      • Bromocriptine의 Prolactin 분비억제에 관한 연구

        Prolactin is a pituitary hormone that plays several roles in the regulation of breastmilk secretion, reproductive function, interaction with steroid hormone. Hyperprolactinemia is an important cause of amenorrhea and female infertility with or without galactorrhea. In this study, prolactin release from pituitary cells was suppressed by the treatment of bromocriptine, however, prolactin mRNA levels as well as intracellular prolactin content were not suppressed significantly. Based on the result of this study, bromocriptine, a drug for hyperprolactinemia, was effective in the suppression of prolactin release of rat.

        대사증후군 구성요소와 우울증, 스트레스 사이의 관계

        The purpose of this study is to provide an academic basis regarding the necessity of managing depression and stress among metabolic syndrome patients by understanding 5 components of metabolic syndrome, perceived stress, and degree of depression, and by investigating their association using the national nutrition survey reference. This study was conducted by using mental health surveys and health screening test data of the 5th (2010∼2012) primitive data of the national health and nutrition survey. A total of 19,599 respondents over 19 years of age were selected for the final analysis. The level of depression and stress was set as the dependent variable to identify its connection with 5 components of the metabolic syndrome. For the stress recognition, none of the metabolic syndrome components showed a significant correlation. For experiencing the depression symptom, the fasting glucose among the factors showed a significant correlation (p <0.05) among the metabolic syndrome factors. When it falls within the criteria of fasting glucose of metabolic syndrome, it has a great probability of falling under the group who experienced greater depression symptoms. As a result of the analysis by controlling cardiovascular and cerebrovascular disease which is tightly related with metabolic syndrome and depression, this study observed that glucose out of 5 metabolic syndrome components is related with depression.

        Clinical laboratory testing plays a crucial role in the detection, diagnosis, and treatment of disease. Clinical laboratory scientists evaluate test results, develop and modify procedures, and establish and monitor programs, to ensure the accuracy of tests. It is clear that over the past decade the role of the clinical laboratory scientists has expanded and this process will be continued into the future. The purpose of this study is to clarify the factors influencing the job performance of clinical laboratory scientists and aim to provide the role and duties of clinical laboratory scientists. To examine the frequency of work context and the importance of task, the surveys were conducted on 168 clinical laboratory scientists. The questionnaire items were used 10 score by Likert scale. According to the results of this study, the important factor affecting the job performance was “Analyze laboratory findings to check the accuracy of the results”, and the next factors were “Establish and monitor quality assurance programs”, “Enter data from analyzer into computer”, “Calibrate and maintain equipment”. And the factors of “repeating same tasks” and “spend time making repetitive motions” were answered the highest. This study will provide strategies for improving work environment, developing education curriculum and method, and role of clinical laboratory scientists. We must continually upgrade our knowledge, and identify the new trends in technology and science and accept changes. .

        Maintenance of Platelet Counts with Low Level QC Materials and the Change in P-LCR according to Hemolysis with XN-9000

        The platelet count in clinical laboratories is essential for the diagnosis and treatment of hemostasis abnormalities, and accurate platelet counting in the low count range is of prime importance for deciding if a platelet transfusion is needed and for monitoring after chemotherapy. Quality control is designed to reduce and correct any deficiencies in the internal analytical process of a clinical laboratory prior to the release of patient results. Fragmented erythrocytes are the major confusing factors for platelet counting because of their similar size to platelets. The authors found that the low range QC values were out of 2SD with a Sysmex automatic analyzer in internal quality control process. Thus far, there has been little discussion on the relationship between hemolysis and the platelet parameters. Therefore, this study focused on the performance of automated platelet counts, including the PLT-F, the PLT-I, and PLT-O methods at the low platelet range using the low level QC materials and compared the 5 platelet parameters with the hemolyzed samples. The results showed that the CV was the smallest with PLT-F and P-LCR increased from 18.4 to 31.9% in the hemolysis samples. These results indicate that a more accurate estimation of the platelet counts can be achieved using the PLT-F method than the PLT-I method at the low platelet range. The use of the PLT-F system improves the confidence of results in low platelets samples in a routine hematology laboratory. The results suggest that P-LCR is a new parameter in assessing samples when the specimen is suspected of hemolysis and deterioration. Nevertheless, further studies will be needed to establish the relationship with P-LCR and hemolysis using human blood specimens.

        Leukemia in pregnancy was first reported by Virchow in 1845, and acute Leukemia that occurs with pregnancy is extremely rare. About 350 pregnancies with leukemia have been reported in literature. The incident of acute leukemia during pregnancy has been reported in one case per 100,000 pregnancies case. A 40-year-old patient with 30 weeks of pregnancy, (by promyelocyte which is contained granules and auer rods in the bone marrow and biopsy) was diagnosed with acute promyelocyte leukemia WITH t (15;17) (q22;q12); PML-RARA. (M3) in peripheral blood and bone marrow examination, and gave a birth to the fetus normally, January 24, 2013, after receiving the complete remission decision from the bone marrow, complete blood cell count, PML-RARA PCR test, showed normal findings until March 2013. The treatment of acute leukemia during pregnancy should be considered as treatment of a pregnant mother and the impact on the fetus. Decisions about when and how birth takes place is difficult and has to consider both mother and fetus. It is preferable to start immediate treatment without delay so that the treatment time to achieve complete remission or full recovery of the pregnant mother is longer. 가톨릭대학교 성빈센트병원 진단검사의학과

      • Evaluation of Enzymatic Method using an Automated Chemistry Analyzers for Homocysteine Measurement

        . In clinical practice, homocysteine has gained popularity because its elevated values are strongly associated with an increased risk of cardiovascular disease. More recently, a new enzymatic colorimetric assay for homocysteine in biological sample, suitable for automated clinical analyzers, has been proposed. To evaluate one of these enzymatic methods and compare the results obtained with this method with those of an immunoenzymatic method, thirty-two samples were analyzed for total homocysteine by HiSensⓇ homocysteine reagent on the automated chemistry analyzers TBA 200FR and compared to the widely used immunoenzymatic method ADVIA Centaur. In TBA 200FR, the within-run CVs of two control materials were 3.23% and 0.92%, respectively; the between run CVs were 4.58% and 2.55%, respectively. And in ADVIA 1650, the within-run CVs were 6.81% and 0.99%, respectively; the between run CVs were 9.0% and 3.9%, respectively. The recovery for homocysteine was 100% (60.8 μmol/L), 99.1% (48.64 μmol/L), 96.3% (36.48 μmol/L), 96.1% (24.32 μmol/L), and 92.1% (12.16 μmol/L). The regression equation of TBA 200FR vs. ADVIA Centaur was y=0.9095x–2.5086 (r=0.9632). And the regression equation for the ADVIA 1650 chemistry vs. Immulite 2000 was y=0.8418x + 0.3207 (r=0.9625). In conclusion, this enzymatic method using automated chemistry analyzer for homocysteine assay shows acceptable analytical performance. I suggest that this assay will be suitable for routine analysis.
