        단세포 겔 전기영동법을 이용한 생쥐 비장 림프구 DNA 손상에 대한 비타민 C 및 시스테인의 방사선 방어효과

        The alkaline comet assay, employing a single-cell gel electrophoresis (SCGE), is a rapid, simple and sensitive technique for visualizing and measuring DNA damage leading to strand breakage in individual mammalian cells. The protecting effect of pretreatment with vitamin C and cysteine on the DNA damage of gamma ray was investigated in mice splenic lymphocytes. Vitamin C and cysteine were administered orally for five consecutive days before irradiation. Four week old ICR male mice were irradiated with 3.5Gγ of y-radiation and were sacrificed 3 days later. Spleens were taken for DNA damage examination by Comet assay and the tail moments of DNA single-strand breaks in the splenic lymphocytes were evaluated. The results show that pretreatment with vitamin C and cysteine were effective in protecting against DNA damage by gamma ray. Administration of antioxidants like vitamin C and cysteine to mice before irradiation was effective in reducing the tail moment of splenic lymphocytes DNA.

      • 흰쥐 조직에서의 카드뮴 결합 고분자량 단백질 [Cd-BP(Ⅱ)]유도

        The study was carried out on the biochemical chracters of Cd-BP(II) after isolation and purification of the protein from the liver of rat ip injection with Cd. A continued study has been doing whether Cd-BP(II) could be induced by Cd or by the other metals such as Zn and Cu. Antisera were made against the antigen of Cd-BP(II) from New Zealand white rabbits. We carried out g-globulin purification, then Ouchterlony test and gel immunodiffusion test. Cd-BP(II) was also found in normal tissues of rat. It was induced up to a considerable level by Cd, whose induced level was higher than that Cu or Zn treatment. The level of induction by Cu or Zn pretreatment plus Cd treatment was lower than that by single treatment of Cu or Zn. Such a result was presumably related to the Cd toxicity.

        We isolated two Cd-binding high molecular weight proteins from intraperitoneally cadmium injected rat liver. Molecular weight of Cd-BP(Ⅰ) purified from Sephacryl S-100 and DEAE Sepharose column chromatography and Cd-BP(Ⅱ) purified from DEAE Sepharose column chromatography and Sulphonyl Sepharose column chromatography was 33,000 and 18,400, respectively. Alcohol dehydrogenase and alkaline phosphatase acitivities were not detected from two purified proteins.

      • 쇠비름 수침액이 방사선 조사에 의한 생쥐의 생존율과 뇨중 아미노산 조성에 미치는 영향

        A study has been conducted to investigate the radioprotective effects of purslane extracts (P) when I.p. injected before irradiation (R). For studying the amino acid composition , three groups of ICR mice (7-week-old) were tested. The first group I.p. injected with purslane extract for 5 days were irradiated with 6 Gy of γ-radiation (P+R). The second group was irradiated with 6 Gy without any pretreatment (R). The third group was non-irradiated control (CT). Each group divided into two groups for the survival rate study, one injected with saline (S) and the other injected with purslane extract (P) for five days and then irradiated with 8 Gy. The amino acid composition of urine samples were analyzed with HPLC for 19 days after irradiation. A few kinds of amino acids such as lysine and methionine in urine from P+R group increased in comparison with those from CT or R group. The survival rate of P group maintained much higher than that of S group during experimental period. The results obtained may support that this plant has radioprotective substances by means of life-lengthening, not of urine amino acid components.

      • 감마선에 조사된 생쥐에 있어서 방사선방어효과 평가를 위한 생물학적 파라메타

        This study deals with the biological changes in mice after γ-irradiation. Four weeks old BALB/c mice were irradiated with 6.5Gy of γ-ray on the fifth day after oral administration of radioprotectants such as ascorbic acid, tocopherol and cysteine. Control group was irradiated with 6.5Gy without pre-administration of radioprotectors. Blood cells and sperm cells were counted and body, testis and spleen were weighed 3 days after irradiation. And also liver antioxidant activity and range of spleen immune cells were measured. Differences in most biological parameters were not clearly distinguished between experimental groups. However, the relative spleen weight, the relative testis weight and the population size of spleen immune cells such as T helper cells, B cells and macrophages measured by means of FACS showed significant difference between irradiated and radioprotectant administered group. It is concluded that the relative spleen weight, the relative testis weight and the population size of spleen immune cells are easy and useful parameters for assessing the effect of radioprotective substances and for quantifying biological damage of radiation, as well.

