        The Effect of Yangkyuk-Sanhwa-Tang on Cytokine Production in the Patients with Cerebral Infarction

        Yangkyuk-Sanhwa-Tang(YST) has been widely used as a formula for the Soyangin cerebral infarction (CI) patients according to Sasang constitutional philosophy. Brain cells produce cytokines and chemokines during the inflammatory process after stroke both in animal models and in patients. Previously, regulation of serum cytokine levels by YSThas been observed in individuals at the acute stage of CI disease, but there have not been other scientific investigations on YST. The author investigated the effect of YST on theproduction of various cytokines using peripheral blood mononuclear cells (PBMCs)from the Soyangin (CI) patients, and Soyangin normal group. The cytokine production was analyzed using enzyme-linked immunosorbent assay (ELISA). The amount of interleukin (IL)-1, IL-1, IL-6, IL-8, and tumor necrosis factor (TNF)- in culture supernatant significantly increased in the LPS-treated cells compared with unstimulated-cells (P < 0.05). However, in LPS-stimulated PBMCs, cytokines level in CI patients group was higher than that of normal group. YST (1 mg/ml) significantly inhibited IL-1, IL-1, and IL-8 production in PBMCs stimulated with LPS (about 85% for IL-1, 87% for IL-1, and 53% for IL-8, P < 0.05), but did not significantly inhibit IL-6 and TNF- production in the CI patients group. We also show that YST significantly increased LPS-induced IL-1, IL-6, and TNF- production in the normal group. Thesedata suggest that YST has a regulatoryeffect on the cytokine production, which might explain its beneficial effect in the treatment of CI.

        Korea makes efforts with all might to strengthen to national competition with "Globalization" as the slogan in recent years. That's because national competition under the globalization period depends on not only the increase of a firm's sales and competition power by the expansion of markets, but also a nation's effective international logistics system. Government regulations to the firm under the globalization area should be lightened, so the nation include the firm to make inroads into domestic market by establishing international logistics system efficiently. That is, it is not too much to say that national competition under the globalization area depends on effective international logistics system. However, for Korea's international logistics system, it is evident that no only inland transportation facilities including harbor facilities, road, and railroad, but also social overhead capital are lack compared to demand. That is more severe in capital bound centering on Seoul and Inchon, and it is situation that traffic congestion and the lack of logistic terminal cause disorder of goods transport. It is needed to change capital bound. Therefore, construction of effective logistics system in this area is the present question. At this point, reorganization of logistics system by constructing Kyong-in canal is reasonable plan for goods transport. Terminal(Seoul/Seohae) which is processed as a canal facility is necessary to deal with demand of logistic terminal in capital bound. It is also necessary to establish transport system for systematic construction of Kyong-in canal, to maintain suitable amount of materials, to install required facilities and to obtain profitable business. So, the purpose of this study is to suggest several points to process effective Kyong-in canal by analyzing cases of operations and facilities in foreign canals, examining necessity of Kyong-in canal in respect of improvement in logistics system and advance of container transport in capital bound and drawing implications. Korea's logistics costs in a year, 1995, appeared to be high, and as I suggest this constitution, transport costs took 55.7% of total costs and management costs of stock had 10.3% of total logistic costs. Therefore, Kyong-in canal propulsion directions are as follow : First, Kyong-in canal should be drived to establish complex logistics facilities contributing to improve goods transport system in capital bound. Second, the vessels which pass through Kyong-in canal should be barge ships in the 9,000 ton class, small container feeder ships in the 2,500 ton class and general cargo boat in the 2,000 ton class. Third, Seohae terminal should function to support the facilities of Inchon harbor which is lack compared to demand. Seohae terminal should be used as export-import container cargo of Kyong-in area and handling harbor of container cargo in the trade with China. Fourth, Seoul terminal should be used as starting point and terminal point of canal use cargo, starting point and terminal point of coastal transport, the path of canal transport and trade cargo with China. Fifth, it is necessary that Kyong-in canal should function as logistic terminal of Seohae and Seoul. That is, Seoul terminal will perform complex function of cargo transport and placement and logistics treatment by having the state-of-the-art cargo handling function. Sixth, the construction of Kyong-in canal is the business that needs considerable investment. Kyong-in canal should be drived to divide into two parts. One is basic facilities and those are canal and lock gate, the facilities that are come alongside the pier, and cargo handling facilities of harbors. The other is supplementary facilities and those are logistics such as warehouse, collection and delivery transport.

