Monosodium iodoacetate에 의해 유도된 골관절염 마우스 모델에서 별갑 물추출물의 효과

This study aimed to investigate the effects of aqueous extract of Pelodiscis Carapax (PCE) in monosodium iodoacetate (MIA)-induced osteoarthritis (OA) mice. PCE (200 mg/kg) was orally administered to C57BL/6 mice for 2 weeks. After MIA injection (75 mg/mL), we performed a behavior test (open field test, rotarod test) followed by micro-computed tomography, histopathological analysis, and real-time quantitative PCR analysis on knee joint tissues. PCE treatment improved general gait locomotor activity, joint morphological features, and histopathological features compared to the MIA-induced mice. In addition, it confirmed that the mRNA expression levels of tissue inhibitor of metalloproteinases (TIMP)-1 in the joint tissues were increased in the PCE-treated group. Our result revealed that PCE treatment phenotypically improved OA symptoms.

Objectives : Samhwangsasim-tang (SST), Hwangryeonhaedok-tang (HHT), Ukgan-san (UGS), Onjunghwadam-hwan (OHH) and Samul-tang (SMT) have been used for the treatment of various diseases. This study was performed to compare the anti-obesity effects of 5 herbal formulas in high fat diet-(HFD) induced obese mice. Methods : The mice were randomly divided into seven groups that were fed a normal diet (ND), a HFD, a HFD plus SST (HFD + SST), a HFD plus HHT (HFD + HHT), a HFD plus UGS (HFD + UGS), a HFD plus OHH (HFD + OHH), or HFD plus SMT (HFD + SMT) at 300 mg/kg/day for 7 weeks. All groups were assayed for body weights, food efficiency ratio (FER), final liver and fat weight and blood biochemical parameters. Results : The increased body weights, food efficiency ratio (FER), and serum total triglyceride were decreased in HFD + OHH group relative to the same measurements in HFD group. Furthermore, the HFD + SST group significantly reduced FER, liver and abdominal subcutaneous fat weight gains, and serum total triglyceride, whereas HDL-cholesterol level was increased compared to HFD group. Conclusions : These results suggested that HFD + OHH and HFD + SST exert anti-obesity effects in HFD-induced obese mice.

This study was investigated to determine the quality monitoring of distributed materials from Glycyrrhizae Radix (26 samples), Atractylodis Rhizoma Alba (24 samples) according to storage period after 1-3 year. We have estimated by identification, purity, loss on drying, ash, acid insoluble ash, extract content, essential oil content, assay and microbial contamination, As a result, Glycyrrhizae Radix (26 samples) were satisfied with the standard of KP. (Korean Pharmacopoeia) and WHO's microbial contamination limit standard. In the Atractylodis Rhizoma Alba (24 samples), 2 samples were not satisfied with the standard of KP.(Korean Pharmacopoeia) and WHO's microbial contamination limit standard. The results make practical application of the basic data for the quality control of herbal medicine in storage.
네트워크 약리학을 기반으로 한 이진탕의 구성성분과 동맥경화 타겟 유전자의 효능 예측

In this study, we performed a network analysis between constituents and atherosclerosis target genes to predict the atherosclerotic effect of Yijin-tang using network pharmacological methodology. Yijin-tang have been used for treatment of circulatory system of blood in Korea and consist of five herbal medicines which is Pinellia ternata, Glycyrrhiza uralensis, Zingiber officinale, Poria cocos, Citurs unshiu. Using the database, we searched compounds in herbal medicines, and related target gene. Total compounds and target genes related with atherosclerosis were twenty-seven and fifty-seven, respectively. The network comprised 1,391 nodes (four medicines, 27 compounds and 57 atherosclerotic targets) and 1,807 edges (compound-target gene interaction). Our results demonstrated that apigenin, β-sitosterol, capsaicin, and limonin were correlated as many target genes (≥ 10). The CASP3, CYP3A4, PTGS2, SRC, UGT1A1, and VEGFA genes have relevance to many compounds (≥5) and their related pathway may be manifested by various mechanisms of action through the regulation of major target genes related to atherosclerosis closely related to the components of Yijin- tang. Network pharmacology- based our results provide a powerful tool for exploring the mechanisms of action of Yijin-tang and novel bioactive compounds.

This study was investigated to determine the quality control of Oriental medicine from stores dealing in Oriental medicine around Seoul and Daegu. We tested total 120 samples that widely used 15 species in herbal medicine (Lycii Fructus, Platycodi Radix, Angelicae Gigantis Radix and 12 others) being collected from Oriental medicine clinic, pharmacy, Oriental pharmacy, Oriental medical hospital and Oriental drug store. We have estimated Oriental medicine by K.P. (Korean Pharmacopoeia), K.H.P(Korean Herbal Pharmacopoeia) and announcement of KFDA. The items of examination were identification, purity, loss on drying, ash, acid insoluble ash, extract content, essential oil content, assay, heavy metal limit, and pesticides residue(BHC, DDT, Aldrin, Endrin, Dieldrin). As a result, 20 samples in total 120 samples were not satisfied with the standard and 7 species in total 15 species were not satisfied with the standard. Identification test, extract content test and pesticides residue(BHC, DDT, Aldrin, Endrin, Dieldrin) were satisfied with the standard. The result will be the basic data for the quality control of Oriental medicine.
