A procedure for extraction and purification of 8 kDa antigen of Brucella abortus RB51 was developed. Bacteria heat inactivated at $60^{\circ}C$, 30 min was extracted by 1% sarcosine and followed by fluid pressure liquid gel filtration chromatography of 2 series, Superose 12 HR 10/30 and Sephacryl S-100. There was produced $71.46{\mu}g/g$(wet) of 8 kDa antigen, and it resisted 1% trypsin, solved 1% triton X-100 higher than distilled water and inactivated 0.1% proteinase K. These results show that 8 kDa antigen may be a lipoprotein existed cell surface of B. abortus RB51. Also, we developed ELISA using purified 8 kDa surface antigen of Brucella abortus RB51 strain, its specificity and sensitivity was 95.0%, 98.6%, respectively. As compared with dot-blot assay using whole cell and ELISA using 8 kDa antigen, its correlation was 93.5%.

As compared with reaction of antibody for sonicated antigen of Brucella abortus strain RB51 and 1119-3 by Western blot analysis, Brucella field positive sera was detected strong reaction at 40~80 kDa LPS of strain 1119-3, but detected very weak reaction at strain RB51 partly. Otherwise, as we analyzed major immunogen of RB51 by antisera bled periodically during 6 months after RB51 vaccination. we detected strong immunological reaction at 17, 18 and 8 kDa antigen of RB51. Especially, reaction of 8 kDa antigen by Western blot coincided with reaction of dot-blot assay in RB51-antibody detection method. We also compared with reaction of field sera by STAT(standard tube agglutination test), dot-blot assay and Western blot (reaction of 8 kDa antigen of strain RB51). 16 sera of 4~5 months after RB51 vaccination are all negative by STAT, and 12 field brucellosis positive serum are all positive, and also 12 of 16 sera vaccinated RB51 are positive by dot-blot assay and reaction of 8kDa antigen by Western blot. but 1 of 15 Brucellosis negative sera reacted nonspecifically dot-blot assay.

This study was performed to investigate the electromyographic(EMG) activity, firing time and sequence of the mandibular elevator muscle on gum chewing. For this study, 28 patients with temporomandibular disorders(TMD), especially internal derangement of TM joint, and 16 dental students without any signs and symptoms in the masticatory system were selected as the patients group and as the normal group, respectively. The patients group was composed of 14 right and 14 left side affected patients. For recording of EMG activity(${\mu}V$) of the anterior temporalis(TA) and the masseter muscle(MM), and measuring of firing time(millisecond) from the start of mandibular opening movement to the firing of the muscles, BioEMG, BioEGN, and Combo program integrated in the Biopak system(Bioresearch Inc., Milwaukee, USA) were used. Gum chewing stroke was performed in both right and left side for several times, and the first and the second chewing strokes were analysed and compared with regard to EMG activity, firing time, firing sequence, correlation between EMG activity and firing time. The data obtained were analysed by SPSS windows program and the results of this study were as follows : 1. In the patients group, chewing on the affected side showed higher EMG activities in the masseter of the side than those of the contralateral side, but chewing on the unaffected side showed higher EMG activity in the masseter and in the anterior temporalis of the side than those of the contralateral side. 2. There were no difference of firing time between both sides on chewing on the affected side, but firing time in the chewing side were earlier than that of contralateral side on chewing on the unaffected side in patients group. 3. In the normal group, EMG activities in the masseter and in the anterior temporalis in the chewing side were always higher than those of the contralateral side, and there were no difference of firing time between the two sides. 4. In general, firing sequence of four muscles of both sides were ipsilateral TA, ipsilateral MM, contralateral TA, and contralateral MM in earlier order of time in both groups. Correlation coefficients between EMG activity and firing time were negative value, and more significant correlation were appeared in the normal group than in the patients group.

This study was performed to investigate the effects of clutch adaptation on the mandibular rotational torque movement in normal people. 69 dental students were selected for the study. Their mean age were 23.6 years and they did not present any signs and symptoms of temporomandibular disorders. $BioEGN^{(R)}$ with $Rotate^{(R)}$ program was used to observe and record the amount of mandibular rotational torque on protrusion, on right excursion, on left excursion, and on comfortable wide opening movement. The natural tooth contact movement and the movement with clutch were performed in the above four each mandibular movement. Clutch was made by the method used in $Pantronic^{(R)}$ clutch fabrication. Distance of slant frontal which was translatory trajectory in frontal plane and degree of rotational torque in horizontal and in frontal plane were recorded. The data obtained were processed with SPSSWIN program and the results were as follows : 1. Distance of slant frontal in each mandibular movement generally increased with clutch. 2. Degree of rotational torque in horizontal and in frontal plane on protrusion and on lateral excursions did not increase with clutch, but the degree on wide opening increased with clutch. 3. Degree of rotational torque in horizontal plane on protrusion and on lateral excursions did not show any difference between right and left side, but the degree in frontal plane on protrusion and on lateral excursions showed significant difference between right and left side. 4. Total amount of rotational torque from right and left sides on protrusion and lateral excursions were not increased with clutch, but the degree on wide opening movement was increased with clutch. And in this case, degree in horizontal plane was larger than that in frontal plane. 5. Correlation between total amount of rotational torque in horizontal plane and that in frontal plane were highly significant on protrusion and on lateral excursions with or without clutch, but the significant correlation on wide opening without clutch became not significant with clutch.

