        Ergostatrien-3β-ol (EK100) from Antrodia camphorata Attenuates Oxidative Stress, Inflammation, and Liver Injury In Vitro and In Vivo

        Hepatic ischemia/reperfusion (IR) injury is a complication that occurs during liver surgery, whereby hepatic tissue is injured by oxygen deficiency during ischemia, then further damaged by a cascade of inflammatory and oxidative insults when blood is resupplied during reperfusion. Antrodia camphorata is an indigenous fungus in Taiwan and an esteemed Chinese herbal medicine with various bioactivities. This study examined the effect of ergostatrien-3β-ol (EK100), an active compound found in both the fruiting body and mycelia of A. camphorata, on IR injury pathologies in rats and cell models of oxidative and inflammatory stress. Male Sprague-Dawley rats were randomly assigned to receive a vehicle or 5 mg/kg EK100 prior to hepatic IR injury induced by 1 h ischemia followed by 24 h reperfusion, or a sham operation. RAW 264.7 murine macrophages and HepG2 hepatocytes were pretreated with EK100, then inflammation was induced with lipopolysaccharides in the former and oxidative stress was induced with hydrogen peroxide in the latter. EK100 decreased IR-induced elevation in serum levels of alanine aminotransferase and aspartate aminotransferase and lowered levels of the inflammatory cytokines tumor necrosis factor-α, interleukin (IL)-6, and IL-1β. In addition, EK100 significantly reduced hepatic mRNA levels of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2, as well as nitrite production and iNOS gene expression in both hepatocyte and macrophage cell lines. We demonstrated that EK100 exhibits potent protection against hepatic IR injury, which may be used to design strategies to ameliorate liver damage during liver surgery.

        Multiple Impairments in Male Reproduction 1 (mimr1), a Novel Male-Sterile Mutant of Arabidopsis thaliana, Shows Several Defects in Male Reproductive Development

        We have characterized a new male-sterile mutant in Arabidopsis that exhibits conditional sterility but has restored fertility when drought-stressed. This mutant,multiple impairments in male reproduction 1 (mimr1),shows pleiotropic defects in both vegetative and reproductive development. Examination with dissecting and scanning electron microscopes revealed that its pollen grains are not effectively released from the anther locule after dehiscence, and anther differentiation is defective. Growth of the style and stamen filaments are also abnormal. Histological analysis demonstrated that these phenomena are due not only to a noticeably reduced extension of the stamen but also greater elongation of the pistil. Genetic analysis indicated that mimr1 is a single locus recessive nuclear mutant. The mutation can be mapped to a locus strongly linked to a 1200-kb region on Chromosome 3. Meta-analysis of expression patterning presented several candidate genes in that region. No mutants with similar phenotypes have previously been reported, suggesting that mimr1 is a novel male-sterile locus. Characterization of MIMR1 will provide further insights into the molecular basis for the development of plant reproductive organs.

        The Moderating Effect of Internal Control on Performance of Cross-Border M&A under the Uncertainty of Economic Policy: Evidence from China

        Purpose - The purpose of this paper is to investigate the relationship between internal control, economic policy uncertainty, and performance of cross-border merger and acquisition (M&A) based on the panel data of Chinese listed firms. The authors expected that internal control has a positive moderating effect on the performance of cross-border M&A and that it mainly occurs during periods when economic policies are relatively stable. In addition, the authors tried to find out the mechanism of internal control affecting cross-border M&A and the corporate performance. Design/methodology - The authors tested the hypotheses by a multivariate regression model based on the panel data of Chinese listed firms from 2009 to 2017. The dependent variable is the change value of business performance (DROA_1,2,3) and the explanatory variables are cross-border M&A (MA), China’s uncertainty of economic policy (EPU), and internal control level (IC) respectively. Findings - The authors find that internal control has a positive moderating effect on the relationship between cross-border M&A and corporate performance. Further, the authors find that the moderating effect is more significant in state-owned enterprises and that it mainly occurs during periods when economic policies are relatively stable. Originality/value - This paper is the leading study that tries to analyze empirically the relationship between internal control, economic policy uncertainty, and performance of cross-border M&A. It provides a new avenue through which internal control might reasonably mitigate the risks of crossborder M&A and correspondingly improve the performance of cross-border M&A. It also confirms the moderating effect of internal control on the performance of cross-border M&A under the uncertainty of economic policy.

        Initial Report on the Oriented-precipitation of T1-Phase in Creep-Aged Al-Cu-Li Single Crystal

        In this work, the precipitation behavior of the T1-phase on each {111}Al habit plane was characterized by transmissionelectron microscopy (TEM), which was observed along two < 110 > Al zone axes in an Al-Cu-Li single crystal during creepaging. The results indicate that the oriented-precipitation of T1-phase in the Al-Cu-Li single crystal was originally determinedduring creep aging. The number densities of the T1-phase on the habit planes of (111)Al and (1−11)Al is noticeablyhigher than those of the T1-phase on the habit planes of (−111)Al and (11−1)Al in the same Al-Cu-Li single crystal when theexternal stress is loaded along the direction close to [111]Al orientation. It implies that the T1-phase tends to preferentiallyprecipitate on the habit planes that are perpendicular to the loading direction during creep aging. This work provides theinitial experimental evidence of the oriented-precipitation of the T1-phase in Al-Cu-Li alloy, and proposes the possibilityof regulating the precipitation distribution of the T1-phase.

