        임상에서 분리한 메치실린 내성 황색포도상구균에 대한 녹나무잎 추출물의 항균활성

        Methicillin-resistant Staphylococcus aureus (MRSA) has been emerging worldwide as one of the most importanthospital and community pathogens. At the same time, because of the difficulty in developing chemical synthetic drugs andbecause of their side-effects, scientists are making more efforts to search for new drugs from plant resources to combatclinical multidrug-resistant microbial infections. Cinnamomum camphora (C. camphora) is a plant of family Lauraceae, andgrown Jeju island in South Korea that are used as a drug to treat neurasthenia, epilepsy, cystitis, pyelonephritis, digitalis,cancer, and diabetes mellitus in folk remedies. In this study, antibacterial activites of 80% ethanol extract of C. camphoraleaves (CCE) were investigated in combination with antibiotics against clinical isolates of MRSA. The results showed thatCCE was determined with MIC and MBC values ranging from 156 to 313 and 313 to 625 ㎍/㎖, oxacillin from 128 to 256and 128 to 512 ㎍/㎖, ampicillin from 4 to 64 and 8 to 128 ㎍/㎖. The combination of CCE with oxacillin or/and ampicillinwere synergistic effect against MRSA 1, 6, 7, 8, 10, 11, 12, 13, and 15/ MRSA 1, 2, 6, and 7. 적 요메치실린내성 황색포도상구균(MRSA)은 가장 중요한 지역 사회병원성 균주의 하나로서 항생제 내성 문제가 세계적으로 대두되고있다. 감염 치료에 대한 부작용과 내성이 적은 새로운 항생제의 개발에 많은 관심과 연구가 필요하다. Cinnamomum camphora는 제주도에서 자생하는 녹나무과의 식물이다. 민간에서는 신경 쇠약,간질, 방광염, 신우 신염, 당뇨, 암과 당뇨병 치료제로 이용되고 있다. 본 연구에서는 80% 에탄올을 이용하여 녹나무잎을 추출물하여임상에서 분리한 메치실린 내성 균주에 대한 항균활성을 연구하였다. 그 결과 녹나무잎 추출물에서 높은 항균활성을 관찰 할 수 있었고, 항생제와의 병용처리 녹나무잎 추출물의 MIC와 MBC의 범위는156-313 ㎍/㎖와 313-625 ㎍/㎖로 판독되었고, oxacillin의 병용처리의 범위는 1288-256 ㎍/㎖와 128-512 ㎍/㎖, 그리고 ampicillin의 병용처리 범위는 ㎍/㎖와 8-128 ㎍/㎖로 판독되었다. 녹나무잎추출물과 항생제의 병용처리는 MRSA 항균활성에 대한 상승효과가 높게 나타났다.

      • 산국 (Chrysanthemum boreale) 휘발성 추출액의 항균 및 항진균활성

        To investigate the antimicrobial and antifungal activity of Chrysanthemum boreale(C. boreale) essential oil, the growth inhibition against Aspergillus nidulans, Fusarium oxysporum, Alternaria mali, Staphylococcus aureus, Streotococcus pyogenes, Streptococcus mutans and Escherichia coli in the presence of the essental oil was monitored by agar dilution method. The antifungal effect of C. boreale on Aspergillus nidulans, Fusarium oxysporum and Alternaria mali as affected at the concentration of 400 ppm. The antibacterial effect of C. boreale on staphylococcus aureus, streptococcus pyogenes and Streptococcus mutans was affected at the concentration of 0.4 μl, while growth of E. coli was not inhibited at the concentration of essential oil. The results of this experiment on microorganisms indicated that naturally occuring chemical substances from C. boreale might have biologically toxic activities, that is, allelopathic potentials to the microorganisms.

        Induction of Apoptosis in Human Oral Epidermoid Carcinoma Cell by TheEssential Oil of Artemisia Scoparia WALDST. et KITAMURA

        Artemisia scoparia (A. scoparia), perennial herb is indigenous to Korea and has been traditionally used in liver damage. We investigated the effect of the essential oil obtained from A. scoparia on apoptosis of KB cells. Cytotoxicity and cellular DNA content were analyzed by MTT assay and flow cytometry, agarose gel electrophoresis, and Hoechst 33258 staining. The caspase-3 and poly (ADP-ribose) polymerase (PARP) proteins were estimated by Western blotting method. We found that the essential oil induced the apoptosis of the KB cells by concentrations of 0.4 to 0.2 mg/ml which was verified by DNA fragmentation, apoptotic bodies, and the sub-G0/G1 ratio. The essential oil also transient caspase-9 and caspase-3 activity and cleavage of PARP in KB cells for 24 h. The essential oil-induced apoptotic cell death was accompanied by up-regulation of Bax and down-regulation of Bcl-2. In conclusion, we demonstrated that the essential oil of A. scoparia induces apoptosis in KB cells.