In Korea, brucellosis has been reported periodically in cattle and rarely in dogs; however, it has not previously been screened in domestic animals such as elk, pigs and goats. To investigate the serological prevalence, serum samples were taken from the aforementioned animals annually during 2007-2013 and screened by the rose-bengal test (RBT) or modified RBT, after which positive sera were evaluated by the standard tube agglutination test (STAT). Finally, RBT and STAT-positive sera were confirmed by competitive-ELISA. Brucella abortus biovar 1 was isolated from three elk that were shown to be positive serologically in 2008. There was no evidence of brucellosis in pigs. Based on serological monitoring and investigation of etiological agents, there is no evidence of outbreak of brucellosis in elk, pigs or goats of Korea since 2008. However, the possibility for brucellosis from cattle to affect these other livestock exists; therefore, extensive and continuous serological monitoring is required to maintain their brucellosis-free status.

Q-fever is a vector-borne (Coxiella [C.] burnetii) zoonotic disease that is an increasing public health concern. To date, some research about Q-fever prevalence in dairy herds and human patients has been reported in Korea, but information about Korean native cattle is scarce. To measure the prevalence rates of C. burnetii in Korean native cattle, a total of 1,095 bovine serum samples collected during 2010~2013 were analyzed with an enzyme-linked immunosorbent assay. Sixty-eight heads of cattle were diagnosed as positive and while 19 heads were suspected (positive rate = 6.2%). Interestingly, Jeju province had a seropositivity rate six times greater than that of other provinces (18.9% vs. 3.2%). High seroprevalence might be caused by wide distribution of ticks in Jeju province compared to other regions. Based on these data, extensive monitoring of C. burnetii infection in cattle, tick distribution, and climate changes is required.

Vaccinations, surveillance, quarantine, and disposal of the infected poultry are the common strategies for prevention and control of the highly infectious poultry diseases; however, many pathogens still persist and are potential causes threatening the nationwide spread of poultry diseases. A strict biosecurity strategy including disinfection is the key to control the spread of avian pathogens, such as the Newcastle disease virus (NDV). It is important to select and use the disinfectants whose efficacy and the effective concentrations against the specific pathogens are known. Therefore, in the present study, we evaluated the virucidal efficacy of five active substances of commercial disinfectants, namely potassium peroxymonosulfate (PPMS), sodium dichloroisocyanurate (NaDCC), glutaraldehyde (GLT), benzalkonium chloride (BZK), and didecyldimethylammonium chloride (DDAC), that are used against NDV. Further, we validated the efficacy of eight pre-approved disinfectants. The minimal virucidal concentrations of the active substances against NDV were as follows: PPMS, 2.50 g/L; NaDCC, 2.00 g/L; GLT, 0.40 g/L; BZK, 2.00 g/L, and DDAC, 1.00 g/L. Furthermore, all the eight disinfectants were found to be effective against NDV at the recommended concentrations, thereby confirming that the active substances are functional against NDV. This is the first study reporting the virucidal activity of the active substances of commercial disinfectants against NDV, in accordance with the Animal and Plant Quarantine Agency guidelines, in South Korea. The results of the virucidal efficacy testing of chemical disinfectants from this study will help poultry industries implement improved strategies for controlling infection.

Inaccurate amounts of active ingredients in disinfectants can result in inefficiency or in potential toxicity to the environment and living organisms. This may lead to disinfection failure, and consequently, biosecurity failure. To ensure the efficiency of disinfectants and their ongoing compliance with safety and quality requirements, continuous and systematic post-market surveillance studies are needed. Herein, the content of ten commercial disinfectants purchased in 2021 was analyzed. Selective analytical techniques, such as automated titration, colorimetry, and high-performance liquid chromatography, were used to evaluate the content of several active ingredients present in disinfectants, such as potassium peroxymonosulfate, benzalkonium chloride, glutaraldehyde, phosphoric acid, citric acid, and malic acid. The obtained values were then compared with the label claims; the active ingredient contents of all disinfectants were within the acceptable range of 90-120% of the label claim.

Brucella (B.) canis is mainly transmitted by direct or indirect contact with aborted fetuses and placenta. It's also known to be able to infect human, which likely results in providing veterinarians and companion animal owners for infectious risk. To develop diagnostic ELISA, we cloned and expressed rp1L gene of B. canis, which encodes the ribosomal protein L7/L12. Using this purified recombinant protein, indirect-ELISA (iELISA) was evaluated using 78 positive and 44 negative sera. The sensitivity and the specificity of iELISA were 94% and 89%, respectively. The results indicated that indirect-ELISA using recombinant ribosomal protein L7/L12 may be useful for diagnosis of canine brucellosis.
