        제주 자생 감국 꽃 추출물 유래 flavonoid 화합물의 항산화 및 항염 활성

        Anti-inflammatory and anti-oxidative activities were examined on the extract of Dendranthema indicum (D. indicum) flowers. The flowers were extracted two times for 24 h each with 70% ethanol. Upon the biological activities screening, the ethanol extract exhibited potent free radical scavenging activities and inhibited the production of nitric oxide on LPS-induced RAW264.7 macrophages effectively without causing cell toxicity. Further purification by medium pressure liquid chromatography (MPLC) and identification of the isolates led to identification of cynaroside (1) and apigetrin (2). The chemical structures of the isolated compounds were elucidated based on spectroscopic data including nuclear magnetic resonance (NMR) spectra, as well as comparison of the data to the literature values. Also, the quantitative analysis of the compounds was perfromed by high-performance liquid chromatography (HPLC). The isolates 1 and 2 were determined to inhibite the nitric oxide (NO) production dose-dependently. Based on these results, it was suggested that D. indicum extract could be useful as anti-inflammatory agents in cosmetics applications. 본 연구에서는 감국 꽃 추출물의 항산화 및 항염활성을 검색하고 유효성분을 분리하여 화학구 조를 규명하였다. 감국 꽃 에탄올 추출물의 DPPH 및 ABTS + 라디칼 소거 활성을 측정한 결과 우수한 라디칼 소거 활성이 있음을 확인하였다. 또한 LPS로 유도된 RAW264.7 대식세포에서 세포독성 없이 우수한 nitric oxide (NO) 저해 활성을 나타내었다. 감국 꽃 에탄올 추출물의 유효성분을 찾고자 medium pressure liquid chromatography (MPLC)를 실시하여 2개의 화합물을 분리하였으며 1 H 및 13 C NMR 데이터 분석및 문헌 비교를 통하여 화학구조를 동정하였다; Cynaroside (1), Apigetrin (2). 분리된 화합물을 HPLC를 통하여 정량분석을 수행하였으며 감국 꽃 추출물의 주요 성분으로 확인되었다. 화합물 1, 2의 항염활성을 확인한 결과 모두 농도의존적으로 우수한 NO 저해활성을 나타내었다. 이러한 연구 결과를 바탕으로 감국꽃 추출물은 천연 항산화 및 항염 소재로써 활용 가능할 것으로 사료된다.

        세토카 가지 정유의 성분 분석 및 생리 활성

        본 연구에서는 세토카 가지 정유의 주요 성분을 분석하고 이들의 항균, 항염 및 세포독성 실험을 진행하였다. 세토카는 제주도에서 널리 재배되고 있는 감귤류의 품종이다. 세토카 나무의 가지는 간벌 작업으로 인해 대부분 폐기되고 있고, 이러한 폐자원의 활용은 최근 많은 관심을 받고 있다. 정유 성분은 세토카 가지의 에탄올추출물을 호호바오일로 처리하여 얻었다. 세토카 정유의 주요 성분은 ethyl linoleate (64.1%), ethyl palmitate (16.5%), neophytadiene (11.1%) 및 β-citronellol (5.1%)임을 확인하였다. 이들의 항균활성을 확인하기 위하여 피부 관련 미생물에 대한 paper disc 확산법을 실시한 결과 Staphylococcus aureus 및 Propionibacterium acnes에서 좋은 항균활성을 보였다. 또한 항염활성을 확인하기 위해 lipopolysaccharide (LPS)로 염증이 유도된 대식세포에서 nitric oxide (NO) 생성량을 측정한 결과 세토카 정유 성분은 농도의존적으로 NO 생성을 저해하였다. WST-1 분석법을 이용하여 세포독성을 측정한 결과 RAW 264.7 macrophage 및 HaCaT 각질형성세포에서 세포생존율이 무처리 대조군과 비슷한 결과를 보였다. 이러한 결과를 바탕으로 세토카 가지 정유는 세포독성이 없으면서 염증억제 및 항균효과가 있음을 확인하였으며, 이를 응용한 화장품소재로써의 개발이 가능할 것으로 여겨진다. This study was designed to analyze the chemical compositions of ‘Setoka’ branch essential oils (SEBO) and to test their biological activities. ‘Setoka’ is a Citrus species widely cultivated in Jeju Island. At the present, ‘Setoka’ branches produced by thinning process were mostly discarded as a waste. Therefore, utilization of this branch waste has received much attention. ‘Setoka’ branch essential oils (SBEO) were prepared by treatment of its ethanol extracts with jojoba oil. SBEO were chemically analyzed using gas chromatograph-mass spectrometry (GC-MS), and following components were identified; ethyl linoleate (64.14%), ethyl palmitate (16.50%), neophytadiene (11.06%) and beta-citronellol (5.09%). The anti-inflammatory activity in the SBEO was examined using RAW 264.7 murine macrophage cells stimulated with LPS. As a result, the SBEO inhibited nitric oxide (NO) productions with a dose-dependent manner. In addition, SBEO showed good anti-microbial activities against drug-susceptible and -resistant skin pathogens such as Staphylococcus aureus and Propionibacterium acnes, which are acne-causing bacteria. Based on these results, we suggest that SBEO has the possibility for use as an anti-inflammatory and anti-microbial agent in cosmetic applications.

