Samyang Genex succeeded in commercialization of anticancer agent-paclitaxel by plant cell culture technology. The core technology of Samyang Genex relating paclitaxel production includes cell line development, cell line preservation, cell culture, scale-up technology, and purification technology. On the basis of the research, Samyang Genex built the factory operated by CGMP (current good manufacturing practice). The paclitaxel-Genexol$^{TM}$-is commercially available in Korea, and it will be launched to world market including USA after approval of US FDA.

To obtain basic data for the better use of Cordyceps pruinosa we reassessed the effect of different medium, culture method, pH, and carbon and nitrogen sources on the mycelial growth properties of four C. pruinosa isolates. The growth of mycelia differed among the four isolates depending on medium type and cultivation days. Among the tested 8 kinds of solid media, the four isolates grew well on PDA and MMMA (mushroom minimal medium agar). While, among the tested 8 kinds of liquid media, all the isolates grew well in SDYM (Sabourand`s dextrose yeast extract medium) . The isolates also grew well in the SDYM with pH from 4.0 to 9.5 without any inhibition. One isolate could best grow at pH 8 to 9.5. Regarding the ability of utilizing carbon source, the difference of mycelia growth among the isolates was the most with xylose. Regarding nitrogen source, three isolates could utilize urea which is new fact in this species. These results provide new points on the growth properties of the fungal mycelium which has not been explored before. Overall, this reassessed study concluded that it is necessary to check in advance the growth properties of mycelium when a new isolate of C. pruinosa is expected to be used for application.
Ethyl Acetate와 Methanol을 이용한 블루베리 추출물 대사체 분석

본 연구에서는 LC-MS/MS를 이용한 블루베리의 methanol과 ethyl acetate 추출 분획에 존재하는 대사체의 분석을 통해 효율적인 대사체 profiling의 가능성을 탐색하였다. LC-MS/MS에서 검출되는 대사체를 통계 처리한 결과, methanol 추출 분획에서는 5-O-feruloylquinic acid, malvidin hexoside, malvidin-3-arabinoside, petunidin-3-arabinoside, delphinidin hexoside, delphinidin, petunidin hexoside와 같은 안토시아닌 계열의 화합물들이 존재하였고, ethyl acetate 분획에서는 chlorogenic acid, chlorogenic acid dimer, 6,8-di-C-arabinopyranosylluteolin, luteolin과 같은 플라보노이드 계열의 화합물이 검출되었다. 본 연구는 기존 연구와 달리 대사체학 기법을 이용한 블루베리 추출물 전체 대사물질의 profiling을 시도한 최초의 연구로서 블루베리에 함유된 유용 성분의 스크리닝등 향후 응용 연구에 유용한 기반으로 이용될 수 있을 것으로 기대된다. Metabolite profiling of blueberry (cultivar "Spartan") was performed by extraction using different solvents, methanol and ethyl acetate, through metabolomic analysis using LC-MS/MS. Unsupervised classification method (PCA) and supervised prediction model (OPLS-DA) provided good categorization of metabolites according to the extraction solvents. Metabolites of the anthocyanin family, including delphinidin hexoside, delphinidin, 5-O-feruloylquinic acid, malvidin hexoside, malvidin-3-arabinoside, petunidin-3-arabinoside, and petunidin hexoside, were mainly detected in methanol fractions, whereas those of the flavonoid family, including chlorogenic acid, chlorogenic acid dimer, 6,8-di-C-arabinopyranosyl-luteolin, and luteolin were successfully prepared in the ethyl acetate fraction. Thus, metabolomic analysis of blueberry extracts allows for the simple profiling of whole and distinctive metabolites for future applications.
액체배양에서의 홍국균의 생장, 색소 및 monacolin K 생산 특성

The hyphal growth, production of color pigments and monacolin K by Monascus strains were investigated in liquid medium. Thirty five different strains were collected and cultured in potato dextrose yeast extract broth (PDYB), potato dextrose broth (PDB) and malt extract broth (MEB) medea at 25℃ for 7 days. The growth rates of most of strains were highest in PDYB medium. Growth rate as well as pigment production were influenced by suspension conditions of mycelia during liquid cultivation. Most of strains producing monacolin K corresponded to strains producing red pigment highly and showing more pH changes of liquid media. Monacolin K produced from strains was detected in culture broth as well as mycelia. Any citrinin was not detected in monacolin K producing strains. These results imply that the selection of the strains producing red pigment highly and showing more pH changes in liquid cultivation could be applied for primary screening of Monascus strains for preparation of red mold rice.