      • 산국 (Chrysanthemum boreale) 휘발성 추출액 및 EDTA가 미생물 및 인체 치은 섬유아세포에 미치는 영향

        Chrysanthemum boreale (C. boreale) has been used for the treatment of furuncles, carbuncles, sores, conjunctivitis, headache, vertigo and swollen throats as a folk medicine in Korea, but there is little report about experiment evidence of antimicrobial activity. To investigate the antifungal and antibacterial activity of Chrysanthemum boreale(C. boreale) volatile substance and EDTA, the growth inhibition against Aspergillus nidulans (A. nidulans), Fusarium oxysporum (F. oxysporum), Alternaria mali(Al. mali), Candida albicans (C. albicans), staphylococcus aureus (S. aureus), Streptococcus pyogenes(S. pyogenes), streptococcus mutans (S. mutans), Escherichia coli(E. coli), Helicobacter pylori (H. pylori) and Porphyromonas gingivalis (P. gingivalis) was monitored by agar dilution and broth microdilution method. The antifungal effect of C. boreale on A. nidulans, F. oxysporum, Al. mali and C. albicans was shown at the concentration of 0.04%. The antibaterial effect of C. boreale on S. aureus, S. pyogenes, S. mutans and H. pylori was shown at the concentration of 0.04%. However, the growth was inhibited at the concentration of 0.04% irrespective of the presence of EDTA 0.125% was much higher than higher than that of several concentration of volatile substance. The minimum inhibitory concentrations of volatile substance were 0.16% against S. aureus, 0.04% against S. pyogenes, 0.02% against S. mutans, 0.40% against E. coli, and 0.18% against C. albicans. The growth of human gingival fibroblasts was not affected by C. boreale volatile substances at the concentration of 0.04%. These results indicate that volatile substance from C. boreale may have biologically toxic activities to the microorganisms.

        근로자의 구강건강영향지수에 미치는 요인

        본 연구는 경남지역 산업체 근로자를 대상으로 구강건강영향지수에 미치는 요인을 알아보고자 272명을 대상 으로 설문지를 자기기입식으로 작성하도록 하였으며 다음과 같은 결론을 얻을 수 있었다. 구강건강영향지수에 영향을 미치는 변인은 전신건강지수, 연령, 식생활 행동, 구강건조증의 4변인인 것으로 나타났다. 변인중 전신건강지수와 식 생활 행동은 구강건강영향지수에 정적 영향을 미쳤으며, 연령과 구강건조증은 부적 영향을 미쳐, 전신건강지수가 높 고, 좋은 식생활 행동을 가질수록 구강건강영향지수가 높으며, 연령이 낮고, 구강건조증이 없을수록 구강건강영향지수 가 높다는 것을 알 수 있었다. 이상의 결과 근로자의 구강건강영향지수에 미치는 요인을 분석하여 구강건강증진을 위한 프로그램 개발에 힘을 써 구강질환이 발생되기 전에 예방하고 나아가 근로자의 건강과 구강건강영향지수를 향상 시켜야 할 것이다. The purpose of the research is to investigate the factors affecting the life quality of oral health according to the knowledge and behavior related with oral health. Total 272 people who work in Gyeong-Nam province participated in this survey. The research shows that major 4 factors are subjective whole body health, ages, dietary pattern and dry mouse. The subjective whole body health and dietary pattern have an positive effect on the life quality of oral health, whereas ages and dry mouse an negative effect on this. As a result, the health and the life quality of oral health should be improved by investigating the factors affecting oral health and thus developing the program enhancing the oral health to prevent oral disease

