Isoquercitrin 함유 연꽃잎 추출물의 피부주름개선 효능 연구

본 연구에서는 제1형 프로콜라겐 분비량 측정을 통해 피부 주름개선 효과가 있는 천연물을 검색하던 중, 연꽃잎추출물(Nelumbo nucifera leaves extract, NLE)이 가장 우수한 효능을 나타냄을 확인하였다. 고압액체크로마토그래피로 NLE를 분석한 결과 isoquercitrin이 함유되어 있었으며, isoquercitrin도 normal human dermal fibroblast (NHDF)에서 제1형 프로콜라겐의 분비를 촉진한다는 것을 확인할 수 있었다. 또한 NLE와 isoquercitrin은 자유 라디컬 소거 활성을 가지고 있었으며, 특히 isoquercitrin은 인간 피부유래 세포주인 HaCaT 및 NHDF에서 활성산소종의 생성을 억제하고 총항산화능을 증가시켰다. 최종적으로 본 연구팀은 isoquercitrin의 함유량을 높이기 위해 정제한 NLE를 1.7% 함유한 크림(isoquercitrin 51 ppm 함유)을 제조하여 눈가의 주름개선 효과를 평가하였다. 30 ~ 65세의 한국인 여성 22명을 대상으로 얼굴의 한쪽 면엔 NLE 함유 크림을, 그리고 다른 한쪽 면엔 대조제품을 매일 2회씩, 8주간 연속 사용하도록 하였다. 그 결과 NLE 함유 크림을 사용하였을 때 피부자극이 발견되지 않았으며, 대조제품과 비교하여 통계적 유의성 있게 눈가의 주름을 감소시키는 것을 확인할 수 있었다. 본 연구결과는 NLE 및 isoquercitrin이 우수한 콜라겐 생성 촉진 및 항산화 효능을 가지며, isoquercitrin 함유 NLE는 안전한 천연 주름개선 화장품 소재로 적용 가능하다는 것을 시사한다. In this study, we conducted research to find anti-wrinkle skin care ingredients from natural sources via the measurement of procollagen type 1 levels in the medium of normal human dermal fibroblast (NHDF) and found that a Nelumbo nucifera leave extract (NLE) was as the best effective ingredient. By the high performance liquid chromatography analysis, we confirmed isoquercitrin was one of the main compounds of NLE. Moreover, it also increased the procollagen type 1 production without cytotoxicity of NHDF. NLE and isoquercitrin exerted free radical scavenging activity. Especially, isoquercitrin exhibited strong intracellular antioxidant capacity in human skin-derived cells, HaCaT and NHDF. Finally, we performed clinical test for the inhibitory effect of NLE on skin wrinkle formation. A randomized study was conducted in 22 healthy female volunteers, aged between 30 and 65 yrs, with moderate to moderately severe facial wrinkles. Volunteer applied a 1.7% NLE cream (isoquercitrin 51 ppm) and a control cream at each facial side (left/right) twice daily for 8 weeks. The 1.7% NLE cream improved skin roughness through reducing the wrinkle of the craw’s feet significantly without any skin side effect. Our results demonstrate that NLE and isoquercitrin can increase the collagen production and exert antioxidant activity. Therefore, we expect that the new non-toxic herbal extract, isoquercitrin-containing NLE will be interesting natural ingredient of cosmetics with anti-wrinkle efficacy.

