        The effects of acid hydrolysis product of Panax ginseng and MeOH extract of Euphorbia humifusa on the growth of human brain tumor cells were evaluated using U-373 MG human astrocytoma and SK-N-MC human neuroblastoma cells as model cellular systems. These plant extracts induced cytotoxicity in both cells in a dose-dependent manner. These cytotoxic effects were significantly inhibited by GSH, an antioxidant, in both cells. BAPTA/AM an intracellular Ca^(2+) chelator, significantly blocked the cytotoxic effects of these extracts in U373 cells, but enhanced these effects in SK-N-MC cells. These results suggest that the plant extracts may be a valuable choice for the studies on the treatment of human brain tumors.

        감꼭지의 항산화 활성 성분

        In this study, in order to search for new functional materials from natural products, was carried out the study ofantioxidant active ingredients in persimmon calyx(calyx of Diospyros kaki Thunberg). I have experimented with the effect ofantioxidant activity of five different extract(MeOH, n-hexane, EtOAc, n-BuOH and H2O extract) obtained from persimmoncalyx. As a result, the butanol extract, that is the main component fraction of antioxidant activity was found. Three compoundswere isolated by silica gel column chromatography from the n-BuOH extract of persimmon calyx. Their structures of compound1, 2 and 3 isolated from n-BuOH extract of persimmon calyx were identified as quercetin, (+)-catechin and gallic acidby using the TLC, 1H-NMR and 13C-NMR.

        수치에 의한 생약의 항산화 활성 및 성분 변화

        This study was conducted to investigate the change of antioxidant activity and ingredient in 4 species medicinal plant by processing. As a result, EtOAc extract and n-BuOH extract obtained from Cistanche deserticola Y. C. Ma and processed Cistanche deserticola Y. C. Ma showed difference on the experiment of antioxidant activity by DPPH method. Change of ingredient also was confirmed by TLC and HPLC analyses. Two main compounds of antioxidant activity change were isolated by column chromatography from the EtOAc extract and n-BuOH extract of Cistanche deserticola Y. C. Ma. The chemical structure of the compound 1 isolated from EtOAc extract of Cistanche deserticola Y. C. Ma was elucidated as 2'-acetylacteoside by means of IR and NMR analyses. Also, the chemical structure of the compound 2 isolated from n-BuOH extract of Cistanche deserticola Y. C. Ma was confirmed as acteoside. The antioxidant activity of processed Cistanche deserticola Y. C. Ma. was declined by 3 time decrements of 2'-acetylacteoside such as 1.89 mg/g to 0.62 mg/g and 2 time decrements of acteoside such as 5.81 mg/g to 2.67 mg/g by processing.

        약용식물의 Tyrosinase, Hyaluronidase 저해효과 및 항산화 활성

        This study was conducted to investigate tyrosinase inhibitory effect, hyaluronidase inhibitory effect and antioxidant activity by DPPH radical scavenging method on the MeOH extract of 50 species medicinal plant for screening of functional properties. As a result, Chaenomeles sinensis Koehne extract among 50 species medicinal plant turned out to be having tyrosinase,hyaluronidase inhibitory effect and antioxidant activity. The major component of tyrosinase and hyaluronidase inhibitory effect was isolated from EtOAc extract of Chaenomeles sinensis Koehne. And the component of antioxidant activity was isolated from n-BuOH extract of Chaenomeles sinensis Koehne. Their structure of compounds were identified as oleanolic acid and (-)-epicatechin by spectroscopic evidence, respectively.

        감꼭지의 Tyrosinase와 Elastase 저해 활성 성분

        This study was conducted to investigate glutathione S-transferase(GST) activity, tyrosinase inhibitory effect and elastase inhibitory effect in persimmon calyx(calyx of Diospyros kaki Thunberg) for screening of functional materials from natural products. As a result, EtOAc extract of persimmon calyx turned out to be having tyrosinase inhibitory effect and elastase inhibitory effect. The active constituents of tyrosinase and elastase inhibitory effect were isolated from EtOAc extract of persimmon calyx. Their structure of compounds were identified as ursolic acid and (−)−daucosterol by spectroscopic evidence,respectively.

        청미래덩굴 잎의 항산화 활성 성분

        this study, we have investigated the antioxidant activities of the leaves of Smilax china Linne in order to findthe antioxidant substances which has an scavenging effect of reactive oxygen species.(ROS) from natural products. As a result,EtOAc and n-BuOH extracts of the leaves of Smilax china Linne exhibited potent antioxidant effect on various antioxidantexperiment. The major components of antioxidant activity were isolated from EtOAc and n-BuOH extracts of the leaves of Smi-lax china Linne. Their structure of compounds were identified as kaempferol-7-O-α-L-rhamnopyranoside and kaempferol-3,7-O-α-L-dirhamnopyranoside by spectroscopic evidence, respectively. Antioxidant activities were observed in kaempferol-7-O-α-L-rhamnopyranoside at 23g and in kaempferol-3,7-O-α-L-dirhamnopyranoside at 27g as concentration of DPPH 50%reduction.Key words Smilax china Linne, reactive oxygen species (ROS), antioxidant, kaempferol-7-O-α-L-rhamnopyranoside,kaempferol-3,7-O-α-L-dirhamnopyranoside

