Background: This study sought to evaluate the skin-protective effects of polysaccharide extracts derived from four natural materials (ginseng, green tea, shiitake, and aloe vera) to determine their potential as raw materials for the cosmetics industry. In a broad sense, we aimed to determine the effects of skin protection on preventing hair loss. Methods and Results: Polysaccharide extracts were prepared from ginseng, green tea, shiitake, and aloe vera. For each polysaccharide extract, the total sugar content was assessed, and antioxidant activity was measured using superoxide dismutase-like activity assay. The efficacy of the extracts at enhancing cell proliferation and hydrogen peroxide-induced apoptosis were assessed in human follicle dermal papilla cells (HFDPC) and keratinocyte (HaCaT) cells. Conclusions: Each of the tested polysaccharides effectively protected dermal papilla cells and keratinocytes by promoting cell proliferation and/or suppressing apoptosis. The protective effect on hair-related keratinocytes and dermal papilla cells is expected to have a positive effect on the prevention of hair loss. Therefore, we propose that ginseng, green tea, shiitake, and aloe vera polysaccharide extracts each show promise to be developed as effective pharmaceutical and/or cosmetic ingredients for preventing hair loss.
Anti-apoptotic Effect of Seoritae Extract on Human Follicle Dermal Papilla Cell

Background: This study investigated the anti-apoptotic effects of an extract from seoritae, a black soybean cultivar, on human follicle dermal papilla cells (HFDPC) and assessed the underlying mechanisms to evaluate its potential as a cosmetic raw material. Methods and Results: The antioxidant effects of the seoritae extract were evaluated using a 2,2- diphenyl-1-picrylhydrazyl radical-scavenging activity assay. In HFDPCs, the results of tests evaluating the enhancement of cell proliferation and protection against apoptosis induced by hydrogen peroxide or dihydrotestosterone (DHT) revealed that seoritae extract exhibited a skin- protective effect. The levels of β-catenin and Bax proteins, which are involved in cell survival and death, were measured to assess the underlying signaling mechanisms that enabled the extract to protect HFDPCs against DHT activity. Conclusions: Our results collectively indicate that seoritae extract can enhance HFDPC proliferation and inhibit cell apoptosis, thus it has the potential to be developed as a cosmetic ingredient with a potential hair loss-alleviating effect on HFDPCs.
시클로 펜타디엔과 구조적으로 회전이 쉬운 Dienophile간의 Diels-Alder 반응에 대한 이론적 연구

Cyclopentadiene과 exo-cyclic 이중결합을 가진 5각고리 화합물간의 Diels-Alder반응을 PM3 방법을 이용하여 연구하였다. Dienophile의 종류가 바뀌어도 전이상태의 구조는 커다란 변화를 나타내지 않았다. 반면에 dienophile의 exocyclic C(=O)-R 결합의 회w전에 의하여 나타나는 두 conformer들은 서로 다른 생성물을 얻을 수 있었으며, 이들의 반응성을 FMO들의 energy gap을 이용하여 설명하였다. 반면에 dienophile의 접근방향에 따른 exo, endo경로의 선택성을 살펴보기 위하여 전이상태에서의 deformation enerhy와 활성와 에너지간의 상관관계를 살펴보았다. 반응의 최저 에너지 경로를 Curtin-Hammett principle을 사용하여 논의 하였다. Dieis-Alder reaction between cyclopentadiene and 5-membered ring compounds which have exo-cyclic double bond has been studied using the PM3 method. Transition states do not show large geometrical change with the variation of dienophiles. Two isomers are possible due to the rotation of the exo-cyclic double bond of a dienophile. The reactivity for the formation of different products are explained using the FMO energy gap. The exo and endo selectivity of the reaction has been also studied from the correlation between the deformation energy and the activation barrier. Minimum energy reaction path is discussed using the Curtin-Hammett principle.
아다만틸을 기반한 N-아릴아미드 신규 안드로겐 수용체 길항제

안드로겐 수용체 길항제로서 N-아릴아미드의 신규한 아다만틸 유도체들을 합성하고 항안드로겐 활성을 평가 하였다. 아다만틸 유도체를 함유하는 N-아릴아미드는 아다만틸 치환체가 없는 유도체보다 더 높은 활성을 가졌다. 아다만틸의 공간부피 및 방향족 고리에 전자밀도 상승효과를 주는 pendant 작용기들이 강력한 길항작용에 결정적인 영향을 미쳤다. 리간드와 수용체 사이의 상호 작용을 설명하기 위해 분자 모델링 연구를 수행 하였다. A novel series of adamantyl derivatives of N-aryl amides as androgen receptor antagonists were synthesized and their anti-androgenic activities were evaluated. The N-aryl amides containing adamantyl derivatives had more activity than those lacking adamantyl substitutions. The synergistic effect of bulkiness of the adamantyl group and modulation of electron density on the aromatic ring by pendant groups was crucial to the potent antagonism. Molecular modeling studies were performed to elucidate the interactions between ligands and receptors.
