Clove extract by methanol increased expression of the tyrosinase gene on B16 mouse melanoma cells containing tyrosinase promoter. $10{\mu}g/mL$ and $100{\mu}g/mL$ of the extract showed expression rate of the tyrosinase gene about 138% and 245%, respectively, compared with control. At $500{\mu}g/mL$, expression rate of the extract was impossible to measurement by high cytotoxicity. The solvent fraction of methylene chloride also exhibited highly expression rate as methanol extract. However, the solvent fractions of butyl alcohol and water showed repressive effect on expression of tyrosinase gene at $500{\mu}g/mL$. In MTT assay, cell survival rate of the extract exhibited similar to expression rate of tyrosinase gene. That is, $10{\mu}g/mL$ and $100{\mu}g/mL$ of the extract showed the cell survival rate about 128% and 187%, respectively.

To estimate the inhibitory effect of Rubus coreanum extract on melanin biosynthesis, we tested its inhibitory effects on tyrosinase promoter in B16 mouse melanoma cells. Rubus coreanum extract by methanol had inhibitory effect approximately 10% on tyrosinase promoter at 100 ㎍/mL. In the MTT assay, the same extract exhibited very low cytotoxicity under 100 ㎍/mL. The fractions of methylene chloride, butyl alcohol and water did not showed the inhibitory effect on tyrosinase promoter, but the fraction of ethyl acetate exhibited inhibitory effect approximately 11% at 100 ㎍/mL.
대두(Glycine maxim) 추출물이 티로시나아제 프로모터 활성에 미치는 효과

The methanolic extract of Glycine maxim increased the expression of the promoter in B16 mouse melanoma cells harboring a tyrosinase promoter. Extract concentrations of 10 μg/㎖ and 100 μg/㎖ resulted in tyrosinase promoter expression rates of approximately 113% and 184%, respectively, as compared to the control. The fraction layers consisting of butyl alcohol and methylene chloride improved expression effects on the tyrosinase promoter. In particular, the butyl alcohol fraction evidenced a high expression rate at 100 μg/㎖. In the MTT assay, the methanolic extract did not evidence cytotoxicity at concentrations under 500 μg/㎖. Therefore, the results observed with the extract of Glycine maxim showed that the substance exerted a positive effect on the tyrosinase promoter.
수종의 버섯 추출물이 티로시나아제 프로모터에 미치는 효과

To estimate the inhibitory effect of some mushroom extract on melanin biosynthesis, we tested its inhibitory effects of five mushroom extracts on tyrosinase promoter in B16 mouse melanoma cells. In five mushroom extracts, Cordyceps militaris and Poria cocos exhibited low repression effect on tyrosinase promoter. However, Ganoderma lucidum, Paecilomyces japonicus, Phellinus linteus showed high repression effect. Especially, Paecilomyces japonicus and Phellinus linteus extracts had very higher repression effect approximately 81~83% at 100 ㎍/mL. In the MTT assay, Paecilomyces japonicus and Phellinus linteus extracts exhibited high cytotoxicity. Therefore, repression effect of the extracts were closely connected with cytotoxicity.
어성초(Houttuynia cordata) 추출물이 Tyrosinase 유전자 발현에 미치는 효과

멜라닌 색소 생성에 관여하는 tyrosinase 유전자의 발현을 억제하는 물질을 탐색하고자 tyrosinase 프로모터를 지닌 B16 mouse melanoma cell에 어성초 메탄올 추출물을 처리한 결과 메탄올 추출물은 10 μg/mL와 100 μg/mL의 농도에서 대조군에 비해서 약 13%와 45%의 억제효과를 나타냈으며, 세포생존율은 1 μg/mL, 10 μg/mL, 100 μg/mL의 농도에서 약 104%, 101%, 74%로서 세포독성이 낮게 나타났다. Butyl alcohol과 물 분획물은 tyrosinase 유전자의 발현을 억제하는 효과가 없었지만 methylene chloride 분획물은 tyrosinase 유전자의 발현을 억제하는 효과를 나타내었으며, 특히 methylene chloride 분획물은 10 μg/mL와 100 μg/mL의 농도에서 각각 약 82%와 48%의 발현율을 나타냄으로써 대조군에 비해 tyrosinase 유전자의 발현을 크게 억제하였다. Chestnut bark extract by methanol repressed the expression of tyrosinase gene of B16 mouse melanoma cell containing tyrosinase promoter. 10 ㎍/㎖, 100 ㎍/㎖, 1 ㎎/㎖ of the extract repressed expression of tyrosinase gene about 38%, 47%, and 78%, respectively, compared with control. In the MTT assay, the same extract exhibited very low cytotoxicity at 1 ㎍/㎖, 10 ㎍/㎖, 100 ㎍/㎖, and 1 ㎎/㎖, respectively. The fractions of Methylene chloride and ethyl acetate did not showed the repressive effect on the expression of tyrosinase gene, but the fraction of butyl alcohol repressed highly at 10 ㎍/㎖ and 100 ㎍/㎖.

Natural extract in liquid phase was adjusted to 0, 0.25, 0.5, 1, 2, and 4% concentration to check microbial changes and to measure 4, 8, 12 o C for refrigeration temperature. In the case of grapefruit extract, the microbial safety was maintained at all the concentrations at 4 o C storage, but the antimicrobial activity was maintained at 12 o C storage and at 8 o C and 21 days storage. In the case of grape seed extract, only the 4% of the culture at 8 o C satisfied the requirement of safety of food distribution for the last 21 days, and the safety criterion was satisfied only at 4% concentration at 12 o C for 18 days. Complex Scutellaria baicalensis extract showed the total number of microbial cells treated by concentration. It was confirmed that microbial flow safety was maintained at low temperature (4 o C). However, at 8 o C and 12 o C, Exceeded the distribution limit. When polylysine was applied to brown rice cake, it showed activity in all groups except 4 o C, but these properties were not observed at 8 o C and 12 o C. At a concentration of 0.5% or more of chitosan, the growth of the microorganism is suppressed by the 21st day very stably, and a similar tendency is observed at 8 and 12 o C, so that it may be an antimicrobial material that inhibits microorganisms. At the first day, the distribution standards for general bacterial counts were exceeded.Ethyl-pyruvate showed that microorganism safety was maintained at 4 o C and 1% concentration, and food safety was stable even at 2 or 4%. Glycine showed very good and stable distribution stability at 4 o C. However, at 8 o C and 12 o C, the shelf life of 14 days could not be maintained as with the addition of other antimicrobial active substances.