        존엄 기반 노인미술치료 체험연구

        The purpose of this study was to explore what elderly people experience receiving for deep understanding of the meaning and nature of those experienced by elderly people in dignity-based art therapy, applied the methodology of the hermeneutic phenomenology by Van Manen. The participants of this study were collected from the elderly over 65 years old residing in the metropolitan area with dignity based geriatric art therapy and in - depth interviews. The interviews were conducted until the data were saturated. As a result of analyzing the data, six essential topics and 19 sub - topics were derived. The research participants experienced experience of new experience by witnessing the sensation through encounter with the art media through dignity - based elderly art therapy, emphasized the relationship acquired through voluntary life retrospect, I have made a serious insight by giving meaning to my life and I have been trying to maintain my daily life by voluntarily giving meaning, forgiveness, active healing and not regretting after life, I had the experience of accepting it as part of my life journey. This study is expected to provide qualitative information on experience contributing to determine additional research or intervention and their direction about the dignity-based art therapy for elderly people and is also expected to be used as basic date in developing art therapy program for dignity therapy. 본 연구는 노인이 존엄 기반 노인미술치료를 통해 어떠한 경험을 하는지 탐구하고, 그 체험의 의미와 본질을 밝혀 심층적으로 이해하는 데 목적이 있다. 이를 위하여, Van Manen(1990)의 해석학적 현상학의 연구방법론 을 적용하였고, 존엄 기반 노인미술치료를 경험한 65세 이상의 노인들을 참여자로 선정하였다. 수집한 자료 를 분석한 결과 6개의 본질적 주제와 19개의 하위 주제를 도출하였다. 본 연구 결과에서 노인은 먼저, 미술 매체를 통해 체득된 경험이 감각을 깨워 뭉클함을 체험하였다. 또한 인생 회고를 거치며 반복적으로 자기 가치의 소중함을 자각하면서 관계에 대한 욕구가 충족되었다. 나아가 자신의 삶에 의미를 부여하며 진지한 통찰을 하였고, 용서와 적극적 치유를 통해 삶 이후에도 후회를 남기지 않으려 하였다. 또한 일상을 유지하고 자 하는 소망을 품으며, 노년기의 일상을 희망을 찾아가는 삶의 여정의 한 부분으로 받아들였다. 이 연구는 이를 토대로 노인에게 노년의 삶은 가치 있고 소중하다는 메시지를 바탕으로 한 존엄 기반 노인미술치료의 접근이 필요함을 확인했다는 것과 노인의 삶에 깊은 이해를 이끌어 냈다는 점에서 의의를 가진다.

        BSH 존재시 중성자 및 ${\gamma}$-ray 조사에 따른 plasmid DNA의 손상

        In this study, the extent of plasmid DNA damage was observed according to concentration of BSH(Boron Sulfhydryl Hydride) and irradiation doses of neutron and ${\gamma}$-ray. The plasmid used was both pBR 322 (2870 bp) and ${\Phi}X174$ RF(5386 bp) DNA. Plasmid DNA damage by irradiation in the presence of BSH was analyzed by agarose gel electrophoresis. In the neutron experiment, DNA damage of both plasmid DNAs was increased according to increasing the concentration of BSH and neutron doses. But in the ${\gamma}$-ray experiment, there appeared no dose dependency as compared to the neutron experiment. The extent of the plasmid DNA damage in the presence of BSH was somewhat different according to irradiation by neutron or ${\gamma}$-ray. 본 연구에서는 boron 화합물인 BSH(Boron Sulfhydryl Hydride)의 농도와 중성자 및 감마선 방사선 조사선량에 따른 plasmid DNA의 손상 정도를 관찰하였다. Plasmid는 pBR 322(2870 bp)와 ${\Phi}X174$ RF DNA(5386 bp)를 사용하였고 조사 후 DNA의 손상 정도는 agarose gel 전기영동 상에서 관찰하였다. 중성자 조사에서는 plasmid DNA의 손상 정도는 BSH의 농도 및 조사 선량이 증가함에 따라 증가하였으나 감마선 조사에서는 조사하지 않은 대조군과 큰 차이를 나타내지 않았다. Plasmid DNA의 손상 양상은 BSH 존재시 중성자 및 감마선 조사에서 다소 다름을 알 수 있었다.

