3 fatty acids (n-3) reduces various physiological complications, including CVDs, nonalcoholic fatty liver disease, and insulin resistance, related to inflammatory responses. In this study, we investigated the partial replacement effects of HFD with beef tallow (BT) on dyslipidemia and endoplasmic reticulum (ER) stress in male db/db mice. The animals were grouped to one of four dietary intervention groups (n = 16 per group): (1) normal diet, (2) HFD, (3) HFD partially replaced with regular beef tallow (HFD+BT1), or (4) HFD partially replaced with beef tallow containing a relatively reduced omega-6 fatty acid (n-6)/n-3 ratio (HFD+BT2) than HFD+BT1. After 6 weeks of dietary intervention, 1 mg/kg of phosphate-buffered saline or tunicamycin (TM) was injected intraperitoneally. HFD+BT2 significantly suppressed the serum total cholesterol and non-high-density lipoprotein cholesterol levels more than HFD and HFD+BT1, and triglyceride levels in the epididymal adipose tissue (EAT) were remarkably decreased. Mice that received HFD+BT2 had elevated protein expressions of phospho-AMP-activated protein kinase (p-AMPK). Moreover, HFD+BT2 effectively inhibited ER stress in the liver and EAT. Consistent with our hypothesis, HFD+BT2 remarkably alleviated dyslipidemia and TM-inducible ER stress, while activating p-AMPK.

      • Ginsenoside Rg3 Reduces Lipid Accumulation with AMP-Activated Protein Kinase (AMPK) Activation in HepG2 Cells

Lee, Seohyun,Lee, Mak-Soon,Kim, Chong-Tai,Kim, In-Hwan,Kim, Yangha Molecular Diversity Preservation International (MD 2012 INTERNATIONAL JOURNAL OF MOLECULAR SCIENCES Vol.13 No.5

Cardiovascular disease (CVD) is one of the main causes of mortality worldwide, and dyslipidemia is a major risk factor for CVD. Ginseng has been widely used in the clinic to treat CVD. Ginsenoside Rg3, one of the major active components of ginseng, has been reported to exhibit antiobesity, antidiabetic, and cardioprotective effects. However, the effect of ginsenoside Rg3 on hepatic lipid metabolism remains unclear. Therefore, we investigated whether ginsenoside Rg3 would regulate hepatic lipid metabolism with AMP-activated protein kinase (AMPK) activation in HepG2 cells. Ginsenoside Rg3 significantly reduced hepatic cholesterol and triglyceride levels. Furthermore, ginsenoside Rg3 inhibited expression of sterol regulatory element binding protein-2 (SREBP-2) and 3-hydroxy-3-methyl glutaryl coenzyme A reductase (HMGCR). Ginsenoside Rg3 increased activity of AMPK, a major regulator of energy metabolism. These results suggest that ginsenoside Rg3 reduces hepatic lipid accumulation with inhibition of SREBP-2 and HMGCR expression and stimulation of AMPK activity in HepG2 cells. Therefore, ginsenoside Rg3 may be beneficial as a food ingredient to lower the risk of CVD by regulating dyslipidemia.

Problems for Foreign Students in the Korean Language Liberal Arts Curriculum, and Suggestions for Improvement - Focusing on the Intensive Korean Language Course (IKC) at Dankook University -

        Lee Seohyun(이서현),Han Jeonghan(한정한) 한국어학회 2019 한국어학 Vol.85 No.-

The purpose of this paper is to look at problems in Korean language undergraduate liberal arts curricula, taking Dankook University’s undergraduate program as an example, and to suggest ways to improve foreign students’ Korean language skills, which are needed to take college courses. In looking a the situations at five universities, we first examined the current status of Korean for Academic Purposes courses aimed at foreign students. The typical problems found here are as follows. First, there were few level-based classes taking place in the mixed classes of domestic and foreign students, such as ‘thoughts and expressions’. Second, the level of textbook development for Korean language courses for foreign students was very poor. Third, the degree of continuity in Korean language courses for foreign students was poor, and has in fact decreased. Fourth, the partnership relationship between the instructors in charge of Korean language courses and the International Affairs Office that is in charge of academic and life management for foreign students does not work well. To solve these problems, starting in the second semester of 2019, Dankook University has taken all Korean language classes scattered in the across the school of liberal arts and incorporated them into an Intensive Korean Language Course (IKC). In addition, IKC instructors were issued a double duty to DKU International Affairs Office to establish strong, integrated management for foreign students. This integration of existing classes into one single curriculum (IKC) helped eliminate many of the aforementioned problems.

