        영구치 치수 기질세포를 이용한 연골 분화 및 분화 시기에 따른 형태학적 변화

        Objectives: The aim was to confirm the stem cell-like properties of the dental pulp stromal cells and to evaluate the morphologic changes during in vitro chondrogenesis. Materials and Methods: Stromal cells were outgrown from the dental pulp tissue of the premolars. Surface markers were investigated and cell proliferation rate was compared to other mesenchymal stem cells. Multipotency of the pulp cells was confirmed by inducing osteogenesis, adipogenesis and chondrogenesis. The morphologic changes in the chondrogenic pellet during the 21 day of induction were evaluated under light microscope and transmission electron microscope. TUNEL assay was used to evaluate apoptosis within the chondrogenic pellets. Results: Pulp cells were CD90, 105 positive and CD31, 34 negative. They showed similar proliferation rate to other stem cells. Pulp cells differentiated to osteogenic, adipogenic and chondrogenic tissues. During chondrogenesis, 3-dimensional pellet was created with multi-layers, hypertrophic chondrocyte-like cells and cartilage-like extracellular matrix. However, cell morphology became irregular and apoptotic cells were increased after 7 day of chondrogenic induction. Conclusions: Pulp cells indicated mesenchymal stem cell-like characteristics. During the in vitro chondrogenesis, cellular activity was superior during the earlier phase (within 7 day) of differentiation.

      • 한국인 제2형 당뇨병 환자에 있어 지속적 피하 인슐린 주입법 치료시 Lispro insulin와 Regular insulin의 효과 비교

        Objective: The Lispro insulin has faster onset of effects and shorter duration than the human regular insulin, an analog of human insulin. The differences in the pharmacodynamics between the two types of insulin in the continuous subcutaneous insulin infusion(CSII) with insulin pump for treating the Korean Type 2 DM patients were analyzed. The effects on the regulation of blood sugar, the amount of insulin injected, and the frequency of hypoglycemia were compared between the lispro insulin and the regular human insulin. Method and Material: One hundred and three Korean Type 2 DM patients were randomly divided into two groups, 49 Lispro insulin and 54 Regular insulin.(Table 1) Both groups were treated for 14 days. Also at a set time before meal, insulin was injected as bolus, using a portable insulin pump, in both groups, 15 minutes before for lispro insulin and 30 minutes before for human insulin The HbA1C, C-peptide, AC/PC, height and weight were measured for each patients at the time of admission. The peripheral blood sugar level and insulin injection profile were recorded 7 times per day for each patients. Results: After treating for 7 days and 14 days, the blood sugar level normalized in both groups. There weren't any significant differences in blood sugar levels in both groups at point of the study, and also there weren't any differences in the frequency of hypoglycemia. In order to maintain the normalized blood sugar level, the lispro insulin group had higher total daily insulin requirement than the regular insulin group after 14 days of treatment (0.90±0.35 vs 0.73±0.39u/kg/day, p<0.05) This difference is probably due to the difference in basement insulin requirements rather than prandial insulin requirements (0.32±0.1 vs 0.43± 0.09u/kg/day, p<0.001). Discussion: In the hospitalized Korean Type 2 DM patients with CSII treatment, continuous subcutaneous insulin infusion therapy using lispro insulin has convenience in usage compared to the regular insulin. However, there were not any differences in the blood sugar levels. Also lispro insulin requires more insulin in order to maintain normal blood sugar level, which is probably due to the increase in the basal requirement. Also there were not any difference in the frequency of hypoglycemia.

        Bi₃zsLa(0.75)Ti₃0_(12) (BLT) thin film have been grown on p-type Si(l00) substrates by a sol-gel spin coating process. The structure and surface morphology of the film were analyzed by X-ray diffraction (XRD) and scanning electron microscopy (SEMI. Furthermore, the electrical properties were examined by capacitance-voltage ( C-V) characteristics by using a LF Impedance Analyzer(HP 4192A). XRD measurement show layered perovskite structure with a single phase in BLT thin film annealed at 700C for 30 min in oxygen. The measured grain size of BLT on Si is about 0.04 - 0.1 pn. From the cross-sectional SEM image, BLT film thickness is measured to be 0.4 pm. The C-V characteristic hysteresis curves show that the NIFS (metal-fexroelectric-semiconductor) structure has memory effect. The memory window decreases, with increasing frequency at MFS structure. And it is found that the memory window increases as the applied voltage to the ferroelectric capacitor is increased. These phenomena may be due to polarization reversals of the BLT ferroelectric film and because of the charge injection from Si substrate into the ferroelectric thin film. The results obtained indicate that the present BLT films are suitable for making ferroelectric field effect transistor(FeFET) memory.

