        Effect of By-Product Gypsum Fertilizer on Methane Gas Emissions and Rice Productivity in Paddy Field

        Rice cultivation in paddy field affects the global balance of methane (CH₄) as a key greenhouse gas. To evaluate a potential use of by-product gypsum fertilizer (BGF) in reducing CH₄ emission from paddy soil, CH₄ fluxes from a paddy soil applied with BGF different levels (0, 2, 4 and 8 Mg ha<SUP>-1</SUP>) were investigated by closed-chamber method during rice cultivation period. CH₄ flux significantly decreased (p<0.05) with increasing level of BGF application. 8 Mg ha<SUP>-1</SUP> of BGF addition in soil reduced CH₄ flux by 60.6% compared to control. Decreased soil redox potential (Eh) resulted in increasing CH₄ emission through a CO₂ reduction reaction. The concentrations of dissolved calcium (Ca) and sulfate ion (SO₄<SUP>2-</SUP>) in soil pore water were significantly increased as the application rate of BGF increased and showed negatively correlations with CH₄ flux. Decreased CH₄ flux with BGF application implied that SO₄<SUP>2-</SUP> ion led to decreases in electron availability for methanogen and precipitation reaction of Ca ion with inorganic carbon including carbonate and bicarbonate as a source of CH₄ formation under anoxic condition. BGF application also increased rice grain yield by 16% at 8 Mg ha<SUP>-1</SUP> of BGF addition. Therefore, our results suggest that BGF application can be a good soil management practice to reduce CH₄ emission from paddy soil and to increase rice yield.

      • 해양동물 눈알고둥 렉틴(LCC-Ⅲ)의 림프구 분열효과 및 면역화학적 특성

        A new lectin was purified from whole body of marine shellfish, Lunella coronata coreensis(LCC) by salt fractionation, anionic and cationic ion exchange chromatography. The results were as follows : LCC lectin was relatively stable at the range of pH4-10 and the temperature of 0-50℃. It was dissociated to a single subunit of 22KD by SDS-polyacrylamide gel electrophoresis. In carbohydrates inhibition assay, lectin activity was strongly inhibited by lactose and lactulose at a concentration 6.25mM. LCC lectins(LCC DE 52 0.1M DE 0.2M) were revealed each other as having partial identity by immunochemical studies. LCC lectin proved to be a potent mitogen for murine splenic lymphocytes. And the optimum mitogenic dose was 7.81㎍/㎖.

        렉틴 MBL과 LCC를 농도와 시간의 다양한 조건에서 PBMC와 반응시켰을 때 IL-1, IL-2, TNFα와 IFNγ등 사이토카인 유전자 발현을 유도함을 RT-PCR로 확인한 바의 결과를 근거로 하여 ELISA방법으로 다양한 반응시간에 따른 TNFα와 IFNγ의 생성양상을 측정하였다. MBL의 경우 TNFα는 사이토카인 생성이 반응 8시간대에서 최대가 되었으나 24시간대로 가면서 급격히 감소되며 그 후는 거의 소멸되었다. IFNγ의 경우, 반응 1시간 경에 급속한 상승을 보이다가 4시간대에서 8시간대까지 감소하였다. 그러나 24시간대부터는 최대치에 가까워지다가 그 후 서서히 감소하였다. LCC로 PBMC를 자극시켜 사이토카인의 생성양성을 살펴본 결과, TNFα는 그 생성이 반응 8시간대에서 가장 높은 농도로 나타났으며 24시간 이후 급격히 감소하다가 72시간 이 후부터는 현저히 소멸됨을 보였다. IFNγ는 1시간부터 생성능이 최대치가 되지만, 4시간대부터 감소되어 8시간대부터 72시간대 까지 크게 변동없이 지속되다가 72시간 이후로는 급격하게 감소하였다. ELISA결과 반응 후 24시간대 이 후부터는 실험대상 사이토카인이 모두 감소하여 동물기원 LCC와 식물기원 MBL은 모두 비슷한 양상을 나타냄을 알 수 있었다.

        Developing new substances for immunosuppress or immunomodulate from natural products is extremely important in the present biomedicine. In this paper, we focused our efforts on the mitogenicity and immunochemical properties of the NI lectins(NIA, NIB, NIC)and APL lectins obtained from Neptunea intersculpta and Asterina pectinifera. But NI lectins are turned out as different molecules through the study of carbohydrate specificity. The activity of APL was inhibited by D(+)-glucosamine, D(+)-galactosamine, stachyose, N-acetyl-galactosamine and methyl-α-D-galactopyranoside. NIC lectin was a potent mitogenic toward murine splenic lymphocytes at a concentration ca. 6.4㎍/㎖. APL was found that it promotes the division of human lymphocytes at a concentration ca. 1.2㎍/㎖.