        국내 재배종버섯의 분자유전학적 분류

        국내 버섯시장의 급속한 확장과 더불어 버섯산업의 구조 가 대규모 농장들을 중심으로 재편되고 있는 추세이다. 반 면 버섯산업은 그 규모에 비하여 연구역량이 크게 부족한 상황인데 이는 주로 버섯재배농가의 인식부족과 학계의 관심부족에 기인한 바 크다 할 것이다. 따라서 버섯산업의 지속적 발전을 위해서는 버섯관련 연구역량의 강화가 필 요하며, 특히 버섯산업의 근간이 되는 신품종 개발연구 분 야에서 민간부문의 연구소 설립 및 학계의 연구인력 양성 이 시급한 실정이다. 그 예로서 국내 재배종 버섯 품종들 은 일부를 제외하면 거의 대부분이 사실상 외래 도입종이 며, 품종의 체계적 관리가 이루어지지 못하여 종균의 퇴 화, 바이러스 및 세균 감염, 동종이명의 종균 유통등의 문 제가 노출되고 있다. 이에 따라 본 연구에서는 국내에 유 통되는 재배종 버섯 품종의 분자유전학적 분류법을 개발 하여 향후 버섯 신품종 개발 및 버섯종균의 체계적 관리에 도움을 주고자 하였다. 버섯들의 분자유전학적 분류를 위 하여 먼저 ITS, IGS, mitochondrial SSU 와 같은 여러가 지 분자마커들의 DNA 염기서열들을 결정하였다. 그 결과 대부분의 마커염기서열들은 버섯 종 수준의 분류에는 사 용될 수 있었지만 동일한 종 내의 품종간의 구분에는 사용 될 수 없었다. 이러한 문제점을 해결하기 위하여, 일반적 으로 genome 의 20% 이상을 차지하는 transposon 과 small sequence repeat (SSR) 사이의 DNA pattern 과 random amplified polymorphic DNA (RAPD) fingerprinting 법을 도입하여 품종간의 구분을 시도하였 다. 이러한 방법을 통하여 새송이 버섯의 경우 경남농기원 보유 24품종을 분류한 결과, 모든 품종들은 크게 5가지 그룹으로 분류되었으며 이는 각 버섯의 자실체 생육특성 과 잘 일치하였다. 이외에도 인천대 버섯균주은행 보유 재 배종 버섯들과 국내의 대규모 농장 및 종균소 보유 버섯 품종들의 분류도 진행하였으며 그 결과를 DB 화 하였다.

        β-Glucosidase 생성 효모 Rhodotorula sp. GYP-1의 분리 및 특성

        Nine microbial strains were isolated from the byproduct of ginseng processing and field of ginseng cultivation. Two strains among them were confirmed. Phylogenetic analysis of these β-Glucosidase strains confirmed that strain GYP-1 belongs to the Rhodotorula and strain GYP-3-3 belong to genus Brachybacterium. Rhodotorula sp. GYP-1 was finally selected due to its high biomass production. The β-Glucosidase activity of Rhodotorula sp. GPY-1 was assessed at 30 ℃, and Higher than 70% of the enzyme activity was maintained at the temperature range of 20-40℃. Although the optimum pH for the highest enzyme activity was pH 5.0, the enzyme was stable throughout the pH range of 5.0-8.0. In addition, Rhodotorula sp. demonstrated antifungal activity against the ginseng root rot disease caused by Botrytis.

