Electron Spin Resonance을 이용한 제주 자생 해조류의 DPPH Free Radical 소거활성 검색

본 실험은 electron spin/paramagnetic resonance(ESR/EPR)를 이용하여 기존의 DPPH free radical 소거 활성을 측정하였고, 이를 해조류 유래 천연자원으로부터 새로운 항산화물질을 탐색하는데 적용해 보았다. 연구 결과, 녹조류 추출물에서는 참홑파래, 납작파래 및 청각의 고온 메탄올 추출물에서만 약간의 라디칼 소거활성을 나타내었고, 이 외의 추출물에서는 소거활성이 거의 보이지 않았다. 갈조류는 총 71개의 추출물 중 37개의 추출물에서 80% 이상의 소거율을 보여 아주 우수한 항산화 물질을 가진 family로 여겨지며, 특히 모자반류 (Sargassum spp.)는 수용성과 메탄올, 고온과 상온의 모든 추출물에서 높은 라디칼 소거 활성을 보여 앞으로의 신소재 물질 후보로 선별되었다. 홍조류에서는 수용성보다는 메탄올 추출물에서 대체적으로 높은 소거활성이 검색되어, 나중에는 유기용매 분획물을 제조하여 라디칼 소거능을 측정 할 수 있을 것이다. 이처럼 해조류는 라키칼 소거 능력을 가지는 성분을 여러 형태로 다량으로 함유 하고 있는 것으로 사료 되며 앞으로의 연구에 기대가치를 더해 줄 것으로 사료된다. Extracts which were prepared by four different extractions - 80% methanol extracts (ME) at high ($70^{\circ}C$) and a room temperature ($20^{\circ}C$), respectively and aqueous extracts (AE) at both temperatures with the residue after the methanol extracts - of 10 green, 19 brown and 25 red seaweeds collected in Jeju Island coast were examined for their DPPH free radical scavenging activity using a ESR (electron spin resonance) spectroscopy. A variety of the extracts showed positive scavenging effect against DPPH free radical (except the green seaweeds). Among the extracts, the brown seaweed extracts exhibited the highest scavenging activity. Especially, Sargassum spp. of the brown seaweeds have remarkable scavenging activities - both methanolic and aqueous at the both temperatures ($20^{\circ}C$ and $70^{\circ}C$). On the other hand, ME showed better scavenging activity than AE in the red seaweed extracts. These results indicate that autogenous seaweeds in Jeju will be potential natural antioxidants for functional food compounds.

This study was conducted to screen in vitro angiotensin - I converting enzyme (ACE) inhibitory activities of methanol (MeOH) and aqueous extracts at 20°C and 70°C, respectively, prepared from twenty-six red algae obtained from the coast of Jeju Island in Korea. Among aqueous extracts at 20°C (20AE) from red algae Lomentaria catenata showed the strongest ACE inhibitory activity and Lithophyllum okamurae recorded the second highest activity. From MeOH extract at 20°C (20ME) Ahnfeltiopsis flabelliformis possessed the strongest ACE inhibitory activity. Remarkable activities from MeOH extracts at 70°C (70ME) were observed in Grateloupia filicina, Sinkoraena lancifolia and Grateloupia lanceolata. However, no significant activity was found in aqueous extracts at 70°C (70AE). The IC50 values, which are concentrations required to inhibit 50% activity of ACE, for ACE inhibitory activities of 20AE from Lithophyllum okamurae and L. catenata were 13.78 and 12.21 μg mL–1, respectively. The IC50 values of 20ME from A. flabelliformis and Laurencia okamurae were 13.84 and 106.15 μg mL–1. Those of the 70ME from Bonnemaisonia hamifera, Grateloupia filicina, Sinkoraena lancifolia, G. lanceolata, Gracilaria vermiculophylla and L. okamurae ranged from 25.82 to 124.69 μg mL–1.

제주 연안에 서식하는 해양 녹조류와 갈조류의 항고혈압 활성을 알아보기 위해 해조류로부터 메탄올과 물을 이용하여 상온(20℃)과 70℃에서 각각 4가지 종류의 추출물을 제조하여 스크리닝하였다. 그 결과, 녹조류에서는 비교적 낮은 ACE 저해활성을 보였으나, 갈조류에서는 70ME 잇바디괭생이모자반, 말미역, 바위주름, 넓패, 톳, 다시마 및 70AE 감태에서 ACE 저해활성이 높게 나타났다. 또한 70AE 감태의 가수분해물은 2.33~3.56 ㎍/mL의 IC50값을 보여 매우 높은 ACE 저해활성을 확인할 수 있었다. 이는 해조류의 ACE 저해활성이 수용성 추출물뿐만 아니라 메탄올 추출물에서도 나타나는 것은 해조류 유래 fucoxanthin, phlorotannin 및 polyphenol계 화합물 등에 의한 영향도 있을 것으로 사료된다. This study was conducted to screen in vitro angiotensin converting enzyme (ACE) inhibitory activities of methanol (MeOH) and aqueous extracts which were prepared by four different extractions-80% methanol extracts (ME) at 20℃ and 70℃, respectively and aqueous extracts (AE) at both temperatures with the residue of the MEs-of ten marine green algae and nineteen brown algae collected along Jeju coast of Korea. Most marine brown algae extracts showed higher capacities than those of marine green algae in ACE inhibitory activity. Particularly, 70℃ MeOH extract (70ME) of Hizikia fusiforme showed the strongest inhibition activity (about 87%) among all the extracts. Also, 70 MEs of Enteromorpha linza, Ishige sinicola, Laminaria ochotensis, Petrospongium rugosum, Sagrassum horneri, Undaria pinnatifida and 20℃ MeOH extracts (20ME) of Myagropsis myagroides, Petrospongium rugosum, 20℃ aqueous extracts (20AE) of Codium contractum, Enteromorpha compressa, and 70℃ aqueous extracts (70AE) of Ecklonia cava, Petrospongium rugosum showed moderate ACE inhibitory activities more than 50% and the other extracts exhibited weak activities. On the other hand, E. cava had the best ACE inhibitory activity among 70AEs. This indicates that 70AE of E. cava contains potential anti?ACE macromolecular. We tried to proteolytic digest 70AE of E. cava to induce production of anti?ACE peptides from E. cava 70AE. The enzymes used are five proteases including Kojizyme, Flavourzyme, Neutrase, Alcalase, and Protamex, which are food grade?commercial enzymes from Novo Co. Flavourzyme?digest of E. cava 70AE showed the highest inhibitory activity about 90%. And the five different enzymatic digests of the E. cava 70AE ranged from 2.33 to 3.56 ㎍/mL, respectively in IC50 values of anti?ACE activity.
Antioxidant Activities of Red Algae from Jeju Island

The aim of the present study was to evaluate the antioxidant activity of red algae in Jeju Island. The algal extracts were obtained with MeOH and fresh water at 20 and 70°C, and screened for antioxidant activities using hydroxyl radical (HO·), superoxide anion (O2–), hydrogen peroxide (H2O2) and DPPH free radical scavenging assays. Among them, Gracilaria verrucosa methanolic extract at 20°C (20ME, 96.85%), G. textorii aqueous extract at 20°C (20AE, 88.01%), Grateloupia filicina 20AE (85.35%), and Polysiphonia japonica 20ME (94.92%) exhibited the highest scavenging activities against HO·, O2–, H2O2, and DPPH free radicals, respectively. Moreover, P. japonica (20ME and 70ME) is correlated between DPPH free radical scavenging activity and polyphenolic contents. These results indicate that some red algae in Jeju Island could be potential candidates for development of antioxidants.