        Researching the effect of medium-nitrogen and hormones on the culture of protoplasts isolated from Mesophyll of Nicotiana tabacum L., protoplasts were cultured on the B5 media supplemented with the various concentration of inorganic nitrogen and hormones for 10 days. After measuring the viability and division rate of protoplast on the various experimental plots, multiple regression analysis was applied to experimental results. The regression constant value of viability is 62.623 and regression coefficient values of nitrate, BAP and incubation time are 0.005, -2.395 and -1.908 respectively. But the effects of ammonium, NAA and 2.4_D are not significant. The regression constant value of division rate is _2.276 and regression coefficient values of ammonium, nitrate and incubation time are 0.032, 0.002 and 1.190 respectively. But the effects of NAA, 2.4_D and BAP are not significant. The viability and division rate of protoplast were high percentage in the B_5 medium supplemented with ammonium 2mM, nitrate 50mM, BAP 1㎎/1 and NAA 3㎎/1. This result implies that ammonium and nitrate are essential components on the culture of tabacco protoplasts and plating efficiency will be increased when ammonium and nitrate concentration are increased in the B5 medium and when BAP concentration is appropriate.

        The lst grade of Nur (Department of Nursing), ACT was higher than REF, but ACT was lower than REF in the 2nd and 3rd grade. SEN was increased in the 2nd grade more than lst grade, but it was decreased in the 3rd grade, VIS as well. The case of SEQ, it developed a tendency to increase as their grade was higher. The Oph (Department of Ophthalmic Optics) showed a tendency to REF, but it changed to propensity of ACT in the 2nd grade. And also propensity of SEN was much higher than INT when they were in the lst grade, but the gap was reduced when they were 2nd grade. As the grade was higher, the students had preference of VIS over VRB and the gap of the preference between VIS and VRB was increased, the propensity of SEQ was increased as well. Therefore the first students in the Department of Nursing showed strong propensity of ACT and they changed to the propensity of REF after. The students of the Department of Ophthalmic Optics showed the opposite propensity. The other learning propensity showed that the propensity of the first grade maintained to upper grade.

        눈알고둥이 생리식염수 용액 추출물은 사람 및 각종 동물 적혈구를 비특이적으로 응집시켰다. 눈알고둥의 새로운 렉틴성분을 생리식엽수용액추출, 황하암모늄침전, 이온교환 크로마토그래피 등의 방법으로 분리 정제하였다.(LCC-I lectin은 35.2배 정체되고, 0.9% 회수되었다) 정제된 렉틴은 전기영동상에서 하나의 띠를 나타냈다. 또한, LCC-I 렉틴의 분자량은 SDS-PAGE에 의해 20,000정도로 추정되었다. 정제된 눈알고둥 렉틴은 pH 변화와 열에 비교적 안정하였다. 적혈구 응집반응은 lactose와 lactulose의 6.25mM 농도에서 저해되어 당 특이성을 나타내었다. 또한 눈알고둥 렉틴은 마우스의 림프구에 대한 분열자극효과를 나타냈다. A molecular weight of LCC-I was calculated as 20,000 by sodium dodesyl sulfate polyacrylamide gel electriphoresis. The purified lectin was relatively stable at various pH and heat. Among the tested sugars, lactose and lactulose inhibited lectin activity at a concentration of 6.25mM, respectively. The lectin was a mitogenic toward murine splenic lymphocytes.

A new lectin was purified by the following procedures: 0.15 M NaCl extraction, salt fractionation, gel filtration, anionic and cationic ion exchange column chromatographies. Through these purification procedures, specific activity of LCC-I was increased from 276 to 9714.3units/mg, And on polyacrylamide gel electrophoresis, LCC-I exhibited one major band. A molecular weight of LCC-I was assumed to be 20,000 by sodium dodesyl sulfate polyacrylamide gel electrophoresis. The purified lectin was relatively stable at various pH and heat. Among the tested sugars, lactose and lactulose inhibited lectin activity at a concentration of 6.25mM, respectively.

        해양동물 눈알고둥 렉틴의 림프구 분열 효과 및 면역화학적 특성

        Developing new substance for immunosuppressor or immunomodulator from natural products is extremely important in the present biomedicine. In this paper, we focused our efforts on the mitogenicity and immunochemical properties of the two lectins (LCC-I, LCC-II) obtained from marine animal Lunella coronata coreensis. Immunochemical techniques were employed to elucidate the structural and/or functional similarities between the LCC lectins. Molecular weight of the LCC lectins, LCC-I and LCC-II were estimated to be around 60 KD and 66-70 KD, respectively. LCC lectins were mitogens for murine splenic lymphocytes, and the optimum mitogenic doses were 31.25mcg/ml and 3.91mcg/ml, respectively. LCC-II lectin was a good mitogen toward human peripheral lymphocytes at a concentration about 31.25mcg/ml.