        제주 재래종 감귤 ‘홍귤’, ‘편귤’ 과피 분획물의 항산화, 항염증 효과

        한약재로 사용되고 있는 제주 재래 감귤 중 홍귤, 편귤 품종 과피 추출물의 기능성 소재로서의 활용 가능성을 알아보기 위해 용매별 분획물이 보유한 항산화 및 항염증 활성을 조사하였다. 홍귤, 편귤 모두 부탄올 분획물에서 총 페놀성 화합물의 함량이 각각 534.4 mg/g, 342.9 mg/g으로 가장 높았으며, 그 다음으로 에틸아세테이트, 헥산, 그리고 물 분획물 순이었다. 총 플라보노이드 함량에 있어서도 부탄올 분획물에서 홍귤 431.8 mg/g, 편귤 415.7 mg/g으로 가장 높게 나타났다. DPPH 라디칼 소거활성은 부탄올 분획물 1 mg/ml 에서 홍귤(89.2%), 편귤(64.2%) 모두 가장 높았으며, ABTS 라디칼 소거활성에 있어서도 두 품종 모두 부탄올 분획물에서 낮은 농도에서도 매우 우수한 활성을 보였으며 그 뒤로 에틸아세테이트, 헥산, 물 분획물 순이었다. 항염증 효과 측정에서 홍귤, 편귤 에틸아세테이트 분획물 모두 NO, IL-6, iNOS 및 COX-2 단백질 생성 억제 효과를 확인할 수 있었다. 특히 홍귤 에틸아세테이트 분획물 100 μg/ml 농도에서 항염증 효과가 우수하였다. This study investigated the anti-oxidative and anti-inflammatory activity of unripe fruit peel solvent fractions of the native jeju citrus ‘Hongkyool’ (Citrus tachibana Tanaka) and ‘Pyunkyool’ (C. tangerina Hort. ex Tanaka). The total polyphenol content and total flavonoid content were highest in the butanol fraction of both ‘Hongkyool’ (534.4 mg/g, 431.8 mg/g) and ‘Pyunkyool’ (342.9 mg/g, 415.7 mg/g). In both cultivars, the butanol fraction showed the strong antioxidant activity by DPPH radical scavenging and ABTS radical scavenging. The DPPH radical scavenging of the butanol fraction from ‘Hongkyool’ and ‘Pyunkyool’ was 89% and 64% at a concentration of 1 mg/ml, respectively. The ABTS radical scavenging of the butanol fraction from ‘Hongkyool’ and ‘Pyunkyool‘ was 94% and 85% at a concentration of 1 mg/ml, respectively. We investigated the effect of the anti-inflammatory activity of the ethyl acetate fraction from ‘Hongkyool’ and ‘Pyunkyool’ on LPS-induced NO production, IL-6, iNOS, and COX-2 protein expression in Raw 264.7 cells. At concentrations of 50 and 100 μg/ml of the ‘Hongkyool’ ethyl acetate fraction, the anti-inflammatory effect was excellent. These results suggest that ethyl acetate and butanol fractions from ‘Hongkyool’ and ‘Pyunkyool’ could be useful functional materials, with anti-oxidative and anti-inflammatory properties.

        가공용 감귤 신품종 하밀감 과피 추출물의 항산화 활성

        The major objective of this study was to investigate the usability as cultivar for processing of new Citrus hybrid 'Hamilgam'. We investigated various antioxidant activities, such as DPPH radical, ABTS radical, Hydroxyl radical and Superoxide anion radical effect, along with total polyphenol content and total flavonoid content of Citrus hybrid 'Hamilgam', C. unshiu, C. natsudaidai. The total polyphenol content and total flavonoid content were the highest in the Hamilgam. The major flavonoids of Hamilgam and Natsudaidai were naringin and neohesperidin, and those of Unshu were narirutin and hesperidin, as determined by HPLC. Especially, the neohesperidin content of Hamilgam showed much higher (100 times over) than that of Unshu. However, the aglycone form was not found. DPPH and ABTS radical scavenging activities were the highest in Hamilgam peels. Hydroxyl radical scavenging activity was high in the order of Unshu, Hamilgam and Natsudaidai. Superoxide anion radical scavenging of 3 cultivars peels displayed low activities compared to DPPH and ABTS radical scavenging. Based on these results, Hamilgam peel extracts possess antioxidant activities and may thus serve as potential sources of functional food, cosmetic products, etc.

