배양용기 내 환기와 광도에 따른 도라지(Platycodon grandiflorum A. DC.) 기내 배양묘의 생장반응

기내 배양환경을 조절하여 건전한 유식물체를 획득하기 위해 도라지를 무균발아시켜 shoot부분을 NAA $0.1\;\cal{mg/L}$ 가 첨가된 MS 기본배지에 치상한 후 배양용기의 뚜껑위에 membrane filter를 부착하여 환기횟수를 달리하고 광도를 33, 66, 99 ${\mu}mol\;m^{-2}s^{-1}$로 처리하여 생장반응을 조사하였다. Membrane filter 부착구의 환기횟수는 $4.9 h^{-1}$이었으며 미부착구는 $0.1 h^{-1}$이었다. 환기횟수 $4.9 h^{-1}$ 처리구에서 $0.1 h^{-1}$ 처리구에 비해 전반적으로 생장이 양호하였으며 $0.1 h^{-1}$에서는 광도가 증가할수록 생장이 저조한 반면 $4.9 h^{-1}$ 처리구에서는 광도의 증가에 따라 생장이 좋은 경향이었다. 동일한 광도에서 환기횟수 $4.9 h^{-1}$ 처리구는 $0.1 h^{-1}$ 처리구에 비해 생체중은 1.9배, 당 함량은 1.4배 이상 높았으며 환기횟수 $4.9 h^{-1}$ 처리구는 광도의 증가에 따라 생체중과 당 함량이 높아졌으나 $0.1 h^{-1}$ 처리구는 광도에 의한 효과가 인정되지 않았다. 함수율은 환기횟수 $4.9 h^{-1}$ 처리구에서 $0.1 h^{-1}$ 처리구에 비해 $4.2\~5.8\%$가 낮았으나 광도 처리에 의한 차이는 없었다. 동일한 광도에서 총 엽록소 함량은 환기횟수 $4.9 h^{-1}$ 처리구에서 $0.1 h^{-1}$ 처리구에 비해 $ 0.27\~0.79\;\cal{mg/g}$ F.W.가 높았으며 환기횟수 $0.1 h^{-1}$ 처리구는 광도의 증가에 따른 효과가 나타나지 않았으나 환기횟수 $0.1 h^{-1}$ 처리구는 높은 광도에서 감소하였다. 환기횟수 $4.9 h^{-1}$는 $0.1 h^{-1}$에 비해 잎의 공변세포가 크고 주변 부세포가 잘 발달되어 있었다. 특히 PPF $99\;{\mu}mol\;m^{-2}s^{-1}$에서 환기횟수 $0.1 h^{-1}$는 부세포의 발달이 미흡하고 기공이 많이 열려 있는 상태인 반면 환기횟수 $4.9 h^{-1}$는 부세포가 잘 발달된 잎을 지니고 있었다. Shoots of balloon flower (Platycodon grandiflorum A. DC.) derived from in vitro germinated seeds were cultured on MS medium containing $0.1\;\cal{mg/L}$ NAA under various photosynthetic photon flux (PPF) 33, 66, and $99\;{\mu}mol\;m^{-2}s^{-1}$ with or without membrane filter. Number of air exchanges per hour (NAEH) of the culture vessel with membrane filter on the lid was $4.9 h^{-1}$ and that without membrane filter was $0.1 h^{-1}$ Plantlets grown in $4.9 h^{-1}$ NAEH showed greater growth than in $0.1 h^{-1}$ NAEH. According to increase of PPF, plantlets growth decreased in $0.1 h^{-1}$ NAEH while it increased in $4.9 h^{-1}$ NAEH. At the same PPF, fresh weight and sugar content in plantlets in $4.9 h^{-1}$ NAEH were above 1.9, 2.0 times higher than those in $0.1 h^{-1}$ NAEH, respectively. Also they were enhanced in $4.9 h^{-1}$ NAEH by increase of PPF whereas no significance in $0.1 h^{-1}$ NAEH. The percentage of water content of plantlets in $4.9 h^{-1}$ NAEH was $4.2\~5.5\%$ lower than those in $0.1 h^{-1}$ and no difference in PPF. The content of total chlorophyll in plantlets in $4.9 h^{-1}$ NAEH was higher $0.27\~0.79\;\cal{mg/g}$ F.W. than that in $0.1 h^{-1}$ NAEH. By increase of PPF, it was decreased in $0.1 h^{-1}$ NAEH while had no significant difference in $4.9 h^{-1}$ NAEH. Guard and subsidiary cells of leaves in $4.9 h^{-1}$ NAEH were more developed than in $0.1 h^{-1}$ NAEH. Especially, in $99\;{\mu}mol\;m^{-2}s^{-1}$ leaves in $0.1 h^{-1}$ NAEH had undeveloped subsidiary cells and wide open stomata whereas those in $4.9 h^{-1}$ NAEH had well-developed subsidiary cells.