        Back ground/Aims: In a genome wide association study, two variants in inosine triphosphatase (ITPA: rs1127354, rs7270101) gene showed to protect against ribavirin induced anemia during early stages of treatment. ITPA gene on chromosome 20, rs1127354 and rs7270101, cause ITPase deficiency and protect from hemolytic toxicity of RBV. In this present study, we aimed to evaluate the clinical significance of ITPA variants who were treated with peg-interferon and ribavirin in Korea. Methods: A total 133 patients who treated with Peg-interferon and ribavirin from January 2008 to December 2011 in Gil Hospital were genotyped at polymorphic site rs1127354 and rs7270101 on chromosome 20. Then we tested for association with Hgb reduction at week 4, 8, 12 and SVR. We investigated the impact of ITPA variants on hemoglobin level and sustained virologic response (SVR). Results: Total of 82 (61%) were genotype 1, and 51 (39%) were non-genotype 1. There was no significant difference in the age, gender, BMI, initial Hgb, ALT, HCV RNA, liver cirrhosis or not between each group. Among 133, 108 patients were CC at rs1127354 (group A) and 25 patients were non-CC at rs1127354 (group B). On the other hands, all of 133 were AA at rs7270101. The mean value of Hgb for group A and B at week 4, 8, and 12 were 12.2 vs. 14.0, 11.8 vs. 13.2, and 11.5 vs. 12.9, respectively (p < 0.05). Overall SVR was achieved in 67.8% (40/59) at genotype 1 and 75% (27/36) at non-genotype 1. As aspects ITPA variants, SVR was achieve in 55% and 50% at genotype 1 (p=0.441), 71% and 66% (p=0.861) at non-genotype 1. SVR was no significant statistical difference between group A and group B. Conclusions: Non-CC at rs1127354 without involvement of those at rs7270101 is strongly associated with protection from RBV-induced anemia. However, ITPA genotype was not associated with SVR.

        Background: Autoimmune hepatitis (AIH) is a progressive inflammatory liver disease of unknown etiology that is reported to be a consequence of aberrant autoreactivity. Several studies which reported the acute presentation of AIH have different clinical course and histologic features. In this study, we compared acute presentation of AIH and chronic presentation of AIH. Methods: We retrospectively reviewed the medical records of patients with autoimmune hepatitis from January 2003 to June 2011 at Gachon University, Gil Hospital. A total of 29 patients were enrolled, 7 patients were diagnosed with acute presentation of AIH. Results: There was no difference between two group in age, gender, and score system of AIH. Patients with acute presentation had higher serum levels of total bilirubin, lower serum levels of albumin in clinical feature (p<0.05), and higher frequency of zone 3 necrosis in histologic feature. The cumulative incidental rate of the normalization of serum AST and ALT levels with prednisolone treatment was similar between patients with acute presentation and chronic presentation in clinical course. Conclusions: Higher AST, ALT and, bilirubin were clinical specific feature, and zone 3 necrosis is a histological characteristic of autoimmune hepatitis with acute presentation.