Green Tea (-)-Epigallotocatechin-3-Gallate Induces PGC-1α Gene Expression in HepG2 Cells and 3T3-L1 Adipocytes

Lee, Mak-Soon,Lee, Seohyun,Doo, Miae,Kim, Yangha The Korean Society of Food Science and Nutrition 2016 Preventive Nutrition and Food Science Vol.21 No.1

Green tea (Camellia sinensis) is one of the most popular beverages in the world and has been acknowledged for centuries as having significant health benefits. (-)-Epigallocatechin-3-gallate (EGCG) is the most abundant catechin in green tea, and it has been reported to have health benefit effects. Peroxisome proliferator-activated receptor ${\gamma}$ coactivator $(PGC)-1{\alpha}$ is a crucial regulator of mitochondrial biogenesis and hepatic gluconeogenesis. The objective of this study was to investigate whether EGCG from green tea can affect the ability of transcriptional regulation on $PGC-1{\alpha}$ mRNA expression in HepG2 cells and 3T3-L1 adipocytes. To study the molecular mechanism that allows EGCG to control $PGC-1{\alpha}$ expression, the promoter activity levels of $PGC-1{\alpha}$ were examined. The $PGC-1{\alpha}$ mRNA level was measured using quantitative real-time PCR. The -970/+412 bp of $PGC-1{\alpha}$ promoter was subcloned into the pGL3-Basic vector that includes luciferase as a reporter gene. EGCG was found to up-regulate the $PGC-1{\alpha}$ mRNA levels significantly with $10{\mu}mol/L$ of EGCG in HepG2 cells and differentiated 3T3-L1 adipocytes. $PGC-1{\alpha}$ promoter activity was also increased by treatment with $10{\mu}mol/L$ of EGCG in both cells. These results suggest that EGCG may induce $PGC-1{\alpha}$ gene expression, potentially through promoter activation.

      • PUFAs Attenuate Blood Lipids and LPS-Inducible Inflammation and ER stress in Adipocyte Tissue in ob/ob Mice

        Seohyun Park,Zheng Jiaxiang,Jihye Lee,Jae-Joon Lee,Jung-Heun Ha 한국식품영양과학회 2021 한국식품영양과학회 학술대회발표집 Vol.2021 No.10

PUFAs has been reported to alleviate diet-induced systemic inflammation, dyslipidemia in mammals. To understand the effects of replacement of saturated fatty acids (SFAs) with PUFAs to obese subjects, we supplemented a normal diet with perilla oil (PO) or corn oil (CO) to genetically obese mice for 12 weeks with LPS injection. 1) PUFAs improved lipid profiles. PUFAs consumption attenuated hepatic TG and LDL-C; however, HDL-C was significantly increased in PO or CO replaced than normal chow consumed group. 2) Intriguingly, PUFAs consumption decreased inflammatory responses in the epididymal adipose tissue(EAT). PUFAs decreased the secretion of adipokines (IL-1β, IL-6 and CXCL1). Attenuation of adipocyte inflammation maybe related to the post-translational modification by decreasing phosphorylation of MAPKs. Prolonged inflammation may trigger endoplasmic reticulum (ER) stress, therefore, we logically postulated that consumption of PUFAs may decrease ER stress. 3) As our expectation, PUFAs consumption attenuated LPS-inducible ER stress in the EAT by attenuation of BiP and CHOP. Therefore, PUFAs decreased hepatic lipids accumulation, inflammation and ER stress in the EAT.
