Objective: The purpose of this study was to determine the effects of feeding ethanol on growth performances, carcass characteristics, and lipid metabolism of finishing Korean cattle (Hanwoo) steers. Methods: Thirty (30) Hanwoo steers (average 25.1 months of age, body weight 660.1 kg) were assigned to three treatments: control (0% ethanol), E-3 (1.44% ethanol for 3 months), or E-5 (0.72% ethanol for 2 months followed by 1.44% ethanol for 3 months). The animals were allotted by treatment group into six pens and fed concentrate and perennial ryegrass. Ethanol (30%, v/v) was supplemented into drinking water twice a day to meet final concentrations based on average water consumption of finishing Hanwoo steers. Results: There were no statistical differences among the groups in final body weight, average daily gain, or carcass yield grade indices such as cold carcass weight, fat thickness, and loin area. The marbling score tended (p = 0.228) to increase with the highest score (6.7) in the E-5 group followed by 6.3 and 6.0 in E-3 and control groups, respectively. The appearance frequencies of quality grades of 1++ (the best), 1+, 1, and 2, were; 30%, 50%, 0%, and 20% for control, 10%, 80%, 10%, and 0% for E-3, and 10%, 80%, 0%, and 10% for E-5 groups, respectively, indicating improvements of quality grades by feeding ethanol. Concentrations of serum glucose tended to decrease whereas those of insulin and non-esterified fatty acid to increase by feeding ethanol (E-3 and E-5; p>0.05). Conclusion: Feeding ethanol directly into drinking water of finishing Hanwoo steers stimulated lipogenesis in intramuscular adipose tissue (marbling) and thereby improved carcass quality grade. The serum metabolites results supported the hypothesis of lipolysis of existing adipose tissue, such as abdominal fats, and lipogenesis in intramuscular adipocytes.

The objectives of this study were to develop polyclonal antibody to adipocyte plasma membrane(APM) proteins isolated from Korean native cattle, and to investigate its tissue specificity. Plasma membrane proteins from adipocyte, liver, heart, spleen, and kidney were isolated using a self-forming percoll gradient. Sheep(40∼50㎏) was immunized three times at three week interval with the purified APM proteins. Blood was taken from immunized sheep at 10(AS-1), 12(AS-2), and 14(AS-3) days after the third immunization. Antisera titers and cross-reactivity against other tissues were determined by enzyme-linked immunosorbent assay(ELISA). Antisera reacted strongly to the APM proteins showing detectable amounts of antibody at 1:81,000 dilution. The antisera reacted to APM proteins specifically whereas they showed no reactivity to any other tissue plasma membrane proteins. Furthermore, tissue specificity of antisera against APM proteins was reconfirmed by immunoblotting using anti-sheep immunoglobulin G-peroxidase conjugate as a second antibody. The antisera to APM proteins showed adipocyte specificity compared to other tissues. In conclusion, polyclonal antibody against APM proteins isolated from Korean native cattle was developed successfully, and these antisera showed the specific reactivity with APM proteins.

본 연구는 돼지 지방세포 원형질막에 대한 특이 항체를 개발하고자 실시되었으며, 궁극적인 목적은 면역학적 방법을 이용하여 저지방 돼지고기를 생산하기 위함이다. 평균체중 4∼5㎏ 정도의 Landrace 교잡종 돼지를 도살하여 피하 지방조직, 심장, 비장, 간장, 및 신장을 채취하였다. 지방세포와 기타 장기로부터 분리한 원형질막을 SDS-PAGE로 확인한 바, 지방세포 원형질막의 경우 기타장기에 비하여 상대적으로 높은 분자량(50∼100kD)의 단백질을 함유하고 있음을 발견하였다. 지방세포 원형질막 단백질을 토끼에 주사한 후 항혈청의 항체 역가를 ELISA법으로 측정한 결과, 32,000배의 희석배율에서도 역가를 측정할 수 있을 정도로 높은 역가를 나타내었으며, 기타 장기로부터 추출한 원형질막 단백질과는 25∼42%(지방세포 원형질막 단백질과의 반응을 100% 기준)의 교차반응을 나타내었다. 이상의 결과는 돼지의 지방세포 원형질막에 대한 항체가 본 연구실에서 성공적으로 개발되었으며, 이는 저지방 육류의 생산을 위한 기초 자료로서 중요한 의미를 지니며, 향후 본 연구의 결과를 이용한 생체 시험이 계속 수행되어야 할 것으로 사료된다.