        Development of a Serum-Free Culture Method for Endothelial Cells of the Stria Vascularis and Their Pro-inflammatory Secretome Changes Induced by Oxidative Stress

        Objectives. Reactive oxygen species in the stria vascularis (SV) of the cochlea may be involved in the pathogenesis of sen-sorineural hearing loss. However, the effects of oxidative stress on SV endothelial cells (SV-ECs) remain largely un-known, and no feasible in vitro cell culture model exists for the functional study of SV-ECs. Methods. We isolated primary SV-ECs from the SV of neonatal mice. The apoptosis-reducing effects of fibronectin in SV-ECs cultured with serum-free medium were determined using β-galactosidase staining and flow cytometry. SV-ECsincubated in serum-free medium were treated with various H2O2 concentrations to evaluate the effects of H2O2 ontheir viability. The secretome of SV-ECs treated with or without H2O2 (100 μM or 500 μM) was analyzed using high-resolution mass spectrometry. The function of the SV-EC secretome was evaluated by a macrophage assay. Results. We successfully isolated and characterized the SV-ECs. Treatment with H2O2 at concentrations up to 500 μM for2 hours and further incubation with serum-free medium in plates precoated with fibronectin showed no significanteffect on apoptosis. Compared to the control SV-ECs, the amount of differential proteins in the secretome of SV-ECsstimulated with 500 μM H2O2 was much higher than in those treated with 100 μM H2O2. Kyoto Encyclopedia ofGenes and Genomes and Gene Ontology analyses suggested that the proteins differentially expressed in SV-ECstreated with 500 μM H2O2 were involved in the regulation of multiple signaling pathways and cellular processes. Thesecretome of H2O2-stimulated SV-ECs exhibited significant pro-inflammatory effects on macrophages. Conclusion. We successfully established an in vitro serum-free culture method, identified the differential proteins releasedby oxidative stress-induced ECs and their functions, and revealed the pro-inflammatory effects of the secretome ofH2O2-stimulated SV-ECs. Therefore, SV-ECs might elicit immunoregulatory effects on bystander cells in the microen-vironment of oxidative stress-induced cochlea, especially cochlear macrophages.

        Removal of phenol from gas streams via combined plasma catalysis

        A hybrid system consisting of non-thermal plasma and perovskite-like catalyst is developed andevaluated for the effectiveness in removing phenol from gas streams. For thermal catalysis,La0.8Sr0.2Mn0.8Cu0.2O3 shows high activity for phenol removal. Further, La0.8Sr0.2Mn0.8Cu0.2O3 is appliedfor combined plasma catalysis (CPC). The results indicate that phenol removal efficiency with CPCremains 100% at applied voltage range of 13–16 kV. Importantly, secondary pollutants (O3 and NOx) andenergy efficiency can be inhibited and increased, respectively, as CPC is applied. Overall, this studydemonstrates that combining non-thermal plasma with perovskite-like catalyst is effective in removingphenol from gas streams.

        Semi-Lagrangian Reproducing Kernel Particle Method for Slope Stability Analysis and Post-Failure Simulation

        Slope stability analyses are often performed using Limit Equilibrium Methods (LEMs) and Finite Element Method (FEM). However, these methods can only model the slope condition up to the point of failure. Meshfree methods, which do not require amesh or a grid in the simulation process, have the potential to model the post-failure slope behavior as mesh tangling would not occurto cause numerical instability and non-convergence. Hence, while retaining the benefits of conventional numerical schemes,meshfree method can be more advantageous when problems with large deformation are encountered. In this paper, Semi-LagrangianReproducing Kernel Particle Method (SLRKPM), which is a type of meshfree method, is extended to analyze geomechanicsproblems such as the stability of a slope and post failure slope behavior. The results from SLRKPM agree well with those fromconvention methods (LEMs and FEM) in terms of factor-of-safety and failure surface. In addition, SLRKPM is able to simulate theslope failure process and successfully capture the development of shear band. This proves that SLRKPM has a significant advantageover FEM when dealing with problems involving large deformation and failure of geomaterials.

        Inhibition of cancer cell growth and migration by dihydroxynaphthyl aryl ketones

        Dihydroxynaphthyl aryl ketones 1-5 exhibit activity as tubulin polymerization inhibitors by targeting the colchicine binding site of microtubules making them potential anticancer drugs. Therefore, analogues 1-5 have been evaluated for their cytotoxic activity against the cancer cell lines DU-145 (prostate), T24 (bladder) and MCF-7 (breast). notable differences in biological activity were observed for compounds 1-5, most likely related to the nature of the aryl substituent bonded to the carbonyl group. among the tested compounds, only compound 5 showed selectivity for cancer cells over healthy, non-transformed cells. T24 cancer cells treated with compound 5 presented a concentration-dependent decrease in cell proliferation and a loss of migration ability. The cytotoxicity of compounds 1-5 on the selected cell-based assays is discussed in terms of it lipophilicity and polarizability parameters.