        비쑥의 휘발성물질에 의한 KB 세포의 세포사멸 유도

        일부 제조업 생산직 남성근로자들의 구강건강상태 및 관련요인

        본 연구는 근로자들을 대상으로 구강건강상태 및 관련요인을 파악하여 구강건강증진을 향상시키기 위하여 경상남도지역 일부 근로자 178명을 대상으로 자기기입식 설문지를 작성하게 하여 다음과 같은 결론을 얻었다. 치아우 식증이 있는 군에서 지난 1년간 치과방문경험이 있거나 치석제거경험이 있다고 응답한자에서 치아우식증이 없는 군 보다 낮게 나타나 유의한 차이가 있었다(p<.05). 치아우식증이 있는 군에서 치아가 쑤시거나 욱신거리며 아픈 경험이 있다고 응답한 자가 51.3%, 잇몸이 아프거나 피가 난 경험이 있다 48.7%, 불쾌한 입냄새 경험이 있다 53.8%로 나타 나 유의한 차이가 있었다(p<.05). 치은염증이 있는 군에서 지난 1년간 치석제거경험이 없거나 치은염증이 없는 군보다 낮게 나타났다(p<.001). 치은염증이 있는 군에서 하루 칫솔질 회수는 3회이상이 9.6%, 취침전 칫솔질을 항상 했다 4.7%, 구강위생보조용품 이용은 대부분 했다 5.9%로 나타나 유의한 차이가 있었다(p<.001) 치아우식증 유무에 영향을 미치는 변수로는 주관적 구강증상, 치실 또는 치간칫솔 사용여부, 구취증상 변인으로 파악되었고, 치은 염증 유무에 영향을 미치는 변수로는 취침직후 칫솔질 유무, 혀통증 증상, 구취증상, 시린치아증상, 스켈링경험여부 변인으로 파악 되었다. 이상 구강검진을 통한 상담과 교육을 통해 근로자들의 구강건강에 대한 의식을 고취시켜 근로자들의 구강건 강을 증진 유지시키는데 도움이 되는 방향으로 개선되어야 될 것이다. The Purpose of this study was to examine the factors affecting oral health status of some workers. Answer sheets for questionnarie for 178 workers at Changwon city, Korea, were collected and analyzed using SPSS 19.0. The number of respondents who have visited dental clinic is lower among group with dental caries than group without dental caries. The respondents who have scaling corresponds to 31.3% in the group with dental caries and a little bit lower percentage is shown in the group without dental cares(p<.05). Among those with dental caries, the respondents who experienced tooth pains are to 51.3% and those who had frequent blooding in gum 48.7% and those who had ordors inside the mouth 53.8%. The number of respondents who had scaling within last one year among the group with gingival inflammation is lower than those without it(p<.001). In the group with gingival inflammation, no smokers are corresponding to 13.9%(p<.01) and those with more than 3 times brush of tooth 4.7% and those with oral hygiene devices are to 5.9%. Important variables which have effects on dental caries are understood as self-evaluation for dental health, dental floss or interdental brush, ordors in the mouth. Also variables related with gingival inflammation are considered as tooth brush before bed time, tongue pain, orders in the mouth, the cold as ice tooth and tooth scaling experience. In summary, Oral health education is needed to increase the motivation of industrial workers to control their basic dental disease.

      • Intracellular Invasion of Staphylococcus aureus against Human Gingival Fibroblasts : A Potential Pathogenic Mechanism in Oral Infection

        Staphylococcus aureus(S. aureus) is one of the important pathogens in acute oral infection, but the pathogenic mechanism of S. aureus is not fully understood. In our previous study, interaction of staphylococcal protein A and cytoskeletal actin filaments is involved in the S. aureus invasion of cultured KB cells, an established human epidermoid carcinoma cell line. To investigate the pathogenic mechanism of S. aureus in oral infection, the invasion of S. aureus in human gingival fibroblasts(HGFs) was studied. Invasion of S. aureus ATCC 25923 and OPT 2 was found to be time-dependent (0-120 min). That was increased linearly when an increasing number of bacteria were added to media (10^2 - 10^7 cfu/mL/well). The invasion of wild type stain (OPT 2) isolated from the patient with acute oral and maxillofacial infection was higher than that of standard strain (ATCC 25923). However the invasion of S. aureus Wood 46, which is deficient in protein A, was notably lower than those of the strains that have protein A (ATCC 25923 and OPT 2). Cytochalasin D that inhibits the organization of actin filaments inhibited the invasion of S. aureus (ATCC 25923 and OPT 2) in HGFs. These results suggest that pathogenic mechanism of S. aureus in oral infection may be associated with intracellular invasion, and interaction of staphylococcal protein A and cytoskeltal actin filament may be involved in the invasion to HGFs.

        Potassium alum과 비우식성 감미료의 병용이 Streptococcus mutans의 성장에 미치는 영향

        Traditionally, Koreans have used potassium alum by mouth rinse when they suffered from stomatitis. This study was carried out to investigate the combined effect of potassium alum with aspartame, sorbitol and xylitol on the growth of Streptococcus mutans (S. mutans). The relative growth ratio of S. mutans JC-2 cultured at different concentrations of potassium alum and different concentrations of aspartame, sorbitol and xylitol was examined. The combined effect of different concentrations of potassium alum with 0.5% and 10.0% of aspartame, sorbitol and xylitol was examined. The lowest minimum inhibitory concentration of potassium alum was 5.0mg/ml against S. mutans JC-2. The relative growth ratio of potassium alum on S. mum JC-2 was affected at the concentration of 2.0mg/ml. The growth of S. mutans JC-2 decreased at the concentration of 10.0% of aspartame and sorbitol. Also, a combination of potassium alum and sugar substitutes was more effective against S. mum than the use of one sugar substitute alone. The inhibitory effect of potassium alum and sugar substitutes on S. mutans may be an important mechanism of caries prevention by potassium alum.