        유채 전초 추출물 유래 항산화 및 항염 활성 성분

        In this study, we investigated anti-oxidative and anti-inflammatory efficacy, and identified their constituents from Brassica napus L. (Korean name: Yuchae) whole plant. Upon the anti-oxidative activities screening, the ethanol extract exhibited potent DPPH and ABTS<sup>+</sup> radical scavenging activities. On the anti-inflammation studies using LPS-induced RAW264.7 cells, the extract inhibited the production of NO and pro-inflammatory cytokines (TNF-α, IL-1β, IL-6) effectively. To identify major constituents of B. napus extract, further purification was performed and led to isolation of two compounds; isorhamnetin 3,7-O-diglucoside(1) and isorhamnetin 3-O-glucoside(2). Quantitative analysis by high pressure liquid chromatography (HPLC) determined the flavonoid 1 as the major constituent. Isolated compounds showed DPPH radical scavenging effects and decreased NO levels without causing cell toxicities. These results indicate that the extract of Yuchae, a rich plant resource in Jeju Island, could be potentially applicable as an anti-oxidative and/or anti-inflammatory ingredients.

        부지화 미숙과 에센셜 오일의 항산화 및 항균 활성 효과

        This study was designed to analyze the chemical composition of essential oil in ‘Shiranuhi’ immature fruitand to test their biological activities. ‘Shiranuhi’ immature essential oils (SIEO) were obtained by steam distillation fromfruits collected from Jeju Island and were analyzed using gas chromatograph (GC)-flame ionization detectors (FID) andGC-MS. Fourteen components were identified in the essential oil. Limonene (75.21%) and terpineol (8.68%) were the majorcomponents in SIEO. Since acne vulgaris is the combined result of a bacterial infection and the inflammatory response tothat infection, we examined whether SIEO possessed antibacterial against skin pathogens. As a result, SIEO showed excel-lent antibacterial activities against drug-susceptible and -resistant Propionibacterium acnes and Staphylococcus epidermidis,which are acne-causing bacteria. In this study, SIEO was examined on DPPH radical scavenging activities, which showedmoderate antioxidant activity (SC50, 15.36µL/mL). In order to determined whether SIEO can be safely applied to humanskin, the cytotoxicity effects of SIEO were determined by colorimetric MTT assays in normal human fibroblasts and kerati-nocyte HaCaT cells. They exhibited low cytotoxicity at 0.5µL/mL in both celllines. Based on these results, we suggest thepossibility that essential oil of ‘Shiranuhi’ maybe considered as an antibacterial and antioxidant agent.

        섬오갈피나무 줄기 유래 테르펜 화합물의 멜라닌 생성 저해 활성

        본 연구에서는 섬오갈피 줄기 추출물 및 용매 분획물의 미백 활성을 검색하고 유효성분을 분리하여 화학구조를 규명하였다. 섬오갈피 줄기 에탄올 추출물 및 용매 분획물의 멜라닌 생성 억제활성을 측정한 결과, 헥산 및 에틸아세테이트 분획물에서 우수한 멜라닌 생성 억제 효과가 있음을 확인하였다. 비교적 세포독성이 적은 헥산 분획물에서 유효성분을 찾고자 크로마토크래피를 실시하여 3개의 화합물을 분리하였으며 <sup>1</sup>H 및 <sup>13</sup>C NMR 데이터 분석 및 문헌 비교를 통하여 화학구조를 동정하였다; kaurenoic acid (1), 16α-hydro-17-isovaleroyloxy-ent-kauran-19-oic acid (2), 16α-hydroxy-17-isovaleroyloxy-ent-kauran-19-oic acid (3). 분리된 화합물에 대한 미백 활성 실험 결과, 화합물 1-3 모두 농도의존적으로 멜라닌 생성을 억제하였다. 또한 세포 내 티로시나제 효소의 활성을 감소시킴을 확인하였으며 이러한 연구 결과를 바탕으로 섬오갈피 줄기 추출물은 기능성 미백 화장품 소재로써 활용 가능할 것이라 사료된다. A. koreanum was investigated to identify the constituents possessing whitening effects. As anti-melanogenesis activities were screened for the ethanol extract and solvent fractions, n-hexane (Hex) and ethyl acetate (EtOAc) fractions showed the most potent activities. Three constituents were isolated from the n-Hex fraction of A. koreanum; kaurenoic acid (1), 16α-hydro-17-isovaleroyloxy-ent-kauran-19-oicacid (2), 16α-hydroxy-17-isovaleroyl-oxy-ent-kauran-19-oic acid (3). The chemical structures of the isolated compounds were elucidated based on the spectroscopic data including <sup>1</sup>H and <sup>13</sup>C NMR spectra, as well as comparison of the data to the literature values. Whitening effects were studied for the isolated compounds. Upon the anti-melanogenesis test using α-MSH stimulated B16F10 melanoma cells, the compounds 1, 2 and 3 inhibited the cellular melanogenesis and intracellular tyrosinase activities effectively. Based on these results, A. koreanum stems extract could be potentially applicable as whitening ingredients in cosmetic formulations.