        생쥐 공장 융모의 감마선 전신조사에 의한 형태학적 변화

        소화기계 암의 효과적인 치료중의 하나로 방사선 치료법을 들 수 있으나 이 방법은 장 점막과 같이 빠르게 증식하는 조직에는 특히 부작용을 초래할 수 있다. 생쥐의 공장 융모가 감마선 전신 조사에 의해 형태학적으로 변화하는 정도를 관찰하였다. 생후 4주된 ICR계 생쥐를 6.5Gy와 12Gy를 전신 조사 3일 후에 공장을 채취하여 탈수 과정을 거쳐 파라핀을 입힌 조직을 절편기로 잘라 슬라이드에 부착시키고 일반적인 hematoxylin & Eosin 염색과정을 거쳐 염색후 광학현미경으로 관찰한 결과, 6.5Gy와 12Gy를 조사 받은 생쥐의 융모는 대체로 정상생쥐와 비교 할 때 융모의 길이가 짧아졌으며 융모의 두께가 비후되었고 내강은 넓어지는 경향을 나타내었다. 따라서 융모가 소화과정에 중요한 역할을 담당하며 방사선에 비교적 민감함을 나타내므로 방사선에 의한 생체영향 평가 뿐만아니라 방사선 방어제를 검색하는데 하나의 실험 파라메타가 충분히 될 수 있음을 인지하였다. Radiotheraphy may be considered as one of the most effective treatments for digestive tumors. This procedure has major side effects, especially in fast growing tissues like intestinal mucosa. For this reason, the morphological changes in mouse jejunal villi after whole body ${\gamma}-irradiation$ were analysed. Four week old ICR male mice were irradiatied with 6.5Gy or 12Gy of whole body ${\gamma}-irradiation$ and were sacrificed 3 days later. Jejunum among intestine were taken for morphology. Samples were carried out dehydration process and sliced sample covered with paraffin was hold on the slide and then light microscopy was observed by Hematoxylin & Eosin staining. villi at both irradiated doses were showed that the length of villi were shortened and thickened and that lumen were expanded in comparison with non-irradiated group. Since willi have an important role in digestion and very sensitive to radiation from this test, it has a role of test parameter for finding radioprotectors as well as evaluating the biological effect by radiaton.

      • 골육종의 도약전이 진단에서 골스캔의 보완검사로서의 FDG-PET/CT: 증례 보고

        골육종에서 드물게 발생하는 도약전이 병소는 불량한 예후 인자로서 중요한 연관성이 있기 때문에 골육종 환자의 광범위 절제술 시에 절제범위에 포함되어야 한다. 따라서 수술 전 검사에서 골육종의 골수 내 침범에 대한 정밀한 평가가 필수적이며, 골스캔 검사는 도약전이의 발견에 있어서는 높은 위음성률을 보일수 있으며 자기공명영상 검사는 종양이 위치한 골 전체를 촬영하지 않는 경우 임상적인 증거가 없으면 간과하기가 쉽다는 단점이 있다. 이에 저자들은 주 종양 부위만을 포함한 국소 자기공명영상 촬영과 골스캔에서 발견되지 않았으나 FDG-PET/CT에서 도약 전이가 진단된 대퇴 원위부의 골육종 증례에 대해 문헌 고찰과 함께 보고하고자 한다. Skip lesion is not uncommon feature in osteosarcoma and considered to be importantly associated with poor prognosis factor, and thus, should be excised with the main mass. The accurate pre-operative evaluation of the intramedullary extent of osteosarcoma is essential, because it determines the level of bone resection. Among the reliable detection methods, bone scan has a drawback of high rate of false negative results and regional MRI has a difficulty to cover the whole involved lesions without clinical suspicion. The authors report a case of osteosarcoma of the distal femur with a proximal skip lesion that was not detected by either regional MR imaging or by bone scan, but which was visualized by FDG-PET/CT.