        Effects of α-lipoic acid on LPS-induced neuroinflammation and NLRP3 inflammasome activation through the regulation of BV-2 microglial cells activation

        Microglial cells are known as the main immune cells in the central nervous system, both regulating its immune response and maintaining its homeostasis. Furthermore, the antioxidant α-lipoic acid (LA) is a recognized therapeutic drug for diabetes because it can easily invade the blood-brain barrier. This study investigated the effect of α-LA on the inflammatory response in lipopolysaccharide (LPS)-treated BV-2 microglial cells. Our results revealed that α-LA significantly attenuated several inflammatory responses in BV-2 microglial cells, including pro-inflammatory cytokines, such as tumor necrosis factor-α and interleukin (IL)-6, and other cytotoxic molecules, such as nitric oxide and reactive oxygen species. In addition, α-LA inhibited the LPS-induced phosphorylation of ERK and p38 and its pharmacological properties were facilitated via the inhibition of the nuclear factor kappa B signaling pathway. Moreover, α-LA suppressed the activation of NOD-like receptor pyrin domain containing 3 (NLRP3) inflammasomes, multiprotein complexes consisting of NLRP3 and caspase-1, which are involved in the innate immune response. Finally, α-LA decreased the genes accountable for the M1 phenotype, IL-1β and ICAM1, whereas it increased the genes responsible for the M2 phenotype, MRC1 and ARG1. These findings suggest that α-LA alleviates the neuroinflammatory response by regulating microglial polarization. [BMB Reports 2019; 52(10): 613-618]

      • Prevalence and characterization of <i>Salmonella</i> in two integrated broiler operations in Korea

        <P><B>Background</B></P><P>Vertical integration of the broiler industry allows producers to combine different biosecurity and sanitation practices, housing technologies, and feeding regimens to improve food safety. The purpose of this study was to investigate the prevalence and distribution of <I>Salmonella,</I> to determine the source of <I>Salmonella</I> contaminants, and to reveal the relationships between isolates at each step in the vertically integrated broiler production system in two representative integrated broiler companies in Korea.</P><P><B>Results</B></P><P>A total of 2148 samples were collected from 2 broiler breeder hatcheries, 14 broiler breeder farms, 3 broiler hatcheries, 16 broiler farms, 8 broiler transporting trucks and 6 slaughterhouses belonging to representative integrated broiler companies, and 205 (9.5%) of these samples were positive for <I>Salmonella</I>. The <I>Salmonella</I> prevalence in broiler hatcheries (34.0%) and broiler transporting trucks (62.5%) was higher (<I>P</I> < 0.05) whereas that in the broiler breeder hatchery (0.8%) was lower (<I>P</I> < 0.05), than the overall prevalence. Nine and 13 different <I>Salmonell</I>a serotypes were isolated from integrated companies A and B, and the predominant serotypes were <I>S.</I> Virchow (39.7%) and <I>S.</I> Hadar (59.2%), respectively. Pulsed field gel electrophoresis patterns of isolates from the two operations showed significant genetic relatedness within a single system.</P><P><B>Conclusions</B></P><P>In a comparison of the two operations that participated in this study, the prevalence of <I>Salmonella</I> differed significantly between the broiler breeder hatchery, and broiler hatcheries and broiler farms.</P>

        Similarities of Aspects of Biological Rhythms between Major Depression and Bipolar II Disorder Compared to Bipolar I Disorder: A Finding from the Early-Onset Mood Disorder Cohort

        Objective The biological rhythm is closely related to mood symptoms. The purpose of this study was to assess the differences in biological rhythms among subjects with mood disorder [bipolar I disorder (BD I), bipolar II disorder (BD II), major depressive disorder (MDD)] and healthy control subjects. Methods A total of 462 early-onset mood disorder subjects were recruited from nine hospitals. The controls subjects were recruited from the general population of South Korea. Subject groups and control subject were evaluated for the Korean language version of Biological Rhythms Interview of Assessment in Neuropsychiatry (K-BRIAN) at the initial evaluation. Results The mean K-BRIAN scores were 35.59 [standard deviation (SD)=13.37] for BD I, 43.05 (SD=11.85) for BD II, 43.55 (SD=12.22) for MDD, and 29.1 (SD=8.15) for the control group. In the case of mood disorders, biological rhythm disturbances were greater than that in the control group (p<0.05). A significant difference existed between BD I and BD II (BD I<BD II, p<0.001) and between BD I and MDD (BD I<MDD, p< 0.001) but no difference was observed between BD II and MDD. Conclusion BD II and MDD are similar to each other but different from BD I in biological rhythm patterns in early-onset mood disorder cases. Biological rhythm disturbances are similar for early-onset major depression and BD II.
