        대학생이 지각한 진로불확실성에 대한 대처: 목표 참여, 목표 이탈과 자기 평가와의 관계

        The purpose of this study is to investigate how to control the motivation of career goals in the face of increasing vocational uncertainty among college students during the school - work transition period, based on the motivational theory of lifespan development (MTD). For this purpose, data on vocational uncertainty, control strategies, and cognitive self - appraisals were collected and analyzed by average and regression analyses for the third and fourth grade students at four - year national and private universities in K region. First, there was no difference in the perception of career uncertainty according to socio - demographic factors. Second, career uncertainty was not related to goal engagement, but was positively related to goal disengagement. Third, situational cognitive assessment which is evaluated as a challenge rather than a threat to the situation of career uncertain, was positively related with the distancing strategy among the goal disengagement strategies. Fourth, it was found that high personal resource evaluation for the uncertain situation of career was positively related with the goal engagement. Finally, the situational cognitive assessment derived to play the controlling role only in the relationship between career uncertainty and goal disengagement. If the situation is evaluated as a challenge rather than a threat, the higher the perception of the uncertainty of the career, the goal disengagement is higher. Based on the results, the research and practice of career counseling to help university students succeed in their school - work transition. 본 연구의 목적은 생애발달동기이론에 기초하여, 교육-직업 전환기에 있는 대학생들을 대상으로 증가하는 직업적 불확실성에 직면하여 어떻게 자신의 진로 목표 동기를 조절하는지를 조사하는 데에 있다. 이를 위해 K지역 내 4년제 국립 및 사립대학에 재학 중인 3, 4학년을 대상으로 진로불확실성과 조절 전략 그리고 개인의 인지적 자기 평가에 대한 자료를 수집하여 평균분석과 회귀분석을 실시하였다. 연구결과 첫째, 사회인구학적 요인에 따라 진로불확실성에 대한 지각에는 차이가 없었다. 둘째, 진로불확실성은 목표 참여와는 관계가 없었으며 목표 이탈과 정적인 관계로 나타났다. 셋째, 진로가 불확실한 상황에 대해 위협보다 도전으로 평가하는 상황인지평가는 목표 이탈 전략 중 거리두기 전략과 정적인 관계가 있는 것으로 나타났다. 넷째, 진로가 불확실한 상황에 대해 높은 개인자원평가는 목표 참여와 정적인 관계가 있는 것으로 나타났다. 끝으로, 상황인지평가는 진로불확실성과 목표 이탈과의 관계에서만 조절의 역할을 하는 것으로 나타난바, 진로불확실성을 높게 지각할수록 상황에 대해 위협이 아닌 도전으로 평가하게 되면 목표 이탈이 더 높아지는 것으로 나타났다. 연구 결과를 토대로 대학생의 교육-직업으로의 전환 성공을 조력하기 위한 진로 상담의 연구와 실제에 대해 논의하였다.

        H460 인체 폐암세포에서 길경 물 추출물에 의해 유도된 세포사멸에서 자가포식의 역할

        Objectives : Recent studies have suggested that Platycodonis Radix has various pharmacological effects such as anti-cancer, antioxidant, anti-asthma, anti-diabetes, anti-obesity, hepatoprotective, and cardiovascular protection effects. The aim of this study was to investigate the role of water extract of Platycodonis Radix (WPR)-induced autophagy in H460 human lung cancer cells. Methods : H460 cells were treated with WPR and cell viability was calculated by an MTT assay. To evaluate changes in apoptosis- and autophagy-related genes, Western blotting was performed. Two kinds of autophagy inhibitors, 3-Methyladenine (3-MA) and bafilomycin A1, were pretreated to confirm the role of WPR-induced autophagy. Results : WPR reduced the viability of H460 cells in a treatment concentration-dependent manner, which was associated with induction of apoptosis. It was also confirmed that WPR induced autophagy based on the formation of specific intracellular vacuoles and changes in the expression of autophagy-related genes. Interestingly, pretreatment with 3-MA and bafilomycin A1 increased WPR-induced cytotoxicity and apoptosis. Conclusions : WPR induced autophagy at low concentrations and early stages of treatment, but promoted apoptosis at high concentrations and late stages. Moreover, WPR-induced autophagy had a cytoprotective role in H460 cells.