This study was conducted to investigate the quality characteristics of instant gruel containing ear mushroom and black rice with the goal of improving the added value and increasing the consumption of ear mushroom. To make ear mushroom instant gruel using the puffing technique, the proportion of white rice to black rice and the amounts of dried ear mushroom added ranged from 75~100% : 25~0% and 0~4%, respectively. Based on rapid visco analysis (RVA) of gruel powder, peak viscosity, holding strength, final viscosity and set back increased as we increased the proportion of dried ear mushroom and decreased that of black rice. The gruel made of 80% white rice : 20% black rice and 3% dried ear mushroom contained $18.53{\mu}g/100g$ vitamin $D_2$, 3.73 g/100 g dietary fiber and is therefore expected to improve bone health and bowel movement. Also, the DPPH free radical scavenging activity for this gruel was 56.7%, which indicates high antioxidant activity. In contrast, using 100% white rice or 75% white rice : 25% black rice adversely affected the taste and flavor of the gruel. 본 연구는 국내 재배면적이 확대되고 있는 목이버섯의 부가가치 향상과 소비량 확대를 위해 팽화기술을 접목한 목이버섯 즉석죽을 제조하고, 품질특성을 조사하여 실버층에 쉽게 적용할 수 있는 제품을 개발하고자 하였다. 백미와 흑미를 75~100%와 25~0%로 혼합한 후 비타민 $D_2$와 식이섬유 함량이 높은 건목이버섯을 0~4%로 첨가하여 호화도, 영양성분, 항산화성 및 관능 등을 조사하였다. 즉석죽 분말의 호화도는 건목이버섯 첨가량이 증가할수록, 흑미 첨가량이 감소할수록 최고점도와 유지강도, 강하점도, 최종점도, 노화점도가 증가하는 경향이었다. 백미 80%와 흑미 20%를 혼합한 후 건목이 버섯 3%를 첨가한 즉석죽은 비타민 $D_2$ $18.53{\mu}g/100g$, 식이섬유 3.73 g/100 g이 함유되어 있어 뼈의 형성과 유지와 배변 활동 촉진 효과가 기대되었다. 또한 DPPH free radical 소거능도 56.79%로 나타나, 항산화성이 높아 실버층의 건강에 기여할 것으로 기대되었다. 본 실험 결과, 목이버섯은 기능성 가공제품 개발의 소재로서 사용가치가 매우 높아 실버층뿐 아니라, 다양한 연령층에 적합한 가공제품 생산에 응용가능하리라 생각된다.
다양한 자외선(UVB) 처리조건에 의한 목이버섯의 품질 특성

To examine the possibility of ear mushroom (EM) as a source of natural vitamin D, the UVB (ultraviolet B) was treated according to sample drying status, drying methods before UVB treatment and harvest time. And then, vitamin D<sub>2</sub> and ergosterol contents were investigated. According to the sample drying status, the vitamin D<sub>2</sub> contents of fresh and freeze-dried EM (whole) increased to 4,634.4~4,780.9 ㎍/100 g D.W. (dry weight) under UVB dose 52.5~70.0 kJ/㎡ and above 18,693.1 ㎍/100 g D.W. under above 105 kJ/㎡, respectively. By drying methods before UVB treatment, vitamin D<sub>2</sub> contents of EM powder (below 500 ㎛) that dried in the vinyl house and freeze-dryer increased to 4,886.2~5,132.9 ㎍/100 g D.W. under above 105 kJ/㎡ and 17,103.7 ㎍/100 g D.W. under 70 kJ/㎡, respectively. Ergosterol content decreased with increasing UVB dose in all experiments. According to the harvest time, vitamin D<sub>2</sub> content under UVB dose 210 kJ/㎡ showed marked difference and in order of June, July, August, October and April. As for the results, the optimum harvest time, drying method before UVB treatment, sample size, UVB dose for the EM contained high vitamin D<sub>2</sub> content were June, freeze-drying, whole, and 105 kJ/㎡, respectively.