S-533 A Case of Posterior Reversible LeukoEncephalopathy Syndrome in a Woman with Renal Artery Stenosis

Posterior reversible encephalopathy syndrome (PRES) is characterized by a clinical and radiological entity with seizure, headache, hypertension, visual loss and encephalopathy with neuroimaging findings of reversible vasogenic subcortical edema. The co-morbid conditions is hypertension, renal disease, autoimmune disease, eclampsia and transplantation. A 42-year-old woman was admitted to our hospital with complaint of sudden onset of seizure. Her blood pressure is 210/110mmHg and mental is drowsy state. After this episode she compained of blindness for a few days. Brain MRI showed multifocal edema in bilateral occipital lobe, frontoparietal lobe. In several hypertensive work-up studies, renal doppler ultrasonography showed stenosis of right renal artery. Segmental stenosis in right renal artery was demonstrated by renal angiography. We performed stent insertion in renal artery narrowing portion. Her blood pressure was maintained less than 140/80mmHg without the antihypertensive drugs. PRES was induced by renal hypertension and it’s cause was right renal artery stenosis. Here we report a rare case of renal artery stnenosis with complication of PRES.

Morinda citrifolia Extract Prevents Alcoholic Fatty Liver Disease by Improving Gut Health

Alcoholic liver disease (ALD) is a major chronic liver disease. Chronic alcohol consumption induces dysbiosis, disruption of gut barrier function, oxidative stress, inflammation, and changes in lipid metabolism, thereby leading to ALD. In this study, we investigated whether the commercial Morinda citrifolia extract Nonitri can ameliorate ALD symptoms through the gut–liver axis. We used mice chronically administered EtOH and found a marked increase in serum endotoxin levels and biomarkers of liver pathology. Moreover, the EtOH-treated group showed significantly altered gut microbial composition particularly that of Alistipes, Bacteroides, and Muribaculum and disrupted gut barrier function. However, Nonitri improved serum parameters, restored the microbial proportions, and regulated levels of zonula occludens1, occludin, and claudin1. Furthermore, Nonitri suppressed inflammation by inhibiting endotoxin-triggered toll-like receptor 4-signaling pathway and fat deposition by reducing lipogenesis through activating AMP-activated protein kinase in the liver. Furthermore, Pearson's correlation analysis showed that gut microbiota and ALD-related markers were correlated, and Nonitri regulated these bacteria. Taken together, our results indicate that the hepatoprotective effect of Nonitri reduces endotoxin levels by improving gut health, and inhibits fat deposition by regulating lipid metabolism.

Genus-Specific Distribution and Pathovar-Specific Variation of the Glycinecin R Gene Homologs in Xanthomonas genomes

Xanthomonas axonopodis pv. glycines produces bacteriocins called glycinecin, and two glycinecin genes, glyA and glyR, were reported previously. In this paper, we describe genomic distribution and variation of the glyR gene revealed by extensive Southern hybridization analysis. In contrast to the glyA gene present only in X. axonopodis pv. glycines, the glyR gene was found to be distributed widely in all the pathovars of Xanthomas genus. It was also found that the glyR gene is a multigene family while the glyA is a single copy gene. Moreover, the copy number and the variation of the glyR multigene are unique to each pathovar of Xanthomonas. The uniqueness can be easily detected by the patterns resulted from Southern hybridization using the genomic digests. Thus, we suggest the glyR gene can serve as a useful genus-specific and pathovarspecific DNA marker for Xanthomonas. One of the glyR homologs was further isolated from X. axonopodis pv. glycines, and analyzed to be functional with strong inhibitory activity against several members of Xanthomonas.

To reveal the spatial and temporal variability in the distribution, growth, and maturation of skipjack tuna Katsuwonus pelamis in the western tropical Pacific, we compared two El Nino-Southern Oscillation (ENSO) signals and the sea surface temperature (SST) in the main fishing area with fishery and biological data. An index of skipjack tuna distribution was calculated using Korean purse seine fishery data from 1985 to 2003. Biological data for skipjack tuna were collected monthly from Korean catches during the 1994-2003 period. The catch was more closely related to the SST in the main fishing area than to the ENSO signals. However, cross-correlated function analysis showed delayed interactions between abiotic and biotic factors. The El Nino events preceded the eastward movement of the fishing center by 2-3 months. El Nino had a positive effect on the skipjack tuna catch, and the change in the Southern Oscillation Index (SOI) preceded the catch fluctuation by ca. 5-7 months. In addition, negative El Nino effects on gonad maturation and the mean length of skipjack tuna were detected with time lags of 12 and 7 months, respectively. The length frequency indicated that the regime-specific growth pattern at each discrete period seemed to be related to the ENSO.