        Generally, The ion exchange membrane, which is a component of VRFB, is important not only for its excellent chemical stability, but also for its excellent electrochemical properties. In this study, we synthesized four cation exchange membrane of multi-block types with different hydrophobic structures based on sulfonated poly(p-phenylene)s in order to investigate the hydrophobic structure which affects durability in VRFB. In addition, among them, we found one hydrophobic structure suitable for VRFB. And to conclude, Its suitable materials demonstrated excellent oxidation stability, outstanding efficiency performance and low vanadium ions permeability through long-term operation of VRFB.

        상황버섯 추출물에 의한 인체혈구암세포의 Telomerase 활성 저하

        Recent studies demonstrated that proteoglycan extracted from Phellinus linteus has a potent anti-tumor and/or anti-inflammatory activity by leading to induction of apoptosis and inhibition of prostaglandin E2production; however, its mechanism of action and its molecular targets on the telomere length regulation in human cancer remain unclear. In this study, we investigated the effect of proteoglycan on the levels of telomere regulatory components and the activity of telomerase in human leukemia U937 cells. Exposure of U937 cells to proteoglycan resulted in growth inhibition and induction of apoptosis as measured by MTT assay and flow cytometry analysis, which was associated with induction of tumor suppressor p53and cyclin-dependent kinase inhibitor p21 (WAF1/CIP1). Proteoglycan treatment markedly inhibited the activity of telomerase, and the levels of and transcription factor c-myc, human telomerase reverse transcriptase (hTERT) and telomerase-associated protein-1 (TEP-1), main determinants of the telomerase enzymatic activity, were progressively down-regulated by proteoglycan treatment in a dose-dependent fashion. Taken together, these findings provide important new insights into the possible molecular mechanisms of the anti-cancer activity of proteoglycan. (Cancer Prev Res 15, 172-177, 2010)

        길경탕의 치밀결합 강화 및 MMPs의 활성 억제를 통한 인체방광암세포의 이동성 및 침윤성의 억제

        Objectives: Gilgyung-tang (GGT) has been used as one of the main multi-herb formulas to treat “Peo-ong” (lung abscess). In this study, we investigated the inhibitory effects of water extracts of GGT on cell migration and invasion, two critical cellular processes that are often deregulated during metastasis, in human bladder cancer 5637 cells.Methods: Effects on cell viability were quantified using an MTT assay. To analyze the anti-metastatic effects, we conducted a wound healing migration assay, an in vitro invasiveness assay, and a measurement of the transepithelial electrical resistance (TER). The expression of protein and mRNA were measured by Western blotting and real-time polymerase chain reaction (RT-PCR), respectively.Results: GGT markedly inhibited the cell motility and invasiveness of 5637 cells within the concentration range that was not cytotoxic. The inhibitory effects of GGT on cell invasiveness were associated with tightening of the tight junctions (TJs), which was demonstrated by an increase in the TER. The RT-PCR and Western blotting results indicated that GGT decreased the levels of claudin proteins. GGT also inhibited the activity and expression of matrix metalloproteinase (MMP)-2 and -9 and simultaneously increased the levels of tissue inhibitor of metalloproteinase-1 and -2.Conclusions: Our findings suggest that GGT reduces both the migration and the invasion of 5637 cells by modulating the activity of TJs and MMPs.