It is essential to isolate and characterize adipocyte specific antigens for the practical application of antibodies to regulate body fat in meat animal. In this study, SDS-polyacrylamide gel electrophoresis(PAGE), 2-dimensional (D) electrophoresis, affinity column chromatography and amino acid sequencing have been done to isolate and characterize adipocyte specific proteins(antigens) in pig. From SDS-PAGE, a protein with molecular weight of 65kDa was suspected as an adipocyte specific antigen, but it was found in the other organs, too, such as liver and heart in 2-D electrophoresis. A single band protein(64kDa) was isolated from affinity column chromatography using proteins isolated from adipocyte, liver, heart, kidney, and spleen and antibodies raised against adipocyte plasma membrane proteins in sheep. N-terminal amino acid sequences of this protein were similar to serum albumin and/or its precursor. It is, however, difficult to make any final conclusion from this study that the protein isolated from affinity column chromatography is exactly same as serum albumin. Further studies to characterize structure and function of this protein using specific antibody raised against this protein are necessary.
Microsatellite 분석을 통한 한우의 유전적 다양성

Six bovine microsatellite loci were analyzed to characterize the genetic structure of Korean cattle (Hanwoo) plus three populations of Yanbian yellow cattle, Japanese black cattle, and Holstein. Allele diversity, observed heterozygosities, expected heterozygosities, polymorphic information contents, gene differentiation (F_(ST)), indirect estimates of gene flow (Nm: migrants per generation), and Nei's DA distance were calculated. The highest genetic diversity was exhibited in Yanbian yellow cattle (Het_E = 0.732) and the lowest in Japanese black cattle (Het_E = 0.450). Korean cattle showed relatively high genetic diversity (Het_E = 0.690). The genetic differentiation values between populations, as measured by pairwise F_ST, Nm, and Nei's genetic distance, indicated Korean cattle showed a close relationship with Yanbian yellow cattle. Taken together, it is concluded that there is extensive gene flow between Korean cattle and Yanbian yellow cattle.
흰쥐 지방세포 원형질막 단백질에 대한 다클른항체의 in vitro 세포 특성효과

The objectives of the current study were to raise polyclonal antibodies in sheep against adipocyte plasma membrane(APM) proteins isolated from Sprague-Dawley rat, to investigate tissue specificity, and to determine cytotoxic effects of antiserum on cultured rat adipocytes. Plasma membrane proteins from adipocyte, heart, kidney, liver, muscle, and spleen were isolated using an iso-osmotic sucrose self-forming gradient. An adult male sheep was immunized three times at three week intervals with the purified Sprague-Dawley rat APM proteins. Antisera were taken from immunized sheep at 10, 12, and 14 days after the third immunization. Antiserum exhibited strong antigen-antibody reactivity against APM proteins determined by enzyme-linked immunosorbent assay(ELISA), and the reactivity could be detected at dilutions in excess of 1:81,000. Antisera showed very low binding affinity with proteins isolated from heart, kidney, liver, muscle or spleen whilst they showed very high reactivity with APM proteins in ELISA. Tissue specificity of the antisera was reconfirmed by Western immunoblotting using anti-sheep immunoglobulin G-alkaline phosphatase conjugate as a secondary antibody. Treatment of antisera on confluent Sprague-Dawley rat adipocytes in culture caused lysis of the cells and release of cytosolic lactate dehydrogenase whereas adipocytes treated with non-immunized serum maintained their integrity.