Suppression of IgE production and modulation of Th1/Th2 cell response by electroacupuncture in DNP-KLH immunized mice

Effects of electroacupuncturc (EA) on Th1/Th2 cell response were investigated in BALB/c mice immunized intraperitoneally with 2,4-dinitrophenylated keyhole limpet protein (DNP-KLH). Successive electroacupuncture stimulation on the ST36 acupoint was performed just after immunization. Serum levels of antigen-specific lgE and total lgE were significantly decreased compared with non-acupunctured controls. Production of the Th2-specific cytokines IL-4 and IL-13 in the anti-CD3 mAb-activated splenocytes was significantly suppressed in ST36 electroacupunctured mice compared with non-acupunctured mice. These results imply that successive electroacupuncture on ST36 can decrease the serum level of antigen-specific lgE and total lgE by suppression of the Th2 lineage development.

코로나19 사태에서 사회복지사가 인식하는 심리방역 자원으로서의 음악치료

이 연구는 코로나19 사태 속에서 사회복지사가 심리방역 자원으로서의 음악치료를 어떻게 인식하고 있는지 파악하고자 Colaizzi의 현상학적 연구 방법을 적용하였다. 연구 참여자는 음악치료서비스를 담당한 경험이 있는 사회복지사 5명이었고, 자료 수집 및 분석은 2020년 6월부터 2021년 5월까지 진행하였다. 연구 결과는 ‘심리방역 자원으로서의 음악치료’, ‘프로그램 중재자로서 느끼는 한계’, ‘코로나19 상황에서 음악치료를 위한 제안’이라는 3가지 범주로 분석되었다. 결과를 통해 사회복지 현장에서 음악치료서비스가 긍정적인 효과가 있다는 경험을 바탕으로 심리방역 자원으로 음악치료가 활용되기를 바라는 인식을 확인할 수 있었다. 그러나 사회적 거리 두기와 방역지침으로 치료서비스가 중단됨에 따라 연구 참여자들은 치료사 역할을 대신 수행하며 역량의 한계가 있음을 경험했다. 이는 사회복지사와 음악치료사간의 긴밀한 교류 및 협업의 필요성을 나타내고, 사회복지 현장의 음악치료 적용에서 체계의 재구성에 관한 시사점을 제공한다. 이 연구는 코로나19와 같은 위기 상황에서 심리방역 자원으로서 음악치료서비스의 대면/비대면 적용 가능성에 관한 사회복지사의 인식을 탐색해본 것에 의의가 있다. This study aimed at identifying how social workers perceive music therapy as a resource for psychological prevention under the COVID-19 global pandemic by applying the Colaizzi phenomenological research method. The participants in the study were five social workers with experience in music therapy services, and the collection and analysis of relevant data were conducted from June 2020 to March 2021. The collected data fell into three categories; ‘music therapy as a resource of psychological prevention’, ‘limitations felt as a program mediator’ and ‘proposals for music therapy’ under the COVID-19 Pandemic. The results confirmed the perception that music therapy is useful as a psychological prevention resource based on the experience that music therapy services have a positive effect in social welfare settings. On the other hand, as treatment services were discontinued due to social distancing and quarantine guidelines, the study participants experienced limitations in their capabilities and took on the role of therapists instead. This indicates the necessity of a close exchange and collaboration between social workers and music therapists and provides inputs for the reorganization of the system for applying music therapy in social welfare settings. This study is meaningful in exploring social workers’ perception of the possibility of face-to-face/non-contact application of music therapy services as psychological prevention resources in crises such as the COVID-19 global pandemic.