        식중독 세균과 주요 축산식품 및 가공품 조합에 대한 위해순위 결정

        This study was performed to determine risk ranking of the combination of pathogen-livestock or livestock products to identify the most significant public health risks and to prioritize risk management strategies. First, we reviewed foodborne outbreak data related to livestock products and determined main vehicles and pathogens according to the number of outbreak and case. Second, expert’s opinion about management priority of pathogen-livestock product pairing was surveyed with 19 livestock experts in the university, research center, and government agency. Lastly, we used the outcome of Risk Ranger (semi-quantitative risk ranking tool) of 14 combinations of pathogen and livestock or livestock products. We have classified the combination of pathogen-livestock products into group I (high risk), II (medium risk), and III (low risk) according to their risk levels and management priority. Group I, which is the highest risk for foodborne outbreak, includes Salmonella spp./egg and egg products, Campylobacter spp./poultry, pathogenic E. coli/meat and processed ground meat. In conclusion, the results of this study will provide the specific guideline of mid- and long-term planning for risk assessment and risk management prioritization of the combination of pathogen and livestock, or livestock product.

        암의 발생과 증식에서 Nrf2와 Keap1의 기능

        Nuclear factor erythroid-derived 2-related factor 2 (Nrf2) is a transcription factor that orchestrates diverse stress signaling and protects cells from oxidative or electrophilic stress by leading various transcriptional programmes. The studies about this regulatory protein and its negative regulator, Kelch-like ECH-associated protien 1 (Keap1) have been conducted internationally and have awoken many people's concern in a key controversy. The controversy is centred around whether Nrf2 acts as a tumor suppressive function or an oncogenic function, leading to the question of whether Nrf2 could be targeted for chemopreventive and anticancer therapeutic method. In abundant publications of chemopreventive drugs, enhancement of the Nrf2 activity may have beneficial effects on suppression of carcionogenesis and prevention a range of other chronic diseases. However, recent many reports have found that enhanced Nrf2 activity possesses oncogenic capacity. Indeed, gain-of-function mutations in Nrf2 and loss-of-function mutation in Keap1 are found in most cancers, although the frequencies of mutations are often low. More importantly, it has been shown that constantly high levels of Nrf2 expression and low level of Keap1 may be related with chemotherapy resistance and poor prognosis. It is therefore debatable whether the activation, or alternatively the inhibition, of Nrf2 is a practical approach to the prevention or treatment of cancer. This review primarily focuses on the various domain structures of Nrf2 and Keap1, the interaction among Nrf2 and Keap1, and then attempts to find the resolution of the paradox.

        Activation of Heme Oxygenase-1 is Involved in the Preventive Effect of Honokiol against Oxidative Damage in Human Retinal Pigment Epithelial Cells

        Previous studies have shown that honokiol, a bioactive compound originated from the Magnolia, has beneficial effects on various chronic diseases. Although our previous study has demonstrated that honokiol effectively protects C2C12 myoblasts against hydrogen peroxide (H2O2)-induced oxidative stress and cytotoxicity, the antioxidant mechanism in human retinal pigment epithelial (RPE) cells has not been reported yet. Therefore, this study investigates whether honokiol attenuate cytotoxicity of H2O2 to human RPE ARPE-19 cells. Our results showed that H2O2-induced loss of cell viability and increase of reactive oxygen species production in ARPE- 19 cells were significantly alleviated by honokiol, which was associated with increased phosphorylation of nuclear factor erythroid 2-related factor 2 (Nrf2) and upregulation of heme oxygenase-1 (HO-1). In addition, honokiol markedly attenuated DNA damage and G2/M phase cell cycle arrest and improved apoptosis in H2O2-treated ARPE-19 cells. Furthermore, mitochondrial dysfunction by H2O2 was reversed by honokiol through preservation of mitochondrial membrane potential, decrease of Bax/Bcl-2 expression ratio, and inhibition of cytochrome c release into the cytoplasm. However, zinc protoporphyrin, an inhibitor of HO-1, significantly abrogated these preventive effects, suggesting a critical role of Nrf2/HO-1 in the antioxidant activity of honokiol. Taken together, these results demonstrate that activation of Nrf2/HO-1 antioxidant signaling by honokiol could rescue ARPE-19 cells from oxidative damage.