        대중교통망 이동성지표 개발 (네트워크 분석을 중심으로)

        교통은 개인의 이동(mobility)에 기원을 둔다. 교통환경에서 이 같은 개인의 이동을 특정 값으로 제시하려는 이동성연구는 여러 각도에서 시행되어 왔으며, 그 결과로 통행자의 통행 서비스수준을 정량적으로 평가할 수 있는 다양한 방안이 제안되어왔다. 그러나 복합대중교통에 대한, 특히 지역적 범위에서 평가할 수 있는, 이동성연구는 많지 않았다. 본 연구는 복합대중교통망에 대한 이동성지표를 지점 대 지점에서 지역간 범위로 확대하여, 대중교통 승객의 접근시간, 주행시간, 환승시간을 고려한 대중교통 이동성지표를 제안한다. 복합대중교통의 이동성(mobility) 평가를 위하여, 대중교통 이동성을 도보수단까지 포함하였으며, 비교 대안수단은 대중교통의 경쟁수단인 개인교통수단으로 설정하였다. 본 연구에서 제안한 대중교통 이동성 분석지표는 지역간 대중교통시설이 개인교통수단에 비해 어느 정도의 경쟁력을 가지는지(지표 1), 도보수단을 포함하여, 출발지역에서 도착지역까지 개인교통수단에 비해 얼마나 편리한지(지표 2), 이용자의 지역간 통행을 지원할 수 있도록 얼마나 많은 경쟁력 있는 다양한 경로가 제공되는지(지표 3)를 평가할 수 있도록 설정하였다. Transportation is from an individual mobility. Various efforts to propose specific values of the individual mobility have been conducted in diverse transportation environment. However, mobility studies for multimodal public transportation are rare especially on not the range of line but area. This study propose a method to calculate transit mobility indices as expanding mobility analysis from point-to-point to area-to-area, considering access time to transit facility, running time and transfer time of passengers. To extract mobility indices, we included walking as a lowest category of mode and set passenger car as a competitive mode to transit mode. This study propose three public transportation mobility indices as 1) how competitive public transportation facility is offered against passenger car 2) how convenient transit mode including walking is provided against passenger car from origin to destination and 3) how many various paths are presented to support passenger's travel between regions. These indices are tested on the Seoul metropolitan area with 10 lines of urban rail and about 420 lines of bus. In addition, we proposed two political applications of proposed mobility indices to increase public transportation mobility between two regions and to maximize the mobility of study area when a line is added in the area.

        A new free-living marine nematode species of the genus Dracogalerus Allen and Noffsinger (Nematoda: Draconematidae) from a shallow subtidal zone of Jeju Island, Korea

        A new species of free-living marine draconematid nematode, Dracogalerus koreanus n. sp., is described from the shallow subtidal coarse sediments and various invertebrates of Jeju Island, Korea. This is the first record of this genus from Korea. Dracogalerus koreanus n. sp. is morphologically most similar to D. cryptocephalus (Irwin-Smith) in having similar head shape (rostrum broadly rounded anteriorly) and eight cephalic adhesion tubes, but differs by the small number of rounded protuberances on the ventral side of non-annulated tail end (5 vs 6), small number of posterior subventral adhesion tubes in male (5-6 vs 8), intermingled somatic setae (5-6 vs absent), spicules (slightly curved and relatively thick vs strongly curved and very slender), and higher c value in male (8.8 vs 7.5).

        세라티아 배양에 의한 세라티오펩티다아제의 생산에 관한 연구

        세라티아 균주의 배양으로부터 소염제로 사용되는 세라티오펩티다아제의 생산에 관한 연구를 수행하였다. 여러가지 탄소원, 질소원 및 유도제가 효소의 생산에 미치는 영향을 조사하였는데, 탄소원은 효소의 생산이나 세포성장에 좋지 못한 기질이었으며, 특히 citrate의 경우 균체성장량은 포도당과 거의 동일하였으나, 세라오펩티다제의 생산에 저해효과가 있음이 밝혀졌다. 세라티오펩티다아제는 아미노산인 leucine을 첨가해 주었을 때 그 생산되는 양이 크게 향상되었으며 leucine의 최적 농도는 0.03였다. An anti-inflammatory agent, serratiopeptidase, was produced from the culture of the Serratia marcescens. The effects of carbon sources, nitrogen sources and inducers on the production were investigated. Citrate was found to be inhibitory in the production of serratiopeptidase. The enzyme was synthesized in the synthetic medium without inducers, albeit low level of synthesis. But the synthesis was increased by the addition of proteinaceous substrate and leucine. Induction of extracellular proteinase by its end-product was discovered, which is not common in the proteinase synthesis in the bacteria. By the glucose fed-batch culture, we found the possible catabolite repression on the production of serratiopeptidase.