        생약복합제 GCSB-5의 품질 표준화를 위한 방풍의 지표성분 탐색 및 HPLC 분석

        GCSB-5 preparation is a purified extract from a mixture of 6 medicinal plants(Acanthopanacis Cortex, Achyranthis Radix, Saposhnikoviae Radix, Cibotii Rhizoma, Glycine Semen Nigra, Eucommiae Cortex) that have been widely used for the treatment of various bone disorders. The aim of this study was to investigate HPLC analysis method and screening of standard compound on Saposhnikoviae Radix for quality standardization of a medicinal crude drug GCSB-5. Standard compound of Saposhnikoviae Radix was decided with cimifugin by isolation and instrumental analysis such as NMR. HPLC analysis method for the simultaneous determination of cimifugin was established for the quality control of the medicinal plants of Saposhnikoviae Radix species, GCSB-5 raw material and preparation. And validation of HPLC analysis methods were conformed for verification of HPLC methods by check to specificity, linearity, intra-day precision, inter-day precision and accuracy followin g ICH guideline.

        마늘의 약물대사효소 CYP3A4 저해 활성

It hasbeen well known that fresh garlic and garlic supplement of comercial preparations have various therapeutic properties includ-ing antimicrobial activity, antiplatelet aggregation, antihypertension, and cholesterol-lowering effects, which contribute to itsdby alliinase cleavage of the naturally occurring alliin upon crushing or mincing of garlic, and is the progenitor of a number ofother products, such as diallyl disulfide. CYP3A4, heme-containing monooxygenase, is a key enzyme responsible for drugmetabolism. Therefor, in the present study, we isolated and examined the compounds with CYP3A4-inhibiting activities fromgarlic. Among EtOAc extracts of garlic, we found that N-p-coumaroyltyramine and N-feruloyltyramine showed remarkableCYP3A4-inhibiting activities, compared to diallyl disulfide.         오배자 성분의 항산화 및 간보호 효과

        활성산소에 기인하는 다양한 성인병을 예방 치료할 수 있는 안전하고 우수한 항산화 물질을 천연물로 부터 개발하기 연구의 일환으로 오배자의 주성분에 대한 항산화효과 및 간보호효과를 연구하여 다음과 같은 결과를 얻었다. 1. Ferric-Thiocyanate 법에 의하여 과산화반응 억제에 따른 항산화 활성을 측정한 결과 모두 tocopherol보다 강한 과산화반응 억제 활성을 보였으며 gallic acid methyl ester, protocatechuic acid와 syringic acid는 tocopherol 보다는 강한 활성을 보였으며 BHA와는 유사 또는 그 이상의 활성을 보였다. 특히 1, 2, 3, 4, 6-penta-O-galloyl-β-D- glucose는 tocopherol과 BHA보다 매우 강한 과산 화반응억제 활성을 보였다. 2. TBA 법에 의한 과산화지질의 생성의 지표인 MDA 억제효과에 의한 항산화효과를 측정한 결과 gallic acid 는 1.6×10-4의 농도에서 BHA 보다 높은 억제율을, protocatechuic acid는 1.6×10-6의 농도에서 BHA 보다 높은 억제율을 보였다. gallic acid methyl ester와 1, 2, 3, 4, 6-penta-O-galloyl-β-D-glucose는 농도가 증가함에 따라 우수한 MDA의 억제활성을 나타내었다. 3. Glycyrrhizin을 비교군으로 하여 간 보호 실험을 한 결과 1, 2, 3, 4, 6-penta-O-galloyl-β-D-glucose의 0.5mg/ml첨가군에서는 GOT의 강한 감소 효과를 보였으며 GPT 실험에서는 각 시료 0.5mg/ml 첨가군에서 감소하는 경향을 나타내었다. The major components were isolated from the n-hexane, EtOAc and BuOH extract of Galla Rhois(Rhus Javanica Linne). Their structures were characterized as syringic acid, gallic acid methylester, protocatechuic acid, gallic acid and 1, 2, 3, 4, 6-penta-O-galloyl-β-D-glucose. This study was carried out to investigate the biological activities of isolated compounds. Five compounds were tested for hepatoprotective effects on CCl4-induced cytotoxicity in primary cultured rat hepatocytes and antioxidative effect on Ferric-Thiocyanate method and TBA method. As a result, isolated five compounds showed stronger antioxidative activity than tocopherol, and the antioxidative activity of gallic acid methylester, protocatechuic acid and syringic acid were similar to that of BHA on Ferric-Thiocyanate method. Specially 1, 2, 3, 4, 6-penta-O-galloyl-β-D-glucose showed stronger effect of lipid-peroxidation inhibition than BHA. Gallic acid appeared stronger inhibitory effect of malondialdehyde on TBA method. Hepatoprotective effect of 1, 2, 3, 4, 6-penta-O-galloyl -β-D-glucose was similar or even higher than that of glycyrrhizin on primary cultured rat hepatocyte cytotoxicity.

        진피의 수치 조건에 따른 성분 및 항산화 활성 변화

        Processing of medicinal plants is one of the processing methods for reducing of toxicity or improving of effect on medicinal plants. In this study, we studied the changes in the constituents and antioxidant activity in accordance with the processing conditions by the salt water of Citrus unshiu Markovich. Changes in antioxidant activity was measured by the DPPH method and changes in the components were analyzed by high performance liquid chromatography. As a result, the content of the main constituents (narirutin, hesperidin) and total polyphenol were increased by increasing the concentration of salt water. Moreover, antioxidant activity was increased gradually in proportion to the increase of the total polyphenol content.
